  • Ultrastructural changes of chloride cells in the gill from seawater-adapted killifish (Orizias latipes) were studied with a transmission electronmicroscope. Chloride cells contain many mitochondria and specifically developed tubular systems. Apical pits, formed by several neighboring chloride cells, were exposed to the environment. Degeneration and death of the chloride cells by apoptosis occurred more frequently than by necrosis. Apoptotic chloride cells shrank, became to apoptotic bodies, and eventually were phagocytosed and digested by the microphages around them. We conclude that apoptosis plays an important role in increased cellular turnover of chloride cells for the osmoregulation caused by changes in salinity of the environment.

The changes of soil salinity in the Pinus densiflora forest after seawater spread using a fire-fight helicopter

  • The east coast of the Korean Peninsula is susceptible to fires because of the low rainfall in winter and spring, and large forest fires have occurred in this area. Lack of fresh water to combat fires has hampered efforts to prevent widespread forest fires in this region. Seawater has not been used as a suppressant because of possible detrimental effects of salt. We investigated the mobility of saline water in the forest soil and their effect on the microbial activity. Using a fire-fighting helicopter, seawater was sprayed over three plots (50 × 100 m) located on the eastern slope of the Baekdu mountain range in South Korea in April, 2011. We sampled the soil in April 4, May 20, and August 5 to determine the amount of salt that remained in the soil. The electrical conductivity value of the soil decreased to <400 μS/cm over a 1-month period. Approximately, four months after the application of seawater, the electrical conductivity value and Na+ content in all treatment plots did not significantly differ to those of the control plot, and total microbial activity also recovered to that of the control. Our results indicate that the amount of rainfall, soil physical-chemical properties, and topological factors may be a critical factor determining the mobility of saline water in forest soil.

  • Harsh seawater environment is subdivided into marine atmosphere, splash zone, tidal zone, submerged zone and bottom of sea depending on the exposed part. Since corrosion rate depends on the conditions of the exposed parts, proper protection and maintenance for each parts are essential for long-term use of steel structures in seawater environment. For steel structures which were installed in Shiwha Lake, a special maintenance system is required to guarantee its long-term durability and safety. As the tidal power plant has recently been operated, the salinity has risen due to the rapid influx of seawater upstream into Sihwa Lake and the corrosion tendency of the structure is variable according to the water level fluctuation. In this study, corrosion properties of steel structures under water level fluctuation was evaluated by corrosion rate measurement, visual inspection and natural potential measurement and their durability and life management were discussed in view of the effect of variation in of seawater level fluctuations in Shihwa Lake.

분산형 해수침투 모델을 이용한 양적 지표 기반의 해안지하수 취약성 평가연구 (Quantitative Assessment of Coastal Groundwater Vulnerability to Seawater Intrusion using Density-dependent Groundwater Flow Model)

  • Extensive groundwater abstraction has been recognized as one of the major challenges in management of coastal groundwater. The purpose of this study was to assess potential changes of groundwater distribution of northeastern Jeju Island over 10-year duration, where brackish water have been actively developed. To quantitatively estimate the coastal groundwater resources, numerical simulations using three-dimensional finite-difference density-dependent flow models were performed to describe spatial distribution of the groundwater in the aquifer under various pumping and recharge scenarios. The simulation results showed different spatial distribution of freshwater, brackish, and saline groundwater at varying seawater concentration from 10 to 90%. Volumetric analysis was also performed using three-dimensional concentration distribution of groundwater to calculate the volume of fresh, brackish, and saline groundwater below sea level. Based on the volumetric analysis, a quantitative analysis of future seawater intrusion vulnerability was performed using the volume-based vulnerability index adopted from the existing analytical approaches. The result showed that decrease in recharge can exacerbate vulnerability of coastal groundwater resources by inducing broader saline area as well as increasing brackish water volume of unconfined aquifers.

  • Corrosion is of greatest concern for metallic materials exposed to corrosive seawater or aggressive marine atmospheres. Marine structures and components made of metallic materials incur an initial cost and additional large costs for corrosion control and maintenance. There have been worldwide efforts to minimize marine corrosion and extend service life of the materials. It is believed that various factors are associated with corrosion of marine grade metallic materials, particularly the temperature of the solution affecting the corrosion rate by changing dissolved oxygen solubility and concentrations of chloride. In the present study, the electrochemical characteristics of S355ML steel are investigated to identify corrosion acceleration tendencies with changes in solution temperature under marine environments. It was found that increasing seawater temperature, promoted not only activation of chloride ion transfer, but also the formation of porous $Fe(OH)_3$ or $Fe_2O_3$, leading to the acceleration of corrosion.

