Selection of Target Materials for GLP Genotoxic Tests by Searching the Mutagenicity Information of Chemicals by Occupational Safety and Health Act (산안법 관리대상물질의 변이원성 검색을 통한 GLP 유전독성 시험대상 후보물질의 선정)

  • Objectives: There is a requirement to select target materials for mutagenicity(Genotoxicity) testing, so we determined to set the test priorities of them by searching the related database. Methods and Results: We searched a number of databases to find information on mutagenicity tests with chemicals under the Occupational Safety and Health Act(OSH Act), such as KOSHANET, National Toxicology Program(NTP), European Chemicals Agency(ECHA), US National Library of Medicine(NLM), and Genetic Toxicology Data Bank(GENE-TOX), as well as ChemIDplus webpage, and presented the information. Also we anticipated their hazards with ACToR sites to confirm the 58 mutagenicity(Genotoxicity) tests we will perform. Conclusions: We presented target materials for mutagenicity testing with specific GLP tests consisting of reverse mutation(Ames), chromosomal aberration and micronucleus test.

Occurence of Chemical Resistance and Control of Dollar Spot Caused by Sclerotinia homoeocarpa in Turfgrass of Golf Course (Sclerotinia homoeocarpa에 의한 잔디동전마름병(Dollar spot)의 약제 저항성균 발생 및 방제)

  • Antifungal activity of 20 chemicals registered to turfgrass diseases was evaluated. Among the chemicals, iprodione, benomyl, iprodione+thiram, pencycuron+tebuconazole, hexaconazole, and iprodione+thiophanate-methyl exhibited high antifungal activity to the dollar spot fungus. All isolates were greatly inhibited by the chemicals at the concentration over 32ppm($\mu\textrm{g}$/ml). However, sensitivity of the isolates to chemicals was varied at the lower concentration as 8 and 16 ppm as follows. The isolate originated from Ora golf course was resistant to iprodione and iprodione+thiophanate methyl, Gonjiam isolate to iprodion, Youngpyung isolate to iprodione+thiram, and Dogo isolate to iprodione+thiram, pencycuron+tebuconazole, and tebuconazole. It was found from this study that the varied chemical resistance among the isolates was positively related to the application time of the chemical in the golf course. Consequently, when a chemical was applied more often times than the others to the golf course, the fungal isolates originate from the field showed higher resistance to the former chemical. Effects of the chemicals on control of the dollar spot was evaluated in the field. All tested chemicals revealed over 70% disease control efficacy, however, mepronil+propiconazole was the best showing 83% control efficacy and followed by fenarimo, iprodione, terbuconazole, thiram, and thiophanate-methyl.

Occupational Exposure to Chemicals for Workers and Levels of Airborne Chemicals in Hairdressing Salon (미용실 작업자의 화학물질에 대한 직업적인 노출과 실내공기오염 수준)

  • Hairdressers may be exposed to many chemicals from hair dye, permanent, shampoo, hair spray and so on. The study was conducted to evaluate occupational exposure to 8 chemicals (isopropanol, ethanol, xylene, toluene, ethylbenzene, acetone, ammonia and formaldehyde) for workers and levels of these materials in airborne of a hairdressing salon. Since compared to ACGIH TLVs these 8 chemicals were very low, it is thought that there has no problem to occupational exposure to these chemicals. Even if hairdressing salon is not enforced on Korean Standards for Indoor Air Quality, level of TOVC was thought to be below the standards. At the aspect of indoor air quality formaldehyde needs to be controlled in the future. The results imply that emission of isopropanol, ethanol, acetone and ammonia are related with hairdressing job, but emission of xylene, toluene and ehtylbenzene is more related with traditional indoor air pollution than hairdressing job. Sources of formaldehyde is thought to be little related with hairdressing job. HVAC system was a little effective on general ventilation.

Prediction of Environmental Fate of Certain Chemicals Using Computer Simulation Programs (Computer Program을 이용한 화학물질의 환경동태 예측)

  • Environmental hazards of a chemical could be assessed by two different approaches : toxicity test and assessment of exposure potentials to human and environmental organisms. For the prediction of environmental fate of chemicals three available computer programs were compared each other and were verified. The results obtained by using these computer programs, PCHEM, EXAMS, and E4CHEM were summarized as follows. The estimated octanol/water partition coefficients by PCHEM were similar to the experimental values in the literature. But the other factors, water solubility and vapor pressure were different from the data in the literature. The simulation results of selected compounds by EXAMS showed similar tendency to the literature results of model field environment. Therefore, this computer program could be utilized to predict the environmental fate of chemicals. E4CHEM program is very simple and this program could predict the ultimate environmental fate of stable chemicals by input of two or three parameters. However, the validity should further be verified in the future field study using more compounds. It is suggested that these approaches could be fully utilized by understanding their limitations to predict the environmental fate of new chemicals under development, to screen the potential environmental pollutants among chemicals already-in use, and to devise measures to minimize the hazards to the environment.

  • Chemical mutagenicity is a major hazard that is important to workers' health. Despite the use of large amounts of allyl chloride, the available mutagenicity data for this chemical remains controversial. To clarify the mutagenicity of allyl chloride and because a micronucleus (MN) test had not yet been conducted, we screened for MN induction by using male ICR mice bone marrow cells. The test results indicated that this chemical is not mutagenic under the test conditions. In this paper, the regulatory test battery and several assay combinations used to determine the genotoxic potential of chemicals in the workplace have been described. Further application of these assays may prove useful in future development strategies of hazard evaluations of industrial chemicals. This study also should help to improve the testing of this chemical by commonly used mutagenicity testing methods and investigations on the underlying mechanisms and could be applicable for workers' health.

