The influence of triploidization on cell and nucleus size characteristics of the same tissues of erythrocyte, retina, kidney, hepatocyte and midgut epithelium in marine medaka, Oryzias dancena has been determined histologically. Induced triploid fish are produced by cold shock treatments. Likewise, the size of horizontal cell nucleus in inner nuclear layer of retina, ganglion cell nucleus in ganglion cell layer of retina, proximal tubule cell of kidney, hepatocytes and nuclear height of midgut epithelium all appear to be significantly larger than diploid (P<0.05). On the other hand, retina thickness is larger in diploid than induced triploid (P<0.05). Induced triploid shows low density of cell number. Results of this study suggest that same characteristics in the induced triploid exhibiting larger cells and nucleus sizes with fewer number of cells than the diploid can be useful criteria for the distinction between diploid and induced triploid, and also the ploidy level in marine medaka.
Diploid and triploid hybrid between female mud loach Misgurnus mizolepis and male cyprinid loach, M. andguillicaudatus were produced, using artificial propagation. The influence of hybridization and triploidization on characteristics of the some tissues of retina optic tectum and trunk kidney in loach was determined histologically. Diploid hybrid have almost size similarities in nucleus and the cell size of some tissues to both parents. In the triploid hybrid the nucleus and/or the cell size in erythrocyte, horizontal cell in inner nuclear layer of retina ganglion cell in ganglion cell layer of retina, neuronal cell in granular layer of optic tectum, and secondary proximal tubule cell of trunk kidney, are much larger than those of the corresponding cells in the diploid hybrid individuals. However, triploid hybrid showed fewer number of cells in outer nuclear layer in retina and nuclei in secondary proximal tubule of trunk kidney than in diploid hybrid. Results of this study suggest that some characteristics in the triploid hybrid exhibiting larger cell and nucleus size with fewer number of cell than diploid hybrid can be useful criteria for the distinction between diploid hybrid and triploid hybrid, and the ploidy level in loach.
Ultrastructural and histochemical studies were carried out on the prostate gland of a Korean slug (Incilaria fruhstorferi) with light and electron microscopes. The results are as follows. The endothelial tissue of the prostate gland is constituted with tall and narrow ciliated columnar cells, irregularly shaped cells and gland cells in which are included the elongated oval or ellipsoid nucleus those are surrounded by curved membrane. The endothelial tissue of the prostate gland is composed by type-A, B, C and type-D gland cells, and the glanules of type-A, B, and D gland cells are certified to neutral mucopolysaccharide because are related by PAS-alcian blue (pH 2.5) , but the other hand type-C gland cell is only weakly reacted to PAS, but strongly reacted by Million reaction. The glanules of type-A gland cell are small size (about $0.4{\mu}m$) and are seen high electron density, but the glanules of type-B gland cell are large size (about $0.7{\mu}m$) and glanule density is same type-A glanules. Long ellipsoid type-C gland cell contained round nucleus which is well developed beterochromatin in, and that small oval glanules (size, about $0.9{\mu}m$) of moderate high electron density which are formed a group of large glanule together with $4\sim5ea$, but type-D gland cell possessed round small nucleus are seen high electron dense glanules (size, $0.8{\mu}m$).
Targeted radionuclide therapy (TRT) is a method of treating tumor cells using radiopharmaceuticals. Cells and nuclei constituting tissues of the human body are composed of spherical and oval shapes, but cancer cells are composed of various cell types. Therefore, this study analyzed the absorbed dose for each organelle according to the change in the size of the cell nucleus for beta-emitting nuclides during targeted radionuclide therapy through the Monte Carlo method. Cells were set in two sphere shapes, 5 ㎛ and 10 ㎛, and the internal structure was divided into cell nucleus, cytoplasm, and cell surface. Next, the absorbed dose according to the increase in the size of the cell nucleus was evaluated. As a result, 177Lu among the target radionuclides showed the highest dose in all cell compartments. As the ratio of the nucleus in the cell increased, the absorbed dose on the cell surface increased, but the absorbed dose in the cytoplasm and nucleus tended to decrease. Accordingly, it is judged that it is important to select a radionuclide considering the size of cancer cells and determine an appropriate amount of radioactivity during targeted radionuclide treatment.
