Chelation of Tannin from Sorghum Extract using Fe(II) (수수 타닌의 Fe(II) 킬레이트)

  • This study investigated the chelation of a sorghum bran extract using iron (Fe) as a new natural colorant. The composition of the sorghum bran extract and chelation conditions were both examined. The thermal properties of the chelated colorants were analyzed using differential scanning calorimetry (DSC) and a thermal analyzer system(TGA). The sorghum bran extract solution showed a maximum absorbance at 281 nm based on UV/Vis spectrophotometry. According to the chelation pH conditions, pH 7.5 was the most effective. The chelation of the sorghum bran extract increased rapidly when increasing the iron concentration up to 2 mg/L, with no further chelation at a higher concentration. The particle size distribution curve for the chelated tannin revealed four groups: $4.5{\sim}17{\mu}m$, $20{\sim}42{\mu}m$, $45{\sim}80{\mu}m$, and $83{\sim}160{\mu}m$. In a DSC analysis, endothermic peaks attributed to the pyrolysis of the extract and chelated tannin were found at $318^{\circ}C$ and $415^{\circ}C$, respectively. In a TGA analysis, the chelation was shown to increase the final degradation temperature from $253^{\circ}C$ to $382^{\circ}C$, confirming that the chelation improved the thermal stability.

Effect of Chelation with Calcium Disodium EDTA on Haemato-biochemical and Trace Mineral Profile in Blood from Lead Exposed Calves

  • An experiment was performed using 20 calves of about one-month old to investigate the effect of chelation therapy with calcium disodium ethylenediaminetetraacetate ($CaNa_2$EDTA) alone or along with antioxidant $\alpha$-tocopherol in lead loaded calves on blood trace minerals, erythrocytic sulfahydryl groups and some haematobiochemical parameters. Fifteen calves were given lead orally at a daily dose of 7.5 mg of 99% pure lead acetate/kg body weight for 28 days. Then the lead was withdrawn on day 28 and the calves were randomly divided into three groups. Each group of five animals was either treated with $CaNa_2$EDTA alone at the dose rate of 110 mg/kg body weight in two divided doses for 4 days or along with $\alpha$-tocopherol at the dose rate of 100 mg/kg body weight orally daily for 7 days, keeping the remaining five calves as lead-exposed untreated controls. Blood samples were collected at the end of the lead exposure (day 0) and thereafter on day 2, 4, 7 and 10 from the start of the chelation treatment. The treatment with EDTA alone led to slow but non-significant improvement in blood copper level, but incorporation of antioxidant $\alpha$-tocopherol in chelation therapy resulted in its significant decline, as recorded on day 7-post treatment. Withdrawal of lead or treatment with $CaNa_2$EDTA alone or along with $\alpha$-tocopherol enhanced the erythrocytic thiol contents and the levels of T-SH and P-SH became statistically (p<0.05) comparable to those of lead-exposed controls by day 7 and 4, respectively. There was no significant (p>0.05) change in serum urea, creatinine, total protein and albumin levels between the treatment groups. It is concluded from the present investigation that treatment with $CaNa_2$EDTA at the present dose rate is safe to be used for chelation in lead loaded calves.

A Comparison Study of Radiostrontium Chelation with Chitin, Chitosan, EDTA and DTPA (카이틴, 카이토산, EDTA, DTPA의 방사성스트론튬(Sr-85) 착화물 형성에 관한 비교 연구)

  • Purpose: Chitin and chitosan are nontoxic natural chelators that chelate radiostrontium effectively. The purpose of this study was to compare radiostrontium chelation of chitin and chitosan with that of well known chemical chelators, namely EDTA and DTPA. Materials and Methods: The chelaton rates of chitin, chitosan, EDTA and DTPA were compared using a column chromatography method (Sephadex G-25M, Sweden). Three kinds of chitins and four kinds of chitosans were used. All of them were water soluble. Results: Phosphated chitosan showed the highest chelation yield of 97% at pH 7. All of chitins, chitosans, EDTA and DTPA showed chelation yield of more than 90% independent of varing pH level. Conclusion: Chitin and chitosan have similar chelation rate as compared with EDTA and DTPA.

