Effect of Combination of Chitosan Coating and Gamma Irradiation on the Foodborne Pathogen Reduction and Nutritional Properties of Chicken Egg (키토산 코팅 및 감마선 조사에 의한 계란에 오염된 위해 미생물 제어 및 영양학적 특성 평가)

  • The effect of combination of chitosan coating and gamma irradiation on Escherichia coli inactivation and nutritional properties of shell egg was investigated. The E. coli inoculated on shell egg was not detected by 2 kGy of gamma irradiation at day 0 and/or chitosan coating (1%, pH 5.0) after 3 days of storage. There was no E. coli detected thereafter. In the contents of moisture, crude fat, crude protein, ash, retinol, phospholipid, and minerals, no difference was found by treatment combination. However, the contents of cholesterol and carotenoids were reduced by irradiation of 2 kGy (P<0.05). Results suggest that the combination of gamma irradiation (2 kGy) and chitosan coating (1%) can be a good method to improve the safety and prolonged shelf-life of egg because of effective pathogen inactivation without significant adverse changes in nutritional quality.

Development of Nondestructive Sorting Method for Brown Bloody Eggs Using VIS/NIR Spectroscopy (가시광 및 근적외선 전투과 스펙트럼을 이용한 갈색 혈란 비파괴선별 방법 개발)

  • The aim of this study was the non-destructive evaluation of bloody eggs using VIS/NIR spectroscopy. The bloody egg samples used to develop the sorting mode were produced by injecting chicken blood into the edges of egg yolks. Blood amounts of 0.1, 0.7, 0.04, and 0.01 mL were used for the bloody egg samples. The wavelength range for the VIS/NIR spectroscopy was 471 to 1154 nm, and the spectral resolution was 1.5nm. For the measurement system, the position of the light source was set to $30^{\circ}$, and the distance between the light source and samples was set to 100 mm. The minimum exposure time of the light source was set to 30 ms to ensure the fast sorting of bloody eggs and prevent heating damage of the egg samples. Partial least squares-discriminant analysis (PLS-DA) was used for the spectral data obtained from VIS/NIR spectroscopy. The classification accuracies of the sorting models developed with blood samples of 0.1, 0.07, 0.04, and 0.01 mL were 97.9%, 98.9%, 94.8%, and 86.45%, respectively. In this study, a novel nondestructive sorting technique was developed to detect bloody brown eggs using spectral data obtained from VIS/NIR spectroscopy.

Identification of a SNP in Chicken CaSR Gene and Its Effect on Economic Traits (닭의 CaSR 유전자내 단일 염기 변이 탐색 및 경제 형질간의 연관성 분석)

  • The Function of the calcium sensing receptor (CaSR) is to control calcium levels by altering PTH (parathyroid hormone) secretion and renal calcium resorption. The influence of calcium on the basal and stimulated release of several hormones from chicken pituitary glands has been determined in vitro. The objective of this study was to identify SNP in chicken CaSR gene and to investigate the effect of the SNP on economic traits. The sequencing analysis method was used to identify nucleotide polymorphisms within chicken CaSR gene. This study identified SNP at position 1949 bp(Genebank accession No : XM_416491) in the exon 1. The SNP changed the amino acid to alanine(GCC) from serine(TCC). This SNP showed three genotypes, AA, AS and SS by digestion with the restriction enzyme NcoⅠ using the PCR-RFLP method. The A963S showed significant effect only on the first lay day (P<0.05) in Leghorn population. Leghorn with the genotype AA had significantly faster the first lay day(137.6) than the genotype AS(143.0, P<0.05). Also, the A963S showed significant effect only on the first lay day(P<0.05) and mean of egg weight(P<0.05) in KNC population. KNC with the genotypes AA ans AS had significantly faster the first lay day (151.0 and 152.6, respectively) than the genotype SS(159.4, P<0.05). And the genotypes SS had significantly heavier the mean of egg weight(50.4 kg, P<0.05) than the genotype AA ans AS (47.5 and 47.8 kg, respectively). According to result of this study, an a allele of the A963S was found to have a significant effect on the first lay day. It will be possible to use this SNP marker on selecting chicken to improve the first lay day.

