The morphological study on the parenchymal cells in the adult planaria performed to observe their cytochemical and ultrastructural characteristics. The results are as follows. Nine types of cells are found in parenchyma. 1. Free parenchymal cell: These cells contain several chromatoid bodies around the nucleus. Heterochromatins are evenly dispersed in large nucleus. These cells are abundant in free ribosomes. 2. Fixed parenchymal cells: These cells have well-developed granular endoplasmic reticulum, mitochondria and Golgi complex but they contain the cytosols exhibiting electron-lucencies. 3. Rhabdite-forming cells: These cells contain the electron-dense rhabdite granules of up to about 0.3 x 0.9 $\mu$m in size. Rhabdite-forming cells have well-developed cell organelles, granular endolplasmic reticulum, mitochondria and Golgy complex. 4. A-type of basophilic granule cells: These cells contain irregularly-shaped granules exhibiting alcianophilia. These granules surrounded by a limited membrane, approximately 1.4 x 0.7 $\mu$m in size, are accumulated in the cytoplasm. 5. C-type of basophilic granule cells: These cells contain electron-dense granules of less than 0.2 $\mu$m in size, which exhibit PAS- positive reaction. This type of granule is also found in the muscle layer of parenchyma. 6. D-type of basophilic granule cells: This type of granule cell occurs only in the parenchyma around reproductive organ. The granules have cytochemical characteristics that they exhibit strongly positive reaction with PAS and weakly eosinophilic property. These electron-dense granules, which are 0.2 to 0.6 $\mu$m in length, have oval shapes. 7. E-type of basophilic granule cells: These cells are found only in the parenchyma around re productive organ. The granules contained in a small number in the cell, exhibit PAS-positive reaction and have an average size of 0. 2pm. 8. Eosinophilic granule cells: These cells contain a large number of eosinophilic granules which have relatively diverse sizes from 0.3 x 0.2 to 0.8 x 0.4 $\mu$m. Most of granules are round or irregularly-shaped and highly electrondense. These cells have an array of well-developed granular endoplasmic reticulum of which cisternae are distened. 9. Transparent granule cells contain electron-lucent granules which exhibit negative reactions with three kinds of cytochemical methods used in this experiment.
The ultrastructure of the gills of Oplegnathus fasciatus was examined by means of light and transmission electron microscopes. The gills have primary and secondary filaments (lamellae). The following cells are identified and described : epithelial cell, pillar cell, chloride cell and mucose cell. The simple epithelial layer consists of squamous epithelium containing a large nucleus and the surface is covered with some of microridges. The lamella pillar structures are characterized by axial microtubules and lateral membrane interdigitations. Chloride cells contain a lot of mitochondria and specifically developed tubular systems. The rough endoplasimic reticulum and golgi complex, and some of mucous granules were observed in immature mucous cells. The mature mucous cells were AB-PAS positive, globular in shape, and had mucous granules of similar size with various electron densities.

It is important to obtain conn cytodiagnosis to classify background, cytoplasm, and nucleus from the diagnostic image. This study mose an algorithm that detects and classifies carcinoma cells of the uterine cervix in Pap smear using features of cervical cancer. It applies Median filter and Gaussian filter to get noise-removed nucleus area and also applies Kapur method in binarization of the resultant image. We apply 8-directional contour tracking algorithm and stretching technique to identify and revise clustered cells that often hinder to obtain correct analysis. The resulted nucleus area has distinguishable features such as cell size, integration rate, and directional coefficient from normal cells so that we can detect and classify carcinoma cells successfully. The experiment results show that the performance of the algorithm is competitive with human expert.

