Micro-Spot Atmospheric Pressure Plasma Production for the Biomedical Applications

  • We are currently conducting studies on culturing and biocompatibility assessment of various cells such as neural stem cells and induced pluripotent stem cells(IPS cells) on carbon nanotube (CNT), on nerve regeneration electrodes, and on silicon wafers with a focus on developing nerve integrated CNT based bio devices for interfacing with living organisms, in order to develop brain-machine interfaces (BMI). In addition, we are carried out the chemical modification of carbon nanotube (mainly SWCNTs)-based bio-nanosensors by the plasma ion irradiation (plasma activation) method, and provide a characteristic evaluation of a bio-nanosensor using bovine serum albumin (BSA)/anti-BSA binding and oligonucleotide hybridization. On the other hand, the researches in the case of "novel plasma" have been widely conducted in the fields of chemistry, solid physics, and nanomaterial science. From the above-mentioned background, we are conducting basic experiments on direct irradiation of body tissues and cells using a micro-spot atmospheric pressure plasma source. The device is a coaxial structure having a tungsten wire installed inside a glass capillary, and a grounded ring electrode wrapped on the outside. The conditions of plasma generation are as follows: applied voltage: 5-9 kV, frequency: 1-3 kHz, helium (He) gas flow: 1-1.5 L/min, and plasma irradiation time: 1-300 sec. The experiment was conducted by preparing a culture medium containing mouse fibroblasts (NIH3T3) on a culture dish. A culture dish irradiated with plasma was introduced into a $CO_2$-incubator. The small animals used in the experiment involving plasma irradiation into living tissue were rat, rabbit, and pick and are deeply anesthetized with the gas anesthesia. According to the dependency of cell numbers against the plasma irradiation time, when only He gas was flowed, the growth of cells was inhibited as the floatation of cells caused by gas agitation inside the culture was promoted. On the other hand, there was no floatation of cells and healthy growth was observed when plasma was irradiated. Furthermore, in an experiment testing the effects of plasma irradiation on rats that were artificially given burn wounds, no evidence of electric shock injuries was found in the irradiated areas. In fact, the observed evidence of healing and improvements of the burn wounds suggested the presence of healing effects due to the growth factors in the tissues. Therefore, it appears that the interaction due to ion/radicalcollisions causes a substantial effect on the proliferation of growth factors such as epidermal growth factor (EGF), nerve growth factor (NGF), and transforming growth factor (TGF) that are present in the cells.

Influences of Water Level and Vegetation Presence on Spatial Distribution of DOC and Nitrate in Wetland Sediments (수심의 정도와 식생의 유무에 따른 인공습지 토양 내 유기탄소와 질산염의 공간적 분포)

  • Wetlands are a well known ecosystem which have high spatial-temporal heterogeneity of chemical characteristics. This high heterogeneity induces diverse biogeochemical processes, such as aerobic decomposition, denitrification, and plant productivity in wetlands. Understanding the dynamics of dissolved organic carbon (DOC) and inorganic nitrogen in wetlands is important because DOC and inorganic nitrogen are main factors controlling biological processes in wetlands. In this study, we assessed spatial distribution of DOC and inorganic nitrogen with relation to the different hydrology and vegetation in created wetlands. Both DOC and nitrate contents were significantly higher in vegetated areas than open areas. Different water levels also affected DOC contents and their quality. Average DOC contents were $0.37mg{\cdot}g^{-1}$ in deep riparian (DR) and $0.31mg{\cdot}g^{-1}$ in shallow riparian (SR). These results appeared to be related to the interaction between carbon supply by vegetation and microbial decomposition. On the other hand, inorganic nitrogen contents were not affected by water level differences. This result indicates that presence/absence of vegetation could be a more important factor than hydrology in the spatial dynamics of inorganic nitrogen. In conclusion, we observed that vegetation and hydrology differences induced spatial distribution of carbon and nitrogen which are directly related to biogeochemical processes in wetlands.

Preparation of Polyurushiol (PUOH) Using Urushiol and Property of LDPE / PUOH Composite Films (우루시올을 활용한 폴리우루시올(PUOH)제조 및 LDPE/PUOH 복합필름 특성에 관한 연구)

  • Urushiol extracted from lacquer tree exhibits good thermal stabilities as well as antimicrobial andantioxidant properties. However, it has been known that the urushiol derivates bring out allergy. In this study, polyurushiol (PUOH) powders were successfully synthesized for the safe and convenient handling of allergic urushiol. First, the as-synthesized PUOH was confirmed by Fourier transform infrared spectroscopy (FTIR), scanning electron microscope (SEM), thermal gravimetric analyzer (TGA), antioxidant test and antimicrobial test. And then, six different LDPE/PUOH composite films were prepared via a twin screw extruder system and investigated their feasibility to use as active packaging materials. Their chemical structures, morphology, thermal optical and antimicrobial properties of the LDPE/PUOH composite films were investigated as a function of PUOH contents. FTIR and SEM results showed that LDPE/PUOH composite films have a weak interfacial interaction and poor dispersion with a high PUOH loading. The thermal properties increased up to 3 wt% as the content of PUOH increases. Compared to the pure LDPE films, LDPE/PUOH composite films are more effective in the UV absorbance and antibacterial activity against E. coli. To maximize the performance of LDPE/PUOH compositefilms as the packaging materials, further researches are required to enhance the dispersion of PUOH powders in the LDPE matrix.

