  • Poly(oxypropylene-oxyethylene glycol) block copolymer(PBC) oil dispersant, which has low toxicity, high biodegradability, and an excellent dispersion efficiency to crude oils and weathered W/O emulsion was prepared by blending PBC, poly(oxyethylene) oleate, and sorbitan monooleate. The dispersing efficiency was measured by swirling flask method. The PBC oil dispersant had an excellent dispersing efficiency to weathered oil products formed as stable W/O emulsion, and the low toxicity, such as 4000 ppm to Oryzias Latipes(24 hr, TLM), Brine Shrimp Artemia(24 hr, TLM).

  • During the serial attempts to identify the chemical and biological characteristics of Ailanthi Cortex Radicis, the root bark of Ailanthus altissima (Simaroubaceae), we find out the serious toxic effect on kidney by chloroform fraction of the methanolic extract of the herb drug. The toxicities were revealed as the increase of urea nitrogen amount in blood and lactate dehydrogenase and ${\gamma}-glutamyltransferase$ activities in urine and the decrease of the concentration of glutathione and both of protein bound and non-protein bound -SH in kidney tissue.

  • In May 2001, the Stockholm Convention on Persistent Organic Pollutants (POPs) for phasing out and eliminating POPs was signed by 90 countries at the Diplomatic Meeting in Stockholm. In 1998, three years before the Convention, the protocols on POPs and heavy metals were adopted by the United Nations Economic Commission for Europe under the Convention on Long-Range Transboundary Air Pollution. Growing attention on POPs and heavy metals during the past 10 years is primarily due to their toxicity in minute quantities. POPs and some metal compounds are even more toxic because of their bioaccumulation potentials associated with a high lipid solubility. Furthermore, owing to their persistence and semi - volatility, they are widely distributed in the environment, traveling great distances on wind and water currents. Recent international cooperation to address POPs and heavy metals has focused on these issues. Long -range transport of those pollutants are particularly concerned since Korea is located downwind of prevailing westerlies from China. In this paper, a review is provided to assess the properties, sources, emissions, and atmospheric concentrations on POPs and heavy metals.

Biopharmaceutical Studies of Lonazolac Acetic Acid Ester and Lonazolac Argininate (로나졸락 초산에스테르 및 로나졸락 알지니네이트의 생물약제학적 연구)

  • Two new prodrugs of lonazolac, lonazolac acetic acid ester and lonazolac argininate, were prepared and examined for physicochemical properties and biopharmaceutical characteristics. The prodrugs were stable in solid state and lonazolac argininate showed higher dissolution rate than lonazolacca in both artificial gastric and intestinal juices. These prodrugs have higher analgegic effect than that of lonazolac-Ca in mice, and increased anti-inflammatory activities in rats. In addition, ulcerogenic effects and acute toxicity of these prodrugs were lower than those of lonaaolac-Ca. Lonazolac acetic acid ester showed larger area under the plasma concentration-time curves (AUC) than that of lonazolac. Therefore, it was suggested that these prodrugs of lonazolac have advantages over lonzolac-Ca for not only enhanced bioavailability but also decreased ulcerogenic and toxic effects.

Pulmonary Fibrosis caused by Asbestos Fibers in the Respiratory Airway

  • Asbestos products had been widely used until 2007 in Korea since the 1930s. A total ban on their production and applications has been imposed because of the toxic effect of asbestos fibers on the human health. The inhaled asbestos fibers increase reactive oxygen species and inflammatory reactions in the respiratory airway including the alveolar sac, resulting in DNA damages and secretion of several inflammatory cytokines or chemokines. These paracrine communications promote the proliferation of fibroblasts and the synthesis of collagen fibers, thereby depositing them into the extracellular matrix at the interstitial space of alveoli. The fibrotic tissue hindered the gas exchange in the alveolus. This reviews describes not only the cytotoxic effects of asbestos fibers with different physical or chemical characteristics but also the interaction of cells that make up the respiratory airway to understand the molecular or cellular mechanisms of asbestos fiber-induced toxicity. In addition, we propose a pulmonary toxicity research technique based on the mini-lung that can mimic human respiratory system as an alternative to overcome the limitations of the conventional risk assessment of asbestos fibers.

