Morphology and Ecological Notes on the Larvae and Pupae of Simulium (Simulium) from Korea

  • Morphological features of larvae and pupae of twelve species of Simulium (Simulium) from Korea are investigated and redescribed. Detailed descriptions of immature stages of each species are provided with photographs. For the Korean members of subgenus Simulium, combinations of the following characteristics can be used to separate them from other Korean subgenera: larval abdomen without a pair of conical-shaped ventral tubercles; postgenal cleft variously shaped, viz., miter shaped, spearhead, bullet shaped or triangular; rectal papillae 3 simple lobe or compound lobes; pupal gill of 6-16 filaments; cocoon shoe, boot or slipper shaped; and head trichomes 3 pairs (1 facial+2 frontal). Habitat information and other ecological aspects of each species are provided. Updated keys to larvae and pupae of the Korean species of the subgenus Simulium are also provided to facilitate species identification. Taxonomic treatment based on external morphologies of larvae and pupae of Korean species of the subgenus Simulium are focused and included in this study. To understand their interspecific relationships, phylogenetic analyses using multiple data from molecule, morphology, and ecology are warranted in future research. Also continued exploration for new character systems is needed to establish more stable boundaries for subgenus and species delimitations.

낙동강 수계 위천에 서식하는 큰납지리(Acheilognathus macropterus) (Pisces : Cyprinidae)의 초기생활사 (Early Life History of Acheilognathus macropterus (Pisces:Cyprinidae) in the Wicheon Stream of Nakdonggang River)

  • The development of the egg, larvae, and juveniles of Acheilognathus macropterus from the Wicheon stream of Nakdonggang River was investigated for early life history characteristics. The fish used in the study were collected from Wicheon stream located in Bian-myeon, Uiseong-gun, Gyeongsangbuk-do. The fertilized egg was 1.88±0.09 mm (n=30) in long diameter and 1.34±0.06 mm (n=30) in short diameter. After fertilization at a water temperature of 20℃, the egg took 31 h and 40 min to hatch. Newly hatched larvae had an average total length of 3.92±0.13 mm (n=30) and possessed a yolk sac the mouth and anus were not developed. Twenty days after hatching, the postflexion larvae had an average total length of 8.08±0.29 mm (n=30), with the tail tip fully bent at 45° and began feeding. Thirty-three days after hatching, the number of fin stems reached an integer with an average total length of 15.0±1.08 mm (n=30). In this study, differences in the egg size, hatching time, melanophore development period, and distribution location were confirmed between A. macropterus and allied species.

코끼리조개의 인공종묘생산에 관한 연구 II. 난발생과 유생의 발달 (Studies on the Artificial Seedling Production of Geoduck Clam, Panope japonica II. Development of Egg and larvae)

  • 코끼리조개의 인공종묘생산 기술개발의 일환으로 성숙된 모패를 절개법에 의하여 인공수정시켜 난 발생 및 유생의 발달과정을 관찰한 결과를 요약하면 다음과 같다. 코끼리조개의 난은 분리침성란으로 나타났으며, 방란직후 알의 모양은 타원형이나 수정이 되면 직경 $70\mu$m의 구형으로 된다. 수정난의 발생은$ 11^{citc}C$에서 4시간 후 4세포기로 되고, 2일이 지나면 담륜자 유생(trochophore larvae), 수정 후 3일째에는 D상 유생, 23일째에는 각정기, 36일째에는 성숙유생으로 되었다. 수온(w)에 따른 각 단계별 소요시간(t)의 관계를 보면, 8세포 : 1/t=0.0209 w-0.1167 (r=0.9967) 포배기 : 1/t=0.0055 w-0.0192 (r=0.9825) 담륜자기 : 1/t=0.0034 w-0.0155 (r=0.9907) D상 유생기 : 1/t=0.0014 w-0.0023 (r=0.9843) 상기 식에서 산출된 코끼리조개의 생물학적 기초수온은 3.82$^{\circ}C$였다.

Influence of plant on distribution of an endangered butterfly, Leptalina unicolor (Bremer & Grey, 1853), in restored riverside areas along the Geum River

  • Background: The dramatic worldwide decline in the butterfly species Leptalina unicolor (Bremer & Grey) is largely the result of continuous habitat decline and disturbance by humans. The discovery of a narrow habitat in riverside wetlands utilized by L. unicolor raises the hope that such restricted key areas could be rather easily protected. Results: Here, we explain the environmental variables and habitat characteristics that primarily influence the distribution of L. unicolor discovered at the riverside areas along the Geum River. L. unicolor larvae were found at 9 of 13 study sites, and their abundance was strongly positively correlated with plant biomass. Our investigation showed that among four plant species (Miscanthus sinensis, Spodiopogon cotulifer, Setaria viridis, and Imperata cylindrica), L. unicolor larvae were the most abundant on the leaves of M. sinensis. They were not abundant on the leaves of S. cotulifer, S. viridis, or I. cylindrica. Interestingly, the number of L. unicolor larvae was positively correlated with the coverage area ($m^2$) of M. sinensis (F = 41.7, $r^2=0.74$, P < 0.0001). Conclusions: It appears that water (e.g., wetlands, ponds, and watersides) located along the riverside areas along the Geum River is important for the constant maintenance and conservation of L. unicolor. This is based on the habitat characteristics (water preference) of M. sinensis, which is used as a habitat by L. unicolor larvae. However, the waterside is dry and terrestrialization is in progress owing to the decreased water levels and water supply caused by an opened weir. Hereafter, this area will likely require management to secure a stable habitat for L. unicolor.

