Enhanced Production of Digoxin by Digitoxin Biotransformation Using In Situ Adsorption in Digitalis lanata Cell Cultures

  • For the enhanced production of a cardiac glycoside, digoxin, using in situ adsorption by biotransformation from digitoxin in plant cell suspension cultures, selection of proper resins was attempted and the culture conditions were optimized. Among various kinds of resins tested, Amberlite XAD-8 was found to be the best for digoxin production in considering adsorption characteristics as well as the effect on cell growth. Adequate time for resin addition was determined to be 36 h from the beginning of biotransformation and the presence of resins should be as short as possible to increase the productivity. In addition, to prevent the cells from direct contact with resin particles, immobilized systems were designed and examined. Immobilization further improved the advantages of in situ adsorption. It was confirmed that the increase of the contact area for mass transfer was an important factor in utilizing an immobilized system to enhance digoxin production.

Effect of glucose and nitrogen sources on production of gellan

  • The gellan was produced by Pseudomonas elodea ATCC 31461 under aerobic condition. Gellan provides various functionalities such as gelling, suspending, stabilizing, emulsifying and binding properties in aqueous systems. In this study, the effect of glucose concentration and nitrogen sources concentration, bacto peptone and $NH_4NO_3$, on the cell growth and the production of gellan were evaluated. The maximum specific yield (g gellan/g cell) of 2.22 was obtained at 1.0% of glucose. Bacto peptone increased cell growth. And nitrogen limitation was essential for higher production of gellan. The highest cell and gellan production were abtained at 0.5 g/${\ell}$ of bacto peptone without $NH_4NO_3$.

영지버섯 다당체의 Nitric Oxide 생성능 및 생성기전 연구 (Nitric Oxide Production Ability and its Formation Mechanisms in Macrophage TIB 71 Cell Line by Polysaccharide Extracted from Ganoderma lucidum)

  • This study was carried out to get infomation on the nitric oxide production ability and its formation mechanisms of polysaccharides extracted from Ganoderma lucidum(PSG) by using murine macrophage cell line. The cultured mycelial cells of Ganoderma lucidum were extracted by alkali, and than neutralized by acid. The extract were passed through the column of DEAE cellulose for more purification. The neutral fraction was concentrated and precipitated with 95% ethanol. The precipitate was lyophilized and PSG was obtained. The immunomodulating effects of PSG on macrophage were performed by using murine macrophage cell line ATCC TIB 71 cells with PSG 0.5mg. PSG alone could not induce the production of nitrite, but it had a significant potential effect on nitrite secretion when the cells were primed and triggered with BCG and Interferon(IFN)-${\gamma}$. Also it was prominent by using calcium channel blocker(verapamil) and adenylate cyclase activator(forskolin).

  • This paper present an effective modeling scheme of SPE cell system for hydrogen production. As oxygen and hydrogen produced by water electrolysis using SPE are high purity, we can use oxygen in biomedical and hydrogen could be used in many ways. Recently, it is under the eye as a surplus power storage system. PSCAD/EMTDC model of SPE cell system for hydrogen production to efficiently utilize solar cell energy is showed in this paper. The simulated results are then verified by comparing them with the actual values obtained from the data acquisition system. Authors are sure that it is a useful method to the researchers who study SPE cell system for hydrogen production.

자율적인 밸런싱을 실현하는 Bucket Brigade 기반의 조립셀 운영방식 (Application of Bucket Brigades in Assembly Cells for Self Work Balancing)

  • Assembly line has been recognized as an efficient production system in mass production. However, the recent production environment characterized as mass customization urges production managers to transform a long assembly line to a number of short assembly cells. To maximize the utilization of resources in an assembly cell, it is important to have the line balanced. This paper presents a bucket brigade-based assembly cell. Bucket brigade is a way of coordinating workers who progressively perform a set of assembly operations on a flow line. Each worker follows a simple rule: perform assembly operations on a product until the next worker downstream takes it over; then go back to the previous worker upstream to take over a new assembly job. In this way, the line balances itself. The bucket brigade assembly cell is analyzed and compared with traditional assembly lines and general assembly cells. The paper also discusses some prerequisite requirements and limitations when the bucket brigade assembly cells are employed.

  • Vaccines, especially for viruses, have been produced from egg-based manufacturing process. The method is simple and easy to set up the manufacturing process. However, the method has many problems in quality control, limit of manufacturing capacity, and ethical issues. Over the last decade, an alternative method, which manufactures vaccines using cell culture-based system, has received great attention to overcome the problems in egg-based vaccine production. This article examines current status and perspectives of cell culture-based vaccine production.

