Lipids, which along with carbohydrates and proteins are among the most important nutrients for the living organism, have a variety of biological functions that can be applied widely in biomedicine. A fatty acid, the most fundamental biological lipid, may be classified by length of its aliphatic chain, and the short-, medium-, and long-chain fatty acids and each have distinct biological activities with therapeutic relevance. For example, short-chain fatty acids have immune regulatory activities and could be useful against autoimmune disease; medium-chain fatty acids generate ketogenic metabolites and may be used to control seizure; and some metabolites oxidized from long-chain fatty acids could be used to treat metabolic disorders. Glycerolipids play important roles in pathological environments, such as those of cancers or metabolic disorders, and thus are regarded as a potential therapeutic target. Phospholipids represent the main building unit of the plasma membrane of cells, and play key roles in cellular signaling. Due to their physical properties, glycerophospholipids are frequently used as pharmaceutical ingredients, in addition to being potential novel drug targets for treating disease. Sphingolipids, which comprise another component of the plasma membrane, have their own distinct biological functions and have been investigated in nanotechnological applications such as drug delivery systems. Saccharolipids, which are derived from bacteria, have endotoxin effects that stimulate the immune system. Chemically modified saccharolipids might be useful for cancer immunotherapy or as vaccine adjuvants. This review will address the important biological function of several key lipids and offer critical insights into their potential therapeutic applications.
MicroRNAs (miRNAs) are a class of small non-coding RNAs that regulate the expression of target messenger RNA (mRNA) complementary to the 3' untranslated region (UTR) at the post-transcriptional level. Hsa-miR-422a, which is commonly known as miRNA derived from transposable element (MDTE), was derived from short interspersed nuclear element (SINE). Through expression analysis, hsa-miR-422a was found to be highly expressed in both the small intestine and liver of crab-eating monkey. AT-Rich Interaction Domain 5 B (ARID5B) was selected as the target gene of hsa-miR-422a, which has two binding sites in both the exon and 3'UTR of ARID5B. To identify the interaction between hsa-miR-422a and ARID5B, a dual luciferase assay was conducted in HepG2 cell line. The luciferase activity of cells treated with the hsa-miR-422a mimic was upregulated and inversely downregulated when both the hsa-miR-422a mimic and inhibitor were administered. Nuclear factor erythroid-2 (NF-E2) was selected as the core transcription factor (TF) via feed forward loop analysis. The luciferase expression was downregulated when both the hsa-miR-422a mimic and siRNA of NF-E2 were treated, compared to the treatment of the hsa-miR-422a mimic alone. The present study suggests that hsa-miR-422a derived from SINE could bind to the exon region as well as the 3'UTR of ARID5B. Additionally, hsa-miR-422a was found to share binding sites in ARID5B with several TFs, including NF-E2. The hsa-miR-422a might thus interact with TF to regulate the expression of ARID5B, as demonstrated experimentally. Altogether, hsa-miR-422a acts as a super enhancer miRNA of ARID5B by collaborating with TF and NF-E2.
Thioredoxin reductase (TrxR) 시스템은 세포 내 산화 환원 반응의 항상성 유지와 신호 전달 경로를 조절하는데 중추적인 역할을 함으로써 세포의 생존과 기능 유지에 필수적이다. TrxR 시스템은 thioredoxin (Trx), TrxR 및 nicotinamide adenine dinucleotide phosphate의 구성요소를 포함하며, TrxR 효소의 촉매 반응에 의해 환원된 Trx는 하위 표적 단백질을 환원시켜 결과적으로 산화적 스트레스에 대한 방어와 세포 분화, 성장 및 사멸을 조절한다. 암세포는 무한한 세포 증식과 높은 대사율로 인해 과도하게 생성된 활성산소종을 소거하기 위해 세포 내 항산화능을 향상시켜 세포의 생존을 유지하는 반면, 항산화 시스템에 대한 의존도 및 민감도가 높아 이를 표적으로 한 항암 활성 연구에서 잠재적인 가능성이 있음을 제시한다. 여러 연구 결과에서 TrxR이 다양한 유형의 암에서 높은 수준으로 발현되고 있음이 밝혀졌고, 또한 TrxR 시스템을 표적으로 한 항암 활성에 대한 연구가 증가하고 있다. 따라서 본 총설에서는 세포 내 TrxR 시스템의 기능과 암의 발달 및 진행에서의 역할을 다루고, TrxR 억제제의 항암 활성 및 기전을 검토함으로써 항암 활성 연구에 대한 전략으로 TrxR 시스템의 타당성과 가치를 논하였다.
