Effects of Artificial CO2 Release in Soil on Chlorophyll Content and Growth of Pinus densiflora and Quercus variabilis Seedlings (토양 내 인위적인 이산화탄소 누출에 따른 소나무와 굴참나무 묘목의 엽록소 함량과 생장 반응)

  • This study was conducted to analyze the responses of chlorophyll contents and growth of Pinus densiflora and Quercus variabilis seedlings on distance from the well and $CO_2$ flux after the artificial $CO_2$ release. From June 1 to 30, 2016, $CO_2$ gas was injected at the rate of $6L\;min^{-1}$ at the study site in Eumseong. Chlorophyll content was analyzed in the middle of July, 2016, and root collar diameter (RCD), height (H), and biomass were measured in May and December, 2016 after planting 2-year-old P. densiflora and 1-year-old Q. variabilis seedlings in May, 2015. The chlorophyll content of P. densiflora seedlings did not show a significant correlation with $CO_2$ flux, whereas the chlorophyll content of Q. variabilis seedlings showed a significant negative correlation with increasing $CO_2$ flux (P<0.05). The RCD and H growth rates of both species showed the significant difference in the distance from the well of the $CO_2$ anthropogenic release treatment. In particular, the RCD and H growth rate of P. densiflora seedlings and the RCD growth rate of Q. variabilis seedlings increased significantly as the seedlings were closer to the well, but the H growth rate of Q. variabilis seedlings decreased significantly. In addition, as the $CO_2$ concentration in the ground increases, ${\Delta}R/S$ ratio increases in both species, suggesting that the high $CO_2$ concentration in the soil promotes carbon distribution relative to the root part. The results of this study can be used as data necessary to monitor the $CO_2$ leakage and physiological and growth responses of both species to leakage of stored $CO_2$ in the future.

Investigation of Physicochemical Properties According to Different Parts and Maturity of Momordica charantia L. (여주열매의 성숙단계 및 부위에 따른 이화학적 특성)

  • Momordica charantia L. is a valuable food and medicinal plant of the gourd family (Cucurbitaceae) that is cultivated in tropical and subtropical regions of the world. Physicochemical properties of M. charantia based on cultivars, parts and growing stage were investigated. Crude protein contents of leaf were 27.5%, 26.9%, and 23.6% in native leaf (NL), cv. Erabu leaf (EL), and cv. Dragon leaf (DL), respectively. In particular, the crude protein content was the highest in leaves. The crude fat content was in the order of developmental stage 1 of cv. Erabu fruit (EF1) and developmental stage 1 of native fruit (NF1) with values of 4.0%, and 3.9%, respectively. There was also high amount of crude fiber in stem of all three cultivars. The crude ash content was in the order of cv. Erabu leaf, cv. Dragon leaf, and developmental stage 3 of native fruit (NF3) with values of 23.2%, 17.4%, and 13.6%, respectively. The major minerals found in M. charantia were K, Ca, and Mg. The potassium contents of developmental stage 3 of native fruit (NF3), developmental stage 3 of cv. Dragon fruit (DF3), cv. Dragon stem (DS), and developmental stage 2 of native fruit (NF2) were 498.37, 339.21, 314.30, and 307.34 ㎎ /100g, respectively, while the calcium contents were decreased of EL, DL, and NL with values of 513.45, 371.69, and 209.43 ㎎/100g, respectively. The calcium content was higher in leaves and stems than fruits. On the otherhand, the highest magnesium content was measured in EL (69.92 ㎎/100g). The highest contents of chlorophyll a, chlorophyll b, and total chlorophyll were found in NL (442.9 ㎎/100g dw), EL (759.6 ㎎/100g dw), and EL (1164.9 ㎎/100g dw), respectively. The vitamin C contents from developmental stage 2 of cv. Erabu fruit (EF2), NF3, developmental stage 3 of cv. Erabu on fruit (EF3), and NF2 were found with values of 2849.9, 2330.5, 1985.1, and 1844.5 ㎎/㎏, respectively, and found to be higher in Korean cultivar and Erabu fruit than in Dragon. The charantin contents of leaf were higher than the fruit found to be 547.71, 506.04 and 395.62 ㎍/g dw in DL, EL and EF2, respectively. According to the results, mineral contents, total chlorophyll and charantin contents of M. charantia were higher in the leaves (EL and DL) than the fruits. And, vitamin C content was the highest in the fruit (EF2 and EF3). Therefore, much more research needs to be undertaken to use of the leaves as well as fruits. The data showed that M. charantia can be considered a good source of nutrient for application in food system.

