Rapid Detection Methods for Food-Borne Pathogens in Dairy Products by Polymerase Chain Reaction (PCR 방법을 이용한 우유 및 유제품에서 발생하는 식중독 균의 신속 검출법)

  • The dairy industry has consistently grown via the expansion of dairy-based food categories. Dairy product consumption is stable since the nutrient composition in dairy products is ideal for human health. However, dairy products are highly susceptible to food-borne pathogens. Controlling the safety of dairy products is thus important when considering the nutrient-rich matrix of this food category. Currently, immunoassays or molecular biology techniques have been used to evaluate the safety of dairy products in Korea. These methods are based on the detection of proteins and thus have low reproducibility and sensitivity. Recent techniques to detect food-borne pathogens have focused on genetic analyses. Rapid detection methods for food-borne pathogens in milk and dairy products using polymerase chain reaction (PCR) techniques, such as conventional PCR, real-time PCR, repetitive sequence-based (rep)-PCR, PCR-denaturing gradient gel electrophoresis (DGGE), and digital PCR, are reviewed in this article. The aim of this review was to contribute knowledge of the relationship between microflora and the quality of dairy products. This study will also assist in the immediate monitoring of food-borne pathogens in milk and dairy products when an outbreak related to this food category occurs.

  • Supply units in each command are multi-tiered and each supply unit keeps a supply level independently, which can cause excess stock, leading false reports to the Logistics Command and increasing difficulties in managing user needs. This causes excess assets and the excess then causes deformation in demand. therefore, the supply support systems of our armed forces have become high-cost/low-efficiency and are insufficient to meet the needs of users in combat units. Civilian corporations and the US Department of Defense are downsizing the aforementioned multi-tiered supply systems thus revolutionizing efficient and effective logistics by adopting Supply Chain Management(SCM), and Prime Vendor policies. Prime Vendor policy is a logistics support method that allows users to directly request and receive supply items from suppliers, based on supply contracts between suppliers and central maintenance organizations like KDA. In other words, it is a system that allows for users to make orders to suppliers directly and suppliers to deliver goods to the users directly, cutting out the middle stage, thus allowing an efficient supply. This is a way forward in finance that cuts costs in net supplies and allows an efficient utilization of civilian assets. which is also known to fasten the speed of logistical support and stripping down the logistical structure. therefore, this report will explore Prime Vendor policies adopted in certain number of units for medical supplies that were taken in consideration of improvements in stock management in civilian organizations and the US Army, and aims to apply such policies for repair parts.

Factors Affecting Corporations Practice of Supply Chain Management: With a Focus on Information Sharing and Performances (기업의 공급사슬관리실행의 영향요인: 정보공유와 성과를 중심으로)

  • Since manufacturing and supplying corporations today come to view their practice of SCM as important, it becomes essential to analyze and control the structural relationship between the information sharing among corporations and the performances resulting from their practice of SCM. It is thus the purpose of this paper to examine the factors that may prompt corporations to implement SCM by means of analyses into the relationship between information sharing and practice of SCM which corporations should lay stress on as well as the relationship between financial and non-financial performances of corporations. The findings of the study can be summed up as follows: First, as for the relationship between information sharing among and practice of SCM by corporations, information sharing among corporations turned out to affect such factors of implementing their SCM as trust, commitment mutual dependence. Consequently, corporations are requested to endeavor to implement SCM itself faithfully if they really aim to achieve their performances by practice of SCM and, at the same time, to make efforts to obtain understanding and support for information sharing among themselves. Second, from the analysis of the relationship between SCM and financial as well as non-financial performances of corporations, it was found that trust, a factor of SCM practice, had influence upon non-financial performances of corporations, but not upon their financial performances, while commitment and mutual dependence affected both financial and non-financial achievements of corporations. Therefore, it was made clear from the analysis that the decision and systematic control of SCM activities which best suit to a corporation play an important role in improving its financial and non-financial performances, because they greatly depend on the implementing extent of SCM factors such as trust, commitment and mutual dependence among corporations.

  • As customers' demands for diversified small-quantity products have been increased, there have been great efforts for a firm to respond to customers' demands flexibly and minimize the cost of inventory at the same time. To achieve that goal, in SCM perspective, many firms have tried to control the inventory efficiently. We present an mathematical model to determine the near optimal (s, S) policy of the supply chain, composed of multi suppliers, a warehouse and multi retailers. (s, S) policy is to order the quantity up to target inventory level when inventory level falls below the reorder point. But it is difficult to analyze inventory level because it is varied with stochastic demand of customers. To reflect stochastic demand of customers in our model, we do the analyses in the following order. First, the analysis of inventory in retailers is done at the mathematical model that we present. Then, the analysis of demand pattern in a warehouse is performed as the inventory of a warehouse is much effected by retailers' order. After that, the analysis of inventory in a warehouse is followed. Finally, the integrated mathematical model is presented. It is not easy to get the solution of the mathematical model, because it includes many stochastic factors. Thus, we get the solutions after the stochastic demand is approximated, then they are verified by the simulations.

A study on prioritizing the application areas for business development of IoT(Internet of Things) (사물 인터넷의 사업화 영역에 대한 우선순위 선정에 관한 연구)

  • This study aims at identifying and prioritizing the application areas for business development of IoT(Internet of Things). In specific, we collected the opinions of the experts on IoT in Korea through the survey based on pairwise comparison, and then we analyzed them using AHP(analytic hierarchy process). As a result, we found that 'Public Safety & Security', 'Automotive & Transportation', and 'Utility & Energy' are the most promising area in realizing IoT-related businesses. Also, we found that the experts had different views according to the positions of their industry in IoT value chain. The analysis showed that the device industry prioritizes 'Public Safety & Security', whereas the network industry does 'Automotive & Transportation'. The experts from the service industry were found to regard 'Utility and Energy' as the most promising commercialization area of IoT.