  • 1980년 9월(月)부터 1981년1월(月)까지 광주시도축장(光州市屠畜)에서 도살(屠殺)된 돈(豚)에서 검출(檢出)된 Anisakidae 유충(幼蟲)과 광주시장(光州市場)에서 구입(購入)한 고등어에서 채취(採取)한 유충(幼蟲)을 사용(使用)하여 그 충체(忠體)의 계측(計測)과 형태학적(形態學的) 구조(構造)를 관찰(觀察)하여 Anisakis I 형(型)으로 동정(同定)하게 된 결과(結果)는 다음과 같다. 1. 충체(蟲體)는 골벽내(骨壁內)에 침입(侵入)하였거나 골점막면(骨粘膜面)에 부착(附着)하고 있었다. 조직내(組織內) 충체(蟲體)는 변성(變性)의 정도(程度)에 따라 그 구조(構造)의 선명도(鮮明度)에 따라 차이(差異)가 있었으며 어느 것은 명확(明確)한 구조(構造)를 나타내었으나 어느 것은 식별(識別)할 수 없을 정도(程度)로 변화(變化)했다. 2. 돈(豚)과 고등어에서 채취(採取)한 충체(蟲體)의 계측평균직(計測平均直)는 각각(各各) 체장(體長)이 18.0mm, 18.7mm, 체부(體副)이 0.30mm, 0.41mm, 식도(食道)가 1.64mm, 1.18mm, 위(胃)가 0.56mm, 0.67mm 미(尾)가 0.13mm, 0.12mm이었다. 선인(先人)의 보고(報告)에서 체장(體長)에 있어서 사람의 예(例)의 24.4mm와 해어류(海魚類)의 예(例)의 28.4mm의 것보다는 적었으나 돈(豚)의 예(例)의 17.5mm하고는 비슷하였다. 3. 두부(頭部)에 천치(穿齒(boring tooth)와 미부(尾部)에 소극(小棘)(Mucron)이 있고 장위(長胃), 단미(短尾), 순원 (純圓)의 미단등(尾端等)의 특징(特徵)은 단위(短尾), 장미(長尾), 염미단(炎尾端), 미단(尾端)에 소극(小棘)이 없는 Anisakis II 형(型)과 구별(區別)되었다. 위측실(胃側室)(ventricular appendix)과 장맹낭(腸盲囊)(intestinal caecum) 등(等)이 없는 점(點)은 Terranova larvae, Contracaecum larvae, Raphidascaris larvae 그리고 Thynnascaris larvae 등(等)과 구별(區別)되었다. 4. 충체(蟲體)의 단면(斷面)은 각피(角皮). 근층(筋層), 배선(背線), 복선(腹線), Y자형(字形) 또는 쌍엽상(雙葉狀)의 측선(側線), 배세선(排泄線), 장(陽)의 단층도주장토피세절구등은 Anisakis I 형(型)의 특징등(特徵等)을 나타내었고 이것들은 변성(變性)의 진행정도(進行程度)에 따라 다양(多樣)한 변화(變化)를 나타냈다. 5.이상의 형태학적(形態學的) 및 현미경적(顯微鏡的)관찰(觀察)로 미루어 Anisakis I 형(型)의 특 유충(幼蟲)이 확실(確實)히 인정(認定)되었다. 6. 한국(韓國), 일본(日本), 대만(臺灣)에서 돈(豚)에의 Anisakis I 형(型)의 자연감염(自然感染)의 예(例)는 한국(韓國), 일본(日本)에서 각각(各各) 2예(二例)가 있을 뿐이고 사람의 증예(症例) 한국(韓國)에서 2예(例)가 있고 일본(日本)에서는 1,000여예(餘例)가 보고되었고, 대만(臺灣)에서는 인(人), 축(畜) 공(共)히도 보고(報告)된 바 없다.

Investigations on Bacteria as a Potential Biological Control Agent of Summer Chafer, Amphimallon solstitiale L. (Coleoptera: Scarabaeidae)

