IL -12 Expression by Cefodizime As an Immuno-modulator

  • Cefodizime has originally been developed for treating infections as antibiotics. However. according to some of recent studies. cefodizime. a third generation cephalosporin. may potentially have the capability of stimulating chemotactic activity of neutrophils and monocytes as well as the strong immuno-modulator. In this study. we studied to learn about the expressive effect of dentritic cells and macrophage. With this background. We have studied to see if cefodizime can be a potential substance inducing an immunological function in dendritic cells and peritoneal macrophages. IL-12 activates NK cell and macrophage, and shows antiviral effect by excreting INF-${\gamma}$. In vitro. total RNAs were extracted from murine dentritic cell at 4, 8, 12, 24hr after the application of 10, 50, 100${\gamma}g$/ml of cefodizime wighout other stimulators. And we analyzed IL-12 mRNA using RT-PCR method. In conclusion. IL-12 mRNA was increased. and the results suggest that cefodizime activate TH1 cell induction, CTL differentiation as well as accelerating the increase of NK. LAK cell.

  • According to recent reports, cefodizime (CEF), a third generation cephalosporin has the capability of chemotactic activity of neutrophils and monocytes and may act as the strong immuno-modulator. This study was planned to demonstrate whether CEF has the proposed effect on rat dendritic cells in vitro. Dendritic cells were taken from rat spleen tissue and cultured for a week. The obtained dendritic cells were treated with 10$\mu$g/ml, 50$\mu$g/ml, 100$\mu$g/ml cefodizime and 10IU/$m\ell$ IFN-Υ+1$\mu$g/ml LPS. (omitted)

  • The in vitro inactivation of gentamicin and tobramycin by four cephalosporins (cefotetan, cefuroxime, cefodizime, cefotiam) in human serum was investigated. Each cephalosporin was added to human serum samples containing gentamicin sulfate or tobramycin sulfate. Blank samples containing only aminoglycosides were used as controls. Samples were stored at -20, 4 and $25^{\circ}C$ and were analyzed for aminoglycoside concentrations by fluorescence polarization immunoassay ($TDxFLx^{TM}$ system) at 0, 2, 4, 8, 12, 24, 48 and 72 hours after mixing. The serum containing cefotiam stored at $25^{\circ}C$ showed significant inactivation of gentamicin by $12\%$ at 72 hours. The results indicate that cefotitan, cefuroxime and cefodizime do not inactivate gentamicin and tobramycin while cefotiam inactivates gentamicin.