Development of a live vaccine strain of duck viral hepatitis using a Korean isolate (국내 분리주를 이용한 오리 바이러스성 간염 생백신주의 개발)

  • Duck viral hepatitis is an acutic, highly infectious viral disease of young ducklings. The most practical means for controlling duck viral hepatitis is the vaccination of ducklings or of a breeding stock. We attempted to develop a vaccine strain of duck hepatitis virus (DHV) using a Korean isolate by serial chicken embryo passages. The propagation of DHV in chicken embryos was carried 140 passages. After the $50^{th}$ passage, of which the virus was non-pathogenic for ducklings, approximately every $20^{th}$ passage of the virus was tested for vaccinal efficacy. Both the $70^{th}$ and $90^{th}$ passage of the virus gave good protection against challenge infection to a DHV-DRL reference strain(type 1) and a virulent Korean isolate. The $110^{th}$, $125^{th}$ and $140^{th}$ passage of the virus were less protective than the $70^{th}$ and $90^{th}$ passage, which means that more than $110^{th}$ passage may lead to over-attenuation of the virus. Ducklings vaccinated with the chicken-embryo-adapted virus by oral, intramuscular or eye drop administration showed earlier resistance to challenge infection from 3 to 7 days postvaccination. Of the above methods, ducklings vaccinated intramuscularly presented the most rapid resistance against challenge. The minimum immune dose of the chicken-embryo-adapted virus in ducklings was also studied. Ducklings inoculated with a dose of $10^{2.0}\;ELD_{50}$ and below were not fully protected against challenge with a virulent DHV, showing a protection rate of 67% to 73%, but ducklings inoculated with a dose of $10^{3.0}\;ELD_{50}$ and over were completely protected. The virus yield of the chicken-embryo-adapted DHV was examined at 24hrs and 48hrs of the incubation time in the allantoic fluid, embryo head and embryo minus head of the embryonating egg. In all three components, the titer of the virus was higher at 48 hours than that at 24 hours after incubation. And the titer of the virus was higher in the embryo minus head, embryo head and the allantoic fluid, in order. Field trials for evaluating the efficacy of the attenuated DHV as a live vaccine were done in duck farms with about 25% mortality of flocks resulting from duck viral hepatitis. After the use of the experimental vaccine, the mortality due to duck viral hepatitis was dramatically reduced in the farms. These results indicated that the attenuated DHV using a Korean isolate could be a good candidate as a live vaccine strain of DHV in Korea.

Studies on Energy Metabolism of Growing Chicken (닭의 에너지 대사(代謝)에 관(關)한 연구(硏究))

  • An energy metabolism study was conducted with two commercial strains of meat-type chickens, Hubbard and Cobb, and two egg strains, Hy Line and Korean-bred Hanhyup 325. The heat production of growing chickens from each strain were measured by the use of an open-circuit gravimetric respiration calorimeter. The data obtained from this study were summarized as fallows. 1. The average body weight of 9-wk-old Hubbard broilers reared in battery cages was 2,570g/bird. The average body weights of 9-wk-old Hy Line chicks and Hanhyup 325 were 777 and 748g/bird, respectively. 2. At 3 weeks of age, the Hubbard broiler chicks consumed two times the feed consumed by Hy Line chicks (54.6g VS. 26.7g/bird/day). These values increased to 151g and 57.2g/bird/day, respectively, at 8 weeks of age, indicating that the difference in feed intake between meat and egg-type chicks tends to increase as they grow older. In terms of water consumption, the 5-wk-old Hubbard broiler chicks drank $226m{\ell}/bird/day$ as compared with $58m{\ell}$ by Hy Line chicks. These values increased to 282 and $70m{\ell}$, respectively, at 8 weeks of age. 3. The excreta outputs of Hubbard broilers and Hy Line chicks were 18.7 and 6.1g DM/bird/day at 4 weeks of age, and 41.5 and 10.0g DM/bird/day at 8 weeks of age, respectively. 4. The energy metabolizability of broiler chicks were 75.4~77.1% compared to 75.0~83.5% by egg-type chicks. 5. The respiratory quotient (RQ) was between 0.78 and 0.97. There seems to be no difference in RQ between meat and egg-type chicks. The RQ tended to decrease when feed intake was low and vice versa. 6. Both meat and egg-type chicks produced $83.1{\sim}123.1Kcal/kg^{\frac{3}{4}}B.W./day$. The considerably low value of $83Kcal/kg^{\frac{3}{4}}B.W./day$ was obtained when the chicks were off the feed under the stressful conditions. The high value of 123.1Kcal was obtained when the chicken chamber temperature rose to $27{\sim}34^{\circ}C$.

