Caveolin-1 inhibits membrane-type 1 matrix metalloproteinase activity

  • Membrane-type 1 matrix metalloproteinase (MT1-MMP) is a zinc-dependent proteinase found in cholesterol-rich lipid rafts on the plasma membrane. MT1-MMP hydrolyzes extracellular matrix (ECM) proteins, activates pro-matrix metalloproteinase-2 (proMMP-2) and plays an important role in ECM remodeling, cancer cell migration and metastasis. The role of caveolin-1, an integral protein of caveolae, in the activation of MT1-MMP remains largely unknown. Here, we show that the expression of caveolin-1 attenuates the activation of proMMP-2, reduces proteolytic cleavage of ECM and inhibits cell migration. We utilized the cytoplasmic tail domain deletion (${\Delta}CT$) or the E240A mutant of MT1-MMP. Co-expression of caveolin-1 with the wild-type or the ${\Delta}CT$ MT1-MMP decreased the proMMP-2 activation and inhibited collagen degradation and cell migration. Caveolin-1 had no effect on the catalytically inert E240A MT1-MMP. Our findings suggest that caveolin-1 is essential in the down-regulation of MT1-MMP activity by promoting internalization from the cell surface.

  • Manufacturing cell formation is one of the most important problems faced in designing cellular manufacturing systems. The purpose of this study is to design effective manufacturing cell systems by developing a method which forms machines/parts into optimal machine cells/part families. The 0-1 data matrix structure is used to form a basis for manufacturing cell formation. In this paper, we propose a CE method to reorder the 0-1 data matrix for manufacturing cell formation. The resulting solutions are shown to demonstrate the effectiveness of the CE method.

Fluorescent and bioluminescent nanoprobes for in vitro and in vivo detection of matrix metalloproteinase activity

  • Matrix metalloproteinases (MMPs) are zinc-dependent endopeptidases that degrade the extracellular matrix (ECM) and regulate the extracellular microenvironment. Despite the significant role that MMP activity plays in cell-cell and cell-ECM interactions, migration, and differentiation, analyses of MMPs in vitro and in vivo have relied upon their abundance using conventional immunoassays, rather than their enzymatic activities. To resolve this issue, diverse nanoprobes have emerged and proven useful as effective activity-based detection tools. Here, we review the recent advances in luminescent nanoprobes and their applications in in vitro diagnosis and in vivo imaging of MMP activity. Nanoprobes with the purpose of sensing MMP activity consist of recognition and detection units, which include MMP-specific substrates and luminescent (fluorescent or bioluminescent) nanoparticles, respectively. With further research into improvement of the optical performance, it is anticipated that luminescent nanoprobes will have great potential for the study of the functional roles of proteases in cancer biology and nanomedicine. Effect of Preparation Methods of a Matrix Retaining Electrolyte on the Characteristics of a Phosphoric Acid Fuel Cell (인산형 연료전지(PAFC)용 전해질 매트릭스의 제조방법이 전극/매트릭스 계면특성에 미치는 영향)

  • The matrices which consisted of SiC whisker, PES(polyesterasulfone) as a binder, span 80(sorbitan monooleate) as a surfactant, TPP(triphenyl phosphate) as a plasticizer and dichloromethane as a solvent, have been prepared by the various methods such as tape casting, rolling, tape cast-coating and roll-coating method. The fuel cells of single stack type using these matrices are characterized by ac impedance spectroscopy and cyclic voltammetry technique. A fuel cell using a matrix prepared by the tape cast-coating method shows the best performance of 466.34 mA/$\textrm{cm}^2$ at 0.6V because it has the lowest polarization resistance at the interface between electrodes and a matrix due to the largest three phase contact region of gases, catalyst and electrolyte.

Sesquicillin, an Extracellular Matrix Adhesion Inhibitor, Inhibits the Invasion of B16 Melanoma Cells In vitro

  • Tumor cell interaction with the extracellular matrix is defined as the critical event of tumor invasion that signals the initiation of a metastatic cascade. Sesquicillin has been identified as an inhibitor of melanoma cell adhesion to the components of the extracellular matrix (ECM) in cultured broth of fungal strain F60063. Sesquicillin strongly inhibited the adhesion of B16 melanoma cells to laminin, fibronectin, and typeIV collagen. It also inhibited B16 melanoma cell invasion of reconstituted basement membrane Matrigel in vitro in a dose-dependent manner. These results suggest that sesquicillin is a new class of nonpeptidic ECM adhesion inhibitor having anti-invasive activity.

