Enterobacter cloacae, an Emerging Plant-Pathogenic Bacterium Affecting Chili Pepper Seedlings

  • A previously unreported bacterial disease on chili pepper (Capsicum annuum L.) seedlings affecting as many as 4% of seedlings was observed in greenhouses in Chihuahua, Mexico (Delicias and Meoqui counties). Initial lesions appeared as irregular small spots on leaves and brown necrosis at margins tips were observed. Later, the spots became necrotic with a chlorotic halo. Advanced disease was associated with defoliation. A Gram negative, rod-shaped bacterium was isolated from diseased chili pepper seedlings. Three inoculation methods revealed that isolated strains produce foliage symptoms, similar to those observed in naturally infected seedlings. Pathogenic strains that caused symptoms in inoculated seedlings were re-isolated and identified to fulfill koch's postulate. Polyphasic approaches for identification including biochemical assays (API 20E and 50CH), carbon source utilization profiling (Biolog) and 16S rDNA, hsp60 and rpoB sequence analysis were done. Enterobacter cloacae was identified as the causal agent of this outbreak on chili pepper seedlings.

Influence of Culture Media Formulated with Agroindustrial Wastes on the Antimicrobial Activity of Lactic Acid Bacteria

  • The discarding of wastes into the environment is a significant problem for many communities. Still, food waste can be used for lactic acid bacteria (LAB) growth. Here, we evaluated three growth media equivalent to de Mann Rogosa Sharpe (MRS), using apple bagasse, yeast waste, fish flour, forage oats, and cheese whey. Cell-free supernatants of eight LAB strains were tested for antimicrobial activity against nine indicator microorganisms. The supernatants were also evaluated for protein content, reducing sugars, pH, and lactic acid concentration. Cell-free supernatants from fish flour broth (FFB) LAB growth were the most effective. The strain Leuconostoc mesenteroides PIM5 presented the best activity in all media. L. mesenteroides CAL14 completely inhibited L. monocytogenes and strongly inhibited Bacillus cereus (91.1%). The strain L. mesenteroides PIM5 consumed more proteins (77.42%) and reducing sugars (56.08%) in FFB than in MRS broth (51.78% and 30.58%, respectively). Culture media formulated with agroindustrial wastes positively improved the antimicrobial activity of selected LAB, probably due to the production of antimicrobial peptides or bacteriocins.

Fat replacement by pecan nut and oregano oil and their impact on the physicochemical properties and consumer acceptability of frankfurters

  • Objective: A study was conducted to determine the physicochemical quality and consumer acceptability of beef frankfurter-type sausages formulated with pecan nut paste and Mexican oregano oil (MO) of two varieties, Poliomintha longiflora Gray (Mexican oregano Poliomintha, MOP) or Lippia berlandieri (Mexican oregano Lippia, MOL). Methods: Frankfurters were processed under six treatment conditions: control (10.67% pork fat), MOP (control + 0.01% MOP), MOL (control + 0.01% MOL), MOP-N (control + 0.01% MOP + 6% pecan nut paste), MOL-N (control + 0.01% MOL + 6% pecan nut paste), and C-N (control + 6% pecan nut paste). The physicochemical properties and the consumer acceptability were determined. Results: The addition of MO and nut did not influence the water holding capacity, drip loss, and luminosity of frankfurters, but those ingredients increased pH and shear force (p<0.05) and decreased redness (p<0.05) of frankfurters. Frankfurters were generally well accepted by consumers. However, general acceptability of sausages decreased with the addition of MO. Control sausages showed the highest acceptability. Conclusion: The MO and pecan nut paste do not affect drastically the quality of frankfurters. These results provide evidence that partial replacement of pork fat by pecan nut in frankfurters maintain a consistent physicochemical quality and its commercialization looks promising given consumers' acceptance.

Characterization and Functional Properties of an Oat Gum Extracted from a Drought Harvested Oat (Avena sativa)

  • An oat gum was extracted from whole seeds of a drought harvested oat (Avena sativa). Oat gum presented a ${\beta}-glucan$ content of 65%(w/w) and an intrinsic viscosity of 141 mL/g. Gelling capability of oat gum at different concentrations was investigated. Gel hardness increased from 0.08 to 0.25 N as the oat gum concentration changed from 5 to 10%(w/v). Whippability, foam stability, emulsion stability, and reduced viscosity of oat gum at different pH were also investigated. Oat gum whippability was maximum at pH 7 (146%), while the higher foam and emulsion stability values were found at pH 9 (88 and 96%, respectively). The gum reduced viscosity increased from 715 to 958 mL/g as the pH changed from 7 to 9. Oat gum shows great potential as a gel forming, thickening, and stabilizing agent.