Risk and Responsibility in Korean Tobacco Litigation: Epidemiology and Causality in Late Modern Risk (한국 담배소송에서의 위험과 책임: 역학과 후기 근대적 인과)

  • Toxic tort cases have increased dramatically since the 1970s, as large technological systems, such as nuclear power plants and chemical factories, or mass-produced, high-tech products, had exposed citizens and consumers to dangerous substances. It was, however, difficult to establish causal connection between exposure and the alleged harms in many of the environmental, pollution, and product liability cases under the framework of tort law conception of causation and responsibility. Science and law was called upon to resolve such 'late modern' legal cases where true causes are hard to find, where no single explanatory factor is sufficient for explaining diseases like cancer. This article examines how plaintiffs in the Korean tobacco litigation mobilized such late modern tools in science and law, such as epidemiology and the allocation of the burden of proof, in the context of the global circulation of science and law. It further shows how a set of the scientific theories and legal arguments developed in order to cope with late modern risk played a central role in establishing a causation between smoking and cancer in 2011. This article suggests that STS scholars can fruitfully examine the interaction between science and law as a way to understand and engage with social and legal issues engendered by late modern risk.

A comparative study of carcass characteristics and meat quality in genetic resources Pekin ducks and commercial crossbreds

  • Objective: The study was aimed to compare carcass traits, physicochemical and textural properties of meat in two different genotypes of Pekin ducks with regard to sex effect. Methods: The study involved 120 Pekin ducks: 30 males and 30 females of strain P33 (Polish native Pekin ducks) and 30 males and 30 females of Star 53 HY (commercial hybrid Pekin ducks). At 49 d of age, 48 birds (12 males and 12 females of each genotype) were selected for dissection. After the dissection, meat samples were collected to determine meat quality traits. Results: The studied Pekin ducks of different genotype showed significant differences in body weight, carcass weight, dressing percentage, as well as percentages of breast muscles, skin with subcutaneous fat, abdominal fat, neck, and remainders of eviscerated carcass with neck. Duck genotype influenced the content of crude protein, crude fat, Na, K, P, Zn, $pH_{24}$, electric conductivity ($EC_{24}$), cooking loss, $L^{\star}$, $a^{\star}$, most textural traits of breast muscle, and also Na, Mg and Fe content, $EC_{24}$, drip loss, cooking loss and $L^{\star}$, $a^{\star}$, and $b^{\star}$ colour coordinates of leg muscles. Regardless of genetic origin, males exhibited higher BW, carcass weight and carcass neck percentage, as well as lower redness, hardness, chewiness and gumminess of breast muscle compared to females. The genotype${\times}$sex interaction was significant for the crude fat content and cooking loss of breast muscle, and for the yellowness of leg muscle. Conclusion: Star 53 HY ducks are more suited for broiler production due to their higher body weight and dressing percentage. Their breast and leg meat are characterized by more beneficial chemical composition but has poorer sensory and textural properties compared to the meat of P33 ducks.

Frequency Dependency of Electrical Property Stabilization during Vulcanization of Modified NR/IR Blends (개질된 NR/IR 블랜드의 가황 반응에서 나타나는 전기적 특성 안정화의 주파수 의존성)

  • Frequency dependency or electrical property stabilization during vulcanization of modified NR/IR composite materials was studied using in-situ electrical property measuring technique. Volume resistivity(p) before and after vulcanization reaction of the sample was measured as the function or frequency in the range or 1Hz to 10kHz at reaction temperatures of 130, 140, 150, and $160^{\circ}C$, respectively. A double stabilization mode of frequency dependency was observed, in which a slow stabilization process of p to a value of ca. $1.0{\times}10^7\;{\Omega}-cm$ occurred after a drastic initial decrease from ca. $9.0{\times}10^7\;{\Omega}-cm$. In addition, notable temperature dependencies of p values were also observed before and after vulcanization reaction, that is, p values at 130 and $140^{\circ}C$ after vulcanization were observed as about 1/3 of those values before vulcanization. All the observed facts were considered as the results from the interaction between the electrode and the bulk sample materials, i.e., electronic charge-discharge, and from the structure change of samples including CB rearrangement by the vulcanization.

