In this study, the vegetative and reproductive morphology and anatomy of two Hawaiian endemic Portulaca species were examined. Specifically, P. molokiniensis and P. sclerocarpa were compared to closely related species in the genus. The comparisons were both qualitative and quantitative, using characteristics of leaves, stems, roots, and fruits. Tissue organizations of vegetative and reproductive parts of the plants were assessed using microtechnique procedures, statistical analysis, and scanning electron microscopy. The most notable features of these two species were (1) the size and frequency of stomata in P. molokiniensis, and (2) the large number of sclerenchymatous cell layers in the thickest fruit walls of P. sclerocarpa. These findings may imply that stomata development in P. molokiniensis and thick fruit wall development in P. sclerocarpa are evolved features of survival. In particular, the development of thickened walls in indehiscent fruits likely has evolutionary implications of ecological tolerance for better adaptation.
The morphology, anatomy and reproduction (tetra/bisporic and male) of Lithothamnion japonicum Foslie was studied, using holotype material and materials collected from Korea. Lithothamnion japonicum is characterized based on the presence of encrusting, warty to fruticose thallus, branched or unbranched short and cylindrical protuberances, conical shaped spermatangial conceptacles, branched (dendroid) spermatangial systems formed on floor, walls, and roof of conceptacle chamber, cylindrical shaped spermatangial conceptacle canal, 9-10 cell layered spermatangial conceptacle roof, raised tetra/bisporangial conceptacles without rims, flattened tetra/bisporangial conceptacle pore plate, 16-50 pores on each pore plate, 6-8 rosette cells surrounded by each pore, pore canal lining filaments composed of tetra/bisporangial conceptacle roof, and buried senescent tetra/bisporangial conceptacles completely infilled with relatively large and irregularly arranged calcified sterile cells. In this study, we report a new record of Lithothamnion japonicum from Korean coasts.

The ultrastructure of germ cells during spermatogenesis and some characteristics of sperm morphology in male Mytilus coruscus, which was collected on the coastal waters of Gyeokpo in western Korea, were investigated by transmission electron microscope observations. The morphology of the spermatozoon has a primitive type and is similar to those of other bivalves in that it contains a short midpiece with five mitochondria surrounding the centrioles. The morphologies of the sperm nucleus type and the acrosome shape of this species have an oval and modified cone shape, respectively. In particular, the axial rod is observed between the nucleus and acrosome of the sperm. The spermatozoon is approximately $45-50{\mu}m$ in length including a sperm nucleus (about $1.46{\mu}m$ in length), an acrosome (about $3.94{\mu}m$ in length) and tail flagellum (approximately $40-45{\mu}m$). The axoneme of the sperm tail flagellum consists of nine pairs of microtubules at the periphery and a pair at the center. The axoneme of the sperm tail shows a 9+2 structure. Some special charateristics of sperm morphology of this species in the genus Mytilus are (1) acrosomal morphology, (2) the number of mitochondria in the midpiece of the sperm, and (3) the existence of a satellite. The axial rod appears in the acrosome and sperm nucleus as one of the characteristics seen in several species of the subclass Pteriomorphia, unlikely the subclass Heterodonta containing axial filament instead of the axial rod. The number of mitochondria in the midpiece of the sperm of this species in the family Mytilidae are five, as one of common characteristics appeared in most species in the family Mytilidae. Most of Mytilus species contain a satellite body which is attached to the proximal centriole in the middle piece of the sperm, as one of common characteristics of sperm morphology in the family Mytilidae.
화장품에 방부제(살균/보존제)로 사용되는 1, 2-hexanediol (HD)로 인한 부작용을 극복하기 위하여, Escherichia coli (E. coli)의 ${\beta}-galactosidase$ (${\beta}-gal$)를 이용하여 transgalactosylation 반응으로 1, 2-hexanediol galactoside (HD-Gal)를 합성하였다. 본 연구에서는 합성된 HD-Gal의 인간 피부세포에 대한 독성이 어느 정도인지를 HD와 비교하여 관찰하였다. HD-Gal과 HD의 세포독성은 인간 피부각질형성세포(HaCaT cell line)에 HD와 HD-Gal을 처리한 후, cell proliferation assay를 이용하여 비교 분석하였다. 또한 이때, 위상차 현미경으로 HD-Gal과 HD로 처리한 세포의 상태를 비교 관찰하였다. 그 결과, HD-Gal은 42.2 mM에서 211 mM의 농도 범위에서 세포독성이 관찰되지 않았으며, 현미경 관찰에서도 큰 변화를 관찰할 수 없었다. 그러나, HD의 경우에는 저농도에서(42.2 mM and 84.4 mM)는 세포독성이 관찰되지 않았으나, 고농도(168.8 and 211 mM)에서 매우 높은 세포독성을 나타내었고, 현미경 관찰에서는 고농도에서는 물론이고, 세포독성이 관찰되지 않은 HD의 저농도에서도 세포모양과 세포 수에서의 변화가 관찰되었다. 앞으로 세포독성이 감소된 HD-Gal이 HD의 대체제로서 안전, 건강 및 웰빙 개념의 새로운 용도로 개발될 수 있을 것으로 생각된다.
