Effect of Ginsenosides on .the Biosynthesis of Low density Lipoprotein Receptor in Cultured Chinese Hamster Ovary(CHO) Cell (인삼사포닌 (ginsenoside) 저밀도 지방단백질 수용체 생합성에 미치는 영향)

  • Effect of the purified ginsenoside $-Rb_1$ and $-Rb_2$ on LDL receptor biosynthesis of CHO cell cultured in a high cholesterol medium was investigated . Cholesterol uptake by CHO cell cultured in a medium containing various amounts of cholesterol was traced and found that the cholesterol uptake was proportional to the concentration of cholesterol in the medium, and the population of LDL receptors were proportionally decreased as the increasing cholesterol level in the cell. However, when the CHO cells were cultured in the medium containing ginsenosides, no significant decrease of LDL receptor population occured. The biosynthesis of protein and RNA of the above cells was higher than that of CHO cells cultured in the absence of the ginsenosides, suggesting that the ginsenosides might stimulate LDL receptor bio-synthesis. It was also observed that the ginsenosides stimulated the biosynthesis of estradiol and progesterone from cholesterol in the CHO cell. From the above results, it seemed that the ginsenosides lowers the cholesterol level by stimulating the cholesterol metablism including steroid hormone biosynthesis, resulting in the lowering of inhibitory action of cholesterol on LDL receptor biosynthesis.

Effects of Herbal Acupuncture(Raphanus Sativus) at $BL_{20}$ on the Hyperlipemia Rats Induced by High Fat Diet (비유에 대한 래복자 약침(藥鍼)이 고지방 식이로 유발된 비만(肥滿) 백서(白鼠)에 미치는 영향)

  • Objectives : This research was performed to investigate the effect of herbal acupuncture(Raphanus sativus) at Bisu($BL_{20}$) on weight gain, food intake, food efficiency, serum of lipid concentrations, liver function and HDL to total cholesterol ratio of rats fed high fat diet for 5weeks. Methods : Experimental groups were divided into high fat diet group(Control), high fat diet and saline in $BL_{20}$ treated group(Saline), high fat diet and Raphanus sativus-herbal acupuncture in $BL_{20}$ treated group(RHA-1, 0.058mg/ml/g), high fat diet and Raphanus sativus-herbal acupuncture in $BL_{20}$ treated group(RHA-2, 0.029mg/ml/g), high fat diet and Raphanus sativus-herbal acupuncture in $BL_{20}$ treated group(RHA-3, 0.011mg/ml/g), Herbal acupuncture was bilaterally treated at 0.2cc per 4days, total 14 times in 56days. Results : Body weight and food efficiency were decreased in saline, RHA-1, RHA-2. Feed intake was increased in saline, RHA-1, RHA- 2, RHA- 3. The level of serum total cholesterol was decreased in saline, RHA-1. The level of serum HDL-cholesterol, LDL-cholesterol were not significant. The level of serum triglyceride, Total Bilirubin were decreased in saline, RHA-1, RHA-2, RHA-3.The level of serum AST was decreased in RHA-1. But the level of serum ALT was increased in RHA-1. The level of atherogenic index was decreased in saline, RHA-1. The level of HTR was increased in saline, RHA-1. Conclusions : Raphanus sativus-herbal acupuncture in Bisu($BL_{20}$) can control body weight, feed intake and feed efficency ratio, total cholesterol, total bilirubin, and prevention cardiovascular risk.