Increased mRNA levels of cancer upregulated gene (CUG)2 have been detected in many different tumor tissues using Affymetrix microarray. Oncogenic capability of the CUG2 gene has been further reported. However, the mechanism by which CUG2 overexpression promotes cancer stem cell (CSC)-like phenotypes remains unknown. With recent studies showing that pyruvate kinase muscle 2 (PKM2) is overexpressed in clinical tissues from gastric, lung, and cervical cancer patients, we hypothesized that PKM2 might play an important role in CSC-like phenotypes caused by CUG2 overexpression. The present study revealed that PKM2 protein levels and translocation of PKM2 into the nucleus were enhanced in CUG2-overexpressing lung carcinoma A549 and immortalized bronchial BEAS-2B cells than in control cells. Expression levels of c-Myc, CyclinD1, and PKM2 were increased in CUG2-overexpressing cells than in control cells. Furthermore, EGFR and ERK inhibitors as well as suppression of Yap1 and NEK2 expression reduced PKM2 protein levels. Interestingly, knockdown of β-catenin expression failed to reduce PKM2 protein levels. Furthermore, reduction of PKM2 expression with its siRNA hindered CSC-like phenotypes such as faster wound healing, aggressive transwell migration, and increased size/number of sphere formation. The introduction of mutant S37A PKM2-green fluorescence protein (GFP) into cells without ability to move to the nucleus did not confer CSC-like phenotypes, whereas forced expression of wild-type PKM2 promoted such phenotypes. Overall, CUG2-induced increase in the expression of nuclear PKM2 contributes to CSC-like phenotypes by upregulating c-Myc and CyclinD1 as a co-activator.
S6K1 is a key regulator of cell growth, cell size, and metabolism. Although the role of cytosolic S6K1 in cellular processes is well established, the function of S6K1 in the nucleus remains poorly understood. Our recent study has revealed that S6K1 is translocated into the nucleus upon adipogenic stimulus where it directly binds to and phosphorylates H2B at serine 36. Such phosphorylation promotes EZH2 recruitment and subsequent histone H3K27 trimethylation on the promoter of its target genes including Wnt6, Wnt10a, and Wnt10b, leading to repression of their expression. S6K1-mediated suppression of Wnt genes facilitates adipogenic differentiation through the expression of adipogenic transcription factors PPARγ and Cebpa. White adipose tissues from S6K1-deficient mice consistently exhibit marked reduction in H2BS36 phosphorylation (H2BS36p) and H3K27 trimethylation (H3K27me3), leading to enhanced expression of Wnt genes. In addition, expression levels of H2BS36p and H3K27me3 are highly elevated in white adipose tissues from mice fed on high-fat diet or from obese humans. These findings describe a novel role of S6K1 as a transcriptional regulator controlling an epigenetic network initiated by phosphorylation of H2B and trimethylation of H3, thus shutting off Wnt gene expression in early adipogenesis.
This experiment was carried out to observe the origin and developmental pattern of somatic embryos of Oenanthe javanica ($B^{L}.$) DC. The experiment included observation of embryogenic cells and their development stages by light microscope, transmission electron microscope and scanning electron microscope. The embryogenic cells, which were smaller than non-embryogenic cells in size with expanded nucleus and dense cytoplasm. When stained with hematoxylin, the embryogenic cells were readily distinguished from the non-embryogenic cells of which cell walls were stained with safranin. It was observed at somatic embryos developed from single cells on the epidermis of developing embryos or in the surface or inside of embryogenic clumps by segmentation pattern. Observation with a transmission electron microscope revealed that the embryogenic cells had dense cytoplasm expanded nucleus, small vacuoles, large amyloplasts containing starch grains, and abundant organelles including lipid bodies. Under a scanning electron microscope, embryogenic callus was shown to consist of very smaller cells than non-embryogenic cells in an orderly arrangement and covered with a net-like structure, while the non-embryogenic callus consisted of large cells, irregular in size and arrangement, and covered with a gelatin-like material.
Diethylnitrosamine (DEN) is known as a potential hepatic carcinogen by single administration. This study was designed to measure the effects of DEN-induced cell damage on the triglyceride and cholesterol concentration in the liver, excluding dietary effects. Fertilized chicken eggs, 10 days before hatching, were randomly divided into three groups (n=20) and each egg was injected 10 ${mu}ell$ of corn oil (vehicle control), 5 $\mu\textrm{g}$ of DEN/10 ${mu}ell$ of DEN/10 ${mu}ell$ into yolk via air sac. After 48 hr and 96 hr incubation, the damage of the chick-embryo liver cell was investigated by electron microscopy and by measuring the concentration of lipid components (total cholesterol, free cholesterol, phospholipid and triglyceride). For eggs administered 10 $\mu\textrm{g}$ of DEN and incuvated 96 hr, in hepatocyte, the nucleus membrane was roughed, the size of nucleolus was apparently increased and euchromatin was accumulated. Mitochondria were condensed and cristae, located mitochondiral inner membrane, were obscured. Additionally, the leaves of triglyceride and cholesterol classes were significantly increased depend on the amount treated with 10 $\mu\textrm{g}$ DEN at 96 hr, but phospholipids component of cell membrane, were decreased with significance. As a conclusion, carcinogen induced hepatic lesion was correlated with the changes in lipid component of liver.

Microbeam is a new avenue of radiation research especially in radiation biology and radiation protection. Selective irradiation of an ionizing particle to a targeted cell organelle may disclose such mechanisms as signal transaction among cell organelles and cell-to-cell communication in the processes toward an endpoint observed. Bystander effect, existence of which is clearly evidenced by application of the particle microbeam to biological experiments, suggests potential underestimation in the conventional risk estimation at low particle fluence rates, such as environment of space radiations in ISS (International Space Station). To promote these studies we started the construction of our microbeam facility (named as SPICE) to our HVEE Tandem accelerator (3.4 MeV proton and 5.1 MeV $^4$He$\^$2+/). For our primary goal, "irradiation of single particle to cell organelle within a position resolution of 2 micrometer in a reasonable irradiation time", special features are considered. Usage of a triplet Q magnet for focussing the beam to submicron of size is an outstanding feature compared to facilities of other institutes. Followings are other features: precise position control of cell dish holder, design of the cell dish, data acquisition of microscopic image of a cell organelle (cell nucleus) and data processing, a reliable particle detection, soft and hard wares to integrate all these related data, to control and irradiate exactly determined number of particles to a targeted spot.
Morphology of the gill in Lateolabrax japonicus was investigated after staining the gill, as a result, we found the gill is composed of gill raker, gill arch, gill filament and gill lamellae. The number of gill raker was 7~10 in the upper and 13~18 in the lower. Ultrastructure of the gill in Lateolabrax japonicus were examined by means of the light and transmission electron microscopes. The fine structure of the hemocytes in Wolf spider, Pardosa astrigera, is discribed and compared with that of similar cells in other spider species and insects. Five hemocyte types are identified in the hemolymph: prohemocyte, plasmatocyte, granulocyte, spherulocyte and adipohemocyte. Prohemocytes are small with a relatively undifferentiated cytoplasm. The nucleus is comparatively large and has a perinuclear space. Plasmatocytes and granulocytes are pinocytotic function in the hemolymph of the body. The plasmatocytes have some coated pits on the plasma membrane and well developed Golgi complex, The granulocytes appear sequence of events in the formation of coated vesicle from a coated pit on its plasma membrane. Golgi complex become well expressed and give rise to small secretory vesicles which fuse to large bodies. The spherulocytes are larger in cell size than other hemocytes. Their cytoplasm is filled with spherules. The spherules contain the floccurent materials and the helical structured materials, which are 220nm in length and 80nm in width. The adipohemocytes are oval shaped and have a number of lipid droplets.