곤충에서 유래한 항균펩타이드 CopA3는 다양한 세포사멸 과정을 차단한다고 알려져 있다. 세균 톡신에 의한 상피세포 세포사멸이나 6-hydroxy dopamine이 야기하는 신경세포 세포사멸 모두를 차단한다. 연구자 등은 최근에 CopA3가 카스파제에 직접 결합하여 그들의 활성형-절단과정을 차단한다고 보고하였다. 하지만 강력한 CopA3의 항세포사멸 효능을 설명하기 위해서는 추가적인 규명이 필요한 실정이다. 본 연구에서는 세포사멸경로의 핵심억제인자인 survivin 발현에 미치는 CopA3의 영향을 확인하였다. 인간 대장상피세포(HT29)에 CopA3를 처리한 뒤 survivin 발현을 추적한 결과, survivin 단백질 양이 유의하게 증가함을 확인하였다. RT-PCR을 통해서 CopA3가 survivin 유전자의 전사를 증가시킴을 확인하였다. 그리고 CopA3 자극이 Sp1 발현을 증가시키는 사실과, Sp1 억제 물질인 tolfenamic acid 처리가 CopA3에 의한 survivin 증가를 차단한다는 결과들을 바탕으로 우리는 CopA3가 Sp1을 통해 survivin 발현을 유도한다는 최종 결론을 도출하였다. 한편 본 연구를 통해서 CopA3의 강력한 항세포사멸 효능을 설명할 수 있는 분자기작을 새롭게 제시하였다고 사려된다.
면역 체계와 대사 체계는 항상성을 유지하는데 중요한 요소이다. 면역 반응과 대사 조절은 연관성이 높아, 정상적인 대사가 교란되면 대사 질환이 발생하며, 면역 반응에도 변화가 발생하였다. 마찬가지로, 비만은 면역 반응과 높은 관련이 있다. 에너지 대사의 불균형으로 발생하는 비만은 인슐린 저항성, 제2형 당뇨병, 지방간 질환, 동맥경화증, 고혈압 등의 대사 질환과 관련이 있다. 알려진 바로는, 비만은 낮은 수준의 염증이 만성화된 상태가 특징이다. 비만 환경에서, 면역세포의 미세 환경은 대식세포, 자연살해세포, T세포 같은 면역세포의 독특한 활성화 표현형에 의해 염증성이 되었다. 또한, 면역 세포는 세포 간의 기전, 사이토카인을 매개하는 기전을 통해 상호작용하여 비만으로 인한 염증 반응을 강화한다. 이러한 현상은 기존의 췌장 리파아제나 알파-아밀라아제 같은 체내 효소의 억제나 지방전구세포의 분화를 억제를 표적으로 하는 일반적인 비만의 약리학적 치료 외에 면역세포 활성화 조절을 표적으로 하는 비만의 약리학적 치료 전략을 시사한다. 본 논문에서는 대식세포, 자연살해세포, T세포의 활성화 표현형과 비만 환경 내이들의 양상에 대해 정리하였다. 또한, 본 논문에서는 현재까지 확인된 면역세포의 활성화 조절을 통한 비만을 완화하는 약리학적 물질에 대해서 정리하였다.
산업용 IoT는 대규모 연결을 통해 데이터 수집, 교환, 분석과 함께 산업 분야의 생산 효율성 개선에 중요한 요소이다. 그러나 최근 산업용 IoT의 확산으로 인해 트래픽이 폭발적으로 증가함에 따라 트래픽을 효율적으로 처리해줄 할당 기법이 필요하다. 본 논문에서는 산업용 IoT 환경에서 성공적인 태스크 처리율을 높이기 위한 2단계 태스크 오프로딩 결정 기법을 제안한다. 또한, 컴퓨팅 집약적인 태스크를 셀룰러 링크를 통해 이동 엣지 컴퓨팅(Mobile Edge Computing: MEC) 서버로 오프로드 하거나 D2D(Device to Device) 링크를 통해 근처의 산업용 IoT 장치로 오프로드 할 수 있는 하이브리드 오프로딩(Hybrid-offloading) 시스템을 고려한다. 먼저 1단계는 태스크 오프로딩에 참여하는 기기들이 이기적으로 행동하여 태스크 처리율 향상에 어려움을 주는 것을 방지하기 위해 인센티브 메커니즘을 설계한다. 메커니즘 디자인 중 McAfee's 메커니즘을 사용하여 태스크를 처리해주는 기기들의 이기적인 행동을 제어하고 전체 시스템 처리율을 높일 수 있도록 한다. 그 후 2단계에서는 산업용 IoT 장치의 불규칙한 움직임을 고려하여 비정상성(Non-stationary) 환경에서 멀티 암드 밴딧(Multi-Armed Bandit: MAB) 기반 태스크 오프로딩 결정 기법을 제안한다. 실험 결과로 제안된 기법이 기존의 다른 기법에 비해 전체 시스템 처리율, 통신 실패율, 후회 측면에서 더 나은 성능을 달성할 수 있음을 보인다.

