Development of Detection Methods for Cellulolytic Activity of Auricularia auriculajudae

  • To obtain basic information on the detection of cellulolytic activity in Auricularia auricula-judae, the influences of dye reagent, pH, and temperature were assessed. Chromogenic dye (congo red, phenol red, remazol brilliant blue, and trypan blue) was individually incorporated into a medium containing either carboxymethyl-cellulose, Avicel, or D-cellobiose as a polysaccharide carbon substrate. The other assessments utilized pHs ranging from 4.5 to 8.0 and temperatures from $15\sim35^{\circ}C$. Overall, when A. auricula-judae species were transferred onto media contained Congo red and adjusted pH 7.0 and then incubated at $25^{\circ}C$ for 5 days, the clear zone indicative of cellulolytic activity was more pronounced.

Selection of KYC 3270, a Cellulolytic Myxobacteria of Sorangium cellulosum, against Several Phytopathogens and a Potential Biocontrol Agent against Gray Mold in Stored Fruit

  • During 2002-2008 in Korea, 455 extracts from myxobacteria consisting of 318 cellulolytic and 137 bacteriolytic myxobacteria were isolated, which were then screened for antifungal activity against the phytopathogens Botrytis cinerea, Colletotrichum acutatum, Penicillium sp., Pyricularia grisea, and Phytophthora capsici. 204 isolates had antifungal activity, causing both a clear zone due to blocked spore germination and inhibition of mycelial growth; most (199) were from cellulolytic (Sorangium cellulosum) and only five were from bacteriolytic myxobacteria. B. cinerea, the best controlled among the five tested pathogens, had a unique group of antifungal isolates of myxobacterial extracts compared to the other pathogens' groups. Among seventy-nine bioactive myxobacteria, four isolates, KYC 3130, KYC 3247, KYC 3248 and KYC 3270, were selected and all were cellulolytic. Liquid culture filtrates of these four myxobacteria were applied to tomato, cherry tomato, strawberry, and kiwi fruits 5 h before inoculation with gray mold conidia; then the treated fruits were placed in an airtight container and the experiment was repeated six to eight times. Incidence (%) of gray mold on fruit of the infected control treatment was 84-98%, whereas it was only 5-21% after the KYC 3270 treatment. After KYC 3270 treatment of the four fruits, mold control was 79-95%, which was highest among the filtrates and statistically the same as treatment with fludioxonil, a registered chemical against gray mold of stored fruits.

Detection of Cellulolytic Activity in Ophiostoma and Leptographium species by Chromogenic Reaction

  • To understand the ability of producing cellulolytic enzyme activity in the sapstaining fungi, four species of Ophiostoma and two species of Leptographium were investigated in the culture media containing each of cellulose substrates such as CM-cellulose, Avicel and D-cellobiose and each of chromogenic dyes such as Congo-Red, Phenol Red, Remazol Brilliant Blue and Tryphan Blue. When the fungi were grown for $5{\sim}7$ days at $25^{\circ}C$, the formation of clear zone by chromogenic reaction around the margin of the fungal colony was demonstrated in all the culture media Congo-Red containing CM-cellulose. There was difference in the formation of clear zone among the dyes. Only Ophiostoma setosum and Leptographium spp. showed cellulolytic activity to the three substrates. Overall, the results of this study show that ophiostomatoid sapstaining fungi can produce cellulolytic enzymes.

Improvement of Cellulolytic Activity of Pleurotus florida through Radiation Mutagenesis

  • A mushroom mutant with increased cellulolytic activity was developed through radiation mutagenesis. The homogenized hypha suspension of Pleurotus florida was exposed to gamma radiation ($^{60}Co$, AECL) at the dose of $LD_{99}$ (0.51 kGy, $D_{10}$; 0.26 kGy). Among 16 mutants, Pf CM4 showed 17.24% more cellulolytic activity than the wild type (p<0.05). It was observed that Pf CM4 can utilize all kinds of carbon sources tested for their mycelia growth. Starch, xylan, and glucose favourably supported the radial mycelia extension. Yeast extract and $NH_4NO_3$ have been recorded as the best organic and inorganic nitrogen sources, respectively. Pf CM4 was found to grow significantly faster, even at high temperature ($30^{\circ}C$), than wild type (p<0.05), and the optimal pH was 5.5~6.5. This study reveals that the mutant Pf CM4 could be employed for the effective recycling of cellulosic wastes, in addition to mushroom farming.

  • 토양으로부터 carboxymethyl cellulase 및 intra-cellular $\beta$-glucosidase생산능력이 높은 방선균 No. 109균주를 분리하고 이 균주의 배양상 내지는 형태적 특성, 생리적 특성 그리고 세포벽 구성성분 등을 조사, 균동정을 행한 결과 No. 109 분리균은 Streptomyces tanashiensis 또는 그 유연균으로 동정되었다.