Effects of Lifestyle and Dietary Behavior on Cardiovascular Risks in Middle-aged Korean Men

  • Lifestyle and dietary behavior intervention as the primary prevention of lipid disorder seems safe and compatible with other treatments of cardiovascular diseases. Cross-sectional associations between lifestyle factors and dietary behavioral factors with plasma lipid and lipoprotein levels were analyzed in 189 middle-aged men in Suwon, Korea. Overnight fasting plasma levels of total cholesterol, high-density lipoprotein(HDL)-cholesterol, triacylglycerol and glucose were analyzed. Blood pressure and anthropometric data were also measured. Lifestyle factors such as smoking status, alcohol consumption and frequency of physical exercise were evaluated by a self-administered questionnaire. Questions regarding dietary behavior were also asked. The subjects were 43.8%${\pm}$7.9 years old, and 23.8%${\pm}$2.6kg/m$^2$. From stepwise regression analyses, significant correlates with total cholesterol level were body mass index(BMI), alcohol intake(negative), age and coffee drinking(model R$^2$=14.3%). BMI, breakfast-skipping, age, and sleeping hours were significant for triacylglycerol level(model R$^2$=15.8%). BMI, alcohol drinking(negative), age, and coffee drinking were significant for low-density lipoprotein(LDL)(model R$^2$=11.7%). Age(negative), BMI(negative), alcohol drinking, stress level(negative), physical exercise, and cigarette smoking(negative) were significant for high-density lipoprotein(HDL)(model R$^2$=12.1%). From stepwise regression analyses, excluding BMI and age as factors in the model, alcohol intake(negative) and coffee drinking were significantly correlated with total cholesterol level(model R$^2$=4.4%) : breakfast-skipping with triacylglycerol(model R$^2$=3.2%) : alcohol intake (negative) with LDL level(model R$^2$=3.4%) : alcohol intake, physical exercise and stress level(negative) with HDL level(model R$^2$=6.3%). The findings suggest that a healthy daily lifestyle and dietary behavior may have an anti-atherogenic effect by altering plasma lipid and lipoprotein levels in middle-aged Korean men.   • The present study was carried out to investigate the antiobesity and hypocholesterolemic effects of Bifidobacteria animalis DY-64 (B. animalis DY-64), a lactic acid bacterium isolated from the human intestine, in rats fed a high-fat/high-cholesterol diet for 4 weeks. Forty male Sprague-Dawley rats were divided into four groups and fed either a normal (N) or high-fat/high-cholesterol (HFC) diet without or with oral administration of B. animalis DY-64 (N-BA, HFC-BA). The gain in body weight, and liver and adipose tissue weights of the HFC group were heavier than that of the HFC-BA group. Serum total cholesterol (TC), LDL-cholesterol, and leptin levels of the HFC group, which were significantly elevated compared to those of the N group, dropped by 19, 18, 21, and 13% in the HFC-BA group, respectively, whereas the serum HDL-cholesterol level markedly increased. However, serum TG, LDL-cholesterol, HDL-cholesterol, and leptin levels were not significantly different among the N groups (N, N-BA) with or without B. animalis DY-64 administration. TC and TG levels of the liver as well as the TG level of the adipose tissue were significantly reduced in the HFC-BA group. In addition, HR-LPL activity in adipose tissue was also lower in the HFC-BA group than in the HFC group. These results suggest that B. animalis DY-64 isolated from the human intestine exerts hypocholesterolemic effects by reducing serum and liver cholesterol levels and plays a role in the prevention of obesity induced by HFC diet.

  • Total saponin and oil fraction of Polygala tenuifolia were tested for their anticholesterolemic properties by a "Three day fasting method" on rabbits. It has been reported that saponin have the anticholesterolemic activity, however this activity is not general property of all kinds of saponin. As shown in Table, Polygala saponin has not significant anticholesterolemic activity on rabbits. On administering oil fraction of Radix Polygala, serum cholesterol level in rabbits increased to 224.7% while the control group 43.2%.oup 43.2%.

