Bile Acid Inhibition of N-type Calcium Channel Currents from Sympathetic Ganglion Neurons

  • Under some pathological conditions as bile flow obstruction or liver diseases with the enterohepatic circulation being disrupted, regurgitation of bile acids into the systemic circulation occurs and the plasma level of bile acids increases. Bile acids in circulation may affect the nervous system. We examined this possibility by studying the effects of bile acids on gating of neuronal (N)-type $Ca^{2+}$ channel that is essential for neurotransmitter release at synapses of the peripheral and central nervous system. N-type $Ca^{2+}$ channel currents were recorded from bullfrog sympathetic neuron under a cell-attached mode using 100 mM $Ba^{2+}$ as a charge carrier. Cholic acid (CA, $10^{-6}M$) that is relatively hydrophilic thus less cytotoxic was included in the pipette solution. CA suppressed the open probability of N-type $Ca^{2+}$ channel, which appeared to be due to an increase in (no activity) sweeps. For example, the proportion of sweep in the presence of CA was ~40% at +40 mV as compared with ~8% in the control recorded without CA. Other single channel properties including slope conductance, single channel current amplitude, open and shut times were not significantly affected by CA being present. The results suggest that CA could modulate N-type $Ca^{2+}$ channel gating at a concentration as low as $10^{-6}M$. Bile acids have been shown to activate nonselective cation conductance and depolarize the cell membrane. Under pathological conditions with increased circulating bile acids, CA suppression of N-type $Ca^{2+}$ channel function may be beneficial against overexcitation of the synapses.

A Comparison of Flow Condensation HTCs of R22 Alternatives in the Multi-Channel Tube (다채널 알루미늄 평판관내 R22와 R134a의 흐름 응축 열전달 성능 비교)

  • Flow condensation heat transfer coefficients (HTCs) of R22 and R134a were measured on a horizontal 9 hole aluminum multi-channel tube. The main test section in the refrigerant loop was made of a flat multi-channel aluminum tube of 1.4 mm hydraulic diameter and 0.53 m length. Refrigerant was cooled by passing cold water through an annulus surrounding the test section. Data were obtained in the vapor qualities of 0.1∼0.9 at mass flux of 200∼400 kg/$m^2$s and heat flux of 7.3∼7.7 ㎾/$m^2$ at the saturation temperature of 4$0^{\circ}C$. All popular correlations in single-phase subcooled liquid and flow condensation originally developed for large single tubes predicted the present data of the flat tube within 20% deviation when effective heat transfer area is used in determining experimental data. This suggests that there is little change in flow characteristics and patterns when the tube diameter is reduced down to 1.4 mm diameter range. Thermal insulation for the outer tube section surrounding the test tube for the transport of heat transfer fluid is very important in fluid heat-ing or cooling type heat transfer experimental apparatus.

Effect of Intersection Angle of the Flow-focusing Type Droplet Generation Device Channel on Droplet Diameter by using Numerical Simulation Modeling (수치해석 모델링을 이용한 교차 흐름 미세유체 액적 생성 디바이스 채널 교차각이 액적 직경에 미치는 영향)

  • In this paper, we studied the effects of intersection angles of the flow-foucusing type droplet generation device inlet channel on droplet diameter using numerical simulation modeling. We modeled different intersection angles with a fixed continuous channel width, dispersed channels width, orifices width, and expansion channels width. Numerical simulations were performed using COMSOL Multiphysics$^{(R)}$ to solve the incompressible Navier-Stokes equations for a two-phase flow in various flow-focusing geometries. Modeling results showed that an increase of the intersection angle causes an increase in the modification of the dispersed flow rate ($v^{\prime}{_d}$), and the increase of the modification of the continuous flow rate ($v^{\prime}{_c}$) obstructs the dispersed phase fluid flow, thereby reducing the droplet diameter. However, the droplet diameter did not decrease, even when the intersection angle increased. The droplet diameter decreased when the intersection angle was less than $90^{\circ}$, increased at an intersection angle of $90^{\circ}$, and decreased when the intersection angle was more than $90^{\circ}$. Furthermore, when the intermediate energy deceased, there was a decrease in the droplet diameter when the intersection angle increased. Therefore, variations in the droplet diameter can be used to change the intersection angle and fluid flow rate.

