Renal Action of SKP-450, $K^+$Channel Opener, in Dog ( $K^+$ Channel 개방제인 SKP-450의 신장작용)

  • SKP-450 which is $K^{+}$ channel opener, When given into duodenum, exhibited the decline of urine flow accompanied with the decrease of glomerular filtration rates (GFR), renal plasma flow (RPF), N $a^{+}$ and $K^{+}$ excreated in the urine ( $E_{Na}$ , $E_{K}$) and the increase of $K^{+}$N $a^{+}$ratios, and then appeared the significant fall of mean arterial pressure (MAP) and unchanged of reabsorption rates of N $a^{+}$, $K^{+}$ in renal tubules ( $R_{Na}$ , $R_{K}$). SKP- 450 injected into the vein elicited the decline of urine flow along with the reduction of $E_{Na}$ , $E_{K}$, and the increase of $R_{Na}$ , $R_{K}$ and $K^{+}$M $a^{+}$ ratios. SKP-450 administered into a renal artery produced diuretic action along with the increase of $E_{Na}$ , $E_{K}$ and the decrease of $R_{Na}$ , $R_{K}$ in experimental kidney, whereas produced the same aspect to intravenous SKP-450 in the control kidney. Above results suggest that SKP-450 possess both diuretic action in the kidney and central antidiuretic action in dog.tic action in dog.tion in dog.

Effect of Methoxyverapamil on Renal Function of Dogs (개의 신장기능에 미치는 메톡시베라파밀의 영향)

  • Methoxyverapamil, $Ca^{2+}$ channel blocker, when given intravenously by means of bolus, produced the transient increase of urine flow, and then methoxyverapamil was infused in this experiments. Methoxyverapamil, when infused into vein, elicited the increase of urine flow ancampanied with the increased glomeralar filtration rate(GFR), renal plasma flow(RPF), excretion amounts of sodium and potassium in urine($E_{Na},\;E_k$) and osmolar clearance(Cosm), wherease produced the no change of free water clearance($C_{H2O}$) and the reduction of reabsorption rates of sodium and potassium in reral tubules($R_{Na},\;R_k$). Methoxyverapamil, when infused into a renal artery, exhibited the diuretic action in only infused Kidney, at this time changes of renal function were the same aspect to that of intravenously infused methoxyverapamil. Methoxyverapamil, when infused into a carotid artery, exhibited the decreased urine flow along with the reduction of Cosm, $C_{H2O}\;and\;E_{Na}$. Above results suggest that methoxyverapamil possess both the diuretic action by direct action in kidney and antidiuretic action through the central function.

  • The effect of Cimicifuga heracleifolia on CC1$_4$-induced hepatotoxicity was investigated. Cimicifuga heracleifolia has been used to diaphoresis, antipyretics and detoxification in oriental remedy. We examined the effect of Cimicifuga heracleifolia methanol extract by blood chemical analysis and histopathologic examination. ALT and AST were decreased by 38% and 67% in pretreatment group of Cimicifuga heracleifolia compared to CC1$_4$control group respectively. There were significant changes neither in total protein, albumin, triglyceride and cholesterol nor in BUN and creatinine. In histopathologic examination, there were severe necrosis and hemorrhage with infiltration of inflammatory cells around the central vein, zone 3 in the liver of CC1$_4$ treated rat. Ballooning degeneration of hepatocytes were frequently noted in the periphery of the hemorrhagic necrosis. In Cimicifuga heracleifolia pretreatment group, we observed mild degree of ballooning degeneration and inflammatory cell infiltration. No gross necrosis was observed. We measured malondialdehyde (MDA) formation by TBA method. It showed that the formation of MDA in Cimicifuga heracleifolia pretreatment group was decreased compared to the $CCl_4$control group. We got the result of the effect of Cimicifuga heracleifolia on CC1$_4$-induced hepatotoxicity by decreasing serum ALT and AST. It seems that the decrease of lipid peroxidation is related to the recovery effect.