The present work describes experiments undertaken to evaluate the usefulness of selected physicochemical indices of semen, cell membrane integrity and sperm chromatin structure for the assessment of boar semen sensitivity to processes connected with pre-insemination procedures. The experiments were carried out on 30 boars: including 15 regarded as providers of sensitive semen and 15 regarded as providers of semen that is little sensitive to laboratory processing. The selection of boars for both groups was based on sperm morphology analyses, assuming secondary morphological change incidence in spermatozoa as the criterion. Two ejaculates were manually collected from each boar at an interval of 3 to 4 months. The following analyses were carried out for each ejaculate: sperm motility assessment, sperm pH measurement, sperm morphology assessment, sperm chromatin structure evaluation and cell membrane integrity assessment. The analyses were performed three times. Semen storage did not cause an increase in the incidence of secondary morphological changes in the group of boars considered to provide sperm of low sensitivity. On the other hand, with continued storage there was a marked increase in the incidence of spermatozoa with secondary morphological changes in the group of boars regarded as producing more sensitive semen. Ejaculates of group I boars evaluated directly after collection had an approximately 6% smaller share of spermatozoa with undamaged cell membranes than the ejaculates of boars in group II ($p{\leq}0.05$). In the process of time the percentage of spermatozoa with undamaged cell membranes decreased. The sperm of group I boars was characterised with a lower sperm motility than the semen of group II boars. After 1 hour of storing diluted semen, the sperm motility of boars producing highly sensitive semen was already 4% lower ($p{\leq}0.05$), and after 24 hours of storage it was 6.33% lower than that of the boars that produced semen with a low sensitivity. Factors that confirm the accuracy of insemination male selection can include a low rate of sperm motility decrease during the storage of diluted semen, low and contained incidence of secondary morphological changes in spermatozoa during semen storage and a high frequency of spermatozoa with undamaged cell membranes.
The application of high pressure on cellular morphology, proliferation and protein expression of Jurkat cells (human T lymphocyte cell line) has been extensively investigated. In the present study, we manufactured a novel pressure chamber that modulates 5% $CO_{2}$, temperature and pressure (up to 3 ATA). Jurkat cells was incubated 2 ATA pressure and analyzed cellular morphology and growth using an electron microscopy and MTT assay. The cells showed the morphological changes in the cell surface, which appeared to cause a severe damage in cell membrane. The growth rate of the cells under 2 ATA pressure decreased as cultured time got increased. Furthermore, a long term exposure of high pressure on Jurkat cells may act as one of the important cellular stresses that leads to inducing cell death. Cellular proteomes were separated by 2-dimensional electrophoresis with pH 3-10 ranges of IPG Dry strips. And many proteins showed significant up-and-down expressions with hyperbaric pressure. Out of all, 10 spots were identified significantly using matrix-assisted laser desorption/ionization-time of fight (MALDI-TOF) mass spectrometry. We and found that 9 protein expressions were decreased and one protein, heat shock protein HSP 60, was increased in Jurkat cells under 2 ATA. Identified proteins were related to lipid metabolism and signal transduction.
The skin structure of Misgurnus mizolepis was studied based on the microanatomical investigation of skin fragments taken from four regions. The epidermis was distinguished by two types of skin glands, a small mucous cell and a large club cell. The mucous cell was acid sulfomucins (some sialomucins) but the club cell did not give any histochemical tests for mucosubstances. The presence of a well defined lymphatic system with small lymphocytes was established in the stratum germinativum layer of the epidermis. A large number of blood capillaries run very close to each other just below the basement membrane, and a definite area giving AB and PAS positive was present between the basement membrane and scale. These structural features of skin in M. mizolepis seem to be closely related with cutaneous respiration.
This study was performed to demonstrate the presence of large granular lymphocyte(LGL) in Sprague-Dawley(SD) rats and morphologically observe NK cell and also establish the method of isolation of natural killer cell in SD rats. By percoll discontinuous density gradients centrifugation, highly enriched LGL population were shown to fraction 2(border line between 44.2% and 50.8%). LGL were shown to bind selectively to YAC1 mouse lymphoma cell. This fraction expressed very high NK cell cytolysis. Therefore, we thought that LGL have NK activity in SD rats. The Morphology of rat LGL is very similar to that of human LGL. These cells have an eccentric kidney-shaped nucleus. Their most distinctive feature was their cytoplasmic azurophilic granules. Another distinguishing feature of rat LGL was their high cytoplasmic : nuclear ratio. It was concluded that LGL played a role part in mediating natural killer activity in this species.
The cell division of Bacillus thuringiensis are studed by a electron microscope. It was observed that when the cell division was occurred, the bacterial transverse septeum was centripetally formed, and the bacterial spore was divided into two daughter cells. The fore spore septum was initiated by invagination from either sides of the cell membranes, and was easily distinguished it from the transverse septum of the vegetative cell division. The large vesicular mesosome was. observed at one end of the cell membrane. The nucleoids were of variously irregular shapes and had no a nuclear membrane. The morphology of the bacteria was visualizd by a scanning electron microscope. The surface of the cell was generally rough and had a single polar flagellum, which was appeared to be $0.2{\mu}$ in width and $13{\mu}$ in length.
The purpose of this study was to evaluate bacteriocin activity against human flora. Lactobiocin, a bacteriocin produced by Lactococcus sp. HY 449, inhibited the growth of Starphylococcus epidermidis, Starphylococcus aureus, Streptoccoccus pyogenes and Propionibacterium acnes. When crude bacteriocin was added to indicator cells during logarithmic growth, the optical density(O.D 650nm) of cells without bacteriocin increase after 5h of incubation. Whereas in the presence of bacteriocin, the O.D of cell suspensions decreased. The similar patterns were observed for absorbance readings at 280 nm and 260 nm. The release of cellular components when cell were treated with Lactobiocin suggests some degree of membrane damage or cell lysis. Scanning electron microscopy of cells following treatments with Lactobiocin in PBS buffer revealed disruptures of cell morphology. These results indicate that bacteriocin appears to cause cell lysis of tested strains. In cytotoxicity on human fibroblast, LD$\_$50/ of Lactobiocin was ca. 50 mg/ml and no change was observed cell proliferation at the same concentration. Any irritation and allergic reaction did not observed when evaluated by human patch test for Lactobiocin.
