Glucocorticoid Receptor Induced Down Regulation of Metalloproteinase-9 (bfMP-9) by Ginseng Components, Panaxadiol (PD) and Panaxatriol (PT), Contributes to Inhibition of the Invasive Capacity of HTl080 Human Fibrosarcoma Cells

  • This study showed the anti-invasive activity of ginseng components, panaxadiol (PD) and panamatrlol (PT) on the highly metastatic HT1080 human flbrosarcoma cell line. PD and PT reduced tumor cell invasion through a reconstitute basement membrane in the transwell chamber. A significant down regulation of MMP-9 by PD and PT was detected by northern blot analysis. However, MMP-2 was constantly expressed. Quantitative gelatin based zymography confirmed a marked reduced expression of MMP-9 but not MMP-2 in the treatment of PD and PT. Since the chemical structures of PD and PT are very similar to that of dexamethasone, a synthetic glucocorticoid, it was investigated whether PD and PT act through GR. Western blot analysis and immunocytochemistry showed that PD and PT increased the GR fraction in the nucleus. These results suggest that ursolic acid may induce repression of MMP-9 by stimulating the nuclear translocation of GR and hence inhibiting the activity of AP-1 to TPA-responsible element of MMP-9 promoter region. In conclusion, we suggest that CR-induced down-regulation of MMP-9 by PD and PT contributes to reduce the invasive capacity of HT 1080 cells.

Evaluation for Contents of Contaminants and Leaching Characteristics of Bottom Ash (바텀애쉬의 유해물질 함량 측정 및 용출특성 평가연구)

  • In this study, we tried to determine any detrimental effects on water quality when bottom ash obtained from a coal-fired power plant intended to be used as a fill material in construction sites. Physical-chemical properties of bottom ash were determined using proximate analysis, elemental analysis, XRD, and XRF. Classification of bottom ash as a waste material and soil contamination due to the use of bottom ash were performed by Korea waste standard leaching test and soil toxicity test, respectively. Results of leaching tests were compared to the regulations for water quality and groundwater quality and no harmful effects on water quality were found. Most of heavy metals in leachate were below detection limits but trace amount of $Cr^{6+}$ was found. However, concentration of $Cr^{6+}$ was below the regulation criteria. Column leaching tests indicated that concentrations of Pb and Zn were slightly higher than regulations but below regulations within 1 PVE, but concentrations of sulfate were 10 times higher than regulation and thus, the required time to reach regulation was almost 8 PVE.

Study on scheme for screening, quantification and interpretation of trace amounts of hazardous inorganic substances influencing hazard classification of a substance in REACH registration (REACH 물질 등록 시 분류에 영향을 주는 미량 유해 무기물질의 스크리닝·정량·해석을 위한 체계도 연구)

  • Substance identification is the first step of the REACH registration. It is essential in terms of Classification, Labelling and Packaging (CLP) regulation and because even trace amounts of impurities or additives can affect the classification. In this study, a scheme for the screening, quantification, and interpretation of trace amounts of hazardous inorganic substances is proposed to detect the presence of more than 0.1% hazardous inorganic substances that have been affecting the hazard classification. An exemplary list of hazardous inorganic substances was created from the substances of very high concern (SVHCs) in REACH. Among 201 SVHCs, there were 67 inorganic SVHCs containing at least one or ~2-3 heavy metals, such as As, Cd, Co, Cr, Pb, Sb, and Sn, in their molecular formula. The inorganic SVHCs are listed in excel format with a search function for these heavy metals so that the hazardous inorganic substances, including each heavy metal and the calculated ratio of its atomic weight to molecular weight of the hazardous inorganic substance containing it, can be searched. The case study was conducted to confirm the validity of the established scheme with zinc oxide (ZnO). In a substance that is made of ZnO, Pb was screened by XRF analysis and measured to be 0.04% (w/w) by ICP-OES analysis. After referring to the list, the presence of Pb was interpreted just as an impurity, but not as an impurity relevant for the classification. Future studies are needed to expand on this exemplary list of hazardous inorganic substances using proper regulatory data sources.

Control of dissolved Oxygen Concentration and Specific Growth Rate in Fed-batch Fermentation (유가식 생물반응기에서의 용존산소농도 및 비성장속도의 제어)

  • A novel control method with automatic tuning of PID controller parameters has been developed for efficient regulation of dissolved oxygen concentration in fed-batch fermentations of Escherichia coli. Agitation speed and oxygen partial pressure in the inlet gas stream were chosen to be the manipulated variables. A heuristic reasoning allowed improved tuning decisions from the supervision of control performance indices and it coule obviate the needs for process assumptions or disturbance patterns. The control input consisted of feedback and feedforword parts. The feedback part was determined by PID control and the feedforward part is determined from the feed rate. The proportional gain was updated on-line by a set of heuristics rules based on the supervision of three performance indices. These indices were output error covariance, the average value of output error, and input covariance, which were calculated on-line using a moving window. The integral and derivative time constants were determined from the period of output response. The specific growth rate was maintained at a low level to avoid acetic acid accumulation and thus to achieve a high cell density. The specific growthe rate was estimated from the carbon dioxide evolution rate. In fed-batch fermentation, the simutaneous control of dissolved oxygen concentration (at 0.2; fraction of saturated value) and specific growth rate (at 0.25$hr^{-1}$) was satisfactory for the entire culture period in spite of the changes in the feed rate and the switching of control input.