  • This paper presents an effective modeling scheme of SPE cell system for hydrogen production. 국내 연안산 질소고정 단세포 남세균 종주의 광생물학적 수소생산력 (Photobiological Hydrogen Production by Korean $N_2$-fixing Unicellular Cyanobacterial Strains)

  • Photobiological hydrogen production by nitrogen-fixing unicellular cyanobacteria has long been considered to be an environmentally sound and very promising method for the future supply of renewable clean energy. We tried to find out the optimum cell concentration for $H_2$ production in each of the two new Korean nitrogen-fixing unicellular cyanobacterial strains to compare with Synechococcus sp. strain Miami BG043511. The two Korean strains, Cyanothece sp. KNU CB MAL-031 and KNU CB MAL-058, were isolated from Korean west coasts. Cell concentrations up to 17 billion cells $ml^{-1}$ were applied to the tests. High cell concentration over 15 billion cells $ml^{-1}$ resulted in drastically reduced $H_2$ production in all the three strains. The two domestic strains, however, produced 2-3 time more hydrogen than Synechococcus sp. Miami BG043511 at cell concentrations of 5-10 billion cells $ml^{-1}$. At lower cell concentrations than 2 billion cells $ml^{-1}$, MAL-031 exhibited highest $H_2$ production followed by Miami BG043511, with far less production in MAL-058. Present result suggests that Cyanothece sp. MAL-CB031 might be one of the ideal nitrogen-fixing unicellular cyanobacterial strains for the photobiological hydrogen production.

식물생장촉진 근권 Pseudomonas의 Siderophore 생성에 관여하는 요인 (Factors Influencing Siderophore Production by Plant Growth Promoting Rhizopseudomonas Strains)

  • 본 논문에서는 철 이외에 사이데로포 생성에 관여하는 요인을 연구하였다. 약산성(pH6)에서 세포의 양은 감소하는 반면에 세포당 사이데로포의 생성은 증가하였다. 사이데로포 생성의 적정온도는 7NSK2 균주가 $12^{\circ}C$이고 ANP15은 $19^{\circ}C$였다. 배지의 C : N 비율에 따른 사이데로포 생성력을 보면 C : N율이 낮은 배지에서는 사이데로포 생성과 세포생장은 감소됐다. 서로 다른 배지에서 사이데로포 생성을 조사한 바에 의하면 citrate를 탄소원으로 했을 경우 세포의 철 흡수능력이 증가했으며 사이데로포 생성도 감소되었다. 그러나 Glucose나 Succiuate에 비해서 citrate 배치의 세포량에는 별 차이가 나타나지 않았다. 이상의 결과로 보아 환경요인으로 작용할 수 있는 pH나 온도, 탄소원 등은 철분 외에도 사이데로포의 생성에 영향을 미칠 수 있음을 확인하게 되었다.

Enhancement of Tissue Type Plasminogen Activator (tPA) Production from Recombinant CHO Cells by Low Electromagnetic Fields

  • Low Electromagnetic Field (EMF) intensity in the range of $1{\mu}T\;to\;10{\mu}T$(Tesla) was found to enhance the growth of CHO cells and the production of tPA in batch and perfusion cultivations. At $1{\mu}T\;intensity,\;1.3{\times}10^7$ viable cells/ml of maximum cell density and 80 mg/l of maximum tPA production were obtained in batch cultivation, compared to $2.8{\times}10^6$ viable cells/ml and 59 mg tPA/1 in unexposed case (control). A similar trend was observed in the perfusion process, where it was possible to obtain $1.2{\times}10^7$ viable cells/ml of maximum cell density and 81 mg tPA/l of maximum tPA production by more than 80 days of cultivation. However, there was not much difference between $1{\mu}T\;and\;10{\mu}T$ in perfusion cultivation, possibly due to better environmental growth conditions being maintained by continuous feeding of fresh medium into the reactor. On the contrary, both cell growth and tPA production were severely inhibited at higher than 1 mT intensity, showing no growth at 10 mT exposure. Specific growth rate was linearly correlated to specific tPA production rate at $1{\mu}T$EMF intensity, which represents a partially growth-related relationship. It was also found that a large amount of $Ca^2+$ was released at low EMF intensity, even though the cell growth was not much affected. Low EMF intensity significantly improved both cell growth and tPA production, and tPA production seemed to be more affected than the cell growth, possibly due to the changes of cell membrane characteristics. It can be concluded that the elaboration of EMF intensity less than $10{\mu}T$ could improve cell growth and tPA production, but mainly tPA secretion through batch or perfusion process in a bioreactor.
