Nuclear Factor-κB Activation: A Question of Life or Death

  • Apoptosis is a mode of cell death that plays an important role in both pathological and physiological processes. Research during the last decade has delineated the entire machinery needed for cell death, and its constituents were found to pre-exist in cells. The apoptotic cascade is triggered when cells are exposed to an apoptotic stimulus. It has been known for several years that inhibitors of protein synthesis can potentiate apoptosis that is induced by cytokines and other inducers. Until 1996, it was not understood why protein synthesis inhibitors potentiate apoptosis. Then three reports appeared that suggested the role of the transcription factor NF-${\kappa}B$ activation in protecting the cells from TNF-induced apoptosis. Since then several proteins have been identified that are regulated by NF-${\kappa}B$ and are involved in cell survival, proliferation, and protection from apoptosis. It now seems that when a cell is attacked by an apoptotic stimulus, the cell responds first by activating anti-apoptotic mechanisms, which mayor may not be followed by apoptosis. Whether or not a cell undergoes proliferation, the survival, or apoptosis, appears to involve a balance between the two mechanisms. Inhibitors of protein synthesis seem to suppress the appearance of protein that are involved in anti-apoptosis. The present review discusses how NF-${\kappa}B$ controls apoptosis.

Decursinol Angelate Ameliorates Dextran Sodium Sulfate-Induced Colitis by Modulating Type 17 Helper T Cell Responses

  • Angelica gigas has been used as a Korean traditional medicine for pain relief and gynecological health. Although the extracts are reported to have an anti-inflammatory property, the bioactive compounds of the herbal plant and the effect on T cell responses are unclear. In this study, we identified decursinol angelate (DA) as an immunomodulatory ingredient of A. gigas and demonstrated its suppressive effect on type 17 helper T (Th17) cell responses. Helper T cell culture experiments revealed that DA impeded the differentiation of Th17 cells and IL-17 production without affecting the survival and proliferation of CD4 T cells. By using a dextran sodium sulfate (DSS)-induced colitis model, we determined the therapeutic potential of DA for the treatment of ulcerative colitis. DA treatment attenuated the severity of colitis including a reduction in weight loss, colon shortening, and protection from colonic tissue damage induced by DSS administration. Intriguingly, Th17 cells concurrently with neutrophils in the colitis tissues were significantly decreased by the DA treatment. Overall, our experimental evidence reveals for the first time that DA is an anti-inflammatory compound to modulate inflammatory T cells, and suggests DA as a potential therapeutic agent to manage inflammatory conditions associated with Th17 cell responses.

Bovine Lactoferricin Induces Intestinal Epithelial Cell Activation through Phosphorylation of FAK and Paxillin and Prevents Rotavirus Infection

  • We investigated the effect of bovine lactoferricin (Lfcin-B), a peptide derived from bovine lactoferrin, on activation of intestinal epithelial cells in IEC-6 intestinal cell, and protection against in vivo rotavirus (RV) infection. Treatment with Lfcin-B significantly enhanced the growth of IEC-6 cells and increased their capacity for attachment and spreading in culture plates. Also, Lfcin-B synergistically augmented the binding of IEC-6 cells to laminin, a component of the extracellular matrix (ECM). In the analysis of the intracellular mechanism related to Lfcin-B-induced activation of IEC-6 cells, this peptide upregulated tyrosine-dependent phosphorylation of focal adhesion kinase (FAK) and paxillin, which are intracellular proteins associated with cell adhesion, spreading, and signal transduction during cell activation. An experiment using synthetic peptides with various sequences of amino acids revealed that a sequence of 9 amino acids (FKCRRWQWR) corresponding to 17-25 of the N-terminus of Lfcin-B is responsible for the epithelial cell activation. In an in vivo experiment, treatment with Lfcin-B one day before RV infection effectively prevented RV-induced diarrhea and significantly reduced RV titers in the bowels of infected mice. These results suggest that Lfcin-B plays meaningful roles in the maintenance and repair of intestinal mucosal tissues, as well as in protecting against intestinal infection by RV. Collectively, Lfcin-B is a promising candidate with potential applications in drugs or functional foods beneficial for intestinal health and mucosal immunity.