Three-dimensional micro photomachining of polymer using DPSSL (Diode Pumped Solid State Laser) with 355 nm wavelength (355nm 파장의 DPSSL을 이용한 폴리머의 3차원 미세 형상 광가공기술)

  • The basic mechanistic aspects of the interaction and practical considerations related to polymer ablation were briefly reviewed. Photochemical and photothermal effects, which highly depend on laser wavelength have close correlation with each other. In this study, multi-scanning laser ablation processing of polymer with a DPSS (Diode Pumped Solid State) 3rd harmonic Nd:YVO$_4$ laser (355 nm) was developed to fabricate a three-dimensional micro shape. Polymer fabrication using DPSSL has some advantages compared with the conventional polymer ablation process using KrF and ArF laser with 248 nm and 193 nm wavelength. These advantages include pumping efficiency and low maintenance cost. And this method also makes it possible to fabricate 2D patterns or 3D shapes rapidly and cheaply because CAD/CAM software and precision stages are used without complex projection mask techniques. Photomachinability of polymer is highly influenced by laser wavelength and by the polymer's own chemical structure. So the optical characteristics of polymers for a 355 nm laser source is investigated experimentally and theoretically. The photophysical and photochemical parameters such as laser fluence, focusing position, and ambient gas were considered to reduce the plume effect which re-deposits debris on the surface of substrate. These phenomena affect the surface roughness and even induce delamination around the ablation site. Thus, the process parameters were tuned to optimize for gaining precision surface shape and quality. This maskless direct photomachining technology using DPSSL could be expected to manufacture tile prototype of micro devices and molds for the laser-LIGA process.

Temperature and microbial changes of corn silage during aerobic exposure

  • Objective: This study was conducted to estimate the temperature and microbial changes of corn silages during aerobic exposure. Methods: Kwangpyeongok (KW) and Pioneer 1543 (PI) corn hybrids were harvested at 29.7% of dry matter and chopped to 3 to 5 cm lengths. Homo (Lactobacillus plantarum; LP) or hetero (Lactobacillus buchneri; LB) fermentative inoculants at $1.2{\times}10^5$ colony forming unit/g of fresh forage was applied to the chopped corn forage which was then ensiled in quadruplicate with a $2{\times}2$ (hybrid${\times}$inoculant) treatment arrangement for 100 days. After the silo was opened, silage was sub-sampled for analysis of chemical compositions, in vitro digestibility, and fermentation indices. The fresh silage was continued to determine aerobic exposure qualities by recorded temperature and microbial changes. Results: The KW silages had higher (p<0.01) in vitro digestibilities of dry matter and neutral detergent fiber than those of PI silages. Silages applied with LB had higher (p<0.001) acetate concentration, but lower (p<0.01) lactate concentration and lactate to acetate ratio than those of LP silages. The interaction effect among hybrid and inoculant was detected in acetate production (p = 0.008), aerobic stability (p = 0.006), and lactic acid bacteria count (p = 0.048). The yeast was lower (p = 0.018) in LB silages than that in LP silages. During the aerobic exposure, PI silages showed higher (p<0.05) temperature and mold than KW silages, while LP silages had higher (p<0.05) lactic acid bacteria and yeast than LB silages. Conclusion: The results indicated that the changes of silage temperature during aerobic exposure seems mainly affected by mold growth, while applied LB only enhanced aerobic stability of PI silages.

Analysis of Macrobenthic Community Structure in an Intertidal Flat in Hakseong-ri, Boryeong, Korea (보령 학성리 갯벌 조간대 대형저서동물 군집구조 분석)

  • This study was carried out to investigate temporal and spatial distribution of macrobenthic community and elucidate effects of environmental factors on change of community structure in an intertidal flat, Hakseong-ri, Boryeong, Korea. Field surveys were seasonally conducted to collect samples of sediment and macrobenthos using can core in triplicate at nine stations in 2016 and 2017. Our results showed that sediment had high mud content (above 60%) in most samples and mean content of loss on ignition was 2.3% in 2016. A total of 79 species was collected in the study site during the study period. Mean density and biomass were 611 ind./m2 and 64.1 gWWt/m2, respectively. Heteromastus filiformis was the dominant species (48.6%, 297 ind./m2) followed by Macrophthalmus japonicus (10.1%, 62 ind./m2) and Upogebia major (6.9%, 42 ind./m2). Three assembly groups resulted from cluster analysis were more distinguished by interaction between organisms and frequency of dominant species than by physical and chemical environment characteristics. In addition, macrobenthic community in the Hakseong intertidal flat showed seasonal changes based on non metric multidimensional scaling using species abundance.

Agro-ecosystem Diversity and Integrated Mite Pest Management in Fruit Orchards: A Review and Future Prospect (농업생태계 다양성과 과수원 응애류 해충 종합관리: 이론적 고찰과 미래 전망)

  • Integrated mite management provided a basic direction to early the fruit tree IPM. The early IPM concept was based on incorporation of the biological control for mite pests with the existing chemical control compatibly. Since then, the hypotheses and mechanisms of the interaction between species diversity and pest population dynamics have provided a broader understanding of mite-centered pest management in fruit tree ecosystems. Based on the principle of the ecosystem, biological control and pest management through habitat modification or manipulation are developing to the concept of agro-ecological engineering. In particular, the natural enemy diversity is dynamically changed according to the different cultivation environment in the management of mite pests, and the species composition of phytophagous mites is also changed by the environment for orchard management. This paper reviews the biological control of mites, which was the basis of apple IPM, and also re-examines the topics of species diversity and pest management, predacious mite diversity in relation to mite control and the change of species composition of mite pests in the sight of ecological engineering. Finally, we suggest a strategy for biological control of spider mites in apple orchards in Korea.