To investigate ultrastructural characteristics of cancer cell nucleus, 29 cases of malignant tumors of 9 different types were examined. The common findings were marked irregularity of nuclear membrane with pronounced infoldings, clumping of heterochromatin along inner nuclear membrane, enlargement and disturbance of configuration and composition of nucleolus, and frequent observations of nuclear bodies and nuclear inclusions. These findings confirm what have been observed by light microscopy, and are also the signes that can be observed in hyperactive cells. Thus, ultrastructural characteristics of cancerous nucleus are the great variability of nuclear size, shape and composition, but none of them appear to be specific.
The image analyser was used for the measurement of the morphological changes of the spermatogenic cells and seminiferous tubules in the pheasant during the breeding and non breeding season. 1. The seminiferous tubules were enlarged 2.57 times during the breeding season than during the non breeding season. Only Sertoli cells and spermatogonia in the seminiferous tubules can be found during the non breeding season. 2. There is no significantly difference in the ratio of nucleus area against that of cell size in the spermatogonium between breeding and non breeding season. The ratio of area was 28.71% and 29.11%, respectively. However, the enlargement of spermatogonium was noticed during the non breeding season. 3. The highest value of the ratio of the nucleus area against that of cell size among the germ cells was measured 37.40% in the pachytene phase of the spermatocyte during the breeding season. 4. The ratio of nucleus area against that of cell size in the spermatid was 22.53%.
Acinous gland cells (A, B, C, D and E-type cells) and duct cell (G-type cell) are observed in acinus and in duct of salivary gland of Korean Slug respectively by electron microscope. The type-A gland cells are numerous and are packed with medium electron dense granules (diameter, $3{\mu}m$) in cytoplasm. The circular shaped nucleolus and evenly developed chromatins are observed in the nucleus of type-B cell, and cytoplasm includes medium electron dense granules (diameter, $2.5X3.7{\mu}m$). The type-C gland cell has a round nucleus, and thin elongated-shaped heterochromatins are evenly distributed in the nucleoplasm and many net shaped endoplasmic reticulums and oval serous granules of middle electron density (diameter, $3.5X5{\mu}m$) fill the cytoplasm. The type-D gland cell is the largest and the most numerous of the gland cells consisting the salivary gland and heterochromatins in nucleus are well developed in the nucleoplasm. Most of granules (diameter, $0.8X2.5{\mu}m$) in cytoplasm are round, and look dark for the high electron density, and cytoplasm is filled with net-shaped endoplasmic reticulums. The type-E gland cells are rarely existent around the salivary gland, and the granules of those cells are irregular in shape and size and are vacuolized in cytoplasm. Intralobular salivary duct is composed of the high electron dense squamus endotheliums, while the other interlobular salivary duct is filled with irregular columnar epitheliums. The interlobular duct cell contains the high electron dense granules (size, $0.3{\sim}1.5{\mu}m$) in cytoplasm and those granules are secreted into cilia of salivary lumen.
The posterior hypothalamus of the hibernating greater horseshoe bats (Rhinolophus ferrumequinum korai Kuroda) were observed with an electron microscope. The posterior hypothalamus is known to be closely related to the reflex responses activated by cold, and the following observations were obtained in the cellular type of nerve cells: there are three types of neurons in the posterior hypothalamus. 1. The first type of neuron was the largest, ovoid or conical in shape, the nucleus was elliptic and the nuclear envelope had many deep invaginations. The cell organelles were well developed, in particular there was an abundance of variously shaped mitochondria, and the Golgi complex and the polysomes were observed in the cytoplasm. 2. The second type of neuron was moderate in size, ovoid or elliptic in shape, the nucleus was located nearer to the plasma membrane and the nuclear envelope had. a few invaginations. The cytoplasm was rich in amount compared with that of the third type of neuron, and the cell organelles, especially the rough endoplasmic reticulum were well developed. Also lipofuscin pigments were observed. 3. The third type of neuron was the smallest in size and round in shape. The nucleus and the nucleolus were observed in the central portion of the cell body and the nuclear envelope had a few invaginations. The cytoplasm was small compared with those of the first and second types, but the rough endoplasmic reticulum, the mitechondria and the polysomes were relatively well developed. The cytoplasm was characterized by the presence of membrane-bound small bodies with a single membrane containing a fine particular substance around the rough endoplasmic reticulum and the Golgi complexes.