  • Curcumin has been known for its anti-proliferative and apoptotic effects on several cancer cells. We examined the inhibitory effects of curcumin on cancer cell adhesion, motility, invasion and matrix metalloproteinase-9 (MMP-9) activity in MDA-MB-231 human breast cancer cells. MDA-MB-231 cells were cultured with 0, 5, 10 or $20{\mu}M$ of curcumin. Curcumin significantly inhibited the adhesion of cancer cells to the fibronectin at $20{\mu}M$ and suppressed the motility and invasion of cancer cells at all concentrations. Also, the MMP-9 activity was inhibited by curcumin, but MMP-9 protein amounts were not affected. Our data indicate that curcumin inhibits motility, invasion and MMP-9 activity of MDA-MB-231 cells. Therefore, curcumin may contribute to the potential beneficial food component to prevent the cancer metastasis in human breast cancer.

Antimetastatic Effects of Capsaicin in Murine B16 Melanoma Cell Lines

  • The present work was undertaken to evaluate the antimetastatic potential of capsaicin (8­methyl-N-vanillyl-6-nonenamide) by measuring its effects on matrix metalloproteinase activity, cell invasion and lung metastasis. Significant inhibition of matrix metalloproteinase-2 activity by capsaicin (100 $\mu$M) was detected by gelatin zymography. In vitro invasion assay showed capsaicin (50, 100 $\mu$M) reduced tumor cell invasion ($28-40\%$). Capsaicin (i.p., 2.5 mg/kg) inhibited development of lung colonization ($58\%$). These results suggest that capsaicin prevents metastasis in part through suppression of invasion of B16F10 melanoma cells by inhibiting matrix metalloproteinase-2 responsible for degradation of extracellular matrix.

  • Tumor cell invasion and metastasis are a complex multistep process that involves the degradation of extracellular matrix proteins by matrix metalloproteinases (MMPs). Tissue inhibitor of metalloproteinase-1 (TIMP-1) acts as a negative regulator of matrix metalloproteinase and thus prevents tumor cell invasion and metastasis by preserving extracellular matrix integrity.(omitted)

  • Effective properties of ceramic matrix plain woven textile composites were calculated using finite element analysis. The considered geometry is a unit cell of plain weave and the analysis was performed by commercial finite element program, ANSYS. The materials for analysis are 3 types for matrix, 1 type for fiber with various volume fraction. The result indicates that the effective properties of ceramic matrix composites can be controlled by the volume fraction. The result can be used for numerical analysis using ceramic matrix composites.

A Study on the Competitive Position and Strategic Exportable Goods of Korea Focused on Korea-China-Japan Fisheries Products Class (한.중.일 수산물부문에 있어 한국의 경쟁력 수준과 수출전략품목 분석에 관한 연구)

  • This study examines the relative competitive position of korean fisheries products market over period of 2001 to 2005 and selects strategic exported goods from its position provide against concluding FTA agreement with China and Japan. The portfolio approach is used to develope competitiveness-market share matrix. The position of each export countries on the competitiveness market share matrix will be in one of nine cells, with differing implications for their role in korean fisheries products market. Based the competitiveness market share matrix, each export countries are divided into first cell type, third cell type and ninth cell type and the items of ninth cell type are chosen as strategic exportable goods. The results of this study are summarized as follows: First, in the case of each country change aspect, China is trending to decrease quantity but shows number of item that increase gradually with high share still, and look trend that increase third cell type item too gradually, and in case of first cell type item is that competitive position is high more relatively than the Korea. In the case of Japan, ninth cell type item is falling gradually, and share does not show big change generally in case of first cell type item. Second, in the case of strategic exportable goods that analyze using domestic competitive position cell type and MCA with competitive position in domestic fisheries products market and export market, was appear by codfish(frozen), cuttle fish(frozen) etc. in case with China, and by mackerel(frozen), other sea bream(frozen), laver(dry), bathing(dry) etc. in case with Japan. And analyzed goods that have all export competitive advantages in both countries are roes of alaska pollack(frozen), other roes of fish(except frozen roes of alaska pollack), squid(frozen) etc.
