Adhesion of Human Osteoblasts Cell on CrN Thin Film Deposited by Cathodic Arc Plasma Deposition

  • Interaction between human osteoblast (hFOB 1.19) and CrN films was conducted in vitro. CrN films were produced by cathodic arc plasma deposition. The surface was characterized by atomic force microscopy (AFM). CrN films, glass substrates and TiN films were cultured with human osteoblasts for 48 and 72 hours. Actin stress fiber patterns and cell adhesion of osteoblasts were found less organized and weak on CrN films compared to those on the glass substrates and the TiN films. Human osteoblasts also showed less proliferation and less distributed microtubule on CrN films compared to those on glass substrates and TiN films. Focal contact adhesion was not observed in the cells cultured on CrN films, whereas focal contact adhesion was observed well in the cells cultured on glass substrates and TiN films. As a result, the CrN film is a potential candidate as a surface coating to be used for implantable devices which requires minimal cellular adhesion.

  • his paper is to develop a GIS application model (GISCELWAB) for the spatial simulation of surface runoff from a small watershed. The model was constituted by three submodels : The input data extraction model (GISINDATA) which prepares cell-based input data automatically for a given watershed, the cell water balance model (CELWAB) which calculates the water balance for a cell and simulates surface runoff of watershed simultaneously by the interaction of cells, and the output data management model (GISOUTDISP) which visualize the results of temporal and spatial variation of surface runoff. The input data extraction model was developed to solve the time-consuming problems for the input-data preparation of distributed hydrologic model. The input data for CELWAB can be obtained by extracting ASCII data from a vector map. The output data management model was developed to convert the storage depth and discharge of cells into grid map. This model enables to visualize the spatial formulation process of watershed storage depth and surface runoff wholly with time increment.

Cell to Cell Interaction Can Activate Membrane-bound APRIL Which Are Expressed on Inflammatory Macrophages

  • Background: APRIL, originally known as a cytokine involved in B cell survival, is now known to regulate the inflammatory activation of macrophages. Although the signal initiated from APRIL has been demonstrated, its role in cellular activation is still not clear due to the presence of BAFF, a closely related member of TNF superfamily, which share same receptors (TACI and BCMA) with APRIL. Methods: Through transfection of siRNA, BAFF-deficient THP-1 cells (human macrophage-like cells) were generated and APRIL-mediated inflammatory activities were tested. The expression patterns of APRIL were also tested in vivo. Results: BAFF-deficient THP-1 cells responded to APRIL-stimulating agents such as monoclonal antibody against APRIL and soluble form of TACI or BCMA. Furthermore, co-incubation of the siBAFF-deficient THP-1 cells with a human B cell line (Ramos) resulted in an activation of THP-1 cells which was dependent on interactions between APRIL and TACI/BCMA. Immunohistochemical analysis of human pathologic samples detected the expression of both APRIL and TACI in macrophage-rich areas. Additionally, human macrophage primary culture expressed APRIL on the cell surface. Conclusion: These observations indicate that APRIL, which is expressed on macrophages in pathologic tissues with chronic inflammation, may mediate activation signals through its interaction with its counterparts via cell-to-cell interaction.

INVESTIGATION OF "STEPPED" DISCHARGE CURVES IN SINTERED TYPE NICKEL-CADMIUM CELL

  • Formation of the nickel-cadmium alloy in the negative electrode of nickel-cadmium cell subjected to continuous charging at elevated temperatures ($40~45^{\circ}C$) is shown to be one of the causes of the 'stepped' discharge curves. The alloy has been characterized by electrode potential measurement and X-ray diffraction method. The potential lowering during discharge is related to discharge of the alloy. X -ray diffraction suggests that the nickel-cadmium alloy can be formed during charge in negative electrode by interaction of the two metals. Addition of Ni $(OH)_2$ into $Cd{\;}(OH)_2$ active material is found to form the alloy more readily than sintered negative electrode alone.ode alone.

  • In this paper, we developed the new non-rubbing liquid crystal (LC) alignment techniques in the cell with slanted non-polarized ultraviolet (UV) light irradiation on polyimide (PI) film. It is shown that the uniform alignment for nematic (N) LC is obtained by using slanted non-polarized UV light irradiation on PI surface. We successfully obtained that the pretilt angle of NLC is generated about 3.3 degree in the cell with slanted non-polarized UV light irradiation with 70 degree on PI surface, for the first time. It is considered that the pretilt angle generation in NLC is attributed to interaction between the LC molecular and the PI, which is broken the polymer by slanted non-polarized UV irradiation. Therefore, we concluded that the uniform LC alignment is attributed to anisotropic dispersion force due to photo depolymerization with slanted non-polarized UV light irradiation on PI surface.

  • We investigated the generation of high pretilt angle for nematic liquid crystal (NLC) in a cell with obilique non-polarized ultraviolet (UV) light irradiation on mixtured polyimide surface. It was found that the monodomain alignment of NLC is obtained with an angle of incidence of 75∼85 degrees on PI surface. We consider that the monodomain alignment of NLC is attributed to anisotropic dispersion force effect to photo-depolymerization of polymer. We successfully observed that the pretilt angle of NLC is generated above 5 degree with an angle of incidence of 75∼85 degrees. It is considered that the pretilt angle generation NLC is attributed to interaction between the LC molecules and the polymer surfaces.

  • In this paper, we investigated the liquid crystal(LC) alignment and generation of pretilt angle by using photo-alignment techniques on two kinds of polyimide(Pl) surface. It was found that the uniform alignment for nematic(N) LC is obtained in a cell with slanted UV light irradiation on PI surface without side-chain. We successfully observed that the pretilt angle of NLC is generated about 3.3 degrees in a cell with an incidence angle of 70 degrees on PI surface without side chain. It is considered that the pretilt angle generation in NLC is attributed to interaction between the LC molecular and the polymer.

  • Interaction between human osteoblast and TiN films was conducted in vitro. TiN films were produced by cathodic arc plasma deposition. The surface was characterized by atomic force microscopy (AFM). TiN films, glass substrates and Ti films were cultured with human osteoblasts for 48 and 72 h hours. Actin stress fiber patterns and microtubules of osteoblasts were found slightly more organized and distributed on TiN films compared to those on the Ti films and the glass substrates. Human osteoblasts also showed slightly higher cell attachment, proliferation, and focal contact adhesion on TiN films compared to those on Ti films and glass substrates. Our results demonstrated that TiN films showed slightly better cellular adhesion of osteoblasts than Ti films and glass substrates in a short-time culture period.

  • In the field of tissue engineering, researches have been actively conducted to regulate stem cell fate by understanding the interaction between cell and materials. This approach is expected as a promising therapeutic method in the future medicine by utilizing differentiation of stem cells into desired cells or tissues using biomaterial. For this regenerative medicine, there exist lots of attempts to construct optimized structures of various shapes and sizes that can regulate the stem cell fate. In this review, we will empathize the topographic effect as stem cell niche on the mesenchymal stem cell (MSC) response (cell attachment, proliferation, and differentiation) according to the shape and size of the structure of the substrates, and comprehensively analyze the importance and the effect of shape and size of the surface topography.
