Every Single Cell Clones from Cancer Cell Lines Growing Tumors In Vivo May Not Invalidate the Cancer Stem Cell Concept

  • We present the result of our research on the tumorigenic ability of single cell clones isolated from an aggressive murine breast cancer cell line in a matched allografting mouse model. Tumor formation is basically dependent on the cell numbers injected per location. We argue that in vivo tumor formation from single cell clones, isolated in vitro from cancer cell lines, may not provide conclusive evidence to disprove the cancer stem cell (CSC) theory without additional data.

The Effect of Cryopreservation on the Mouse Embryos at Various-pronuclear Stages

  • This study was carried out to establish an appropriate condition for the efficient cryopreservation of the mouse pronuclear embryo. In vitro cryopreservation of pronuclear embryos was carried out by slow freezing or vitrification methods and development rate of 2-cell, blastocyst and hatched blastocyst was measured as well as survival rate of the thawed pronuclear embryo. After slow freezing, vitrification and thawing of mouse pronuclear embryos, the survival rate and blastocyst development rate for the vitrification group was 97.3 and 53.4%, respectively, which was significantly higher as compared to the slow freezing group with 88.6 and 23.9%, respectively (p<0.05). Blastocyst developmental rate in each experimental group was significantly higher for 21 h in the post-hCG group at 40.5-57.0% than the 24 h post-hCG group at 40.5% (p<0.05). ICM (Inner cell mass) cell numbers of blastocyst-stage embryos during the different stages of mouse pronuclear embryos, slow freezing and vitrification period in the control and vitrification groups were 22.1${\pm}$2.7 and 17.0${\pm}$3.1-22.0${\pm}$3.2, respectively; hence, the slow freezing group (10.2${\pm}$2.0) had significantly higher cell numbers than those of the other two groups (p<0.05). Trophoblast (TE) cell number in the control group, 65.8${\pm}$12.6, was significantly higher than in the slow freezing group, 41.6${\pm}$11.1 (p<0.05). The total cell numbers in the control group and 21 h post hCG group were 87.9${\pm}$13.6 and 81.8${\pm}$14.1, respectively, and were significantly higher than for the slow freezing group (51.8${\pm}$12.6; p<0.05).

Forced Expression of HoxB4 Enhances Hematopoietic Differentiation by Human Embryonic Stem Cells

  • HoxB4 has been shown to enhance hematopoietic engraftment by hematopoietic stem cells (HSC) from differentiating mouse embryonic stem cell (mESC) cultures. Here we examined the effect of ectopic expression of HoxB4 in differentiated human embryonic stem cells (hESCs). Stable HoxB4-expressing hESCs were established by lentiviral transduction, and the forced expression of HoxB4 did not affect stem cell features. HoxB4-expressing hESC-derived CD34+ cells generated higher numbers of erythroid and blast-like colonies than controls. The number of CD34+ cells increased but CD45+ and KDR+ cell numbers were not significantly affected. When the hESC derived CD34+ cells were transplanted into $NOD/SCID{\beta}2m-/-$ mice, the ectopic expression of HoxB4 did not alter their repopulating capacity. Our findings show that overexpression of HoxB4 in differentiating hESCs increases hematopoietic colony formation and hematopoietic cell formation in vitro, but does not affect in vivo repopulation in adult mice hosts.

Measurement of Thermal Flow in a Hele-Shaw Convection Cell Using Holographic Interferometry and PIV Technique (홀로그래픽 간섭계와 PIV를 이용한 Hele-Shaw Convection Cell 내부 열유동 해석)

  • Variations of temperature and velocity fields in a Hele-Shaw Convection Cell (HSC) were measured using a holographic interferometry and PIV technique with varying Rayleigh number. Experimental results show a steady flow pattern at low Rayleigh numbers and a time-dependent periodic flow at high Rayleigh numbers. Two different measurement methods of holographic interferometry, double-exposure method and real-time method, were employed to measure the temperature field variations of HSC convective flow. In the double-exposure method, unwanted waves can be eliminated and reconstruction images are clear, but transient flow structure cannot be observed clearly. On the other hand, transient flow can be observed and reconstructed well using the real-time method. PIV results show that flow inside the HSC is periodic and the oscillating state is well matched with the temperature field results. The holographic interferometry and PIV techniques employed in this study are useful for analyzing the unsteady convective thermal fluid flows.