In vitro Antioxidant Activity of Ogae (Korean Native Black Fowl) Egg White Protein Hydrolysates Fractionated by Ultrafiltration (오계란 단백질 가수 분해물 제조 및 한외여과 분획물의 in vitro 항산화 활성 특성)

  • Protein hydrolysates derived from plants and animals having antioxidant, suppression of hypertension, immunodulatory, alleviation of pain, and antimicrobial activity has been known as playing important role like hormone. This study was fractioned to hydrolysis of Ogae egg white protein using the ultrafiltration. The antioxidant activity of the produced peptides was analyzed. As a result, the maximum value of DPPH radical scavenging was 1 kDa(70.83 %), hydroxy radical scavenging was 5 kDa(47.01 %), superoxide anion radical scavenging was 5 kDa(40.57 %), and $Fe^{2+}$ chelation ability was 5 kDa(29.87 %). Furthermore, the antioxidant Inhibition concentration ($IC_{50}$) of peptides was evaluated for each fraction. As a result, the maximum value of HDS was superoxide anion radical scavenging($IC_{50}$, 5.42 mg/ml). 1 kDa was $Fe^{2+}$ chelation ability($IC_{50}$, 1.67 mg/ml), 5 kDa was $Fe^{2+}$ cheating ability($IC_{50}$, 2.09 mg/ml), 10 kDa was $Fe^{2+}$ cheating ability($IC_{50}$, 2.61 mg/ml), papain was $Fe^{2+}$ cheating ability($IC_{50}$, 4.53 mg/ml). Therefore, we expect that peptides produced from Ogae egg white protein using 5 kDa fraction are useful as an antioxidant functional food ingredients.

  • Diverse p-terphenyl compounds, named curtisians, have been isolated from the fungus Paxillus curtisii, and degradation of wood by this fungus is thought to be progressed by iron chelation of p-terphenyl curtisians. In this study, the iron chelation ability of p-terphenyls has been proved by chrome azurol S (CAS) assay, reducing power, and UV-visible spectroscopic analyses. The catechol moiety of p-terphenyl is an essential factor for the potent iron chelation ability, and thus deacylated curtisian with a tetrahydroxyl moiety in the central ring of p-terphenyl is more effective than acylated curtisians.

Bio-guided Isolation of Natural Iron Chelators from Mangifera indica Leaves and their Comparative Study to Desferal®

  • Through bio-guided isolation, two natural iron chelators were isolated from Mangifera indica L. leaves, identified as mangiferin (1) and iriflophenone-3-C-𝛽-D-glucoside (2). Their iron-chelating activity was compared to that of Desferal® using bipyridyl assay and EDTA as a standard. Mangiferin showed the highest activity with IC50 value of 0.385 mM (162.85 ㎍/mL). Furthermore, two combinations of mangiferin with Desferal® (M-D) and iriflophenone-3-C-𝛽-D-glucoside (M-I) were evaluated. The results showed that mangiferin potentiated the iron chelation activity of Desferal® about 46%, also that M-I combination is a promising candidate formula for iron chelation therapy. In addition, mangiferin and Desferal-iron complexes were prepared and characterized by IR, UV, and Mass spectra to compare their mode of chelation to iron. Their structural stability was studied by DFT calculations. Furthermore, they displayed increased ABTS antioxidant activity when bound to iron as compared to their free form, which enhances their pharmacological importance.

Color Stability of the Bioplastic containing Sorghum Extract Chelated by Fe(II) and Cu(II) (Fe(II)와 Cu(II)에 의해 킬레이트화 된 수수추출물 함유 바이오플라스틱의 색상 안정성)

  • To improve the color stability of the bioplastic containing sorghum extract, sorghum extract was chelated by a metal ion. The chelating activity was quantitatively evaluated under the various conditions. Chelation of sorghum extract by Cu(II) was determined by reaction with pyrocatechol violet, whereas Fe(II) chelation was investigated by forming complexes with ferrozine. Chelation of sorghum extract was increased rapidly with increasing concentrations of metal salt and sorghum extract. At a 0.1g/L metal salt addition level, the chelating activity of Fe(II) and Cu(II) were 66.7% and 54.2%, respectively. According to the chelation pH conditions, the sorghum extract was chelated almost 100% by Fe(II) above the pH 6.5. It was confirmed that Fe(II) was a strong chelator of sorghum extract than Cu(II). The sorghum extract chelated with metal salt exhibit higher thermal stability. The bioplastic containing chelated sorghum extract showed relatively less color change than the control.

Influence of pH on Chelation of BaCl2 and EDTA Using Isothermal Titration Calorimetry (등온적정열량계를 이용한 BaCl2와 EDTA 킬레이션 결합 반응의 pH 영향)

  • Isothermal titration calorimetry (ITC) is a useful technique to obtain thermodynamic binding properties such as enthalpy, Gibbs free energy, entropy, and stoichiometry of the chelation reaction. A single independent binding site model was used to evaluate the thermodynamic binding properties in BaCl2 and ethylenediaminetetraacetic acid (EDTA) in Trince and HEPES buffers. ITC enables us to elucidate the binding mechanism and find an optimal chelation condition for BaCl2 and EDTA in the pH range of 7~11. Chelation of BaCl2 and EDTA is a spontaneous endothermic reaction. As pH increased, entropic contributions dominated. The optimal pH range is narrow around pH 9.0, where 1:1 binding between BaCl2 and EDTA occurs.