Effects of Dietary Supplementation of Copper-soy Proteinate on the Performance of Laying Hens (Copper-soy Proteinate의 첨가가 채란계의 생산성에 미치는 영향)

  • This study was conducted to investigate the effects of dietary supplementation of copper-soy proteinate (Cu-SP) on the performance of laying hens. A total of 1,000 Hy-Line Brown laying hens of 32 wks old were assigned to one of the following 5 dietary treatments: Control, Cu-SP 50, 100, 150, 200 (50, 100, 150, 200 ppm Cu supplementation as Cu-soy proteinate). Each treatment was replicated 4 times with fifty birds per replication, housed in 2 birds cages. Fifty birds units were arranged according to randomized block design. Feeding trial lasted 5 wks under 16L : 8D lighting regimen. There were no significant differences among treatments in hen-day and hen-house egg production, egg weight, broken & soft egg production. Feed intake and feed conversion rate (FCR) were significantly (P<0.01) lower in Cu-SP treated groups than Control. Eggshell thickness was significantly (P<0.01) higher in Control than Cu-SP 100, 150 and 200. Egg yolk color index was significantly (P<0.01) higher in Cu-SP 200 than other treatments. Egg shell color index was significantly (P<0.05) higher in Cu-SP 150 and Cu-SP 200 than Cu-SP 100. Concentration of copper, iron and zinc of the egg yolk were not significantly influenced by treatment. There were no significant differences in the level of leukocytes and erythrocytes in the chicken blood. The result of this experiment showed that dietary supplementation of Cu-SP at the level of 50ppm of Cu can reduce feed intake and FCR.

Significance of a Highly Specific and Sensitive Enzyme Linked Immunosorbent Assay on Evaluation of Environmental Toxicant-Mediated Allergic Responses

  • Enhancement of antigen-specific IgE is a hallmark of allergic hyperresponsiveness, therefore it is necessary to adopt or develop a highly sensitive and specific assay for determination of allergen-specific IgE levels in vivo. In this presentation, we introduce an ELISA (enzyme linked immunosorbent assay) system developed to measure the levels of chicken egg ovalbumin (OVA)-specific IgE in serum. The ELISA method uses a commercially available purified rat anti-mouse IgE as a capture Ab and biotinylated OVA as a detection reagent. Avidin-peroxidase with its substrate is used for color development resulting in optical density measurement at 405 nm. The ELISA system produces a highly sensitive dose-response relation-ship between optical density levels and the dilution titer of the OVA-IgE standard serum but no cross-reaction with unrelated IgE or IgG. It is believed that the system is an Efficient tool to delineate an adjuvant effect of environmental pollutants on development of asthmatic and atopic responses.

The Present and Future of Pheasant Raising in Korea (꿩의 생산기술 현황과 생산전망)

  • Though pheasants (Korean ring-necked pheasant) have been raised for several decades, their behavior and wild nature are far from domestication. The pheasant is a seasonal breeding species and lays a limited number of eggs in a breeding season. The growth rate and feed efficiency of pheasants are very low as compaired with those of chicken for meat purpose. In addition, the breeder's access to one's herd for care is not easy. From these reasons, pheasants seem to be unsuitable for meat production in a large flock at present. However, pheasant raising is expected to increase slowly in accordance with rising demand for special poultry meat. Therefore, it is necessary to improve techniques about raising, feeding and rnanagement, so that the consumer price of pheasant meat can be lowered down reasonably.

  • This study was designed to establish Korean food culture by analizing 17 sets of Jinyounuigue(진연의궤) Jinchanuigue(진찬의궤), and Jinjarkuigue(진작의궤) which were the records of royal party procedures in Yi dynasty. Side dishes were classified into 20 groups in this study ; Tang (場) 19, Jungol (전골) 3, J'im 18, Jun (전) 20, Jock 14, Pyunuk (片肉) 14, Cho 12, Hyae 17, Po 8, Chae 3, Bung 1, Nanri 1, Sooran 1, Sookran 1, Jaban 1, Kimchi 2, etc. all of 140 different kinds of side dishes. There was no tendency in omission or addition of food materials. Food materials were beef, pork, lamb, chicken, duck, peasant meat, dock's egg, fish, shellfishes, mollusca, curstacea, seaweeds, vegetables, fruits, beancurds, muk (a starch jelly), d'ock, muchrooms, etc. Seasonings were soysauce, pepper, sesame oil, ginger, green-onion, garlic, bean paste, ginger powder, red pepper powder, red pepper paste, salts, vinegar, honey, sesame power, etc.

  • Infectious coryza is one of the important acute respiratory diseases causing a significant egg drop and retarded growth in chicken. An attempt for the isolation of etiologic agent was made by utilizing SPF sentinel birds housed in commercial farms and the results obtained are as follows. Fifteen isolates of Haemophilus gallinarum were tested for their biological and serological characteristics with reference strains, 221 and Modesto and subsequently classified into two serotypes. Of them, isolates immunolocally identical to the standard strains were also selected as vaccine strains for future studies.