  • Studying the bacteria of hazardous insects allows the opportunity to find potentially better biological control agents. Therefore, in this study, bacteria from summer chafer (Amphimallon solstitiale L., Coleoptera: Scarabaeidae) we isolated and identified the insecticidal effects of bacteria isolated from A. solstitiale and Melolontha melolontha L. (common cockchafer, Coleoptera: Scarabaeidae) and the mixtures of these bacterial isolates were investigated on A. solstitiale larvae. Crystals from Bacillus sp. isolated from M. melolontha were also purified, and tested against the second and third-stage larvae of A. solstitiale. The bacterial isolates of A. solstitiale were identified as Pseudomonas sp., Pseudomonas sp., Bacillus cereus and Micrococcus luteus, based on their morphology, spore formation, nutritional features, and physiological and biochemical characteristics. The insecticidal effects of the bacterial isolates determined on the larvae of A. solstitiale were $90\%$ with B. cereus isolated from A. solstitiale, and $75\%$ with B. cereus, B. sphaericus and B. thuringiensis isolated from M. melolontha within ten days. The highest insecticidal effects of the mixed infections on the larvae of A. solstitiale were $100\%$ both with B. cereus+B. sphaericus and with B. cereus+B. thuringiensis. In the crystal protein bioassays, the highest insecticidal effect was $65\%$ with crystals of B. thuringiensis and B. sphaericus isolated from M. melolontha within seven days. Finally, our results showed that the mixed infections could be utilized as microbial control agents, as they have a $100\%$ insecticidal effect on the larvae of A. solstitiale.

Insecticidal Characterization of Thirteen Bacillus thuringiensis Isolates from Soil (III)

  • Thirteen strains of Bacillus thuringiensis were isolated from soil in Korea and characterized. The all strains produced parasporal crystals and spores in their cells. Two strains had bipyramidal crystals, seven strains contianed round ones and four strains had unregular ones. Only minor biochemical characteristics of the thirteen isolates were different and distinctive, however general characteristics were similar to the known serotypes of B. thuringiensis. Two strains were resistant to ampicilin. Three strains were resistant to bacitracin, six strains were resistant to cephalothin, two strains were resistant to colistin, HL-68 strain was resistant to gentamycin, HL-67 strain was resistant to kanamycin and HL-71 was resistant to tetracycline. Two strains were resistant to penicillin G. Four strains were toxic to Bombyx mori larvae and eleven strains were toxic to Culex pipiens larvae.

Reproductive characteristics, egg and larval development of short ninespine stickleback, Pungitius kaibarae

  • In this study, the reproductive behavior and embryonic and larval development of the short ninespine stickleback Pungitius kaibarae was described and illustrated based on observations during spawning, hatching, and larval rearing trials. Adult P. kaibarae were collected downstream in Jinhae during the reproductive season (April-May). Males had nuptial coloration on their entire black bodies, with blue dorsal spines and yellow eyes, whereas females had a brown spotted pattern on their bodies. Males built nests on the stems of water weeds and attracted females. Fertilization occurred in the nest immediately after spawning, and males guarded the eggs until hatching. The fertilized eggs of P. kaibarae were spherical, demersal, adhesive, and transparent, and each egg measured 1.43 ± 0.07 mm in diameter. The morula, blastula, and gastrula stages, as well as hatching began at 5, 18.5, 21.5, and 96 post fertilization (HPF), respectively, at 20.0 ± 0.5℃. The newly hatched larvae had a total length (TL) of 5.67 ± 0.50 mm, with a yolk volume of 0.583 ± 0.059 mm3. Their mouths and anuses had not yet opened. At 2 days posthatching (days post hatching, DPH), the yolk was completely absorbed and the larvae began to feed exogenously. Pigmentation was observed in freshly hatched larvae 4 h after hatching, with the presence of eight areas with a dotted pattern on the dorsal surface of the larvae and dispersed spots on the head and yolk sac. At 30 DPH, the TL of the juveniles was 21.34 ± 1.70 mm. The nest area and number of eggs were 259.56 ± 101.39 mm2 (75.18-506.04) and 155.33 ± 114.12 (0-437), respectively.

Characterization of bacillus thuringiensis isolates form soil in wonju area

  • Three strains (KW-1, KW-14, KW-15) of Bacillus thuringiensis were isolated from soil in Wonju area and characterized. The three strains produced parasporal inclusion bodies (crystals) and spores in their cells. The KW-1 strain produces spherical crystals. The crystals of strain KW-14 are bipyramidal crystal. The KW-15 strain harbors irregular crystals. Only minor biochemical characteristics of the three isolates were different and distinctive, however general characteristics were similar to the known serotypes of B. thuringiensis. Three strains were resistant to penicilin G, oxacillin and cephalothin. Three strains were highly toxic to Bombyx mori larvae, but not to the Culex pipiens larvae.

서해연안 해역에 분포하는 부유성 난 및 자치어의 분포특성 (Distribution of ichthyoplankton in the West coastal waters of Korea)

  • The ichthyoplanktons were sampled by Bongo net to study the distribution of the eggs and larvae in the West coastal waters of Korea during the study period (February, May, August and October of 2017). Collected fish eggs were divided into Engraulis japonicus and unidentified species. The most dominant species, Engraulis japonicus accounted for 79.52% of the total fish egg, which was followed by unidentified species (20.48%). The total of 36 taxa in fish larvae were identified based on morphological and molecular characteristics. Fish larvae were dominant by Gobiidae species. 48.30% of the total catch in abundance, which was followed by Engraulis japonicus (31.86%). These two taxa composed 80.16% of the total collected fish larvae. The seasonal diversity index of species from the collected fish larvae in the West coastal waters in 2017 was 0.625-1.883 and the evenness index was 0.380-0.865. In addition, dominance index was 0.618-0.920 and richness was 0.573-3.189.