심정용 해수펌프 개발을 위한 수치해석 연구 (Numerical Study for Development of Submerged Seawater Lift Pump)

  • Seawater lift pump systems are responsible for maintaining open canal levels to provide the suction flow of circulating water pumps at the set point. The objective of this paper is to design a 2-stage mixed flow pump (for seawater lifting), investigate the new impeller modeling method, and performance improvements of the impeller by using a commercial CFD code. The rotating speed of the impeller is 1,750 rpm with a flow rate of 2,700 m3/h. A finite volume method with a structured mesh and realized k-${\varepsilon}$ turbulent model is used to guarantee a more accurate prediction of turbulent flow in the pump impeller. The performance variables such as the static head, brake horsepower, and efficiency of the mixed flow pump are compared based on changes in the impeller blade shape.

A continuous-flow and on-site mesocosm for ocean acidification experiments on benthic organisms

  • Mesocosm experiments conducted for ecological purposes have become increasingly popular because they can provide a holistic understanding of the biological complexities associated with natural systems. This paper describes a new outdoor mesocosm designed for $CO_2$ perturbation experiments of benthos. Manipulated the carbonate chemistry in a continuous flow-through system can be parallelized with diurnal changes, while irradiance, temperature, and nutrients can vary according to the local environment. A target hydrogen ion activity (pH) of seawater was sufficiently stabilized and maintained within 4 h after dilution, which was initiated by the ratio of $CO_2$-saturated seawater to ambient seawater. Specifically, pH and $CO_2$ partial pressure ($pCO_2$) levels gradually varied from 8.05-7.28 and $375-2,691{\mu}atm$, respectively, over a range of dilution ratios. This mesocosm can successfully manipulate the pH and $pCO_2$ of seawater, and it demonstrates suitability for ocean acidification experiments on benthic communities.

은연어의 해수 순치에 따른 스트레스 반응의 변화 (Serum Stress Responses during Seawater Acclimation in Coho Salmon, Oncorhynchus kisutch)

  • Stress response to the seawater acclimation in coho salmon (Oncorhynchus kisutch) smolt was investigated. Salt concentration of rearing water was gradually increased for 3 days from freshwater to seawater (30 ppt salt level). The changes of serum concentrations of cortisol as a primary stress indicator, and as secondary indicators, glucose (GLC), lactate (LAC), triglyce.ide (TG), cholesterol (CHOL), sodium ion $(Na^+)$, chloride ion $(Cl^-)$ and enzyme activities (alanine aminotransferase, ALT: aspartate aminotrasferase, AST; lactate dehydrogenase, LDH) were quantified during the acclimation experiment. Among them, cortisol, LAC, TG, CHOL, ALT, AST concentrations showed rapid increase at the first exposure to the 10ppt salt level (day 1), and began to decrease to the constant values after day 2 of adaptation at 20ppt salt level. However, LDH concentration tended to decrease during the whole experimental period. $Na^+\;and\;Cl^-$ showed slight decrease at day 1, and increased to a little bit higher values after day 2 rather than those in freshwater. All the fishes started on taking a food after day 4 of seawater adaptation. From these results, to reduce osmotic shock inducible stress to fish in seawater acclimation, gradual increase of salt levels is recommended.

바이오플락 및 일반 해수에서 질산염의 넙치 (Paralichthys olivaceus)에 미치는 급성 독성 및 혈액학적 성상의 변화 (Lethal Toxicity and Hematological Changes Exposed to Nitrate in Flatfish, Paralichthys olivaceus in Biofloc and Seawater)

