Effects of Photooxidation and Chlorophyll Photosensitization on the Formation of Volatile Compounds in Lard Model Systems

  • Effects of chlorophyll and visible light exposure on the volatile formations and headspace oxygen content were studied in lard model systems at $55^{\circ}C$. Samples with or without addition of chlorophyll under light underwent photosensization or photooxidation, respectively. Total volatiles (TI) in lard with 5 ppm chlorophyll photosensization were 19 times higher than those in visible light photooxidized samples for 48 hr while TI in lard with chlorophyll in the dark were not significantly different from those in photooxidized samples (p>0.05). Headspace oxygen content in photosensitized lard decreased from 21 to 15% for 48 hr but that in photooxidized lard or that in lard with chlorophyll in the dark did not change significantly (p>0.05), which indicates that lard system used in this study is a photosensitizer-free model system and the presence of chlorophyll accelerated the lipid oxidation only under visible light. Oxidation mechanisms of photooxidation with or without presence of photosensitizers under visible light were not the same based on the difference of oxidized volatile profiles and headspace oxygen depletion.

Effect of Bradyrhizobium japonicum on Chlorophyll Content, Nodulation, and Plant Growth in Soybean

  • Study on effectiveness of Bradyrhizobium japonicum on soybean [Glycine max (L.) Merr.], local cultivar 'Sathiya' were carried out in the garden soil of Trib-huvan University. Different parameters like nodulation, chlorophyll content in fresh leaves and growth of plant in inoculated and uninoculated plant was studied. Pot experiment was conducted in the green house to evaluate the effectiveness of B. japonicum on soybean. It was observed that B. japonicum inoculation increased the number of nodules, shoot length of plant and total chlorophyll content in fresh leaves of soybean plant. However, root length was decreased in all inoculated plants.

규산 함유 액상비료 시비에 따른 크리핑 벤트그래스의 생육과 품질 변화 (Growth and Quality Changes of Creeping Bentgrass by Application of Liquid Fertilizer Containing Silicate)

  • 본 연구는 규산 함유 액비(LFSi)의 시비에 따른 잔디의 생육과 품질의 변화를 확인하기 위해 잔디의 가시적 품질, 줄기 밀도, 뿌리 길이, 엽록소 함량, 예지물 및 양분 함량을 조사하였다. 처리구는 대조구(CF), LFSi 1,000배 처리구(SiF-1), LFSi 500배 처리구(SiF-2) 및 LFSi 250배 처리구(SiF-3)로 구분되었다. LFSi 처리 전후에 토양화학성은 차이를 나타내지 않았다. LFSi 처리구와 대조구를 비교하였을 때, 잔디 품질은 7월, 10월 및 11월 조사에서 높았으며, 엽록소 함량과 잔디 예지물은 10월 27일 조사에서 증가하였다. 줄기 밀도, 뿌리 길이, 잎조직 내 질소 함량 및 칼륨 함량은 LFSi 처리구에서 증가하였다. 잔디 잎 조직 중 규소 함량은 칼리 함량이나 줄기 밀도와 정의 상관성을 보였고, 줄기 밀도는 잔디 품질이나 엽록소 함량과 각각 정의 상관성을 나타내었다. 이 결과들을 종합해 볼 때, 크리핑 벤트그래스에서 규산 함유 액비의 시비는 잔디의 잎 조직 중 칼륨 함량이나 줄기 밀도가 증가되어 가시적 품질이 향상되는 것을 확인 할 수 있었다.

비파괴적 엽색분석을 통한 케일 잎의 내적품질 평가 (Evaluation of Internal Quality of Kale Leaf by Non-Destructive Color Measurement)

  • 케일 잎의 내적 품질 중 비타민 C, 질산염, P, Ca, Mg, Fe 함량은 기존의 보고와 유사하였고, 이중 채소 품질로서 가장 큰 관심이 되고 있는 질산염은 100g 생체 중 139${\~}$429 mg으로 나타났으며, 비타민 C 함량은 역시 생체 100g당 106${\~}$203 mg으로 나타났다. 상대적 엽록소 함량과 케일 잎의 내적품질과 엽색과의 관계를 분석해 본 결과 상대적 엽록소와 b 수치가 내적 품질 중 질산염, 마그네슘, 철과 같은 엽록소와 관계 깊은 요인, 그리고 비타민 C 함량과 높은 상관관계를 보였다. 이 중 질산염 함량은 상대적 엽록소 함량과 r = $0.910^{{\ast}{\ast}}$ (질산염 함량 =$5.907^{{\ast}{\ast}}$ 상대적 엽록소 함량 + 21.55), 그리고 b 수치와는 r = $-0.901^{{\ast}{\ast}}$의 고도의 음의 상관관계를 나타내었다. 비타민 C 함량도 상대적 엽록소와 r = $-0.858^{{\ast}{\ast}}$의 고도의 상관관계를 보였다. 내적 품질간의 상관관계에서는 질산염 함량이 적을수록 비타민 C함량이 높아져 r = $-0.795^{{\ast}{\ast}}$의 고도의 음의 상관관계를 보였다. 따라서 재배중 비파괴적인 상대적 엽록소 측정으로 케일 잎의 질산염 등의 내적 품질의 예측이 가능하며 이를 통한 시비관리로 보건적 가치가 높은 저 질산염, 고 비타민 C의 케일생산이 가능해 질 것이라 생각된다.