Effects of Citro Reticulatae Viride Pericarpium on the Vascular Endothelial Growth Factor(VEGF) in the bronchial asthma mouse model (청피(靑皮)가 천식유발 백서에서 VEGF에 미치는 영향)

  • Objective : Author evaluate the effects of Citri Reticulatae Viride Pericarpium for the bronchial asthma using assesment of the vascular endothelial growth factor after Citri Reticulatae Viride Peicarpium was intravenously administered Ovalbumine(OVA)-sensitized and -challenged mice. Material and Methods : Twenty-four female mice, 8-10 weeks old and free of murine specific pathogens, were sensitized by intraperitoneal injection of OVA. Of the sensitized mice, ten mice didn't administrate Citri Reticulatae Viride Pericarpium and the other ten mice administrated Citri Reticulatae Viride Pericarpium. Mice were sensitized on the first and fourteen days introspectional injection of 20 ${\mu}g$ OVA. After$21^{st}\;22^{th}\;and\;23^{rd}$, the initial sensitization, the mice were challenged for 30 minutes with an aerosol of 1% OVA in saline. Citri Reticulatae Viride Pericarpium administered 200mg/kg in the tail of the mouse, one a day, for 7 days and beginning 14 days after first sensitization. Bronchoalveolar lavage was performed 72 hours after the last challenge, and total cell numbers in the BAL fluid were count. Also, lever of VEGF in the BAL fluid was measured by Enzyme immunoassays and Western blot analysis. Results: Total cell numbers in BAL fluid were significantly greater than from 72 hrs after OVA inhalation compared with cell numbers in the control group. However, there was no difference of the total cell numbers between OVA-challenge groups without Citri Reticulatae Viride Pericarpium, and OVA-challenge with Citri Reticulatae Viride Pericarpium. Enzyme immunoassay revealed that VEGF levels in the BAL fluids were significantly increased 72 hrs after OVA inhalation compared with levels in the control group. After administration of the Citri Reticulatae Viride Pericarpiu, the levels of the VEGF in BAL fluids 72 hrs after OVA inhalation reduced dramatically. Western blot analysis revealed that VEGF protein levels were increased in the all mice which were challenge with OVA, without administered Citri Reticulatae Viride Pericarpium, compared the normal mouse. However, in the groups of the administered Citri Reticulatae Viride Pericarpium, the VEGF protein level markedly decreased. Conclusion: Citri Reticulatae Viride Pericarpium might affect the treatment of the bronchial ashma as a inhibition of the VEGF.

Microwave Drying of Food Waste (음식물 쓰레기의 마이크로파 건조)

  • The food wastes from a refectory and an eating house were heated in domestic microwave oven(700W) equipped with a fan and the drying rates and destruction of microorganisms were investigated. The drying rate was decreased with the size of food waste and the food wastes in polypropylene basket were dried faster than that on glass dish. The rate was increased with lower initial moisture content. Death rate of microorganisms was also decreased with the size of food waste. Ninety eight percent of reduction in viable cell numbers for the 400g of food waste could be achieved in 240sec of microwave irradiation. The growth of microorganisms in food wastes after microwave irradiated was observed at $32^{\circ}C$ and 95% relative humidity after 7days and the cell numbers in microwave irradiated food wastes were found to be 1/2 ~ 1/20 of the numbers in untreated wastes in accordance with the mass and the length of exposed time to microwave. To minimize the moisture and microorganisms in food wastes, the use of microwave oven are recommended.

Effects of Serum and Gonadotropins in In-Vitro Maturation Medium on Nuclear Maturation, Development and Cell Numbers of Korean Native Cow Embryos (체외성숙용 배지에 혈청과 호르몬의 첨가가 한우 난포란의 핵성숙과 배발달 및 배반포의 세포수에 미치는 영향)

  • The main objective of this study was to examine the effects of serum and gonadotropins supplement during in vitro maturation(IVM) of bovine oocytes on nuclear maturation and embryo development, and we also examine the cell number. 1 . The first polar body(PB) extrusion rates of Korean native cow(KNC) oocytes matured in medium with FBS or gonadotropins were similar among treatment groups. The development rate to the blastocyst stage was significantly higher in the group of both supplement FBS and gonadotropins(26.0%) than in the group of non-supplement(9.9%) and gonadotropins (12.0%). The numbers of inner cell mass (ICM) and trophectoderm (TE) cells and total cell numbers of blastocysts were highest in the group of both supplement FBS and gonadotropins, and the number of ICM cells was increased by FBS supplementation (p<0.05). 2. The PB extrusion rates of KNC oocytes matured in medium with FBS in the different duration of IVM was significantly higher in the 0-18hr(63.1%) and in the 9-18hr(63.4%) group than in the 0-9hr.(37.4%) group (p<0.05). The embryo development rates did not differ among treatment groups. The numbers of TE cells and total cell numbers of blastocysts were similar among treatment groups, but the number of ICM cells of the 0-18h. group were significantly higher than the other treatment groups (p<0.05). The results indicate that although TCM199 alone can support bovine oocyte maturation and development to the blastocyst stage, a high quality of blastocysts can be produced from oocytes matured in medium containing serum and gonadotropins.