Effect of Air Pollution on Cherry Tree (Prunus yedoensis Matsumara) Planted on National Roadsides between Cheonju and Kunsan City(II) - Chlorophyll, Magnesium and Sodium Contents in the Leaves - (대기오염(大氣汚染)이 전주(全州)-군산간(群山間) 국도변(國道邊) 왕벚나무에 미치는 영향(影響)(II) - Chlorophyll, Mg 및 Na 함유량(含有量)을 중심으로 -)

  • This report succeds the previous paper, the source of materials and statistical designs used were not altered. Under the hypothesis of maleffect of atmospheric pollutions emitted mainly by traffic automobiles on 20-year-old, Prunus yedoensis roadside trees planted along national road between Cheonju abd Kunsan, chlorophyll a and b contents, magnesium and sodium contents of leaves were analysed on September 15. Besides control sites considered to be a pollution free district, 20 polluted road-side sites, 10 on left roadside and 10 on right roadside, oppositely faced in pairs were selected. The leaves collected from 5 trees at each sampling site were bulked to eliminate the individual tree variation. Chlorophyll were extracted by Mackinney and Arnon method. The results obtained are as follows : 1. The planting belt width between road shoulder and paved road face edge for cherry trees, 160~170cm, was considered too narrow for the growing space. 2. On an average, the total chlorophyll content between Cheonju and Iri($8.60{\sim}9.31ml/cm^2$) was lower than that for between Iri and Kunsan($9.24{\sim}10.74ml/cm^2$). 3. The chlorophyll b content showed the difference, higher on right roadside and lower on left side. However this difference could be confirmed by matched pair T-test at 10% level. 4. Without exception, the chlorophyll contents of control sites were higher than those of road-side trees, implying the maleffect of atmospheric pollutants on cherry tree growth. 5. On the contrary, magnesium and sodium contents of the leaves of road-side trees were higher than that of control site trees.

Effect of Salinity Stress on Growth, Yield, and Proline Accumulation of Cultivated Potatoes (Solanum tuberosum L.) (염 스트레스에 따른 감자 품종 (Solanum tuberosum L.) 간 생육, 수량 및 proline 함량 변이)

  • This study evaluated the responses of 18 potato cultivars to three levels of salinity stress (electrical conductivity, EC: 1.0, 4.0, and $8.0dS{\cdot}m^{-1}$). Stem, leaf, root, chlorophyll, tuber yield, and proline content were investigated and statistically analyzed using analysis of variance (ANOVA) and correlations. Stem number and stem diameter were not affected by salinity, but stem length and aerial weight showed highly significant responses to salinity. Aerial weight decreased with increasing salinity levels in most cultivars, while it increased in some the cultivars 'Daejima', 'Goun', 'Haryeong', and 'LT-8'. Leaf number, leaf area index, and leaf weight were most significantly affected by salinity and the cultivar ${\times}$ salinity interaction. Root length, root weight, total chlorophyll and chlorophyll a were affected by salinity, but not by the cultivar ${\times}$ salinity interaction. The opposite trend was shown in chlorophyll b. Although there was great variability among cultivars, tuber yield decreased in all cultivars, and was most significantly influenced by salinity and the cultivar ${\times}$ salinity interaction. 'Superior', 'Kroda', 'Romana', and 'Duback' gave better tuber yields under salinity at EC 4.0 and $8.0dS{\cdot}m^{-1}$ than the cultivars with better aerial weights. Proline content was increased by salinity in all cultivars, and was more remarkable in the cultivars with better aerial weights than in cultivars such as 'Superior' and 'Kroda' with better tuber yields. Leaf number, leaf area index, leaf weight, and root length parameters were considered to be useful criteria in the evaluation of salt tolerance because of their high positive correlation with tuber yield; however, given its negative correlation with tuber yield under high salinity, proline content was not. Salinity tolerances varied greatly among potato cultivars. The low correlation between growth and yields of aerial parts under high salinity suggests that, in commercial agriculture, it might be more practical to compare relative yields to controls. Additionally, 'Superior', 'Kroda', 'Romana', and 'Duback' might be very useful cultivars to use in breeding programs to develop salinity-tolerant potatoes, as well as for sustainable potato production in saline areas.

Effect of Light Quality on the Growth of Panax ginseng in a Phytotron With Natural Light (자연광기상실에서 인삼생육에 대한 광질의 영향)

  • Ginseng plants in the 2nd year were grown under the shade (90-95%) of colored cellophane films at $15^{\circ}C$ in a natural light phytotron. Relative root grown (final/initial) was in decreasing order of blue, green, white, yellow and red. The growth of aerial part, especially stem length was poorest in blue light with relative root growth negatively correlated with stem length. In this relation each color showed specific domain. Stem length showed positive linear correlation with leaf area per plant in each and among light though it was not significant in red light. The content and ratio of chlorophyll a and b in leaf were in the decreasing order of White, Blue, Green, Red and Yellow. There is positive correlation between chlorophyll a and chlorophyll b, and between total chlorophyll and chl:a/b ratio. Blue color shade is expected to be beneficial for ginseng production.