  • A hemolysin producing strain was isolated from Kum rivet estuary located in west part of Korea. In the process of identification the isolated strain was similar to V. mimicus but did not show characteristics of known Vibrio species; therefore, the strain was designated as Vibrio sp. D9 ( V. kumkang) tentatively and further identification study was carried out by comparing its bacteriological characteristics, Morphologically Vibrio sp. D9 was a typical straight roe with a polar flagellium. Among known Vibrio species no identical strains were found when using automatic bacteria identification system ($MicioLog^(TM)$system, release 4.0, Biolog Inc., USA) which evaluated the ability of metabolizing 95 kinds of carbon and nitrogen sources. Vibrio sp. D9 showed 18 and 13 different responses as compared to V. mimicus and V. cholerae, respectively. Clear hemolysis zones were observed with the strain against human and sheep blood agar plate, Hemolytic toxicity was confirmed by strong vascular permeability and fatal toxicity against mouse was also observed. Thus the strain was a pathogenic vibrio. Growth conditions for Vibrio sp. D9 were salinity of $0{\~}5.0{\%}$, pH of $6.4{\~}9.8$, temperature of $15{\~}41^{\circ}C$, respectively.

Ecotoxicity Assessment of Silver Nanomaterials with Different Physicochemical Characteristics in Diverse Aquatic Organisms (다양한 특성의 은나노물질이 수생생물에 미치는 독성영향평가)

  • Silver nanomaterials have been intensively applied in consumer products of diverse industrial sectors because of their strong biocidal properties and reported to be hazardous to aquatic organisms once released in the environment. Nanomaterials including sliver, are known to be different in toxicity according to their physicochemical characteristics such as size, shape, length etc. However studies comparing toxicity among silver nanomaterials with different physicochemical characteristics are very limited. Here, toxicities of silver nanomaterials with different size (50, 100, 150 nm), length (10, $20{\mu}m$), shape (wire, sphere), and coating material (polyvinylpyrrolidone, citrate) using OECD test guidelines were evaluated in aquatic species (zebrafish, daphnia, algae) and compared. On a size property, the smaller of silver nanomaterials, the more toxic to tested organisms. Sphered type of silver nanomaterials was less toxic to organisms than wired type, and shorter nanowires were less toxic than longer ones. Meanwhile the toxic effects of materials coated on silver nanomaterials were slightly different in each tested species, but not statistically significant. To the best of our knowledge, it is first investigation to evaluate and compare ecotoxicity of silver nanomaterials having different physicochemical characteristics using same test species and test guidelines. This study can provide valuable information for human and environmental risk assessment of silver nanomaterials and guide material manufacturers to synthesize silver nanomaterials more safely to human and environment.

Efficacy and Tolerability of Weekly Docetaxel, Cisplatin, and 5-Fluorouracil for Locally Advanced or Metastatic Gastric Cancer Patients with ECOG Performance Scores of 1 and 2

  • Background: Docetaxel, cisplatin, 5-fluorouracil (DCF) given every three weeks is an effective, but palliative regimen and significantly toxic especially in patients who have a low performance score. Here, we aimed to evaluate the efficacy and tolerability of a weekly formulation of DCF in locally advanced and metastatic gastric cancer patients. Materials and Methods: 64 gastric cancer patients (13 locally advanced and 51 metastatic) whose ECOG (Eastern Cooperative Oncology Group) performance status (PS) was 1-2 and who were treated with at least two cycles of weekly DCF protocol as first-line treatment were included retrospectively. The weekly DCF protocol included $25mg/m^2$ docetaxel, $25mg/m^2$ cisplatin, and 24 hours infusion of $750mg/m^2$ 5-fluorouracil, repeated every week. Disease and patient characteristics, prognostic factors, treatment response, grade 3-4 toxicity related to treatment, progression free survival (PFS) and overall survival (OS) were evaluated. Results: Of the patients, 41 were male and 23 were female; the median age was 63 (29-82) years. Forty-one patients were ECOG-1 and 23 were ECOG-2. Of the total, 81.2% received at least three cycles of chemotherapy. Partial response was observed in 28.1% and stabilization in 29.7%. Overall, the disease was controlled in 57.8% whereas progression was noted in 42.2%. The median time to progression was 4 months (95%CI, 2.8-5.2 months) and median overall survival was 12 months (95%CI, 9.2-14.8 months). The evaluation of patients for grade 3-4 toxicity revealed that 10.9% had anemia, 7.8% had thrombocytopenia and 10.9% had neutropenia. Non-hematologic toxicity included renal toxicity (7.8%) and thrombosis (1.6%). Conclusions: In patients with locally advanced or metastatic gastric cancer who were not candidates for DCF administered every-3-weeks, a weekly formulation of DCF demonstrated modest activity with minimal hematologic toxicity, suggesting that weekly DCF is a reasonable treatment option for such patients.

