Cyclophosphamide로 유도된 빈혈 흰쥐에서 참당귀 열수추출물이 혈액학적 빈혈지표에 미치는 영향 (Hematological Effect of Water Extracts of Cham-Dang-Gui on Cyclophosphamide Induced Anemic Rat)

  • Anemia, the condition of the diminished concentration of hemoglobin per erythrocyte is common in patients with cancer and is a frequent complication of myelosuppressive chemotherapy. Cham-Dang-Gui (Angelicae Gigantis Radix) has been used in traditional Korean medicine to treat hematologic deficiencies. In this study, Cyclophosphamide (CYP), an alkylating agent that has a broad spectrum of anti-cancer activities, was intraperitoneally injected into the experimental animals to suppress the bone marrow thereafter, causing anemia. The hemopoietic effects of Cham-Dang-Gui were examined using anemic rat model. Rats were divided into five groups: CON (control group), ANS (CYP-injected + normal diet), AND (CYP-injected + normal diet + Cham-Dang-Gui), ALS (CYP-injected + low iron diet), and ALD (CYP-injected + low iron diet + Cham- Dang-Gui) groups. CYP (30 mg/kg) was intraperitoneally injected to rats for 3 days to induce anemic condition. Saline or Cham-Dang-Gui was administrated orally during the entire experimental period. CYP injection decreased body weight gain and food consumption in comparison with CON group. Oral administration of Cham-Dang-Gui extract with normal iron diet significantly prevented the lower body weight gain. The blood level of hemoglobin, iron status (serum iron, transferrin, ferritin and TIBC) and blood level of vitamin B-12 in Cham-Dang-Gui treated groups were significantly higher than those of Cham-Dang-Gui untreated groups regardless of amount of iron in the diet. Taken together, it could be concluded that the Cham-Dang-Gui extract could improve anemic condition induced by CYP injection by improving hematological value, iron status and vitamin B12 status in rats.

참당귀(Angelica gigas)의 DPPH Radical 소거 활성과 항산화 효과 (DPPH Radical Scavenger Activity and Antioxidant Effects of Cham-Dang-Gui (Angelica gigas))

  • 본 연구의 결과를 요약하면 참당귀추출물 1 mg/mL의 농도에서 열수추출물의 free radical소거 효과는 메탄올추출 물보다 높게 나타났으며 농도 의존적으로 DPPH free radical 소거 효과가 상승하는 것을 관찰하였다. 또한 참당귀 투여에 의하여 화학요법으로 발생된 지질과산화물의 생성을 감소시켰으며, 단백질 산화를 유의적으로 억제함으로써 참당귀추출물은 CYP를 투여로 생성된 free radical을 감소시킴으로써 독성을 감소시켜, 혈액학적 빈혈지표가 더 감소되는 것을 막았다고 사료되며, 항산화제로서의 가능성이 있는 한약재로서 활용성이 높을 것으로 사료된다 항산화효소인 SOD 활성은CYP투여에 의하여 활성이 증가하였으며 참당귀추출물의 투여에 의한 효과가 나타나지 않았으나 감소하는 경향을 보였고 catalase 활성은 CYP 투여에 의하여 활성이 증가하였으나 참당귀의 효과는 차이가 없었다. 결론적으로 참당귀추출물은 free radical 소거능을 갖고, 화학요법으로 발생된 지질과산화물의 생성을 낮춰 화학요법에 의한 부작용으로 발생되는 혈액학적 수치의 감소를 막음으로써 항산화제로의 활용성을 기대해본다.

식이로 유도된 철 결핍성 빈혈 흰쥐에서 참 당귀 열수추출물이 혈액학적 빈혈지표에 미치는 영향 (Hematological Effects of Water Extracts of Cham-Dang-Gui on Dietary Induced Iron Deficient Anemia Rat)

  • 본 연구에서는 무철분식이로 철분결핍성 빈혈 유발을 시킨 흰쥐에게 3주간 무철분식이를 섭취시키면서 saline, 참당귀 추출물 및 철분보충제를 투여한 후, 무철분식이를 일반식이로 대체하여 다시 3주간 같은 처리를 한 후 혈액학적 빈혈지표를 조사하여 결과를 요약하면 다음과 같다. 1) 실험동물의 체중은 무철분식이군에서 일반식이군에서 보다 유의적으로 낮게 나타났다. 무철분식이로 인한 저체중은 참당귀 추출물과 철분 보충제에 의해서 회복되지 않았다. 2) 혈액분석 결과 무철분식이는 모든 혈액 빈혈지표를 저하시켰으며, 철분보충제의 투여는 헤모글빈, 헤마토크릿, 평균 혈구 부피, 평균 혈구혈색소, 평균 혈구혈색소 농도, 총철결합능력이 유의적으로 회복시켰다. 무철분식이와 함께 투여한 참당귀 추출물은 혈액지표를 개선시키지 못하였다. 한편, 일반식이와 함께 섭취하였을 경우, 헤모글로빈, 헤마토크릿, 평균 혈구부피, 평균 혈구혈색소, 평균 혈구혈색소 농도, 총철결합능력 모두 유의적인 회복효과를 보였으나 일반식이만을 섭취한 군에서도 같은 회복효과를 보였다. 결론적으로, 장기간에 걸쳐 철분 섭취가 불량할 경우, 체중증가가 지연되며 후에 철분을 보충할 경우에도 체중의 회복은 개선되지 않았다. 철분결핍으로 인한 저체중의 경우, 철분보충제의 복용은 체중을 만회시키지 못하였으나 혈액학적 빈혈 지표들은 회복시키는 것으로 나타났다. 철분결핍 빈혈에서 식이철분이 부족할 경우, 참당귀의 회복효과는 미비한 것으로 나타났다. 향후 철분결핍 빈혈의 치료를 위한 생약제의 다양한 작용을 밝히기 위하여서는 영양학적인 관점에서 체내 이용률에 관한 심도 깊은 연구가 이루어져야 할 것으로 사료된다.