Effect of Toluene Application to Skin on the Liver Injury in Rats

  • To investigate an effect of the topical toluene application to .at skin on the liver injury, toluene (35 mg/$cm^2$) was sequentially applied for 3 or 5 days to rat skin and then the animals were sacrificed. 5 day toluene-treated rats showed the slight increase of live. weight per body weight(%) compared with control. Serum levels of xanthine oxidase and alanine aminotransferase activity were significantly increased both in 3 days and 5 days toluene-treated animals compared with control. In the histopathological findings, cytoplasmic degeneration of hepatocytes around the central vein was noted in the liver of rats applied with toluene to the skin. These results indicate toluene application to rat skin feds to somewhat slight liver injury. On the other hand, the hepatic benzylalcohol or aldehyde dehydrogenase activities were significantly decreased by toluene application to rat skin. In conclusion, the liver min was induced by toluene application to rat skin, and it can be hypothesized that accumulation of benzaldehyde in liver cell may be responsible for liver injury.

  • When endothelial cells isolated isolated from human umbilical venis were cultred for 6dary using 96-well microplates, the final cell density in each was fiund not to be the same although the medium composition of each well was exactly the same. Cell growth in the wells located at the periphery of a microplate was low, while that in the central area of the plate was high. A possible cause for different rate of growth was proposed as the uneven concentration of oxygen in the culture medium of each well.

Renal Action of TNPA, a Dopamine $D_2$Receptor Agonist, in Dog (Dopamine $D_2$Receptor 효능제인 TNPA의 신장작용)

  • The dopaminergic receptors were consisted of two distinct subtypes, $D_1$and $D_2$, each having different function. The present study was attempted to investigate the effects of R(-)-2,10,11-trihydroxy-N-n-propylnoraporphine (TNPA), a dopamine $D_2$receptor agonist, on renal function in dog. TNPA (5.0~15.0 $\mu$g/kg), when given into the vein, produced a dose-dependently antidiuresis along with the decrease in osmolar clearance ( $C_{osm}$) and urinary excretion of sodium and potassium ( $E_{Na}$ , and $E_{K}$). It also increased reabsorption rates of sodium and potassium in renal tubules ( $R_{Na}$ , $R_{K}$) without any changes in glomerular filtration rate (GFR), renal plasma flow (RPF) and free water clearance ( $C_{H2o}$). TNPA (0.5~1.5 $\mu$g/kg/min) infused into a renal artery decreased urine flow both in the experimental and the control kidneys. TNPA (1.5~5.0 $\mu$g/kg) administered via the carotid artery also greatly exhibited antidiuresis even at intravenously ineffective doses. Changes of renal function by TNPA given into both the renal artery and the carotid artery were almost the same aspect to those induced by intravenous TNPA. These results obtained from the present study suggest that TNPA produces antidiuresis by increasing the reabsorption rates of electrolytes in renal tubules, mainly distal tubule, through changing of central function.unction.

  • Glibenclamide(GLY)(1.0 and 3.0 mg/kg), an ATP-dependent $K^+$ channel blocker, when given into the vein in dogs, produced the diuretic action accompanied with the increase of osmolar clearance($C_{osm}$), urinary excretion of $Na^+$ and $K^+$ ($E_{Na}$, $E_K$), and with the decrease in reabsorption rates for $Na^+$ and $K^+$ in renal tubules ($R_{Na}$, $R_K$), and then ratios of $K^+$ against $Na^+$($K^+$/$Na^+$) were decreased. GLY did not affect mean arterial pressure at any doses used. At a low dose(0.1 mg/kg), GLY injected into a renal artery brought about the diurectic action in both experimental and control kidney, however at a higher dose(0.3 mg/kg), GLY appeared significant diuretic action in the control kidney, but not in experimental kidney and the decrease of glomerular filtration rates(GFR), renal plasma flow(RPF), $E_K$, and the increase in $E_{Na}$. In the control kidney, these changes in renal function exhibited the same aspect as shown in intravenous experiments. In experiments given into carotid artery of GLY(0.5 and 1.5 mg/kg), changes in all renal function included the increase in urine volume were the same pattern as shown in intravenous experiments. The above results suggest that glibenclamide produces diuretic action through central function and the action site of the GLY in kidney is the renal distal tubules in dogs.

