Effects of Dietary Lobster Shrimp Chitosan on Lipid Metabolism in Diet-Induced Hyperlipidemic Rats (가시발새우 키토산 식이가 식이성 고지혈증 흰쥐의 지질대사에 미치는 영향)

  • This study was designed to investigate effects of lobster shrimp chitosan on serum and liver lipid levels in rats fed high fat and cholesterol diets. Male Sprague-Dawley rats of 4-weeks old, weighing 70$\pm$5g, were fed experimental diets for 3 weeks: basal diet+5% cellulose(group 1), high lipid diet+5% cellulose(group 2, control group), high lipid diet+5% balloon flower root(group 3), high lipid diet+5% cellulose(group 4, high lipid diet+5% lobster shrimp chitosan(group 5). Concentrations of total cholesterol, HDL-cholesterol, LDL-cholesterol, free cholesterol, cholesteryl ester, triglyceride, phospholipid in serum and liver were determined. The results obtained were as followes: Serum total cholesterol concentration were significantly reduced in all supplemented group compared with control. HDL-cholesterol concentrations were higher in groups 1, 4, and 5 and the ratios of HDL-cholesterol to total cholesterol concentration were higher in groups 1, 4, while the atherogenic-indices were lower in groups 1, and 4 than control. Concentrations of LDL, LDL-cholesterol and free cholesterol, cholesteryl ester were significantly lower in all supplemented groups than the control group. Concentrations of phospholipid and triglyceride were lower in groups 3, 4, 5, while activities of GPT and GOT were significantly lower in groups 4, 5 than the other group. Concentrations of calcium, magnesium and phosphorous were high in all treatment groups compared to the control group. Liver contents of total cholesterol, free cholesterol and cholesteryl ester were lower in the groups 1, 3, 4, and 5 than in the control group, especially lower in groups 1, and 5. Liver triglyceride and phospholipid concentrations were significantly lower in the groups 1, and 5 than in the other groups. The results from the data suggested that the experimental diet containing 5% lobster shrimp chitosan was effective to reduce the serum and liver lipids.

Effects of Cynanchum wilfordii Extract on Serum Lipid Components and Enzyme Activities in Hyperlipidemic and Streptozotocin-Induced Diabetic Rats (백하수오 추출액이 고지혈증 및 Streptozotocin 유발 당뇨성 흰쥐의 혈청 지질성분 및 효소활성에 미치는 영향)

  • The purpose of this study was designed to observe the effects of the feeding Cynanchum wilfordii extract on the improvement of the blood glucose, lipid components in the serum of dietary hyperlipidemic and streptozotocin(STZ) -induced diabetic rats(S.D. strain, ♂) fed the experimental diets for 5 weeks. Concentrations of total cholesterol, atherosclerotic index, LDL, LDL-Cholesterol, free-cholesterol. cholesteryl ester, TG, PL and blood glucose in serum were significantly higher in the cholesterol administration groups((group 2(cholesterol+water), 4(cholesterol+Cynanchum WIlfordii 3.5g% extract)) than those in the control group (group1 , basal diet+water). But the concentrations of total cholesterol. atherosclerotic index, LDL, LDL- cholesterol. free-cholesterol, cholesteryl ester, TG, PL and blood glucose in serum were remakably lower in the group 4 than those in the group 2. In the STZ(55mg/kg B.W.)-induced diabetic groups((group 3(STZ, IP.)+water), 5(STZ(IP.)+Cynanchum WIlfordii 3.5g% extract? the serum total cholesterol, atherosclerotic index, LDL, LDL-cholesterol, free-cholesterol. cholesteryl ester, TG, PL and blood glucose concentrations actions were rather lower in the group 5 than those in the group 3. In the ratio of HDL -cholesterol concentration to total cholesterol and HDL-cholesterol concentration, Cynanchum wilfordii extract administration groups were higher percentage than III the groups 2 and 3. The activities of aspartate aminotransferase(AST), alanine aminotransferase(ALT), alkaline phosphatase (ALP) and lactate dehydrogenase(LDH) in serum were rather lower in the Cynanchum wllfordii extract administration groups (group 4,5) than in the cholesterol diet group(group 2) and STZ-induced diabetic group (group 3). From the above research, the physiological activity substances in Cynanchum wllfordii were effective on the improvement of the blood glucose, lipid compositions in serum of dietary hyperlipidemic and STZ-induced diabetic rats. And particularly, physiological activity substance in Cynanchum wilfordii was more effective therapeutic regimen for the control of metabolic derangements in adult disease.