  • A comparative study on 'Dang-gui' and 'Kang-whal' the two species of Angelica growing in Korea was carried out about the morphology and cytology. The somatic chromosome number of Cham dang-gui, Ill dang-gui, Badi-namul was found to be 2N=22. The somatic chromosome number of Kang-whal was found to be 2N=110, that of Chili kang-whal to be 2N=44, that of Mul kang-whal to be 2N=22. Stoma of species of Dang-gui were same in size. Those of Chili kang-whal and Kang-whal were strikingly larger than the other. Chili kang-whal and Kang-whal were verified to be polyploid in the genus Angelica.

Anti-Oral Microbial Effect of Ethanol Extract of Angelica gigas Nakai

  • Background: The Korean name for Angelica gigas Nakai (AGN) is Cham-dang-gui, which grows naturally or is cultivated, and its dried roots are used in traditional herbal medicines. The AGN root exert various pharmacological effects. Despite the various pharmacological effects of the AGN root, there are no reports on its anti-oral microbial effects. The purpose of this study was to reveal the anti-oral microbial effect and the microbial and biochemical changes in oral microorganisms according to the concentration of the ethanol extract of AGN (EAGN) root, and to confirm the possibility of using EAGN as a plant-derived functional substance for controlling oral infectious microorganisms. Methods: Disk diffusion test, growth measurement, biofilm formation assay, and measurements of acid production and buffering capacity were performed to confirm the antibacterial effect of EAGN. Results: EAGN showed anti-oral bacterial effects against Streptococcus mutans and Aggregatibacter actinomycetemcomitans at all concentrations, with S. mutans showing a more susceptible effect at concentrations above 5.0 mg/ml and A. actinomycetemcomitans at 3.75 mg/ml. EAGN treatment significantly reduced A. actinomycetemcomitans growth at all concentrations tested. Biofilm formation was significantly reduced at concentrations above 3.75 mg/ml for S. mutans and 2.5 mg/ml for A. actinomycetemcomitans. Acid production in S. mutans and A. actinomycetemcomitans was significantly increased by treatment with EAGN, and the buffering capacities of S. mutans and A. actinomycetemcomitans increased from an EAGN concentration of 3.75 mg/ml and above. Conclusion: EAGN showed anti-oral bacterial effects against both S. mutans and A. actinomycetemcomitans at concentrations above 3.75 mg/ml, which were thought to be related to the inhibition of their growth and biofilm formation. Therefore, EAGN can be used as a safe functional substance derived from medicinal plants owing to its antibacterial effects against S. mutans and A. actinomycetemcomitans.

신약 개발을 위한 참당귀(Angelica gigas Nakai) 추출 Decursin과 Decursinol Angelate의 약리 작용 (Pharmacological Effect of Decursin and Decursinol Angelate from Angelica gigas Nakai)

  • Traditionally, Cham dang-gui (Angelica gigas Nakai) is one of the most popular herbal medicines in Asian countries including Korea. A. gigas has been used as a functional food product for treatment anemia, women's health care, a sedative, an anodyne or a tonic agent. Decursin and decursinol angelate isolated from the roots of A. gigas are pyranocoumarin compounds. Recently, as the global herbal medication market is increasing, investigations about pharmacological effects of decursin and decursinol angelate are rapidly increasing. We summarized previous studies about pharmacological effects of decursin and decursinol angelate, and reviewed relation with pharmacological effects of decursin and decursinol angelate on human disorder, focused on the approach for new drug development. Pharmacological effects of decursin and decursinol angelate were classified as anti-tumor activity, anti-bacterial activity, improvements of the circulating system, inhibition of cytochrome P-450 activity, anti-inflammation activity, anti-oxidant activity and cognitive-enhancing activites. The activity of A. gigas with improvement of the circulating system may have wide therapeutic potential for circulatory diseases, including diabetes, hyperlipidemia and atherosclerosis. Also, anti-inflammation activity A. gigas may be beneficial for the treatment and prevention of asthma, atopic dermatitis and rheumatism arthritis. This relation could potentially lead to the development of herbal new drugs. In order to development a new drug containing decursin and decursinol angelate, it is also necessary to consider the safety profile, and the information in this review would contribute to development a new drug from herbal medicine.