A case of Candida albicans-induced fungal keratitis in a Chihuahua dog - with a focus on optical coherence tomographic features

  • A 4-year-old Chihuahua dog was referred for bilateral corneal ulcers. Slightly raised white fluorescein-positive plaque-like corneal lesions in both eyes appeared as intense hyperreflective areas with posterior shadowing on optical coherence tomography (OCT). Based on corneal cytology and culture, Candida albicans-induced fungal keratitis was diagnosed. Despite treatment, on OCT, endothelial plaques, increased stromal infiltration thickness, vertical shapes of the ulcer edge, and necrotic stromal space were judged to be aggravation of the disease, and surgery was performed. Conjunctival grafting surgery with topical 1% voriconazole effectively resolved fungal keratitis. OCT can provide detailed and objective information related to the disease prognosis.

Prevalence of dog erythrocyte antigens 1 and 7 in eleven canine breeds in the Republic of Korea

  • Blood type in dogs is based on the antigen present on the red blood cell surface. Dog erythrocyte antigen 1 is a crucial red blood cell antigen in dogs, whereas the dog erythrocyte antigen 7 has been studied in limited dog breeds worldwide. To assess the prevalence of dog erythrocyte antigens 1 and 7 in 11 breeds in the Republic of Korea, 624 dog blood samples were examined for antigen detection. Overall, 520 dogs (83.3%) showed dog erythrocyte antigen 1 expression. The distribution varied from 50.0~100.0% according to the breed. Dog erythrocyte antigen 1-positive blood type was the highest in Chihuahua (100%), followed by Jindo dog (98.5%), and Sapsaree (95.3%). Dog erythrocyte antigen 7 was positive in 125 dogs (20.0%), and the positivity varied from 5.0~42.9% according to the breed. Dog erythrocyte antigen 7-positive blood type was the highest in Beagle (42.9%), followed by Chihuahua (37.5%), and Jindo dog (27.8%). The high prevalence of dog erythrocyte antigen 1 is because of the high proportion of Jindo dog and Sapsaree breeds that were mostly positive for the antigen. The high abundance of these breeds could be due to inbreeding and local breeding in the Republic of Korea. To our best knowledge, this study is the first to report on the prevalence of dog erythrocyte antigens 1 and 7 among various canine breeds in the Republic of Korea. The prevalence data obtained from this study may contribute to baseline information on veterinary transfusion medicine in small animal practice.

Analgesic Effect of Combination of Ketamine Hcl/Propionyl promazine HCI and Tiletamine HC1/Zolazepam HCl for Scaling in Dog (개의 치석제거를 위한 마취에 있어 Ketamine HCl/Propionyl promazine HCI과 Tiletamine HCl/Zolazepam HCl의 투여효과)

  • To study effective dossage and administration route for scaling, ketamine HCl/propionyl promazine HCl(ketamine) combination and tiletamine HCl/zolazepam HCl(zoletil) were administered in one hundred six dogs. The dogs were toy poodle, Yorkshire Terrier, Pekingese and Chihuahua. Scaling and polishing time, possible treatment time after the first injection of anesthetics, the number of anethesia added, presence of tongue movement during anesthesia, the presence of swaying sign during recovery and respiration were evaluated. The possible treatment time after the first Injection of anesthetic in toy poodle were 26.3${\pm}$3.0 minutes with intravenous(IM) treatment of ketamine 10mg/kg, and 21.4${\pm}$6.6 minutes with intramuscula(IM) treatment of zoletil 8mg/kg, In Yorkshire Terrier were 19.51: 1.7 minutes with IV treatment of ketamine 10mg/kg. 19.0${\pm}$5.2 minutes IM and 20.8${\pm}$6.1 minutes with IM treatment of zoletil 5mg/kg,24.8${\pm}$3,5 minutes with IM treatment of zoletil 8mg/kg. In pekingese were 27.5${\pm}$2.1 minutes with IM treatment of ketamine 10mg/kg,28.0${\pm}$4.2 minutes with IM treatment of zoletil 8mg/kg. In Chihuahua were 19.5${\pm}$1.9 minutes with IV treatment of ketamine 7mg/kg, 17.5${\pm}$1.7 minutes with IM treatment of ketamine 10mg/kg and 20.3${\pm}$3.8 minutes with IM treatment of zoletil 5mg/kg, 21.2${\pm}$5.5 minutes with IM treatment of zoletil 8mg/kg. Swaying sign was observed in all group during recovery time, espically, in toy poodle and Yorkshire Terrier which administered zoletil 8mg/kg IM showed more severe swaying sign. The present results suggested that injection of zoletil 8mg/kg IM might be relatively effective for scaling in Chihuahua Within 20 minutes treatment for scaling in Yorkshire Terrier and Chihuahua, IM treatment of ketamine 7 to 10mg/kg is recommended.