A Screen for Dual-protection Molecules from a Natural Product Library against Neuronal Cell Death and Microglial Cell Activation (신경세포 사멸과 미세아교세포활성화 억제 동시 가능 천연물질 탐색 연구)

  • Natural products and natural product structures play a general and highly significant role in drug discovery and development process because it has various merits and potentials for new drug source that have extensive clinical experience, development time contraction, excellent stability and safety. In several neurological disorders, neuronal death and excessive activation of microglia (neuro-inflammation) are observed. A number of drug discovery-related neuronal cell death and neuro-inflammation was studied from natural products, respectively. However, until now, it has not been possible to study dual-protection molecules recorded in the Natural Product library. In the present study, using the natural product-derived library of the Institute for Korea Traditional Medical Industry, we investigated dual-protective molecules against glutamate (a classical excitatory neurotransmitter)-induced oxidative stress mediated neuronal cell death and LPS-induced excessive activated microglial cells (immune cells of the brain). Chrysophanol, extracted from Rheum palmatum, had dual-protective effects against both glutamate-induced neuronal cell death and LPS-induced NO production, triggering proinflammatory cytokines and microglia activation and resulting in neuroinflammation. Flow-cytometry analysis revealed that chrysophanol had a scavenger effect, scavenging glutamate- and LPS-induced reactive oxygen species (ROS) produced by neuronal and microglial cells, respectively. Based on the present study, chrysophanol may have an important protective role against neuronal cell death and neuroinflammation in the brain. The results may be helpful for studying drug development candidates for treating central nervous system disorders.

Antibody Induced by the JY-Pol Pneumococcal Conjugate Protects Mice Against systemic Infection Due to Streptococcus pneumoniae (JY-Pol 접합백신으로 유도된 항페렴구균 항체의 보호효과)

  • We previously reported that Streptococcus pneumoniae capsule attached to the surface protein (JY-Pol) was protective to systemic pneumococcal infection. The JY -Pol antigen induced IgM, IgG, and IgA in mice and provoked cell-mediated immunity. In this current study, we investigated the effect of anti JY-Pol antiserun and monoclonal antibody C2 (Mab C2) specific for the JY-Pol antigen against the pneumococcal disease. Mice that were given the antiserum survived longer than mice that received antiserum pre-absorbed with S.pneumoniae cells or DPBS as a negative control. Heat-treated anti JY-Pol antiserum resulted in survival rates similar to intact fresh JY-Pol antiserum. Mab C2 isolated from JY-Pol-immunized mice also enhanced resistance of naive mice against the pneumococcal diseaser. This protection by Mab C2 appeared to be mediated by opsonization as determined in a RAW 264.7 monocyte/macrophage cell line. Epitope analysis showed that Mab C2 epitope consisted of glucuronic acid and glucose that blocked the interaction of JY-Pol to the C2. Taken together, these data indicate that the antiserum induced by the JY-Pol, a naturally pneumococcal conjugate formula, mediated the protection by passive transfer, which was confirmed by protective effect of Mab C2.

Effect of a 60Hz electromagnetic field on the frequency of bleomycin-induced HPRT gene mutation and 1,2,4-benzenetriol-induced sister chromatid exchanges in CHO cell

  • The interaction of low density extremely low frequency magnetic field (ELF MF) in the frequency of hypoxanthine-guanine phosphoribosyltransferase (HPRT) mutation induced by bleomycin and on the frequency of sister chromatid exchanges (SCEs) induced by 1,2,4-benzenetriol(BT) was demonstrated. CHO cells pretreated with bleomycin or 1,2,4-benzenetriol were exposed for 24hrs to a sinusoidal 0.8mT magnetic field at 60Hz. Frequency of HPRT mutation and SCEs were determined. ELF MF exposure led to a two-fold increase of the frequency of HPRT mutation induced by bleomycin. No increase of mutation frequency was observed by ELF MF alone ELF MF also increased the frequency of SCEs induced by BT while no Increase of SCE frequencies were observed by ELF MF alone. These results suggest that low density ELF MF field would art as an enhancer rather than as an initiator of mutagenic effects in CHO cell.