Development & Assessment of Alkyl Chain Modified Aptamers as Potential PET Radiotracers for Lymphoma Diagnosis

  • The Td05 and Sgc8c, DNA-based aptamers, are well-known to target internalized surface markers (IGHM and PTK7) of Burkitt's lymphoma and acute lymphoblastic leukemia (ALL). Thus, Td05 and Sgc8c labeled with metallic radioisotope 64Cu can be evaluated as potential diagnostic PET imaging agents. In this study, we modified the carbon chain length of the last adenosine of aptamer (n = 3, 6, 12) to increase tumor cell uptake and select the best candidate among six types of aptamer analogues and one adenosine of aptamer. After labeling of 64Cu, [64Cu]Cu-DOTA-aptamer analogues were evaluated in vitro studies (serum stability, Log P values, cell uptake, biodistribution). Then, we evaluate in vivo PET imaging study for two candidates (64Cu-DOTA-C12-Sgc8c, 64Cu-DOTA-C6-Td05). PET images clearly visualize tumors at 24 h post-injection rather than at an early time point and the tumor-to-background ratio also increases at the delay time point. 64Cu-DOTA-C12-Sgc8c and 64Cu-DOTA-C6-Td05 could be used as potential radiotracers for lymphoma.

Comparative Evaluation of Loop-Mediated Isothermal Amplification (LAMP) and Conventional PCR for Detection of Shiga-Toxin-Producing Escherichia coli (STEC) in Various Food Products

  • In this study, polymerase chain reaction (PCR) and loop-mediated isothermal amplification (LAMP) were compared in terms of their ability to detect shiga-toxin-producing Escherichia coli (STEC). Various foods were artificially inoculated with STEC to evaluate the limit of detection (LOD), limit of quantification (LOQ), sensitivity, specificity, and efficiency of PCR and LAMP. The LODs were ≤104 and ≤103 CFU/mL for PCR and LAMP, respectively. The LOQs did not differ between PCR and LAMP. However, of the four considered food types, the sensitivities differed by a maximum of 11.1% for seasoned meat and by a minimum of 8.1% for ground beef. LAMP had higher sensitivity than that of PCR and 100% specificity for all four food types. Therefore, LAMP is a reliable molecular method for detecting STEC as comparable to PCR assay, and its specificity and sensitivity are superior to those of PCR, depending on the food type.

Monitoring of the Source of Gelatin in Dietary Supplement Capsules Sold on the Internet

  • Determining the origin of the components in commercially available gelatin, a purified protein derived mostly from pig skin and bovine tissue, is a challenge, leading to concerns on the grounds of religious beliefs and health. Therefore, regular monitoring of labeling compliance by food control authorities is also necessary. In this study, we monitored the origin of gelatin capsules from 181 commercial dietary supplements that were available for purchase on the internet, using species-specific PCR assays. Fifty five products were labeled correctly, declaring that they used bovine-, fish- and plant-derived gelatin, whereas the other 126 capsules were labeled "gelatin" without specifying the origin. Gelatin in these capsules was obtained from cattle (n = 51), pigs (n = 31), or both (n = 44). Therefore, it is important to declare all of the raw materials used to produce gelatin capsules on the labels to best protect consumers' rights, religious beliefs, and health.

  • Foodborne pathogenic bacteria in various food samples in Korea were monitored and the obtained data was statistically analyzed. A total of 1,240 food samples including 280 sashimi, 244 processed frozen products, 258 kimbab (cooked rice wrapped with seaweed), 337 soybean pastes were obtained from 7 cities including Seoul in Korea. Microorganisms tested were Bacillus cereus, Salmonella spp., Staphylococcus aureus, Escherichia coli, E. coli O157:H7, Vibrio parahaemolyticus, Yersinia enterocolitica, Listeria monocytogenes, Campylobacter jejuni, and Clostridium perfringens. The contaminated microorganisms in food samples were comprised of 10.55% B. cereus, 2.7% S. aureus, 2.0% V. parahaemolyticus, 0.8% C. perfringens, 0.2% Y. enterocolitica, and 0.1% of L. monocytogenes, respectively. Salmonella spp., C. jejuni, and E. coli O157:H7 were not detected in any of the food samples. Particularly, B. cereus that harbors the enterotoxin gene was detected in various foods and regions in Korea, therefore it should be a given special consideration not to allow the hazardous level of contamination.

A Study on the Anchoring Safety Assessment of E-Group Anchorage in Ulsan Port (울산항 E 집단정박지 묘박안전성 평가에 관한 연구)

  • This study suggests the minimum critical external forces based on the assessment of anchoring safety to single anchor situation for representative 8 number of ships in E-group anchorage of Ulsan port. Assessment of anchoring safety is compared holding powers of anchor with external forces of wind, wave and current. Holding powers was reflected materials of seabed, equipment numbers regarding anchor and chain weight, also external forces acting on a hull was calculated considering projected wind area and wetted surface area to the full and ballast conditions respectively. The results of anchoring safety assessments to single anchor showed that the minimum criteria of dragging anchor is a little different from ship's type, size and loading conditions. Bulk carrier can be dragged over the 15m/s of winds and Tanker can be dragged over the 13m/s of winds in case of less than 2knots of currents speed.
