Chemical compositions and biological activities of marine invertebrates from the East Sea of South Korea

  • Background: Marine invertebrates are well known as pivotal bioresources with bioactive substances such as anti-inflammatory sterols, antitumor terpenes, and antimicrobial peptides. However, there are few scientific reports on chemical compositions and bioactivities of marine invertebrates from the East Sea of South Korea. Methods: In this study, chemical compositions and biological activities were evaluated on both 70% EtOH and hot water extracts of 5 species of marine invertebrates (Crossaster papposus japonicus, Actinostola carlgreni, Stomphia coccinea, Actinostola sp., and Heliometra glacialis) collected from the East Sea of South Korea. The antioxidant activities were measured by ABTS radical scavenging assay. The cytotoxicity and anti-inflammatory activity were evaluated using MTT and Griess reagents. Moreover, the antibacterial effect was evaluated using paper disc assay and minimum inhibitory concentration (MIC) assay. Results: In the results of antioxidant activities, 70% EtOH extract of A. carlgreni showed the highest activity ($IC_{50}\;0.19{\pm}0.03mg/ml$) compared to other extracts. Moreover, 70% EtOH extract of A. carlgreni could significantly suppress the nitric oxide (NO) production in lipopolysaccharide-induced RAW 264.7. All extracts treated under $400{\mu}g/ml$ have no cytotoxic effects on RAW 264.7 macrophages. In the antibacterial test, both 70% EtOH extracts of C. papposus japonicus and H. glacialis showed a significant antibacterial effect on Staphylococcus aureus. The MIC values were evaluated at 256 and $512{\mu}g/ml$, respectively. Conclusions: These results suggested the bioactive potentials of marine invertebrates from the East Sea of South Korea in pharmaceutical and nutraceutical applications.

Effect of Chemical Compositions on Microstructure and Mechanical Properties of Base Metal and HAZ of Bainitic Steel Plates (베이나이트계 후판강의 모재 및 열영향부의 미세조직과 기계적 특성에 미치는 화학 조성의 영향)

  • In this study, three kinds of bainitic steel plates are manufactured by varying the chemical compositions and their microstructures are analyzed. Tensile and Charpy impact tests are performed at room and low temperature to investigate the correlation between microstructure and mechanical properties. In addition, heat affected zone (HAZ) specimens are fabricated by a simulation of welding processes, and the HAZ microstructure is analyzed. The base steel that has the lowest carbon equivalent has the highest volume fraction of acicular ferrite and the lowest volume fraction of secondary phases, so the strength is the lowest and the elongation is the highest. The Mo steel has a higher volume fraction of granular bainite and more secondary phases than the base steel, so the strength is high and the elongation is low. The CrNi steel has the highest volume fraction of the secondary phases, so the strength is the highest and elongation is the lowest. The tensile properties of the steels, namely, strength and elongation, have a linear correlation with the volume fraction of secondary phases. The Mo steel has the lowest Charpy impact energy at $-80^{\circ}C$ because of coarse granular bainite. In the Base-HAZ and Mo-HAZ specimens, the hardness increases as the volume fraction of martensite-austenite constituents increases. In the CrNi-HAZ specimen, however, hardness increases as the volume fraction of martensite and bainitic ferrite increases.

Sintering and grain growth in binary forsterite(Mg$_2$SiO$_4$)/spinel (MgAl$_2$O$_4$) system

  • The binary forsterite($Mg_2SiO_4$)/spinel ($MgAl_2O_4$) system, a possible refractory for industrial applications, is investigated for their density and grain growth the same firing conditions as the each component material between $1400^{\circ}C$ and $1700^{\circ}C$ ($1650^{\circ}C$). The forsterite grain growth exponent is established to be equal to 5 for all compositions within this binary system. Generally; the spinel addition to forsterite inhibited the forsterite grain growth. The activation energies for the forsterite grain growth of the eight compositions(weight ratio of forsterite/spinel) within the binary system are determined to be: 952$\pm$79(95/5), 363$\pm$37(90/10), 219$\pm$21(80/20), 220$\pm$44(70/30), 112$\pm$16(50/50), 112$\pm$23(30/70), 198$\pm$26(10/90), and 121$\pm$12(5/95) KJ/mol. The more forsterite is contained within the binary system, the higher value the activation energy for forsterite grain growth. It is considered that the forsterite grain growth at the higher forsterite compositions are more inhibited by spinel than that of the lower forsterite compositions.

  • Flame retardant halogen containing compounds have been replaced as environmentally safe material which does not contain hazardous materials generating toxic gas. Self-extinguishing epoxy resin compositions have been studied in order to produce eco-friendly epoxy molding compound, which is used as insulating materials in semiconductor. We developed self-extinguishing epoxy resin compositions which do not contain halogen compounds with new epoxy resin (E3). The new epoxy molding compound (EMC-1) showed high flame resistance (UL-V0) and high thermal resistance ($451.9^{\circ}C$ at 5 wt% loss) enough to use as eco-friendly material.

  • Understanding of chemical composition and energy contents in tree is important to develope strategies of renewable energy policy to cope with climate change. Residual biomass as renewable energy source was evaluated and focused on the bark-containing branches. Chemical analysis studies were conducted for different part of yellow poplar (Liriodendron tulipifera), which were partitioned to inner bark, outer bark, small branches, medium branches, big branches and trunk. The variations in hydrophobic extractives, hydrophilic extractives, lignin, carbohydrate compositions, energy contents (higher heating value) and the ash content were determined. The inner and outer bark had higher ash content, hydrophobic and hydrophilic extractives content, and higher energy content than those of tree trunk. Polysaccharides content in inner and outer bark was quite lower than those of stem or branches. Based on the energy content of residual biomass, replacement of fossil fuel and greenhouse gas emission abatement were calculated.