High-temperature ultrasonic thickness monitoring for pipe thinning in a flow-accelerated corrosion proof test facility

  • In order to monitor the pipe thinning caused by flow-accelerated corrosion (FAC) that occurs in coolant piping systems, a shear horizontal ultrasonic pitch-catch waveguide technique was developed for accurate pipe wall thickness monitoring. A clamping device for dry coupling contact between the end of the waveguide and pipe surface was designed and fabricated. A computer program for multi-channel on-line monitoring of the pipe thickness at high temperature was also developed. Both a four-channel buffer rod pulse-echo type and a shear horizontal ultrasonic waveguide type for high-temperature thickness monitoring system were successfully installed to the test section of the FAC proof test facility. The overall measurement error can be estimated as ${\pm}10{\mu}m$ during a cycle from room temperature to $200^{\circ}C$.

Flow Characteristics for Guide Tube of Circular Irradiation Hole in HANARO (하나로 원형 조사공의 안내관 유동특성)

  • The HANARO, a multi-purpose research reactor of 30 MWth, open-tank-in-pool type, has been under normal operation since its initial criticality in February, 1995. The HANARO is composed of inlet plenum, grid plate, core channel with flow tubes and chimney. The reactor core channel is located at about twelve meters (12 m) depth of the reactor pool and cooled by the upward flow that the coolant enters the lower inlet of the plenum,. rises up through the grid plate and the core channel and comes out from the outlet of chimney. A guide tube is extended from the reactor core to the top of the reactor chimney for easily un/loading a target under the reactor normal operation. But active coolant through the core can be quickly raised up to the top of the chimney through the guide tube by a jet flow. This paper describes an analytical analysis that is the study of the flow behavior through the guide tube under reactor normal operation and unloading the target. As results, it was conformed through the analysis results that the guide jet is suppressed under the top of the chimney after modifying the orifice diameter of 37.5 mm to 31 mm.

Prandtl-Meyer Expansion Through a Small Wavy Wall of Supersonic Flow with Condensation in a Channel (유로내에서 응축을 수반하는 초음속 유동의 미소진폭 파형벽에 의한 Prandtl-Meyer 팽창)

  • The characteristics Prandt1-Meyer expansion of supersonic flow with condensation through a wavy wall in a channel are investigated by experiment and numerical direct marching method of characteristics. In the present study, for the case of moist air flow in the type of indraft supersonic wind tunnel, the dependency of location of formation and reflection of the oblique shock wave generated by the wavy wall and the distribution of flow properties, on the specific humidity and temperature at the entrance of wavy wall and the attack angle of the wavy wall to the main stream is clarified by schlieren photograph, distribution of static pressure and Mach number, and plots of numerical results. Also, we confirm that the wavy wall plays an important key role in the formation of oblique shock wave, and that the effect of condensation on the flow field appears apparently.

Computer Simulation Study of the Potential Anti-arrhythmic Properties of Paeonol (Paeonol의 잠재적인 항부정맥 효과의 컴퓨터 시뮬레이션 연구)

  • Paeonol is a major component found in the Paeoniaceae family such as Paeonia suffruticosa Andrews. Paeonia suffruticosa Andrews has traditionally been used to enhance blood flow and relieve joint pain in east Asian countries including China, Korea and Japan. Current research has shown that paeonol blocked the voltage-gated sodium channel and L-type calcium channel. However, there is a lack of research to reveal the relation between cardiac function and blockade of ion channels by paeonol. Therefore, the aim of this study is to investigate whether paeonol has anti-arrhythmic effects via modulating cardiac ion channels. It is collected that the effects of paeonol on multiple ion channels such as the fast sodium channel and L-type calcium channel from published papers. To incorporate the information on multi-channel block, we computed the effects using the mathematical cardiac model of the guinea-pig and rat ventricular cells (Noble 1998 and 1991 model) and induced early after-depolarizations (EADs) to generate an arrhythmia in the whole heart. Paeonol slightly shortened the action potential duration in the normal cardiac ventricular action potential by the inhibition of sodium channel and L-type calcium channel. Paeonol presented the protective effect from EADs by the inactivation of sodium channel but not L-type calcium channel. Paeonol did not show any changes when it treated on normal ventricular cells through the inhibition of sodium channel, but the protective effect of paeonol through sodium channel on EADs was dose-dependent. These findings suggest that paeonol and its original plant may possess anti-arrhythmic activity, which implies their cardioprotective effects.