Effect of Ganoderma Lucidum on the Liver Function and Lipid Metabolism in Alcohol-Consuming Rats (영지가 알코올 섭취한 흰쥐의 간기능 및 지질대사에 미치는 영향)

  • The effects of Ganoderma Lucidum consumption (GL) on the liver function and lipid metabolic disorder induced by alcohol consuming were studied in rats using GL powder(1%, 2%, 4%) and 10% ethly alcohol. Thirty-five rats(Sprague-Dawley, male) were divided into five groups and fed experimental diets for seven weeks. The concentrations of serum cholesterol, HDL-cholesterol and triglyceride were the highest in the alcohol group, while the levels were apparently reduced by in groups fed GL. The atherogenic indices(AI) also tended to be higher in the alcohol group. Glutamic oxaloacetic transaminase(GOT) activities were significantly lower in body alcohol + 2% GL and alcohol + 4% GL groups, and glutamic pyruvic transaminase(GPT)activities lower only in the alcohol + 2% GL compared with the alcohol group. Alkaline phosphatase(ALP) activities were not significantly different among the groups. The elevated levels of liver cholesterol and triglyceride due to alcohol consumption, were apparently reduced by GL supplementation. On microscopic observation of liver tissues, fat droplets appeared extensively on the liver-lobule in the alcohol group, while fat droplets appeared only on the central vein in the alcohol + 1% GL group. However, in groups administered 2% and 4% GL, fat droplets appeared similar to the alcohol group. The concentrations of fecal bile acid were significantly increased in groups fed GL. These results indicate that GL exerted some beneficial effects on lipid metabolic disorder caused by long-term alcohol consumption. Thus, it seems that GL may be effective in preventing or curing some aspects of alcohol toxicity. More detailed studies are needed in order to determine proper dietary levels of GL for combating alcohol toxicity.

Hypereosinophilic Syndrome: CT Findings in Patients with Hepatic Lobar or Segmental Involvement

  • Objective: The purpose of this study was to describe the CT findings of hepatic hypereosinophilic syndrome in which hepatic lobes or segments were involved. Materials and Methods: Seven patients with hypereosinophilic syndrome with hepatic lobar or segmental involvement were included in our study. In all seven, diagnosis was based on liver biopsy and the results of corticosteroid treatment. CT findings were retrospectively reviewed by three radiologists, who reached a consensus. Biopsy specimens were examined, with special reference to portal and periportal inflammation. Results: CT demonstrated well-defined, homogeneous or heterogeneous low attenuation with a straight margin limited to a hepatic lobe (n = 2), segments (n = 3), or subsegments (n = 2), particularly during the portal phase. Where there was subsegmental involvement, lesions were multiple, ovoid or wedge-shaped, and showed low attenuation. In two patients with lobar or segmental involvement, segmental portal vein narrowing was observed. Histopathologic examination disclosed eosinophilic infiltration in the periportal area, sinusoids and central veins, as well as portal phlebitis. Conclusion: Hypereosinophilic syndrome may involve the presence of hepatic lobar, segmental, or subsegmental low-attenuated lesions, as seen on CT images. Their presence may be related to damage of the liver parenchyma and to portal phlebitis.

Pathological study on experimental hemorrhagic and hemolytic anemia in Nile tilapia, Oreochromis niloticus (나일틸라피아(Oreochromis niloticus)의 실험적 출혈성과 용혈성 빈혈의 병리학적 연구)

  • This study was conducted to know the differences in pathology between artificially induced hemorrhagic anemia (EHA) and hemolytic anemia (ELA) during the anemic and recovery course, using Nile tilapia (Oreochromis niloticus). EHA were induced by repeated bleedings with a volume of about 1% of body weight through caudal vein. ELA were induced by intraperitoneal injections of 1% phenylhydrazine for 2 times. A period of 16 to 49th day was arbitrarily taken as a recovery phase. There were no prominent clinical signs and gross findings except for pale gills during the anemic state of both. Characteristic erythrocytes with a weekly stained cytoplasm started appearing on the 12th day in both and were still noted on the 49 and 20th day respectively in EHA and ELA. EHA and ELA were normocytic hypochromic and macrocytic normochromic in type respectively, although both were normocytic hypochromic during the recovery phase. In liver, fatty degeneration around central vein on the 12-38th day in EHA and hyaline degeneration around central vein on the 12-26th day in ELA were found. In head kidney, increased hemopoiesis was observed on the 12-26th day in EHA and on the 2-12th day in ELA, and macrophages engulfing erythrocytes were observed on the 16-38th day in EHA and on the 2-12th days in ELA. In spleen, activated ellipsoids on the 12-26th day in EHA. and on the 2-20th day in ELA. In ELA, severe accumulation of hemosiderin in both spleen and head kidney were constantly noted from the 2-49th day. On the 49th, Ht was recovered but Hb was still lower than that of control in both anemia.