Background and Objectives: This study aims to clarify the systems underlying regulation and regulatory roles of hydrogen combined with 5-Aza in the myogenic differentiation of adipose mesenchymal stem cells (ADSCs). Methods and Results: In this study, ADSCs acted as an in vitro myogenic differentiating mode. First, the Alamar blue Staining and mitochondrial tracer technique were used to verify whether hydrogen combined with 5-Aza could promote cell proliferation. In addition, this study assessed myogenic differentiating markers (e.g., Myogenin, Mhc and Myod protein expressions) based on the Western blotting assay, analysis on cellular morphological characteristics (e.g., Myotube number, length, diameter and maturation index), RT-PCR (Myod, Myogenin and Mhc mRNA expression) and Immunofluorescence analysis (Desmin, Myosin and 𝛽-actin protein expression). Finally, to verify the mechanism of myogenic differentiation of hydrogen-bound 5-Aza, we performed bioinformatics analysis and Western blot to detect the expression of p-P38 protein. Hydrogen combined with 5-Aza significantly enhanced the proliferation and myogenic differentiation of ADSCs in vitro by increasing the number of single-cell mitochondria and upregulating the expression of myogenic biomarkers such as Myod, Mhc and myotube formation. The expressions of p-P38 was up-regulated by hydrogen combined with 5-Aza. The differentiating ability was suppressed when the cells were cultivated in combination with SB203580 (p38 MAPK signal pathway inhibitor). Conclusions: Hydrogen alleviates the cytotoxicity of 5-Aza and synergistically promotes the myogenic differentiation capacity of adipose stem cells via the p38 MAPK pathway. Thus, the mentioned results present insights into myogenic differentiation and are likely to generate one potential alternative strategy for skeletal muscle related diseases.
도처에 분포하는 peroxiredoxins (Prxs)은 세포 내 방어신호전달 과정에서 다양한 기능을 하는 것으로 나타났다. Prxs는 크게 typical 2-Cys Prx, atypical 2-Cys Prx와 1-Cys Prx의 세 부류로 분류되는데, 이것들은 cysteine 잔기의 수와 촉매기전에 따라 구분된다. 세 종류의 단백질 중, N-말단에 peroxidatic cysteine 잔기를 포함하는 typical 2-Cys Prx는 $H_2O_2$ 분해과정 동안 과산화물-의존적인 sulfenic acid로의 산화와 thiol-의존적 환원과정이 순환되어 일어난다. Sulfenic acid는 고농도의 $H_2O_2$와 Trx, Trx reductase와 NADPH를 포함하는 촉매 요소의 존재하에 cysteine sulfenic acid로 과산화 될 수 있다 과산화된 2-Cys Prx는 ATP 의존성 효소인 sulfiredoxin의 작용에 의해 천천히 환원된다. 세포가 강력한 산화나 열 충격 스트레스에 노출되면, 2-Cys Prx는 LMW 단백질에서 HMW complex로 구조를 변화시켜 peroxidase에서 chaperone으로 기능의 전환을 일으킨다. 2-Cys Prx의 C-말단 부분 역시 이러한 구조적 전환에 중요한 역할을 한다. 따라서, C-말단이 잘려진 단백질은 과산화가 되지 않고 단백질의 구조와 기능이 조절될 수 없다. 이러한 반응들은 활성 자리인 peroxidatic cysteine 잔기에 의해 일차적으로 유도되며, 그것은 세포에서 '$H_2O_2$ sensor' 로서 작용하다. 2-Cys Prx의 가역적인 구조와 기능 변화는 세포가 외부자극에 적응하는 수단으로 작용하며, 아마도 세포내 방어신호체계를 활성화 시키는 것으로 생각된다. 특히, chloroplast에 존재하는 식물 2-Cys Prx는 촉매반응 동안 주된 구조적인 변화를 나타내는 역동적인 단백질 구조를 가지고 있어서, 산화-환원 의존적으로 super-complex를 형성하고 가역적으로 thylakoid membrane에 부착한다.