Chlorophyll content and the expression pattern of ERF transcription factor gene in leaves and roots of wild corn under flooding treatment

  • The origin of wild corn (teosinte) is distributed in the Northwest coastal pacific area of Central America, including Mexico, which is a wetland area of 5 to 6 months per year. Depending on these climate characteristics, wild corn is genetically resistant to flooding condition. In order to evaluate the availability of flooding resistant genes of these wild corns, we examined the physiological responses after the flooding treatment in the early stages of the growth of various wild corns. The difference of chlorophyll content between flooding untreatment and flooding treatments (untreated chlorophyll content - humidified chlorophyll content) was the highest in chlorophyll content in the case of B73, the common corn. In the middle leaf, $\underline{Zea}$ mays subsp. Parviglumis, Zea mays subsp. Mexicana, Zea mays subsp., Zea perennis decreased significantly. In the lower leaves, Zea mays sub and Zea nicaraguensis showed the lowest content compared to B73. PCR analysis was performed using 34 primers divided into two groups, top and bottom. In the wild corn, pyruvate decarboxylase 2 in root and alcohol dehydrogenase 1 in shoot showed the difference in the reaction.

Ecophysiological characteristics of Rosa rugosa under different environmental factors

  • Background: Ecophysiological characteristics of Rosa rugosa were analyzed under different environmental factors from May to October 2022. Photosynthesis, chlorophyll fluorescence, chlorophyll content, leaf water content (LWC), osmolality, carbohydrate content, and total ion content were measured to compare the physiological characteristics of R. rugosa at two study sites (i.e., in large pots and in the Goraebul coastal sand dune area). Results: When R. rugosa was exposed to high temperatures, photosynthetic parameters including net photosynthetic rate (PN) and stomatal conductance (gs) in both experiment areas declined. In addition, severe photoinhibition occurs when R. rugosa is continuously exposed to high photosynthetically active radiation (PAR), and because of this, relatively low Y(II) (i.e., the quantum yield of photochemical energy conversion in photosystem II [PSII]) and high Y(NO) (i.e., the quantum yield of non-regulated, non-photochemical energy loss in PSII) in the R. rugosa of the pot were observed. As the high Y(NPQ) (i.e., the quantum yield of regulated non-photochemical energy loss in PSII) of R. rugosa in the coastal sand dune, they dissipated the excessed photon energy through the non-photochemical quenching (NPQ) mechanism when they were exposed to relatively low PAR and low temperature. Rosa rugosa in the coastal sand dune has higher chlorophyll a and carotenoid content. The high chlorophyll a + b and low chlorophyll a/b ratios seemed to optimize light absorption in response to low PAR. High carotenoid content played an important role in NPQ. As a part of the osmotic regulation in response to low LWCs, R. rugosa exposed to high temperatures and continuously high PAR used soluble carbohydrates and ions to maintain high osmolality. Conclusions: We found that Fv/Fm was lower in the potted plants than in the coastal sand dune plants, indicating the vulnerability of R. rugosa to high temperatures and PAR levels. We expect that the suitable habitat range for R. rugosa will shrink and move to north under climate change conditions.

Estimation of chlorophyll and pheophytin contents of rice (Oryza sativa L.) leaf in seedling bed using CIE chromaticity diagram

  • Leaf colors of rice can be used to identify stress level due to its adaptation to environmental change. For most leaves green-related colors are sourced from chlorophyll a and b. For most leaves green-related colors are consisted of chlorophyll a and b. Chlorophyll concentration is normally measured using a spectrophotometer in laboratory. In some remote observation fields, it is impossible to collect the leaves, preserve them, and bring them to laboratory to measure their chlorophyll content. The measurement of chlorophyll content is observed through its color. Using CIE chromaticity diagram leaf color information in RGB is transformed into wavelength (in nm). Pheophytin contents were also analyzed in 95% ethanol extracts. In the process of leaf development of rice young seedling, both pigments were compared. Leaf samples from different rice seedling bed is taken, their colors and RGB values are recorded using Photoshop Image Analysis. SPAD-502 values were also measured. The chlorophyll and Pheophytin contents were fully estimated by ${\rightthreetimes}_{avg}$ on CIE chromaticity diagram.

