Rhodamine Based Fluorescent Chemosensors for Hg2+ and its Biological Application

  • Two new chemosensors, rhodamine 6G derivative bearing hydroxyethyl group (1) and rhodamine base derivative bearing 15-crown-5 group (2) were synthesized and their sensing behaviors toward various metal ions were investigated by UV/Vis and fluorescence spectroscopies. Addition of $Hg^{2+}$ ion to a $CH_3CN$ solution of 1 and 2 gave visual color changes as well as fluorescent OFF-ON observations. Selectivity and sensitivity of 1 towards $Hg^{2+}$ are excellent enough to detect micromolar level of $Hg^{2+}$ ion, even in equeous media and biological sample (HeLa cell).

  • Since heavy metal pollution is a significant global environmental problem and very dangerous to human health, the improved methods for detecting heavy metals are required recently. Colorimetric chemosensors are now considered as one of the most effective analytical method used in the environment monitoring. New direct dyes having the function of colorimetric chemosensors were synthesized. When metal ions such as $Al^{3+}$, $Ca^{2+}$, $Cd^{2+}$, $Cr^{3+}$, $Cu^{2+}$, $Fe^{2+}$, $Fe^{3+}$, $Hg^{2+}$, $Li^+$, $Mg^{2+}$, $Na^+$, $Ni^{2+}$, $Pb^{2+}$ and $Zn^{2+}$ were added each solution of new direct dyes, the color of solution was changed and can be easily detected with naked eyes without expensive experimental equipment such as atomic absorption spectrometer (AAS) or inductively coupled plasma?mass spectrometer (ICP-MS). The new benzidine analogues were diazotized and reacted with couplers such as H-acid, J-acid, Chromotropic acid, Nevill-winther acid and gamma acid to synthesize new direct dyes. The structures of the new direct dyes were confirmed by high resolution mass spectrometer (FAB ionization) and evaluated with UV-Vis spectroscopy. The UV-VIS spectroscopy was measured for the dye solutions by adding various concentrations of metal ions. It was observed that the absorbance in UV-Vis spectra was changed as the heavy metal ions were added.

New 7-Hydroxycoumarin-Based Fluorescent Chemosensors for Zn(II) and Cd(II)

  • Five new 4- or 8-substituted-7-hydroxycoumarin derivatives (1-5) were synthesized as fluorescent sensors for metal ions. Fluorescent changes and selectivity for metal ions were compared based on the introduction of different ligands and/or testing with different substitution positions of 7-hydroxycoumarin in aqueous solution. Especially, probes 2, 3 and 5 displayed large fluorescence enhancements with $Zn^{2+}$ and $Cd^{2+}$. Probes 2 and 3 showed moderate selectivity for $Zn^{2+}$ over $Cd^{2+}$. On the other hand, probe 4 showed large fluorescence quenching effects upon the addition of $Ag^+$ and $Hg^{2+}$.

  • Considering the number of chemosensors that have been developed for the sensing of metal ions, only a few chemosensors for fluoride anion have been described in the literature that are based on fluorescent or chromogenic responses. We performed colorimetric anion sensing based on the binding of anion analytes with hydrogen donor group in polymer backbone resulting in naked-eye color change and fluorescent quenching. Our challenges using hydrogen donor moiety was designed effectively are continuing in order for high selectivity and sensitivity for ultimate applications such as fluid solution sensing in biomolecules and gas vapor sensing.