NUMERICAL SOLUTIONS OF AN UNSTEADY 2-D INCOMPRESSIBLE FLOW WITH HEAT AND MASS TRANSFER AT LOW, MODERATE, AND HIGH REYNOLDS NUMBERS

  • In this paper, we have proposed a modified Marker-And-Cell (MAC) method to investigate the problem of an unsteady 2-D incompressible flow with heat and mass transfer at low, moderate, and high Reynolds numbers with no-slip and slip boundary conditions. We have used this method to solve the governing equations along with the boundary conditions and thereby to compute the flow variables, viz. u-velocity, v-velocity, P, T, and C. We have used the staggered grid approach of this method to discretize the governing equations of the problem. A modified MAC algorithm was proposed and used to compute the numerical solutions of the flow variables for Reynolds numbers Re = 10, 500, and 50000 in consonance with low, moderate, and high Reynolds numbers. We have also used appropriate Prandtl (Pr) and Schmidt (Sc) numbers in consistence with relevancy of the physical problem considered. We have executed this modified MAC algorithm with the aid of a computer program developed and run in C compiler. We have also computed numerical solutions of local Nusselt (Nu) and Sherwood (Sh) numbers along the horizontal line through the geometric center at low, moderate, and high Reynolds numbers for fixed Pr = 6.62 and Sc = 340 for two grid systems at time t = 0.0001s. Our numerical solutions for u and v velocities along the vertical and horizontal line through the geometric center of the square cavity for Re = 100 has been compared with benchmark solutions available in the literature and it has been found that they are in good agreement. The present numerical results indicate that, as we move along the horizontal line through the geometric center of the domain, we observed that, the heat and mass transfer decreases up to the geometric center. It, then, increases symmetrically.

  • Experimental studies were performed to investigate the effect of Houttuyniae herba Aqua-acupuncture on the growth of Melanoma 816 tissue and Mononuclear cell subsets in mice. The results are as follows : 1. In the study of lymphocytes numbers in mice circulating blood, Houttuyniae herba Aqua-acupuncture showed inhibitory effect, but without stastistical significance. 2. In the study of $CD4^+$ T cell percentage in mice circulating blood, Houttuyniae herba Aqua-acupuncture showed inhibitory effect, but without stastistical significance. 3. In the study of $CD8^+$ T cell percentage in mice circulating blood, Houttuyniae herba Aqua-acupuncture showed inhibitory effect, with stastistical significance. 4. in the study of lymphocytes numbers in mice spleen tissue, Houttuyniae herba Aqua-acupuncture showed inhibitory effect, but without stastistical significance. 5. In the study of $CD4^+$ T cell percentage in mice spleen tissue, Houttuyniae herba Aqua-acupuncture showed inhibitory effect, but without stastistical significance. 6. In the study of $CD8^+$ T cell percentage in mice spleen tissue, Houttuyniae herba Aqua-acupuncture showed inhibitory effect, but without stastistical significance. 7. In the measurement of melanoma tissue weight, Houttuyniae herba Aqua-acupuncture showed increasing effect, but without stastistical significance. According to above results, it is concluded that Houttuyniae herba Aqua-acupuncture is effective upon immune responses in Melanoma 816 inoculated mice.

Measurement of Viable Cell Number in Mixed Culture Based on Microbial Respiration Rate (미생물 호흡속도에 기초한 혼합배양중의 생균수 측정)

  • A simple method to determine viable cell numbers of each species in mixed culture was developed. The oxygen uptake rate (OUR) equals to the product of the specific OUR and the size of the microbial population. In a mixed culture, the OUR is a result of the respiration activities of each sub-population. The OUR was determined from the slope of the linear relationship between time and the decrease of dissolved oxygen concentration when aeration was stopped. The specific OUR was calculated from the slope of the viable cell number versus OUR curve. These values for C. lusitaniae at 20 and $30^{\circ}C$ were $1.36{\times}10^{-9}$ and $3.90{\times}10^{-9}$ and those for P tannoPhilus at 20 and $30^{\circ}C$ were $0.59{\times}10^{-9}$ and $1.86{\times}10^{-9}$ [(%/s)/(cells/ml)J. respectively. Using these values, viable cell numbers were calculated after the OURs of mixed culture at two temperatures were measured. A good agreement between the viable cell numbers determined by this method and by plate count was obtained.