  • Chemical compositions and lead isotope ratios for four glass bead samples of Seokga-tap were analyzed and the results were organized. Among 4 glass beads found in the Seokga-tap, 3 pieces were lead glass. Manufacturing method was to firstly grind pebbles finely and mix lead ore to be melt at $740{\sim}760^{\circ}C$. The mixed ratio of silica and lead was 3:7. Moreover, The evaluation on the lead isotope ratio indicated that two lead glass pieces used lead ore from northern Korea. One piece has the direction of southern Korea lead ore, but it requires a further review. One glass bead of Seokga-tap was brown and it was potash lead glass ($K_2O-PbO-SiO_2$) System. The mixed ratio was approximately 50:10:40 for silica, natural saltpeter, and lead, respectively. Lead isotope ratio data fell within the lead ore from northern China. Therefore, it was concluded that potash lead glass found in the Seokga-tap was produced in northern area of China at the end of $10^{th}$ century and transferred to the Seokga-tap.

Fabrication of La2O3-TiO2-SiO2 System Glass Derived from a Sol-Gel Process

  • $La_{2}O_{3}-TiO_{2}-SiO_{2}$ glass, a type that could not obtained so far by the conventional melting method, was prepared successfully using a sol gel process. Glass derived with the sol-gel process has compositions of $5La_{2}O_{3}-5TiO_{2}-90SiO_{2},\;5La_{2}O_{3}-10TiO_{2}-85SiO_{2}$, and $5La_{2}O_{3}-20TiO_{2}75SiO_{2}$. The UV-visible absorption edge of all glass compositions was below 400 nm. The measured density is in the range of 2.55-2.89, and was nearly identical to the calculated density and the refractive index of the glasses derived from the sol-gel ranges from 1.545 to 1.645. The molar additive coefficient of $TiO_{2}$ measured in this ternary system is lower than the calculated value, while the value of $La_{2}O_{3}$ is higher.

Rheological anomalies of the poly(ethylene 2, 6-naphthalate) and poly(ethylene terephthalate) blends depending on the compositions

  • The effects of the transreactions on the rheological properties have been found in the poly(ethylene 2, 6-naphthalate) (PEN) and poly (ethylene terephthalate) (PET) blends. The rheological properties were very much dependent on the blend compositions, which, in turn, were related to extent of the reactions. In particular, a blend with 50/50 wt% composition exhibits an unusual and remarkable decrease in complex viscosity and it may be related to the randomness of the copolymer structure through transreactions. It has been identified by investigating the extent of transreactions and block length of the copolymer from the (ethylene 2, 6-naphthalate) (EN) and (ethylene terephthalate) (ET) units from $^1{H}$ n.m.r. spectra.

Characterization of Molecular Composition of Bacterial Melanin Isolated from Streptomyces glaucescens Using Ultra-High-Resolution FT-ICR Mass Spectrometry

  • In this study, the chemical composition of bacterial melanin isolated from the Streptomyces glaucescens strain was elucidated by ultra-high-resolution Fourier transform ion cyclotron resonance (FT-ICR) mass spectrometry. Ultra-high-resolution mass profiles of the microbial melanin product were acquired using a 15 Tesla FT-ICR mass spectrometer in positive and negative ion modes via electrospray ionization to obtain more complete descriptions of the molecular compositions of melanin-derived organic constituents. A mass resolving power of 500,000 (at m/z 400) was achieved for all spectra while collecting 400 scans per sample with a 4 M transient. The results of this analysis revealed that the melanin pigment isolated from S. glaucescens predominantly exhibits CHON and CHO species, which belong to the proteins class of compounds, with the mean C/O and C/N ratios of 4.3 and 13.1, thus suggesting that the melanin could be eumelanin. This analytical approach could be utilized to investigate the molecular compositions of a variety of natural or synthetic melanins. The compositional features of melanins are important for understanding their formation mechanisms and physico-chemical properties.

Solid Dispersion of an HIV Protease Inhibitor

  • LB71350 is an HIV protease inhibitor with poor aqueous solubility and extensive first pass effect. The purpose of the present study was to test the feasibility of solid dosage form of LB71350 with improved bioavailability utilizing solid dispersion. Three different compositions with varying ratio of (LB71350: Gelucire 44/14: Tween 20) were studied. Capsule filling of these solid dispersion compositions was tested using a semi-automatic capsule filling system. Oral bioavailability in dog was tested. Chemical and physical stability at 4, 25 and $40^{\circ}C$ was monitored by HPLC assay, dissolution test, powder XRD and microscopy. The capsule filling system yielded uniform products of drug loading up to 10%. Oral bioavailability in dog was improved compared to the aqueous suspension of crystalline LB71350. Capsules were chemically stable for up to 6 months at $40^{\circ}C$. However, there were temperature and composition dependent physical changes. Decrease in dissolution rates after storage at $40^{\circ}C$ was due to the polymorphic change. In conclusion, manufacturing process, bioavailability, and physico-chemical stability have been considered to propose a solid dispersion capsule formulation for the HIV protease inhibitor with poor physico-chemical properties. A new less soluble crystalline form identified during the physical stability test warrants further study.