Effects of Korean Citrus junos and Medicinal Herbs on Liver Protection and Lipid Metabolism of Alcohol Fed Rats

  • In order to investigate whether or not the alcohol-treated rat liver cells can be protected by Korean Citrus junos and medicinal herbs, We compared the serum biochemistry of rats administered both alcohol and the complex of Korean Citrus junos and medicinal herbs to control rats treated with alcohol alone. The activities of Aspartate aminotransferase (AST) and Alanine aminotransferase (ALT) in the citron 3 (Citron 3, less mellowed citron which was ripened for three months)+Phellinus linteus, Alnus japonica, Dendropanax morbifera and citron 4 (Citron 4, completely mellowed citron which was ripened fer four months)+Phaseolus radiatus, Cordyceps militaris group were significantly low when compared with the negative control group (p<0.05). The levels of triglyceride (TG) in all experimental groups were significantly lower than the negative control group (p<0.05). The concentrations of total cholesterol in the citron 3+Phellinus linteus, Atnus japonica, Dendropanax morbifera and citron 4+Phaseolus radiatus, Cordyceps militaris, Phellinus linteus were lower than the negative control group (p<0.05). The activities of alcohol dehydyogenase (ADH) in all experimental groups were significantly high when compared with the normal control group (p<0.05). These results suggest that the complex of Korean Citrus junos and medecinal herbs could be an excellent candidate for protecting vat liver cell damage induced by alcohol.

Studies on medicinal protection with KH-19 against side effect of 5-fIurouracil and its mechanism (KH-19의 5-flurouracil 부작용 억제 효력 및 기전 연구)

  • The objective on this study was to investigate medicinal protection with KH-19, which was composed of 9 kinds of oriental herbs tonifying the blood, against side effect of chemotherapy in mice and its mechanism through microarray. Not only WBC (white blood cell), and PLT (platelet) as a hematopoiesis toxicity indicator but also spleen weight as an immune toxicity indicator was reduced significantly 7 day after. 5-flurouracil (FU) treatment. However, reduction of WBC, PLT, and spleen weight after 5-FU treatment was significantly recovered by KH-19. For mechanic study on KH-19 action, gene expression in mouse spleen treated 5-FU was compared with gene expression in mouse treated 5-FU and KH-19. The result indicates that 23 genes were increased. in expression level over 2 fold and 41 genes were decreased in expression level more than 2 fold at mouse spleen by KH-19 treatment. KH-19 mechanism may be complicated more than other drug which of mechanism were composed of single compound because KH-19 was composed of various compounds.

Forgery Protection System and 2D Bar-code inserted Watermark (워터마크가 삽입된 이차원 바코드와 위.변조 방지 시스템)

  • Generally, the copy protection mark and 2D bar-code techniques are widely used for forgery protection in printed public documents. But, it is hard to discriminate truth from the copy documents by using exisiting methods, because of that existing 2D-barcode is separated from the copy protection mark and it can be only recognized by specified optical barcord scanner. Therefor, in this paper, we proposed the forgery protection tehchnique for discriminating truth from the copy document by using watermark inserted 2D-barcord, which can be accurately distinguished not only by naked eye, but also by scanner. The copy protection mark consists of deformed patterns that are caused by the lowpass filter characteristic of digital I/O device. From these, we verified the performance of the proposed techniques by applying the histogram analysis based on the original, copy, and scanned copy image of the printed documents. Also, we suggested 2D-barcord confirmation system which can be accessed through the online server by using certification key data which is detected by web-camera, cell phone camera.

The Simplified PWM Method using Serial Communication in Cascaded H-Bridge Multilevel Inverter (직렬통신을 이용한 H-브릿지 멀티레벨 인버터의 PWM 구현방법)

  • As h-bridge multilevel inverter is connected with series of single phase power cell, so it obtain high voltage using low voltage power semi-conductor and output voltage similar to sine wave. In this topology, the number of power cell increases in proportion to the output voltage level. Therefore, there are drawbacks that are responsibility against operating ability of main controller and signal wire increase. However, we can overcome this problems by the substitution of serial communication for the PWM signal in power cell control. Additionally, it has merits of reliability and maintenance. This paper deals with the synchronization and phase-shift method of power cell PWM using CAN(Controller Area Network) communication interrupt in H-bridge multilevel inverter. The advantages of proposed method are signal-line simplification using serial communication between main controller and cell controller, burden reduction in main controller, modularization of power cell, easy protection of each power cell, expandability improvement and reliability increase of control signal and power cell. This paper establishes propriety and reliability of proposed method through experiment of 13-level H-bridge multilevel inverter.