  • In order to develop a high cellulolytic direct-fed microorganism (DFM) for ruminant productivity improvement, this study isolated cellulolytic bacteria from the rumen of Holstein dairy cows, and compared their cellulolytic abilities via DM degradability, gas production and cellulolytic enzyme activities. Twenty six bacteria were isolated from colonies grown in Dehority's artificial (DA) medium with 2% agar and cultured in DA medium containing filter paper at $39^{\circ}C$ for 24h. 16s rDNA gene sequencing of four strains from isolated bacteria showed that H8, H20 and H25 strains identified as Ruminococcus flavefaciens, and H23 strain identified as Fibrobacter succinogenes. H20 strain had higher degradability of filter paper compared with others during the incubation. H8 (R. flavefaciens), H20 (R. flavefaciens), H23 (F. succinogenes), H25 (R. flavefaciens) and RF (R. flavefaciens sijpesteijn, ATCC 19208) were cultured in DA medium with filter paper as a single carbon source for 0, 1, 2, 3, 4 and 6 days without shaking at $39^{\circ}C$, respectively. Dry matter degradability rates of H20, H23 and H25 were relatively higher than those of H8 and RF since 2 d incubation. The cumulative gas production of isolated cellulolytic bacteria increased with incubation time. At every incubation time, the gas production was highest in H20 strain. The activities of carboxymethylcellulase (CMCase) and Avicelase in the culture supernatant were significantly higher in H20 strain compared with others at every incubation time (p<0.05). Therefore, although further researches are required, the present results suggest that H20 strain could be a candidate of DFM in animal feed due to high cellulolytic ability.

Optimal Medium Conditions for the Detection of Cellulolytic Activity in Ganoderma lucidum

  • To determine the optimal medium conditions for the detection of the cellulolytic activity in Ganoderma lucidum, we varied three media conditions: dye reagent, pH, and temperature. First, we evaluated the use of four dyes, Congo Red, Phenol Red, Remazol Brilliant Blue, and Trypan Blue. To observe the effect of pH on the chromogenic reaction, we also made and tested various media spanning acidic and alkaline pHs, ranging from 4.5 to 8.0. Furthermore, in order to research the effect of temperature on the clear zone and the fungus growing zone, we tested temperatures ranging from 15 to $35{^{\circ}C}$. On the whole, the best protocol called for Ganoderma lucidum transfer onto media containing Congo red with pH adjusted to 7.0, followed by incubation at $25{^{\circ}C}$ for 5 days. Our results will be useful to researchers who aim to study extracellular enzyme activity in Ganoderma lucidum.

  • Alkaline cellulolytic enzymes are prepared from Coprinus cinereus 2249. Recovery method of enzyme protein from cultured medium and effect factors on enzyme activity of protein were investigated. The results could summarized as follows, \circled1 Amount of enzyme protein from cultured medium was highest in incubation with shaking and addition of skim milk. \circled2 Protein from cultured medium was alkaline enzymatic protein which shows the highest activity at pH 9.0. \circled3 The most effective recovery method of enzyme protein was the precipitation of protein by addition of cultured medium of protein in ethanol. \circled4 The enzyme activity was enhanced by tween-80 and decreased with $Al_2(SO_4)_3$, $H_2O_2$et al, and was little changed with metal ions except $Hg^{++}$.

전통식품 유래 유산균의 해조류 발효 및 Probiotic 특성 (Seaweed Fermentation and Probiotic Properties of Lactic Acid Bacteria Isolated from Korean Traditional Foods)

  • 해조류의 발효가 가능하고 probiotic 특성이 우수한 유산균을 분리 선발한 후 이들의 미역과 다시마 발효능을 검토하였다. 미역 및 다시마 발효가 가능한 균주를 김치 젓갈, 된장으로부터 331 균주를 순수 분리하여 해조류 구성 다당(alginate, cellulose) 분해능, 균의 생육, 항균 활성 등을 비교 검토한 결과 4균주(stain No. 162, 164, 192, 196)가 우수하였다. 선발 균주 모두 인공위액, 인공담즙액, NaCl에 높은 생존율을 나타내었고 이들 4균주 중 No. 192가 가장 우수하였으며 Enterococcus faecium으로 동정되었다. 미역과 다시마를 이용하여 선발 유산균을 배양한 결과 No. 192 균주가 발효특성이 가장 양호하였으며, No. 162, 164, 196 균주도 양호하였다. 선발 유산균을 이용한 미역과 다시마에서 성장이 가능하였으며 발효 후 미역과 다시마 발효물의 항산화 활성이 증진되었다.

Cellulose Utilization and Protein Productivity of Some Cellulolytic Fungal Co-cultures

  • Protein productivity by the cellulolytic fungi, Trichoderma viride(MTCC 800), Chaetomium globosum and Aspergillus terreus was compared in co-culture and mixed culture fermentations of cashewnut bran. Co-cultures were more effective in substrate saccharification, which ranged between $85{\sim}88%$ compared to the $62{\sim}67%$ saccharification shown by the monocultures. Maximum saccharification was induced by T. viride and C. globosum co-culture resulting in the highest 34% release of reducing sugars. The maximum 16.4% biomass protein and the highest protein productivity(0.58%) were shown by T. viride and A. terreus co-culture. A. terreus performed better in co-culture in the presence of T. viride rather than with C. globosum. Among the cellulolytic enzymes, FPase(Filter Paper Cellulase) activity was significantly higher in all the co-cultures and in the mixed culture than in their respective monocultures. Mixed culture fermentation involving all the three fungi was not effective in increasing the per cent saccharification or the biomass protein content over the co-cultures.