Effects of Candida norvegensis Live Cells on In vitro Oat Straw Rumen Fermentation

  • This study evaluated the effect of Candida norvegensis (C. norvegensis) viable yeast culture on in vitro ruminal fermentation of oat straw. Ruminal fluid was mixed with buffer solution (1:2) and anaerobically incubated with or without yeast at $39^{\circ}C$ for 0, 4, 8, 16, and 24 h. A fully randomized design was used. There was a decrease in lactic acid (quadratic, p = 0.01), pH, (quadratic, p = 0.02), and yeasts counts (linear, p<0.01) across fermentation times. However, in vitro dry matter disappearance (IVDMD) and ammonia-N increased across fermentation times (quadratic; p<0.01 and p<0.02, respectively). Addition of yeast cells caused a decrease in pH values compared over all fermentation times (p<0.01), and lactic acid decreased at 12 h (p = 0.05). Meanwhile, yeast counts increased (p = 0.01) at 12 h. C. norvegensis increased ammonia-N at 4, 8, 12, and 24 h (p<0.01), and IVDMD of oat straw increased at 8, 12, and 24 h (p<0.01) of fermentation. Yeast cells increased acetate (p<0.01), propionate (p<0.03), and butyrate (p<0.03) at 8 h, while valeriate and isovaleriate increased at 8, 12, and 24 h (p<0.01). The yeast did not affect cellulolytic bacteria (p = 0.05), but cellulolytic fungi increased at 4 and 8 h (p<0.01), whereas production of methane decreased (p<0.01) at 8 h. It is concluded that addition of C. norvegensis to in vitro oat straw fermentation increased ruminal fermentation parameters as well as microbial growth with reduction of methane production. Additionally, yeast inoculum also improved IVDMD.

A Case of Traumatic Brain Injury in a Chihuahua Dog: Serial Clinical and Computed Tomographic Findings (치와와에서 발생한 외상적 뇌손상 증례; 연속적인 임상적 및 전산화 단층영상 소견)

  • A dog (Chihuahua, 2-year-old, intact female) was referred to us because of cluster seizure. She had history of falling from height few days before presentation. Brain computed tomography (CT) results demonstrated fracture line on right temporal bone and hypodense, edematous changes of the adjacent brain parenchyma on right cerebral parenchyma. Based on history, clinical signs, and diagnostic imaging findings, this patient was diagnosed to traumatic brain injury. After diagnosis, the patient was well controlled with anti-inflammatory drug and anti-epileptic drugs. When 30, 480, and 1260 days after initial brain CT examination, we performed serial brain CT rechecks. This case report describes serial clinical and brain CT findings after traumatic brain injury.

Surgical Treatment of Sliding Hiatal Hernia by Incisional Gastropexy at Fundus and Pylorus in a Chihuahua Dog (치와와견에서 위저부 및 유문부의 절개성 위고정술을 통한 축성열공허니아의 수술적 치료)

  • A 1 year 2 months old, male Chihuahua dog weighing 0.92 kg was presented with a history of intermittent vomiting after eating since two weeks ago. Based on the history, clinical signs, physical examination, and contrast radiographs, the diagnosis of sliding hiatal hernia was made. Diaphragmatic plication, esophagopexy and left-sided gastropexy concurrently required for surgical correction but in this case, diaphragmatic plication and esophagopexy could not be applied because of the patient's special condition. Instead, left-sided gastropexy was performed with additional pyloropexy. To treat reflux esophagitis, cimetidine and sucralfate were administrated and patient was fed in an upright position. Two days after the surgery, the patient showed normal activity and after then there was not found recurrence or complication.