  • Transgenic tobacco plants were produced by the transformation of ginseng CAB gene using Agrobacterium tumefaciens LBA4404. The presence of CAB gene in the second generation of transgenic tobacco plant was confirmed by genomic PCR. The photosynthetic ability of transgenic plants was higher than normal tobacco plants and the maximum photosynthetic point of transgenic and normal tobacco plants was 500 $\mu$mol m$^{-2}$ s$^{-1}$ . The photosynthesis of C7, C11, 1, C14 cell lines was higher than normal plants at all the light intensities investigated. The photosynthesis of C2, C11, C14 cell lines in 90% dark condition was higher than normal plants. The chlorophyll contents of transgenic tobacco plants were almost same as normal plants. The % of dry weight, nicotine content, total sugar and nitrogen contents of harvested transgenic tobacco plant leaves were almost same as normal plants.

Relationship between Phycoerythrin and Nitrogen Content in Gloiopeltis furcata and Porphyra yezoensis

  • Seasonal changes of pigment compositionin two intertidal red algae, Gloiopeltis furcata (Postels et Ruprecht) J. Agardh, and Porphyra yezoensis Ueda, were investigated. Chlorophyll α and phycoerythrin levels were high during winter, but decreased in late spring or summer, with accompanying discoloration from deep red to green or yellow. This discoloation corresponded closely to the fluctuationof phycoerythrin content. Nevertheless, photosynthesis capacity was maintained by the increasing water temperature in the field, suggesting that large amounts of phycoerythrin are not necessary for photosynthesis. Phycoerythrin conten correlated significantly with nitrogen content in both species when the nitrogen level was greater than the level of critical content (1.30% DW in G. furcata, and 2.26% DW in P. yezoensis), indicating that phycoerythrin plays a more important role in the nitrogen status as a nitrogen pool than that of nitrogen critical content. Furthermore, the dependence level of the alage on phycoerythrin as a nitrogen pool was greater in P. yezoensis than in G. furcata because of the remarkable increase of phycoerythrin content in P. yezoensis with increasing jnitrogen content.

Effects of Salt Stress on Protein Content, ATPase and Peroxidase Activities in Tobacco. (염스트레스가 담배식물의 Protein, ATPase 및 Peroxidase 활성에 미치는 영향)

  • The analysis of biochemical changes in tobacco plant as increase of NaCl concentraion was conducted. Total protein content and soluble protein content were decreased as NaCl concentration was increased, in that steady decreased until 120mM NaCl and largely decreased at 150mM NaCl. The expression of 74Kd subunit was increased until 60mM NaCl. However, the amount of 74Kd protein was decreased from 90mM NaCl. There was no difference for expression of other protein subunits. Chlorophyll a content was significantly decrease as NaCl concentration was increased, but chlorophyll b content was not much decreased. The slow increase up to 120mM NaCl and large increase at 150mM NaCl for ATPase and peroxidase activities indicated that 120mM NaCl could be a limiting concentration for salt injury.

Comparison of Growth and Essential Oil Composition in Two Hydroponically Grown Species of Thymes at Different Nutrient Solution Strength (수경재배시 배양액 내 이온농도에 따른 두 종의 백리향 생육 및 정유 성분 비교)

  • The growth and quality differences between common thyme(Thymus vulgaris L.) and lemon thym (Thymus$\times$citriodorus) were investigated. They were grown in a hydroponics culture system (DFT) with a nutrient solution developed by the European Vegetable R&D Center in Belgium. Nutrient solution ionic strength used were 1.2, 2.4, 4.8 and 7.2 mS.cm-1. The dry matter of two thymes were increased with increasing ionic strength, while shoot length, root length and fresh weight were decreased. Chlorophyll content was higher in lemon thyme, while vitamin C content and essential oil content were higher in common thyme. The essential oil content showed a similar trend as those of growth. The main compostion of essential oil were thymol and carvacrol in common thyme, and geraniol and $\alpha$-citral in lemon thyme. The highest content of these compounds was obtained in 1.2mS.cm-1 treatment. Consequently, the ionic strength of 1.2mS.cm-1 was suitable for the production of quality thymes.

Comparison of Chlorophyll Contents and Patterns of Korean and Japanese Powder Green Tea (한일 말차(抹茶)의 클로로필 함량 및 패턴 비교)

  • This research used High Performance Liquid Chromatography(HPLC) to analyze and compare the content and the pattern of chlorophyll to find the difference in quality, water temperature, and the stirring time between the three different types of Korean and Japanese powder green tea, which were currently on the market. Chlorophyll contents of Japanese-made medium quality tea (S5: 8.34mg/g), and low quality tea (S6: 8.23mg/g) were relatively high, and that of Korean-made medium quality tea (S2 ; 4.87mg/g) was lower. The water temperature of 85$^{\circ}C$ and 95$^{\circ}C$ were high except in Korean-made medium and low quality powder green tea (S2 and S3). Japanese-made powder tea required 60 seconds of stirring time and Korean-made tea required 15 seconds, but 30 seconds seem to be the ideal stirring time for both of them. Chlorophyll a, b, pheophytin a, b, and pheophorbide were confirmed from the chlorophyll pattern. There were 1.5∼2 times more of chlorophyll a in Japanese-made one compared to that of Korean-made one; also 1.5∼2 times more of pheophytin a in Korean-made one than Japanese-made one. It might be concluded that Japanese-made powder green tea was higher in quality.
