High-Level Expression of Recombinant Human Interleukin-2 in Chinese Hamster Ovary Cells Using the Expression System Containing Transcription Terminator

  • Many biological properties and the clinical potential of human interleukin-2 (hIL-2) draw much attention to its high-level expression in mammalian cells. Recombinant human IL-2 (rhIL-2) was expressed in Chinese hamster ovary (CHO) cells, using the recently developed expression system which confers position-independent expression. Stable CHO cell lines carrying several hundred amplified copies of the rhIL-2 gene were easily obtained and rhIL-2 was expressed at high levels after selection with increasing concentrations of methotrexate. Interestingly, the insertion of the transcription terminator of the human gastrin gene into the downstream region of the gene for rhIL-2 considerably increased rhIL-2 expression. Using the expression system with the transcription terminator, it was possible to get a CHO cell line expressing the rhIL-2 at a very high level, about $11.4\mug/10^6$ cell/day, which is about 6 times higher than that previously reported. The biological activity of the rhIL-2 protein purified from the cell line was also confirmed by the cell proliferation assay.

  • The inherent performance of Ultrasonic Motor(USM) which is on of highlighted a directly-driven positioning servo motor/actuator. In this paper, the speed of control USM based on neural network control. The neural network control can roughly be classified as the direct control and indirect control schemes. An indirect control scheme is adopted for Ultrasonic Motor speed control. A back propagation algorithm is used to train neural network controller. The Simulation results show that this neural network control system can provide good dynamical responses.

  • "Dong Ui Bo Gam" was written by Huh jun that was influenced by Cho Sun Sung Confucianism's environment had been receptive to rich idea. "Dong Ui Bo Gam" "Shin Hyung Mun" was composed of the base on a full understanding in taoist idea. It had a real systemical structure, was help to taoist training. The taoist idea of "Dong Ui Bo Gam" was made up of the extension of mature theory and training in Cho Sun Dan Hak. Specially, The taoist idea of "Dong Ui Bo Gam" was had an effect on "Euibangyoochui" better as compared with "Dong Ui Bo Gam" "Shin Hyung Mun" in contents "Shin Hyung Jang Bu DO" generalized the whole contents of "Dong Ui Bo Gam", was influenced by taoist idea, also peculiar method that was not searched in other medical books. 'Heart 心' was handled importantally in "Dong Ui Bo Gam" "Shin Hyung Mun" under the influence of taoist idea and Cho Sun Sung Confucianism.

Korean Medical Treatment for Prolonged Central Hyperthermia Following Pontine Hemorrhage: A Case Report (뇌교 출혈 이후 발생한 지연된 중추성 고체온증에 대한 한방치료 1례)

  • A patient had central fever following pontine hemorrhage. Central hyperthermia caused by stroke is a rare case, and it is difficult to control. There are few case reports about central hyperthermia in Korean medical treatment (KMT). The patient suffered central hyperthermia accompanied by tachycardia, dyspnea, and irritability. However, there was no evidence of infection. Thus, hypnotics, sedatives, and a minor tranquilizer (Lorazepam and Midazolam) was prescribed. Despite a temperature peak of $39.9^{\circ}C$, most of the symptoms were alleviated. The patient's average body temperature was about $37^{\circ}C$, which is higher than most people at $36.5^{\circ}C$. His symptoms were diagnosed as ascendant hyperactivity of liver Yang (肝陽上亢), and the patient was prescribed Shihogayonggolmoryo-tang. During the 44 days of KMT, there was no change in his average body temperature and no central hyperthermia over $39^{\circ}C$. This case report demonstrates the possibility of controlling central hyperthermia caused by pontine hemorrhage using KMT.