The morphological study on different types of cells of reproductive organ including spermatogenesis in the adult planaria was performed to observe their cytochemical and ultrastructural characteristics. 1. Spermatogenesis The circular luminated material appears immediately inside the nuclear envelope of early spermatid and is found also in the nucleus of sperm, but typical acrosomal structures cannot be observed. Approximately ten of small-sized mitochondria occur around the nucleus in the transitional phase from primary spermatocyte to secondary spermatocyte, but in sperm a long mitochondrion is closely associated with nucleus, parellel to long axis of it. The sperm has a relatively long head connected with two tails via hollow neck. 2. Reproductive organ The penis bulb and the bursa stalk were observed. (1) Penis bulb The cells constituted penis bulb are classified into six types on the basis of ultrastructure of the cells and cytochemistry of the cytoplasmic granules. 1) A-type cells: These cells exhibiting low electron density are mainly occupied by large nucleus. These cells possess two different types of granules: highly electron-dense round granules with an average size of $0.9{\mu}m$, and electron-dense granules exhibit PAS-positive reaction. 2) B-type cells contain PAS-positive granules with the size of about $0.4{\mu}m$. They are rich in free ribosomes and mitochondria. 3) C-type cells are found to be dark cells due to high electron-density. These cells are largely occupied by large nucleus. 4) D-type cells: These cells are seen as light cells which have poorly developed cell organelles. 5) E-type tells: These cells contain a large number of glycogen granules which occupy most of cell. 6) F-type cells: These arc parietal epidermal cells surrounding the genital antrum. These cells are characterized by their finger-like shapes and the presence of a number of electron-dense, irregularly-shaped structures inside cells. The relatively large electron-lucent granules can be also found. The F-type cells possess numerous microvilli on their free surfaces. (2) Bursa stalk The cells constituted bursa stalk are classified into 3 types on the basis of cell shapes and presences of electron-dense or electron-lucent granules. 7) G-type cells with a long cytoplasmic process. They have large nuclei and poorly developed cell organelles. 8) H-type cells: These cells are characterized by the presence of a long cytoplasmic process and relatively highly electron-dense cytoplasmic profile. They have poorly developed cell organelles. 9) I-type cells contain large electron-lucent granules which exhibit negative reactions with three kinds of cytochemical staining methods used in this experiment. The fine electron-dense structures can be found inside these granules.

The influence of triploidization on histological characteristics of retina, trunk kidney, liver and midgut tissue, and cell cycle of tail fin and gill tissue in far eastern catfish, Silurus asotus were analyzed. In the infertile triploid fish, the nucleus and/or cell size of secondary proximal tubule cells of trunk kidney, hepatocyte and midgut epithelium are much larger than those of the corresponding cells in the diploid fish (P<0.05). However, triploid tissue showed fewer number of outer nuclear layer in retina and nuclei in secondary proximal tubule of trunk kidney than those for diploid tissue. The mean percentages of the $G_l-$, the S- and the $G_2+M-phase$ fractions were 92.5%, 3.2% and 4.3% in tail fin tissue of diploid, and 93.4%, 2.6% and 4.0% in those of triploid, respectively. There were no significant differences in the percentages of each cell cycle fraction between diploid and triploid. The mean percentages of each phase fractions were 75.1%, 11.1% and 13.8% in gill tissue of diploid and 85.2%, 8.9% and 5.9% in those of triploid, respectively. The differences of cell cycle between tail fin tissue and gill tissue were statistically significant in diploid and triploid (P<0.05). Also, the differences between diploid and triploid were statistically significant in tail fin tissue and gill tissue (P<0.05). Cyclin D1 and cyclin E expressions were not significantly difference between gill tissue and tail fin tissue, and protein expressions of induced triploid were higher than those of diploid. Results from this study suggest that some characteristics in the triploid exhibiting larger cell and nucleus size with fewer number of cell than diploid can be used as an indicator in the identification of triploidization and ploidy level in far eastern catfish.
