  • Bacillus sp. A56 was studied, because of its high flocculating activity. The flocculating substance produced by this strain was purified by ethanol precipitation, cetylpyridinium chloride (CPC) precipitation, and gel permeation chromatography (GPC). The FT-IR spectrum of the purified bioflocculant, designated as BF-56, showed typical characteristics of polysaccharides. The non-sugar substituents, and sugar components of BF-56 containing glucose, fucose, glucuronic acid, and galactose in an approximate molar ratio of 2.76:1.10:1:0.12, suggested that it was a novel bioflocculant with an estimated molecular mass of over $7{\times}10^3$ kDa. Rheological analysis of BF-56 revealed that it was a pseudoplastic that had higher apparent viscosity rate at dilute concentrations than those of zooglan. The solution of bioflocculant BF-56 exhibited non-Newtonian characteristics and it was compatible to high concentrations of salts such as KCl, NaCl, $CaCl_2,\;or\;FeCl_3.$ The present results suggested strong possibility of bioflocculant BF-56 to be fully applicable to industries such as wastewater treatment.

  • A bacterial strain WN9, which produced a new type of extracellular polysaccharide, was isolated from soil samples. By morphological, physiological, biochemical, and phylogenetic studies, strain WN9 was identified as a Paenibacillus sp. and it was named as Paenibacillus sp. WN9, which produced a high molecular extracellular polysaccharide from glucose. The molecular weight of the exopolysaccharide (EPS-WN9) was estimated to be about 31.5 mega-Da. The FT-IR spectrum of EPS-WN9 revealed typical characteristics of polysaccharides. EPS- WN9 consisted of D-glucose and D-mannose with a molar ratio of 1:1.4 being identified as a neutral sugar component. The acidic component of EPS- WN9 was tyrosine. Rheological analysis of EPS- WN9 revealed that the pseudoplastic property and its apparent viscosity remained stable at various temperatures and pHs.

  • The equation to predict the soil temprature of Pusan and Chinju city as a function of time and soil depth for the geothermal energy utilization system and agriculture was devised. The equation was $T(x,t)\;=\;Tm\;-\;To{\cdot}ExP(-{\xi}){\cdot}cos{{\omega}{\cdot}[t-to-x/(2{\cdot}{\alpha}{\cdot}{\omega})^{0.5}]}$ with the soil thermal diffusivity, ${\alpha},\;of\;0.4\;\textrm{m}^2/day,\;0.0375\;\textrm{m}^2/day$ and phase zero point, to, of 24 days, 22.4 days in Pusan and Chinju city, respectively, during ten years from 1987 to 1996. The predicted and measured soil temperatures agreed well with the coefficient of determination of 0.95 at the soil depth of 0.0, 0.5, 1.0, 3.0, 5.0 m. The maximum and minimum temperature in Pusan 3.7, $30.1^{\circ}C$ at soil surface and 14.3, $18.0^{\circ}C$ at the depth of 5.0 m. The total mean temperature of soil in Pusan and Chinju city was about 16.3, $16.0^{\circ}C$, respectively.

  • Porcine epidemic diarrhea virus (PEDV) strain Chinju99, which was previously isolated from piglets suffering from severe diarrhea was used to characterize the membrane (M) protein gene to establish the molecular information, and the results will be useful in elucidating concepts related to molecular pathogenesis and antigenic structures of PEDV isolates. The Chinju99 M gene generated by reverse transcription and polymerase chain reaction (RT-PCR) consisted of 681 bases containing 22.3% adenine, 22.3% cytosine, 23.1% guanine and 32.3% thymine nucleotides, and the GC content was 45.4%. It had some nucleotide mismatches from M gene of other PEDV strains, such as CV777, Br1/87, KPEDV-9, JMe2, JS2004-2 and LJB-03 with 97-99% nucleotide sequence homology to these strains. Also, it encoded a protein of 226 amino acids, which had some mismatches from those of CV777, Br1/87, KPEDV-9, JMe2, JS20004-2 and LJB-03, as the amino acid sequence homology showed a 97-98% to these strains. The Chinju99 had a very close relationship to the Japanese strain JMe2 for the nucleotide and amino acid sequences of the M gene. The amino acids predicted from Chinju99 M gene consisted of mostly hydrophobic residues and contained three potential sites for asparagine (N)-linked glycosylation, two serine (S)-linked phosphorylation sites by protein kinase C, and two S- or threonine (T)-linked phosphorylation sites by casein kinase II.

  • To provide information for the molecular pathogenesis and antigenic structures of Korean isolates of porcine epidemic diarrhea virus (PEDV), the small membrane (sM) protein gene of Chinju99 strain, which was previously isolated from piglets suffering from severe diarrhea was characterized and further analyzed with other PEDV strains. The sM gene of Chinju99 generated by reverse transcription and polymerase chain reaction had a single open reading frame with 231 bases consisting of 24.2% adenine, 18.6% cytosine, 18.1% guanine and 39.0% thymine nucleotides. Nucleotide sequence of the gene revealed 97.8% homology to those of Belgian strain CV777 and British strain Br1/87, and 97.0% to Chinese strain LZC. The gene encoded a protein with 76 amino acids, and putative amino acid sequence of the gene revealed 98.7% homology to those of CV777 and Br1/87, and 96.1% to LZC. The amino acids of Chinju99 sM gene consisted of mostly hydrophobic residues, and there were one potential N-myristylation site and one potential threonine (T)-linked phosphorylation site recognized. Also, there was a transmembrane region with 46 amino acids, and Chinju99 was more close to CV777 and Br1/87 than to LZC in phylogenetic analysis on the sM amino acid sequences.

  • We attempt to get a probabilistic model of a queueing system in the maximum entropy condition. Applying the maximum entropy principle to the queueing system, we obtain the most uncertain probability model compatible with the available information expressed by moments.

  • The protective effects of a Platycodi Radix (Changkil: CK), the root of Platycodon grandiflorum A. DC (Campanulaceae). on carbon tetrachloride-induced hepatotoxicity and the possible mechanisms involved in this protection were investigated in mice. Pretreatment with CK prior to the administration of carbon tetrachloride significantly prevented the increased serum enzymatic activities of alanine and aspartate aminotransferase in a dose-dependent manner. • This study was conducted to investigate physicochemical property changes from July 1998 to August 2001 in Mt. Bukhansan National Park. Four water sampling points were selected to measure the quality of stream water in the northeastern part of the Mt. Bukhansan National Park. The results were summarized as follows; In spring, the average pH of stream water was below the first class of the river water quality standard, while it was normal level in summer. The average electrical conductivity was about 2.3~3.3 times higher in downstream water than in upstream water during spring and summer. The contents of anions($Cl^-$, $NO{_3}^-$, $SO{_4}^{2-}$) were about 1.1~7.4 and 0.4~11.4 times higher in downstream than in upstream water, respectively. These results indicate that water quality was poorer in downstream than in upstream water. We suggest that stream water in the Mt. Bukhansan National Park should be protected from impacts of snow melting mineral particles in spring season and human impacts like wastewater of point source in summer season.
