Microprocessor Based Laser Induced Fluorometry I. Development of System and Application to Liquid Chromatography

  • An analytical applicability of the fluorescence detection with an optical multichannel analyzer to organic dyes was studied in this work. Continuous acquisition of spectra was possible with the use of a microcomputer. The maximum acquisition rate of a spetrum with 70-point average was about 3 seconds. Floppy discs were used to store data for later use in processing. Laser induced fluorescence detector in High Performance Liquid Chromatography was chosen for an application. Fluorescein below $10^{-15}g$ was detected satisfactorily using this system. With the help of microcomputer, three dimensional chromatograms of time-wavelength-intensity were obtained.

  • 이송식 직류 열플라즈마를 이용하여 ZrVFe 합금모재로부터 복합 나노 입자를 제조하여 플라즈마 가스 유량이 제조된 입자의 특성에 미치는 영향을 분석하였다. 입자의 특성은 전계방출 주사전자 현미경(FE-SEM), 입도 분석기(PSA), X선 분광기(EDS), X선 회절계(XRD), Brunauer-Emmett-Teller (BET) 비표면적 측정기를 사용하여 분석하였다. 플라즈마 가스 유량을 20 L/min에서 40 L/min으로 증가시키면 평균입자크기가 91 nm에서 55 nm로 감소하며 입도분포의 기하학적 편차가 줄어들었고 비표면적은 $200m^2/g$에서 $255m^2/g$으로 증가하였으며 제조된 입자의 조성에는 큰 영향을 미치지 못했지만 결정성이 향상되었다.

  • Silicone Rubber (SR)-based potassium- and sodium-selective solid-state electrodes have been developed for the portable blood electrolyte analyzer system. The electrochemical performance of these electrodes have been evaluated with a static experimental setup and with the newly developed blood electrolyte analyzer system (model; HS603). It has been shown that their potentiometric properties are essentially comparable to those of PVC-based ion-selective electrodes, but with greatly improved lifetime (200 and 40 days for potassium and sodium sensors, respectively) and potential stability (within $\pm0.1$ mV). Clinical tests have been performed with real serum samples and the results have been compared with those obtained from Ciba-Corning BGA 288 system; correlations were excellent, proving its practical utility as a new commercial system.

분쇄방법에 따른 구리프탈로시아닌 입자크기 및 결정구조 변화 (Effect of Grinding Methods on Particle Size and Crystalline Structure of Copper Phthalocyanine)

  • Wyler법으로 제조된 구리프탈로시아닌(Cupc)을 kneader, attritor, SC-mill 및 acid pasting법으로 미분쇄하여 입자 크기, 모양, 결정구조를 비교하였다. Cupc 표준물질은 산업적으로 우수한 acid pasting, kneader법으로 제조하여 사용하였고, attritor, SC-mill로 분쇄한 후, 입도분석기, 전자현미경(SEM), X-선 회절스펙트럼(XRD)으로 입자특성을 분석하여 상호 비교하였다. SC-mill의 경우, 분쇄시간에 따라 입도분석기와 전자현미경으로 측정하였을 때, 90 min간의 분쇄시간 까지는 입자크기가 감소하였으나 그 이후부터는 Cupc의 강한 응집으로 평균입자크기가 증가하였다. Attritor로부터 건식 분쇄한 Cupc는 강한 응집에 의해 입도분석기로 입자크기를 측정할 수 없었지만, XRD peak intensity로부터 최적분쇄시간을 간접적으로 예측할 수 있었다.

자동화학 분석기 Toshiba 120 FR의 평가 (Evaluation of Autochemical Analyzer Toshiba 120 FR)

  • The lower limit of detection, precision, setting method of target value, reportable range determination, recovery, linearity, and comparison study with another equipment was evaluated for the Toshiba-120FR chemistry autoanalyzer which was newly introduced at the Daejeon Veteran Hospital in Dec. 2003. Nineteen kinds of test for AST, ALT, ALP, LDH, GGT, TP, ALB, GLU, T-cho, T-bil, TG, UA, CAL, IP, AMY, HDL-C, LDL-C, Cre and BUN were performed to evaluate the lower limit of detection, precision, setting method of target value, reportable range determination, recovery, linearity, and comparison study with other equipment according to the NCCLS guidelines(EP5-A, EP6-P, EP9-A). The Toshiba-120FR autochemical analyzer showed good precision for all tested items. The data concerning the lower limit of detection, precision(total CV 0.47%~3.65%), setting method of target value, reportable range determination, recovery(93%~111%), linearity($R^2=0.997{\sim}0.999$), and comparison study(r=0.977~0.999) with other equipment was acceptable for all tested items. The results of evaluation for the Toshiba-120FR autochemical analyzer showed that this equipment could be used as an alternative to other equipment.