  • 본 연구는 넙치 치어 (평균 체장 $19.8{\pm}2.6cm$, 평균 체중 $19.8{\pm}2.6cm$)를 바이오플락에 각 농도별 0, 62.5, 125, 250, 500, 1,000, $1,500mg\;L^{-1}$의 질산염과 일반 해수에 각 농도별 0, 62.5, 125, 250, 500, $1000mg\;L^{-1}$의 질산염을 96시간 노출시킨 후 생존율 및 혈액학적 변화를 비교하였다. 본 연구에서 넙치 치어의 반수치사농도 ($96h\;LC_{50}$, 96시간 노출 후 총 개체수의 50%가 폐사하는 농도)는 바이오플락 내의 질산염 노출 실험구에서는 $1,226mg\;NO_3L^{-1}$, 해수에서는 $597mg\;NO_3L^{-1}$ (P<0.05)였으며, 이는 넙치 유생이 높은 질산염에 노출되었을 경우, 같은 농도에서도 바이오플락 사육수 보다 일반 해수에서 생물에 독성의 영향이 크게 나타난다고 볼 수 있다. 혈액학적 분석 결과에서는 일반 해수에서 넙치의 hematocrit는 $250mg\;L^{-1}$ 이상의 농도에서 대조구에 비해 통계적으로 유의한 증가를 나타낸 반면, 바이오플락에서 넙치의 hematocrit는 $1,000mg\;L^{-1}$의 질산염 노출에서만 대조구 대비 통계적으로 유의한 증가를 나타냈다 (P<0.05). 그러나, hemoglobin은 일반 해수와 바이오플락 모두 질산염 농도 의존적 변화가 나타나지 않았다. 질산염 노출에 따른 혈장 효소 성분 분석 결과에서는 혈장 AST와 ALT의 경우, 바이오플락과 일반 해수 모두 각 농도별 질산염 노출에 따라 유의하게 증가하였으나, ALP의 경우 두 실험구에서 모두 질산염 농도 증가에 따른 유의한 차이를 나타내지 않았다. 본 연구에서는 바이오플락 환경에서 사육된 넙치가 일반해수에 사육된 넙치에 비해 단기간 질산염 노출에 대한 한계농도 및 혈액학적 성상에서 더 높은 질산염 내성을 가지는 것으로 나타났다. 이는 일반 해수보다 바이오플락 내의 질산염 노출이 상대적으로 생물에게 낮은 독성을 가지며, 이와 같은 효과는 양식에 있어 바이오플락을 이용함으로써 잠재적인 질산염 독성 영향을 경감시켜줄 수 있을 것이다.

틸라피아의 해수순치에 관한 생리학적 연구 I. 내분비학적 변화 (Physiological Studies on Adaptation of Tilapia(Oreochromis miloticus) in the Various Salinities I. Endocrine Changes)

  • This study was taken to examine external changes, behavioral changes, and endocrine changes such as estradiol-17$\beta$, progesterone, T4 and T3 of female Oreochromis niloticus living in 0$\textperthousand$, 10$\textperthousand$, 20$\textperthousand$, and 30$\textperthousand$ salt concentrations, respectively. The results obtained in these experiments were summarized as follows. In seawater obtained in these experiments were summarized as follows. In seawater challenge test, any fish didn't die in each group such as 10$\textperthousand$, 20$\textperthousand$ and 30$\textperthousand$. When fish were adapted from 0$\textperthousand$ to 10$\textperthousand$, 20$\textperthousand$ and 30$\textperthousand$, external body color of fish changed from dark-striped to light-grey color. At the same time, thyroxine and triiodothyronine concentrations significantly(P<0.05) increased, and then were at the highest level in 30 salinity. When fish were adapted from 0$\textperthousand$ to 10$\textperthousand$, feed intake of fish started from the fourth day. From 0$\textperthousand$ to 10$\textperthousand$, 20$\textperthousand$ and 30$\textperthousand$, estradiol-17$\beta$ levels were increased gradually. When fish was adapted from 0$\textperthousand$ to 10$\textperthousand$, 20$\textperthousand$ and 30$\textperthousand$, the levels of each progesterone didn't show significant change, and especially showed the lowest peak in 20$\textperthousand$. The greatest thyroxine activity(T4) was observed in 30$\textperthousand$. The levels of and triiodothyronine(T3) significantly changed in all salinities, and its level was at the highest peak in 30$\textperthousand$ salinity. Correlation coefficients between serum progesterone and triiodothyronine in 10$\textperthousand$ and 30$\textperthousand$ were +0.677 and +0.843, respectively. Correlation coefficient of serum thyroxine(T4) and triiodothyronine(T3) individuals in 10$\textperthousand$ was +0.768, and +0.843, respectively.
