We displayed four types of Solanum nigrum metallothionein (SMT) for the first time on the surface of Saccharomyces cerevisiae using an α-agglutinin-based display system. The SMT genes were amplified by RT-PCR. The plasmid pYES2 was used to construct the expression vector. Transformed yeast strains were confirmed by PCR amplification and custom sequencing. Surface-expressed metallothioneins were indirectly indicated by the enhanced cadmium sorption capacity. Flame atomic absorption spectrophotometry was used to examine the concentration of Cd2+ in this study. The transformed yeast strains showed much higher resistance ability to Cd2+ compared with the control. Strikingly, their Cd2+ accumulation was almost twice as much as that of the wild-type yeast cells. Furthermore, surface-engineered yeast strains could effectively adsorb ultra-trace cadmium and accumulate Cd2+ under a wide range of pH levels, from 3 to 7, without disturbing the Cu2+ and Hg2+. Four types of surfaceengineered Saccharomyces cerevisiae strains were constructed and they could be used to purify Cd2+-contaminated water and adsorb ultra-trace cadmium effectively. The surface-engineered Saccharomyces cerevisiae strains would be useful tools for the bioremediation and biosorption of environmental cadmium contaminants.
The removal of Cd(II) and Cu(II) from aqueous solution by an agricultural solid waste biomass prepared from Moringa oleifera bark (MOB) was investigated. The biosorbent was characterized by Fourier transform infrared spectroscopy and elemental analysis. Furthermore, the effect of initial pH, contact time, biosorbent dosage, initial metal ion concentration and temperature on the biosorption of Cd(II) and Cu(II) were studied using the batch sorption technique. Kinetic studies indicated that the biosorption process of the metal ions followed the pseudo-second order model. The biosorption data was analyzed by the Langmuir, Freundlich, Dubinin-Radushkevich, and Temkin isotherm models. Based on the Langmuir isotherm, the maximum biosorption capacities for Cd(II) and Cu(II) onto MOB were 39.41 and 36.59 mg/g at 323 K, respectively. The thermodynamic parameters, Gibbs free energy (${\Delta}G^o$), enthalpy (${\Delta}H^o$), and entropy (${\Delta}S^o$) changes, were also calculated, and the values indicated that the biosorption process was endothermic, spontaneous and feasible in the temperature range of 303-323 K. It was concluded that MOB powder can be used as an effective, low cost, and environmentally friendly biosorbent for the removal of Cd(II) and Cu(II) ions from aqueous solution.

In this research, biological uptake of heavy metals(Cd(II), Cu(II), Zn(II)) was measured under various conditions ; pH, initial heavy metal concentration, temperature, contact time and the amount of biomass through batch test. From this research, it was found that heavy metals might be removed through adsorption and accumulation in activated sludge process. Heavy metals were highly concentrated by microbial floc in activated sludge. Also, the removal efficiency was reached up to 80~90% within and after 1 hour the increase of removal efficiency was minimal. The order of accumulation efficiency was Cu(II)>Zn(II)>Cd(II), and the bonding strength between heavy metals and microbial floc may be expressed in order of Cu(II)>Zn(II)>Cd(II).
The concentrations and wet deposition flux into the sea of heavy metals of precipitation in Pusan area were measured and estimated. The samples were collected by polyethylene bottle(30ι) from January to November in 1996, and heavy metals were analyzed by atomic absorption spectrometer. The concentration order of heavy metals was Al >Fe >Zn >Pb >Mn >Cu >Ni >Cd >Co, and they were high at inland sites and low at coastal sites. The enrichment factors for some metals(Zn, Cu, Pb, Cd), based on crustal Al, were significantly greater than unity, and the order was Cd > Pb > Zn > Cu. This evidence suggests Cd and Pb are derived predominantly from non-crustal sources. Al, Fe and Mn contents showed good correlation with each other. Therefore this enrichment factor indicates similar geochemical behavior of these elements. The annual wet depositional flux(mg/ $m^2$ /yr) from Pl site was as follows: Al(121.1). Fe,(177.2), Zn(12.9), Mn(6.19), Pb(14.4), Cu(0.64), Ni(1.03), Cd(1.02) and Co(1.01).
The E.M.F of the galvanic cell with fused salt was measured to determine the activities of zinc at 700-820K over the entire composition range of liquid Zn-Cd alloys. The cell used was as follows: (-) W | Zn(pure) $Zn^{2+}(KCI-LiCl)$ | Zn(in Zn-Cd alloy) | W (+) The activities of zinc in the alloys showed positive deviation from Raoult's law over the entire composition range. The activity of cadmium and some thermodynamic functions such as Gibbs free energy, enthalpy, entropy were derived from the results by the thermodynamic relationship. The comparison of the results and the literature data was made. The liquid Zn-Cd alloy is found to be close to the regular solution. The concentration fluctuations in long wavelength limit, $S_{cc}(o)$, in the liquid alloy was calculated from the results.