A Study on the Fire Risk of Car Interior Materials (자동차 내장재의 화재위험성에 관한 연구)

  • In this study, we have performed several tests for composite plastic materials to be applied on interior materials of a vehicle to identify their combustion characteristics using cone calorimeter, smoke density chamber and toxicity index chamber. We have prepared a total of 12 samples for 4 major parts of a vehicle wherein each major part has 3 different materials. The results of cone calorimeter test showed ignition time of PVC sheet and PVC leather were 2s. The 8 samples showed under less than 10s of ignition time. The sample comprising Nylon and PE had the biggest maximum heat release rate of 635 $kW/m^2$. The sample comprising Rubber showed the smallest maximum heat release rate but with the biggest total heat release. The results of smoke density chamber test showed the sample that is made up with Rubber had the biggest specific optical smoke density. The sample comprising PVC leather and PUR showed the biggest VOF4 which enables the initial smoke production. The results of toxicity index test showed that all samples contained carbon dioxide content exceeding its lethal concentration. The sample comprising PVC showed high content of hydrogen chloride and hydrogen bromide. The PVC sheet showed the biggest toxicity index calculated by using lethal concentration and test results. Toxicity index of all sample wes over 1.

Phytochemical, Antidiabetic, Antioxidant, Antibacterial, Acute and Sub-Chronic Toxicity of Moroccan Arbutus unedo Leaves

  • Objectives: Moroccan Arbutus unedo is an essential medicinal plant; however, little is known about the biological properties of its leaves mentioned in Moroccan traditional medicine. Methods: Various standard experiments were performed to evaluate the phytochemical, antidiabetic, antioxidant, antibacterial, and acute and sub-chronic toxicity characteristics of A. unedo leaves. Results: Phytochemical screening led to the identification of several phytochemical classes, including tannins, flavonoids, terpenoids, and anthraquinones, with high concentrations of polyphenols (31.83 ± 0.29 mg GAEs/g extract) and flavonoids (16.66 ± 1.47 mg REs/g extract). Further, the mineral analysis revealed high levels of calcium and potassium. A. unedo extract demonstrated significant antioxidant and anti-diabetic activities by inhibiting α-amylase (1.350 ± 0.32 g/mL) and α-glucosidase (0.099 ± 1.21 g/mL) compared to the reference drug Acarbose. Also, the methanolic extract of the plant exhibited significantly higher antibacterial activity than the aqueous extract. Precisely, three of the four examined bacterial strains exhibited substantial susceptibility to the methanolic extract . Minimum bactericidal concentration (MBC)/minimum inhibitory concentration (MIC) values indicated that A. unedo harbor abundant bactericidal compounds. For toxicological studies, mice were administered with A. unedo aqueous extract at single doses of 2,000 and 5,000 mg/kg. They did not exhibit significant abnormal behavior, toxic symptoms, or death during the 14-day acute toxicity test and the 90-day sub-chronic toxicity test periods. The general behavior, body weight, and hematological and biochemical status of the rats were assessed, revealing no toxicological symptoms or clinically significant changes in biological markers observed in the mice models, except hypoglycemia, after 90 days of daily dose administration. Conclusion: The study highlighted several biological advantages of A. unedo leaves without toxic effects in short-term application. Our findings suggest that conducting more comprehensive and extensive in vivo investigations is of utmost importance to identify molecules that can be formulated into pharmaceuticals in the future.