광경화형 Dipentaerythritol 변성 폴리메타아크릴레이트의 합성과 물성 (Synthesis and Properties of Photocurable Dipentaerythritol Modified Polymethacrylates)

  • Dipentaerythritol 유도체(DPET)와 acrylic acid를 반응하여 6관능 광경화형 변성 폴리아크릴레이트(PA-1)를 제조하였으며, dipentaerythritol 유도체(DPET)에 trimellitic anhydride와 glycidyl methacrylate를 반응시켜 12관능 광경화형 변성 폴리메타아크릴레이트(PA-2)를 제조하고 경화된 물성을 조사하였다. 관능기 수가 증가할수록 중량 평균 분자량은 증가하였으며, TGA (Thermogravimetric Analyzer)로 측정한 UV 경화필름의 열안정성은 (메타)아크릴레이트의 관능기 수가 많을수록 증가하였다. 또한 (메타)아크릴레이트의 관능기 수가 많을수록 경도, 내마모성, 인장강도가 우수하였으며, 내후성 테스트에서는 (메타)아크릴레이트의 관능기 수가 많을수록 황변 지수 값인 yellow index 값이 높아지는 결과를 보여주었다.

  • 두 종류의 4관능 폴리에스터 폴리올에 대해 trimellitic anhydride와 glycidyl metacrylate를 반응하여 두 종류의 8관능 광경화형 폴리에스터 폴리올 변성 하이퍼브랜치 메타아크릴레이트를 합성하고 광경화된 필름의 물성을 조사하였다. Thermogravimetric analyzer (TGA)로 측정한 UV 경화필름의 열안정성은 HBMA-2 (hyperbranched methacrylate-2)가 HBMA-1 (hyperbranched methacrylate-1)보다 우수하였다. 또한 HBMA-2가 HBMA-1보다 경도, 내마모성, 인장강도가 우수하였으며, 내후성 테스트에서도 HBMA-2의 yellow index 값이 높은 결과를 보여주었다.

Styrenic Polymer/Organoclay Nanocomposite Prepared via in-situ Polymerization with an Azoinitiator Linked to an Epoxy Oligomer

  • An azoinitiator linked to an epoxy oligomer, which could easily diffuse into the organoclay gallery and swell it, was used as an initiator to enhance the delamination of an organoclay, Cloisite 25A, in a matrix of styrenic polymers, poly(styrene-co-acrylonitrile) and polystyrene, during the preparation of a nanocomposite via an in-situ polymerization method. X-ray diffraction results and transmission electron microscopic observation of the morphology showed that the epoxy segment enhanced not only the delamination but also the extrication of ammonium cations from the organoclay gallery into the polymer matrix. The latter phenomenon induced the structural change of the alkyl group of ammonium cations in the gallery from a bilayer to monolayer structure, and also decreased the glass-rubber transition temperature as measured by a differential scanning calorimeter and dynamic mechanical analyzer.

  • $BaTiO_3/SrTiO_3$ and $PbTiO_3/PbZrO_3$ superlattice thin films were fabricated on $Pt/Ti/SiO_2/Si$ substrate by the pulsed laser deposition process. The morphologies and physical properties of deposited films were characterized by using X-ray diffractometer, HR-SEM, and Impedance Analyzer. XRD data and SEM images of the films indicate that each layer was well deposited alternatively in the superlattice structure. The dielectric constant of $BaTiO_3/SrTiO_3$ superlattice thin film was higher than that of individual $BaTiO_3$ or $SrTiO_3$ film. Same result was obtained in the $PbTiO_3/PbZrO_3$system. The dielectric constant of a superlattice film was getting higher as the number of layer is increased.

Preparation of cross-linked silk fibroin film by γ-irradiation and their application as supports for human cell culture

  • This study described about preparation of the cross-linked silk fibroin (SF) film by ${\gamma}$-irradiation of the casted SF film, which is fabricated from aqueous solution regenerated via fibers of cocoons and their application as supports for human cell culture. The properties of cross-linked SF film were evaluated by FT-IR spectroscopy, contact angle, solubility to water, thermal analysis, surface area analyzer, and morphology via scanning electron microscopy (SEM). The cross-linked SF films were not dissolved in water and exhibited the rough surface morphology, large surface area, and good thermal properties. The human fibroblast cell (CCD-986sk) and embryo kidney-ft cell were well growed on the surface of cross-linked SF film supports prepared by ${\gamma}$-irradiation. The cross-linked SF film prepared by ${\gamma}$-irradiation can be used as biomaterials for human cell culture.