콩기름 에멀션의 광산화에 대한 산나물과 허브 추출물의 첨가 효과 (Effect of Sannamul and Herb Extract Addition on the Photooxidation of Soybean Oil Emulsion)

  • Purpose: This study was performed to evaluate the effects of sannamul or herb extract addition to a soybean oil-in-water emulsion during photooxidation in the presence of chlorophyll. Methods: The emulsion mainly consisted of purified soybean oil and citric acid buffer (pH 4.0) at a weight ratio of 4 to 6, with chlorophyll a addition at 6 mg/kg. Ethanol extract of daraesoon, samnamul, basil, or peppermint was selectively added to the emulsion at 400 mg/kg, and emulsions in glass serum bottles were oxidized at $5^{\circ}C$ under 2,600 lux light for 48 hours. Lipid oxidation of the emulsions was evaluated based on determination of headspace oxygen content, peroxide value, and p-anisidine value. Pigments and antioxidants were also monitored. Results: The emulsion with added samnamul extract with high contents of polyphenols and low chlorophyll content showed lower oxygen consumption, peroxide values, and p-anisidine values, whereas basil and peppermint extracts with high chlorophyll contents increased photooxidation. Chlorophylls were degraded during photooxidation of the emulsions, and the degradation rate was highest in the emulsion with added samnamul extract. Conclusion: The high antioxidant activity of samnamul extract in the photooxidation of soybean oil-in-water emulsion could be due to low chlorophyll content, high concentrations of polyphenol compounds in the extract, as well as rapid degradation of chlorophylls during oxidation.

THE EFFECT OF HEAT TREATMENTS ON THE CHLOROPHYLL IN GREEN PEPPER

  • Chlorophyll을 함유(含有)하고 있는 녹색 식물(植物)의 조리(調理)에 의(依)한 색깔 변화(變化)는 대단히 중요(重要)하므로, 본실험(本實驗)은 각각(各各) 다른 열처리(熱處理)와 시간(時間)이 풋고추의 chlorophyll 함량(含量)에 미치는 영향을 조사하였다. 풋고추는 boiling water steam 그리고 hot air 를 사용(使用)하여, $100^{\circ}C$에서 5 10 15 분간(分間) 가열(加熱)했고, microwave oven에서는 1 2 3 분간(分間) 처리했다. 결과(結果)에 의(依)하면 chlorophyll 함량(含量)은 일반적(一般的)으로 가열시간(加熱時間)이 길어짐에 따라 감소하는 경향이었으며, 가열처리중(加熱處理中) steam이 가장 큰 영향을 주었고, boiling water 가 그 다음이었다. Hot air와 microwave 처리한 풋고추는 아주 적은 양의 chlorophyll이 변화(變化)하였다. 일반적으로 pheophytin 생성(生成) chlorophyll 함량(含量)이 감소함에 따라 증가(增加)하였다.

Growth and Chlorophyiil Biosynthesis of Vigna angularis under Lead Stress

  • The effect of various supplies of lead singly and in combination with aluminium on growth and chlorophyll biosynthesis was investigated in 7-day-old Vigna angularis seedlings. Expose to 50 uM Pb or more drastically reduced root elongation rate. Significant depressions in root growth was observed within 1 day and no recovery of growth was seen over the duration of treatment period. Root elongation decreased depending on the Pb concentrations. Root growth inhibition was stronger than shoot growth inhibition. The initiation of lateral roots appeared to be more sensitive to Pb than the growth of main roots. Inhibition of root and shoot elongation by Pb was lessened by combined exposure of Pb and Al, suggesting that the presence of Al reverse the inhibitory effect of Pb alone. With the histochemical sodium rhodizonate method the rate of Pb uptake was dependent on the Pb concentration and exposure time of the roots to Pb salts. Pb was first deposited on the root surface and then translocated radially in the root cap cells. During a longer Pb administration (up to 72 h) Pb penetration was nonuniform, with accumulation within the cortex or endodermis. There was drastic reduction in chlorophyll content by Pb. The Pb inhibition of chlorophyll synthesis was concentration dependent. $\delta-Aminolevulinic$ acid dehydratase (ALAD) activity exhibited distinct inhibition from control. Reduction in chlorophyll content was accompanied by proportional changes in ALAD activity. Chlorophyll content and ALAD activity were less affected by combined exposure of Pb and Al, suggesting that Al has a protective effect against the inhibiting action of Pb on photosynthetic activity.