  • This study was performed to investigate the effects of HBM(Healthy Body Maker) on the weight loss and serum cholesterol level of rats. Twenty male Sprague-Dawley rats weighing 200$\pm$5g were grouped into 2. Rats were raised for 6 weeks with diet containing 0% and 5% of HBM. 1. There were no clinical signs for all the experimental period(6 weeks) in 0% and 5% groups. 2. At 3 and 6 weeks. body weight of 5% HBM group was significantly lower than that of control group. respectively(p<0.05, p<0.01). 3. At 4 weeks. the level of MCHC of 5% HBM group was significantly higher than that of control group(p<0.01) And at 6 weeks the level of RBC and Hct of 5% HBM group were significantly higher than those of control group(p<0.05) 4. At 6 weeks. the level of serum cholesterol of 5% HBM group was significantly lower than that of control group. 5. Absolute liver weight of 5% HBM group was significantly lower than that of control group(p<0.05). 6. Minor subaceous sickness of liver cell in the control group appeared but no symptom of liver cell in 5% HBM group. It is concluded that HBM was a safe and effective diet food which aided in inducing weight loss in rat without any observed harmful clinical side effects.

  • Elevated level of serum cholesterol in humans is a risk factor correlated with the development of coronary heart disease. We have screened lactic acid bacteria from pig and human feces for the development of probiotics which have an anti-cholesterol effect. We have used special media to isolate only lactic acid bacteria and they were subjected to the experiments such as oxgal test, carbohydrate fermentation test. Results from the acid tolerance test and growth test in the presence of oxgal demonstrated that some strains would likely survive in thuman stomach, where acidity is high, and in small intestine, where bile fluid is present. In conclusion, we were able to screen lactic acid bacteria which were tolerant against bile acid and supposed to be prominent to lower the cholesterol level in human serum.

  • This study was conducted to investigate the effets of chitosan and beef tallow at different level on glucose and lipid metabolism in rats. Dietary fot level was 20% and 40%, and chitosan was given at levels of 0%, 3%, and 5%(wt/wt) of diet. Chitosan supplement tended to decrease the serum total lipids, total cholesterol, and triglycerides. HDL cholesterol and HDL cholesterol : total cholesterol ratio tended to increase with 5% chitosan supplementation. LDL cholesterol and VLDL triglyceride tended to decrease with chitosan supplementation. Lipid concentration of liver and epididymal fat pad(EEP) tended to decrease with medium dietary fat and chitosan treatment. fecal excretion of total lipid and triglyceride exhibited a tendency to increase with high fat levels and chitosan. Length of small intestine and gastrointestinal transit time were not affected by dietary fit levels or chitosan supplements. Therefore, it could be suggested that chitosan supplement had beneficial effects on lipid metabolism. (Korean J Nutrition 31(7) : 1112-1120, 1998)

Cholesterol, Statins, and Brain Function: A Hypothesis from a Molecular Perspective

  • There is evidence that cholesterol in the brain plays an important role in the neurotransmitter release. A decrease of the cholesterol level severely hampers the activity of the membrane fusion machinery, thereby inhibiting the release. Meanwhile, the results from several clinical studies suggest that a low cholesterol level is linked to the dysfunction of some brain activities. Because the neurotransmitter release underlies the basic brain function, the combined results lead to a testable hypothesis that the cholesterol-lowering drugs may inhibit the neurotransmitter release at the synapse. Such inhibition of the release could result in impaired brain function for a limited group of people. A molecular basis for the hypothesis is discussed.

Changes of Cholesterol and Selenium Levels, and Fatty Acid Composition in Broiler Meat Fed with Garlic Powder

  • Effect of garlic on cholesterol and selenium (Se) levels, and fatty acid composition in the broiler chicks (Indian River) fed diet containing 0, 2, 4, 6, and 10% lyophilized garlic powder (GA) for 6 weeks were determined. Supplementation of garlic powder significantly decreased cholesterol level in broiler compared to the control (p<0.05). Se levels were not significantly different (p>0.05) among treatment groups. Percentages of $C_{16:0}$ and $C_{18:1}$ were gradually decreased ($C_{16:0}$;20.62% of total fatty acid in the control to 17.71 % in 10% GA; $C_{18:1}$;34.08% in the control to 30.71 % in 10% GA), while that of $C_{18:2}$ was increased from 28.69% in the control to 35.89% of 10% GA diet (p<0.0l). These results demonstrate some active components of garlic reduce cholesterol level and affect fatty acid metabolism.