Effect of Ethanol Extract of Herbal Medicine on Crop Growth Inhibitor (한약재 주정 추출물이 종자발아와 유묘생장에 미치는 영향)

  • BACKGROUND: Herbal extracts have been screened for their inhibitory effect of seed germination and root development on weeds, but there is a scarcity of reports for crop growth regulation. The objective of this research was to develop a growth inhibitor on Brassica campestris, and its effective extraction method from herbal medicine extract. METHODS AND RESULTS: Eighty four herbal medicine extracts were tested for their plant growth inhibition activity on B. campestris. The alcohol extracts of Artemisia annua, Cinnamomum cassia, and Mentha arvensis inhibited over 30% of germination and the extract of A. annua, and C. cassia inhibited over 70% of radicle growth at 0.1 % w/w treatment. The partially purified extracts of A. annua, and C. cassia with dichloromethane and hexane showed stronger radicle growth inhibition than the crude extracts on B. campestris. The diethyl ether extract of A. annua showed a similar 50% radicle growth inhibition ($RI_{50}$ = 45 mg/L) to its partially purified extract with dichloromethane or hexane, but the diethyl ether extract of C. cassia showed a worse $RI_{50}$ than the purified extract. CONCLUSION: The alcohol extracts of A. annua, and C. cassia showed potent radicle growth inhibition properties on B. campestris. Diethyl ether proved to be a good solvent for simple extraction from A. annua.

  • A flue gas emission known to cause an acid rain, containing sulfur dioxide, is tightly controlled by a regulation. The membrane contactor could be one of the alternatives which might be useful technologies for better performance of desulfurization. The fundamental sulfur dioxide separation data were obtained by a flat membrane contactor so that they could be utilized for the full scale membrane contactor. The separation of sulfur dioxide was measured in terms of the concentration of NaOH absorbent, the concentration of sulfur dioxide, the feed flow rate and different membrane materials. As the concentration of absorbent increased, the removal efficiency increased. By increasing the concentration of sulfur dioxide, the removal efficiency decreased. As the feed flow rate increased, it was observed that the removal efficiency decreased. Finally, the effect of the membrane material on separation was found to be negligible.

  • Lactulose is well known for functional component in the food and pharmaceutical field and utilized in a wide variety of foods as a bifidus factor or functional ingredient for intestinal regulation. Lactulose synthesis can be classified into chemical and biological methods. In chemical methods, lactulose is synthesized by alkaline isomerization, but it has many disadvantages such as including product purification, lactulose degradation, side reactions and waste management. Therefore, the enzymatic synthesis methods were recently studied to solve these problems. ${\beta}$-galactosidase is a important enzyme in the dairy industry, because of the production of lactose-hydrolyzed products. Also, ${\beta}$-galactosidases can be utilized to synthesize lactulose from lactose by a trans-galactosylation reaction, using fructose as a galactosyl acceptor. However, the synthesis of lactulose from lactose is economically not suitable due to high levels of lactose price. This review summarizes the current state of lactulose production by chemical and biological processes.

Ring Opening and Polymerization of Alpha-Lipoic Acid (알파 리포산의 개환 및 중합)

  • Alpha-lipoic acid (ALA) synthesized in the body has virtues such as anti-oxidation, blood sugar regulation, appetite suppression, and anti-obesity, etc. ALA, which is also used as a drug, has a five-membered ring including disulfide and so easily losses bioavailability due to ring opening and subsequent polymerization by heat or ultraviolet. This report studies various conditions for ring opening polymerization. The ring opening starts above the melting point of ALA, but there was no temperature dependence above it. At $70^{\circ}C$, the degree of ring opening was proportional to reaction time and inversely proportional concentration. The degree of ring opening in acetic acid with UV for 1 hour reached the maximum conversion (70%). Most cleaved ALA changed into disulfide polymers, and the molecular weight of the polymers increased as the amount of ring opening increased.

  • Environmental issues on the waste emission and its treatment are of great interest in these days. In order to resolve the pollution problems, recycling the waste materials is generally recommended. Especially, emission of waste acids in designated sources is increasing every year. In this study, we focused on the effective recycling of the waste acids rather than treatments. Management systems of the waste acids are not systematically designed, and the quality of the waste regulation on the recycling product is unclear in domestic and foreign countries. We surveyed the present conditions on domestic waste emission and recycling of waste acids. For the final analysis of the recycling products, iron chloride, iron sulfate, copper cyanide, copper oxides, and cement copper are selected as candidates. We expect that this article would help establishing the systematic management system on treating the waste acid materials.

Effect of Dissolved Oxygen on the Production of Epothilone in Bioreactor Cultures Sorangium cellulosum (Sorangium cellulosum의 생물반응기 배양에서 용존산소가 epothilone의 생산에 미치는 영향)

  • The biological production of a potent anticancer agent, epothilone, by Sorangium cellulosum was carried out using flask and fermentor cultures. Soluble starch was selected as the main carbon source and the concentrations of lactose and yeast extract were optimized at 4 and 0 g/L, respectively, when using the flask cultures. In the fermentor cultures, the cells were cultivated at a high DO level of more than 80% of air saturation in the growth stage and then the DO level was controlled at about 50, 20 or 1-2% when the carbon source was exhausted. The epothilone production increased with decreasing DO level after the exhaustion of the carbon source, and the maximum concentration of epothilone was 5.4 mg/L. It was found that the DO level had significant regulation effects on the epothilone production.