The Crystal and Molecular Structure of Cholesteryl Pentanoate (콜레스테릴 펜타노에이트의 결정 및 분자구조)

  • Cholesteryl pentanoate $(C_{32}O_2H_{54})$ is orthorhombic, space group $P2_12_12_1$, with a = 21.930(3), b = 21.404(3), c = 6.419(5) $\AA$, Z = 4, V = 3012.8(5)$\AA$$^3$, $D_c$ = 1.04 g$cm^{-3}$, ${\lambda}(Mo\; K{\alpha}$ = 0.71069 $\AA$, $\mu$ = 0.58 $cm^{-1}$, F(000) = 1048, T = 298, R = 0.086 for 1502 unique observed reflections with I > 1.0 $\sigma$ (I). The structure was solved by direct methods and refined by cascade diagonal least-squares refinement. The C-H bond lengths and the methyl groups are fixed and refined as their ideal geometry. A comparison with other cholesteryl esters gives normal structure for the tetracyclic ring, while the tail regions of the side chain and the ester group which stands on end, show a variation from their normal values, presumably due to thermal effects. The molecules are stacked together by non-bonded van der Waals forces with the shortest intermolecular distance of 3.529 $\AA$.

Effects of Varied Mushroom on Lipid Compositions in Dietary Hypercholesterolemic Rats (고콜레스테롤혈증 유발 흰쥐에 있어서 버섯류가 지질 성분에 미치는 영향)

  • For the investigation of the effects of mushrooms on prevention of hypercholesterolemia, dietary hyperc-holesterolemic rats were fed for 2 weeks with basal diet containing 5% G. lucidum, 5% L. edodes, 5% A. judae and 10% G. lucidum, 10% L. ededos or 10% A. judae mushroom. Concentrations of total cholesterol, HDL-cholesterol, LDL-cholesterol, triglyceride, phospholipid, free-cholesterol, cholesteryl ester, VLDL and chylomicron in serum were analyzed. The result obtained are as follows : Concentrations of total cholesterol, LDL, LDL-cholesterol in serum were lowest in the group 6 (10% L. edodes mushroom) and HDL-cholesterol concentration in serum was significantly higher than the concentrations of the control group. Concentrations of triglyceride, phospholipid, free-cholesterol, cholesteryl ester in serum were lower than the concentration of the control group, and the concentrations of triglyceride, cholesteryl ester in group 6 were lower than those of the other groups. Concentrations of phospholipid and free-cholesterol in group 3 were lower than those of the other groups.

Thermotropic Behavior of Hydroxypropyl Chitosans Bearing Cholesteryl and Acryloyl Groups (콜레스테릴과 아크릴로일 그룹을 지닌 하이드록시프로필 키토산들의 열방성 거동)

  • A new hydroxypropyl chitosan capable of forming a thermotropic liquid crystalline phase and two kinds of derivatives based on the hydroxypropyl chitosan (6-cholesteryloxycarbonylpentoxypropyl) chitosans (CHPCTs) and acrylic acid esters of CHPCT (CHPCTEs) were synthesized. The crosslinked films with liquid crystalline order were also prepared by photocrosslinking CHPCTE in mesophase. The liquid crystalline properties for all the samples and the swelling behavior of the crosslinked samples in acetone were investigated. In contrast with the hydroxypropyl chitosan, all the uncrosslinked cholesteryl-bearing samples farmed monotropic cholesteric phases with left-handed helicoidal structures and exhibited reflection colors over the full cholesteric range. This is the first report of a thermotropic cholesteric liquid crystalline chitosan derivative with reflection bands in the visible region. Both the optical pitches (λ$\_$m/'S) of CHPCT and CHPCTE decrease with temperature or with cholesteryl content at a given temperature. However, the λ$\_$m/ of CHPCT was larger than that of CHPCTE at the same temperature and at the same cholesteryl content. All the crosslinked samples did not display reflection colors, indicating that the cholesteric structure of CHPCTE significantly changes upon crosslinking. The two-dimentional anisotropic swelling characteristic of liquid crystalline networks was observed for all the crosslinked samples.