본 연구에서는 비수리 추출물의 피부 상재균에 대한 항균 작용과 항산화 활성, tyrosinase 저해 효과 등에 관한 조사를 수행하였다. 피부 상재균에 대한 항균활성 측정결과, P. ovale에 대한 ethyl acetate 분획의 MIC는 0.125%로 나타났으며, 이는 methyl paraben보다 큰 항균활성을 나타내었다. 추출물의 free radical(1,1-diphenyl-2-picrylhydrazyl, DPPH) 소거활성($FSC_{50}$)은 aglycone 분획(14.63 ${\mu}g$/mL)에서 가장 큰 활성을 나타내었고, 또한 Luminol-의존성 화학발광법을 이용한 $Fe^{3+}$-EDTA/$H_2O_2$ 계에서 생성된 활성산소종(reactive oxygen species, ROS)에 대한 비수리 추출물의 총항산화능은 aglycone 분획(0.07 ${\mu}g$/mL)에서 가장 큰 활성을 나타내었다. 비수리 추출물에 대하여 rose-bengal로 증감된 사람 적혈구의 광용혈에 대한 억제 효과를 측정하였고, 농도 의존적(1 ~ 50 ${\mu}g$/mL)으로 세포보호 효과를 나타내었다. Tyrosinase의 활성 저해 효과($IC_{50}$)는 비수리 추출물의 ethyl acetate 분획과 aglycone 분획이 각각 104.83 ${\mu}g$/mL, 27.55 ${\mu}g$/mL으로 나타났다. 이상의 결과들은 비수리 추출물이 $^1O_2$ 혹은 다른 ROS를 소광시키거나 소거하고, ROS에 대항하여 세포막을 보호함으로써 생체계, 특히 태양 자외선에 노출된 피부에서 항산화제로서 작용할 수 있음을 시사한다. 또한 tyrosinase 저해활성과 피부 상재균에 대한 항균작용으로부터 항산화, 항노화 및 항균성 화장품 소재로서의 응용 가능성을 확인하였다.
발암물질의 해독에 관여하는 2상 효소계의 지표효소인 QR을 활성화시키는 암예방성분의 존재여부를 탐색한 이전의 연구에서 들깨박의 메탄올 추출물이 hepa1c1c7 cell에서 높은 QR 유도활성을 나타내었다. 본 연구에서는 탈지공정에 앞서 실시되는 볶음과정에서 비활성상태로 존재하던 QR inducer가 활성상태로 유리되는 것으로 가정하고 산가수분해와 볶음처리를 한 후, 각각의 메탄올 추출물에 대한 QR 유도활성을 측정하였다. $100^{\circ}C$에서 30, 60, 120분간 산가수분해시킨 날들깨박의 메탄올 추출액은 가수분해시간이 증가할수록 세포의 QR 활성유도능이 증가하는것으로 나타났다. $180^{\circ}C$와 $200^{\circ}C$에서 5, 10, 20분간 볶은 들깨박의 경우, QR유도 활성은 날들깨박에 비해 높았으며, 묶음시간이 걸어질수록 증가하는 경향을 보였다. 볶은 들깨박의 메탄올 추출액은 농도가 증가함에 따라 비례적으로 QR 효소 활성을 증가시켰다. 탈지들깨박이 동물조직의 QR 효소활성에 미치는 영향을 생쥐를 이용하여 확인한 결과, 볶은 들깨박의 메탄올 추출물은 QR 효소활성을 간과 위에서 유의적으로 증가시켰고, 날들깨박은 간과 폐에서 유의적으로 효소활성을 증가시켰다. 이렇듯 들깨박은 동물의 여러기관에서 암예방의 지표효소인 QR을 유도하는 것으로 나타나, 발암 물질로부터 생체를 보호하는 물질을 함유하고 있을 가능성이 높은 것으로 추정된다.

제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