Transverse variability of flow and sediment transport in estuaries with an estuarine dam

  • Estuarine dams are dams constructed in estuaries for reasons such as securing freshwater resources, controlling water levels, and hydroelectric power generation. These estuarine dams alter the flow of freshwater to the coastal ocean and the tidal properties of the estuaries which has implications for the estuaries' circulation and sediment transport. A previous study has analyzed the effect of estuarine dams on 1D (along-channel) circulation and sediment transport. However, the effect of estuarine dams on the transverse variability of along-channel and across-channel circulation and sediment transport has not been studied and is not known. In this study, a coupled hydrodynamic-sediment dynamic numerical model (COAWST) was used to analyze the transverse variability of along-channel and across-channel flow and sediment transport in estuaries with estuarine dams. The estuarine dam was found to change the 3D structure of circulation and sediment transport, and the result was found to depend on the estuarine type (i.e., strongly stratified (SS) or well-mixed (WM) estuary). The SS estuary had inflow in the channel and outflow over the shoals, consistent with estuarine circulation. Longer discharge interval reduced the estuarine circulation. The WM estuary had inflow over the shoals and outflow in the channel, consistent with tide-induced circulation. As the estuarine dam was located nearer to the estuary mouth, the tide-induced circulation was reduced and replaced with estuarine circulation. The lateral circualtion was the greatest in the tide-dominated estuaries. It was reduced and changed direction due to differential advection change as the dam was located nearer the mouth. Overall, the WM estuary transverse flow structure changed the most. Lateral sediment flux was important in all estuaries, particularly for transporting sediments to the tidal flats.

  • The uniform gas distribution between anode channels of the indirect internal reforming type molten carbonate fuel cell (MCFC) is crucial design parameter because of the electric performance and the durability problems. A three-dimensional computational fluid dynamics (CFD) analysis is performed to investigate flow characteristics in the anode channels and manifold under different pressure drop and channel temperature conditions. The combined meshes consists of hexadral meshes in the channels and polyhedral meshes in the manifold are adopted and chemical reactions inside the MCFC system are not included because of computational difficulties associated with the size and geometric complexity of the system. Results indicate that the uniformity in flow-rate is in the range of $\pm$ 0.048 % between the anode channels when the pressure drop of anode channel is about 150 Pa. A gas flow-rate uniformity decreases as the pressure drop of anode channels decreases and as the temperature difference between indirect internal reforming (IIR) channels and anode channels increases.

Development of a Linear Stability Analysis Model for Vertical Boiling Channels Connecting with Unheated Risers

  • The characteristics of two-phase flow instability in a vertical boiling channel connecting with an unheated riser are investigated through the linear stability analysis model. Various two-phase flow models, including thermal non-equilibrium effects, are taken into account for establishing a physical model in the time domain. A classical approach to the frequency response method is adopted for the stability analysis by employing the D-partition method. The adequacy of the linear model is verified by evaluating experimental data at high quality conditions. It reveals that the flow-pattern-dependent drift velocity model enhances the prediction accuracy while the homogeneous equilibrium model shows the most conservative predictions. The characteristics of density wave oscillations under low-power and low-quality conditions are investigated by devising a simple model which accounts for the gravitational and frictional pressure losses along the channel. The necessary conditions for the occurrences of type-I instability and flow excursion are deduced from the one-dimensional D-partition analysis. The parametric effects of some design variables on low quality oscillations are also investigated.