  • The anti-platelet activity of ilexoside D isolated from the roots of Ilex pubescens Hook. et Arn. was investigated in in vitro and ex vivo models of platelet aggregation induced by ADP, thrombin or collagen in rats. In vitro ilexoside D inhibited more effectively platelet aggregation induced by ADP and thrombin than by collagen as compared with aspirin. Ex vivo ilexoside D also inhibited platelet aggregation induced by ADP and collagen, but not by thrombin, and the inhibitory action of ilexoside D was more effective than that of aspirin. However, in vitro ilexoside D inhibited very poorly the generation of malonyldialdehyde, which is known to be concomitantly released with thromboxane $A_2$ during platelet aggregation. These results suggest that the anti-platelet activity of ilexoside D may not be responsible for prostaglandin synthesis in platelets.

  • In this study, we try to determine the distributive property in southwest coastal waters, such as K wang-yang bay, Ka-mak bay, Yeo-ja bay, Wan-do, Hea-nam, Young-gwang, and Mok-po, using simultaneous analytical method of 310 chemicals. The results were detected tens of the organic pollutants in sampling sites, and the major chemicals detected were CH type chemicals such as aliphatic, polycyclic compounds and were CHN(O) type chemicals such as aromatic amines, nitro compounds, In particular, pesticides were mainly detected in summer, phenols and phthalate esters were not seasonal effect. Also, a number of aromatic chemicals were detected in Kwang-yang bay, From the results of this study, we confirmed that it is mainly contaminated in summer and the pesticide chemicals are the major pollutants in southwest coastal waters.

Development of Handling Guidelines for the Safety and Health of Transporters of Hazardous Chemicals - Focusing on Safety Containers and Packaging for Delivery of Hazardous Chemicals Used for Reagents - (유해화학물질 운반자의 취급안전보건 관리를 고려한 취급기준 마련 - 유해화학물질 시약 등 택배 안전용기 및 포장기준을 중심으로 -)

  • Objectives: In this study, we analyzed the current state of delivery containers and packages and established handling guidelines to safely transport delivery containers and packages for use in research, testing, and examination reagents. Methods: Handling guidelines were revised in such categories as maintenance of the handling facilities, storage, loading and unloading, containers and packages, transportation, etc. In addition, we analyzed the current state of domestic sales for hazardous chemicals used for research, testing, and examination reagents, and investigated the handling guidelines related to delivery transportation in the USA, EU, and Japan by chemical property. Results: There are 6,160 companies selling hazardous chemicals. Among them, the 476 companies selling reagents for use in research, testing, and examination were investigated. Total amounts handled reached 425,000 tons, contributing to 0.2% of the total. For delivery transportation, internal containers and packaging was specified for chemical properties as follows: within 1 L for flammable gas, within 5 L for flammable liquid, and within 18 L for others. In addition, the maximum size of the outer package was set within 130 cm for total length, width, and height, and no dimension of the packaging could exceed 60 cm. Sixty-four hazardous chemicals with explosiveness or acute inhalation toxicity were prohibited for delivery transportation. Conclusion: Specified handling guidelines for inner and outer containers as well as packaging were regulated for delivery transportation of hazardous chemicals used for reagents. In addition, 64 hazardous chemicals were prohibited for delivery transportation. These are designed to prevent transportation accidents involving hazardous chemicals for reagents and thus protect the safety and health of transporters who handle hazardous chemicals.

Study on Inhalation Toxicity of Halogen Compound Using Rats (랫드를 이용한 할로겐 화합물의 흡입독성 연구)

  • Inhaled halogen compound was examined through micronucleus tests and toxicity tests using SD (Sprague-Dawley) rats with repeated dosages for six hours a day, five days a week, during four weeks. In four-week repeated exposure, no specific sign caused by the $CF_3I$ compound was observed on the clinical symptoms, body weight variation, feed consumption, and urinalysis data in the testing groups with reference to the control group. In hematological and biochemical blood tests of the testing groups, the significant, but in their normal ranges, value dependencies of glucose, aspartate aminotransferase, alanine aminotransferase and etc., on the halogen carbohydride concentration was observed. In histo-pathological tests, no specific lesion or concentration dependent change due to the $CF_3I$ compound dosage was observed in both sexes of the female and male in the tested animals. But, micronucleus tests on marrow cells extracted from the tested animals which were repeatedly exposed in the $CF_3I$ compound during four weeks, the frequencies of micronuclei were significantly increased dose-dependently compared to the control groups.

  • To develope of the inexpensive anti-stain chemicals, it was conducted to investigate the inhibitory effectiveness of mycelial growth and anti-mold effectiveness against test fungi causing surface mold of wood, iron corrosivity and hygroscopicity of six fluoride-based chemicals made from industrial by- product.Among the six chemicals. RNF-3 and RNF-4 were the most effective with respect to a mycelial growth control and anti-mold effectiveness. For the wood treatment with these chemicals, the optimum concentration was about 2% and there was no difference in the effectiveness of chemicals between wood species. The wood treated with synthesized chemicals showed a relatively high iron corrosion rate for corrosive F component whereas, in the treatment with RNF-3 and RNF-4, there was no difference from the untreated wood when the concentration was less than 2%. The hygroscopic property of wood was not effected by treatment of these chemicals. To prove the feasibility for practical using of RNF-3 and RNF-4 chemicals, it is necessary to test of anti-mold effectiveness in the sawmill by field test.