In Vivo Effects of CETP Inhibitory Peptides in Hypercholesterolemic Rabbit and Cholesteryl Ester Transfer Protein-Transgenic Mice

  • We previously reported that cholesteryl ester transfer protein (CETP) inhibitory peptides (designated $P_{28}$ and $P_{10})$ have anti-atherogenic effects in hypercholesterolemic rabbits (Biochim. Biophys. Acta (1998) 1391, 133-144). To further investigate those effects, we studied rabbit plasma that was collected after 30 h of a $P_{28}$ or $P_{10}$ injection. We found that there is a strong correlation between the in vivo CETP inhibition effects and alterations of lipoprotein particle size distribution in rabbit plasma, as determined on an agarose gel electrophoresis and gel filtration column chromatography. In vivo effects of the peptide were observed again in C57BL/6 mice that expressed simian CETP. The $P_{28}$ or $P_{10}$ peptide ($7\;{\mu}g/g$ of body weight) that was dissolved in saline was injected subcutaneously into the mice. The $P_{28}$ injection caused the partial inhibition of plasma CETP activity up to 50%, decreasing the total plasma cholesterol concentration by 30%, and increasing the ratio of HD/total-cholesterol concentration by 150% in the CETP-transgenic (tg) mice. The CETP inhibition by the $P_{28}$ or $P_{10}$ made alterations that modulated the size re-distribution of the lipoproteins in the blood stream. Particle size of the very low (VLDL) and low density lipoproteins (LDL) from the peptide-injected group was highly decreased compared to the saline-injected group (determined on the gel filtration column chromatography). In contrast, The HDL particle size of the $P_{28}$-injected group increased compared to the control group (saline-injected). The expression level of the CETP mRNA of the $P_{28}$-injected CETP-tg mouse appeared lower than the saline-injected CETP-tg mouse. These results suggest that the injection of the CETP inhibitory peptide could affect the CETP expression level in the liver by influencing lipoprotein metabolism.

Lecithin : Cholesterol Acyltransferase Activities in Rats Fed Cow량s Milk with Different Levels of Cholesterol (식이 콜레스테롤의 섭취량에 따라 우유가 흰쥐의 Lecithin: Cholesterol Acyltransferase 활성에 미치는 영향)

  • The effect of milk in low and high cholesterol diets were investigated on serum cholesterol esterification. Weanling male Sprague-Dawley rats were divided into low(0.01% w/w) and high (1.01% w/w) cholesterol-diets groups. Bothlow and high cholesterol groups were consisted of three groups : control , LM(low heat milk), and HM (high heat milk) groups. After feeding these experimental diets for six weeks, serum cholesterol (free cholesterol and cholestryl ester) concentration and activity of lecithin.: cholesterol acyltransferase (LCAT) were measured, and serum lipoprotein profile was examined using gel column chromatography. According to the result, activity of LCAT was elevated independently by intakes of high cholesterol and milk , which resulted in the increase of daily turnover of serum cholesteryl ester. However, the turnover of HDL-cholesteryl ester increased only by milk. LCAT activity was moderately correlated with levels of total-and HDL-free choelsterol. It is concluded from the present study that milk had the cholesterol-lowering effect which partly appears to be mediated through facilitated reverse cholesterol transport.

Severely modified lipoprotein properties without a change in cholesteryl ester transfer protein activity in patients with acute renal failure secondary to Hantaan virus infection

  • Patients with hemorrhagic fever with renal syndrome (HFRS) often exhibit altered serum lipid and lipoprotein profile during the oliguric phase of the disease. Serum lipid and lipoprotein profiles were assessed during the oliguric and recovery phases in six male patients with HFRS. In the oliguric phase of HFRS, the apolipoprotein (apo) C-III content in high-density lipoproteins (HDL) was elevated, whereas the apoA-I content was lowered. The level of expression and activity of antioxidant enzymes were severely reduced during the oliguric phase, while the cholesteryl ester transfer protein activity and protein level were unchanged between the phases. In the oliguric phase, electromobility of $HDL_2$ and $HDL_3$ was faster than in the recovery phase. Low-density lipoprotein (LDL) particle size was smaller and the distribution was less homogeneous. Patients with HFRS in the oliguric phase had severely modified lipoproteins in composition and metabolism.