To improve the solubility and dissolution rate of acyclovir (ACV), which is low oral bioavailability due to its properties of slight solubility in water and incomplete gastrointestinal absorption, the solid inclusion complexes of ACV with ${\alpha}CD$, ${\beta}CD$, $DM{\beta}CD$ in molar ratio of 1:1 were prepared by the freeze-drying method. The inclusion complexes were investigated by solubility study, UV, IR and DSC. The dissolution rate of ACV was significantly increased by ACV-CDs inclusion complex formation in artificial intestinal fluid at pH 6.8. The enhanced dissolution rate of ACV could be due to an increase of solubility and the formation of an amorphous structures through inclusion complexation with CDs. Especially, $ACV-DM{\beta}CD$ inclusion complex enhanced the maximum plasma concentration levels and AUC following oral administration compared to those of ACV alone. The present results suggest that $ACV-DM{\beta}CD$ inclusion complex serves as a potential carrier for improving the solubility, the dissolution rate and the bioavailability of ACV.
The possibility of heavy metal pollution by contaminated roadside soils was studied under controlled conditions. The soil samples from roadside and those from a retention pond consisting of settling particles were characterized by the XRD analyses and the sequential extraction experiments. Characterization by sequential extraction, for roadside soil indicates elevate total concentrations of heavy metals. The leaching behavior of the samples under different pH and time conditions were also studied. Differences between both types of samples result mainly from the buffering effect of carbonates, present in roadside soils and lacking from settling particles. Acid leaching of the settling particles is equivalent to the sum of FI+FII+FIII, while the amounts leached from roadside soil are lower probably from kinetic reasons. The buffering effects of carbonates were found to greatly delay the onset of the leaching reactions and the extent of dissolution in most metals except for Ca and Mn. The study of leaching kinetics at pH of 6.5 and 5 showed that Cd and Zn reached the maximum possible concentration within 3 days, while Pb did not show any sign of dissolution at both ph values. The absolute amounts of dissolved Cd and Zn increased by 7 to 9 times by decreasing the pH from 6.5 to 5, indicating slightly acidified rain may result in significant metal dissolution. As deduced from both sequential extraction and leaching experiments, the relative mobility of heavy metals is found to be : Mn=Cd>Zn>>Pb>Fe, in spite of large differences in heavy metal contents and localizations.
The interaction of cadmium (II) ion with quercetin, qurecitrin and (+)-catechin was investigated in aqueous solution at various pH. The flavonoid/cadmium stochiometries for cadmium (II) binding to flavonoid have been determined by UV-visible spectroscopy. 1:1 Cd(II)-Flavonoid complex had a maximum absorbance and showed the bathochromic shift of the long-wavelength band of the UV-vis spectra in the alkaline pH, that occurs upon complexation, is due to a ligand-tometal charge transfer. The optimal concentration of Cd(II)-flavonoid complexes showed that complexation reaction could be classified in the following way: 55.27 ${\mu}M$ catechin > 54.72 ${\mu}M$ quercetin > 53.52 ${\mu}M$ quercitrin at the chelating site level. These results suggest that Cd(II)-flavonoid complex has the optimal condition of chelation in 0.2 M $NH_3$ - 0.2 M $NH_4Cl$ (pH 8.0) solution.
Using a murine T cell hybridoma, activation-induced cell death (AICD) was studied. As an in vitro model system for the AICD, 1 cell hybridoma expressing TCR/CD3 complex was incubated onto the immobilized purified anti-CD3 antibody. The immobilized anti-CD3 antibody induced AICD effectively up to 40%. At 1-100 $\mu$M range of SNP, an exogenous source of nitric oxide (NO), the cell proliferation was not affected, but at 1 mM SNP, cell proliferation was significantly reduced. The AICD of T cell hybridoma was inhibited by exogenous NO at non-cytotoxic concentration, In the cells undergoing AICD, the expressions of caspase-3 and FasL were detected, but not iNOS. Similar result was recognized in the apoptosis induced by dexamethasone, an apoptosis-inducing agent. However, the conversion from the inactive form of caspase-3 (32 kDa) to the active form (17 kDa) was significantly reduced in the cells in AICD induced by anti-CD3 antibody, With the result of increased PARP cleavage in the cells, we propose that another PARP cleavage pathway not involving caspase-3 may function in the anti-CD3 antibody induced AICD in the T cell hybridoma.
Salt, as food, is the most essential element for human survival due to its significant physiological functions. Here, we report the simultaneous detection of Pb and Cd in sea salt by square wave anodic stripping voltammetry (SWASV). Stripping voltammetric measurements were conducted using a manufactured rotating disk electrode system (MRDES). The detection limit was $3.6{\pm}0.18{\mu}gL^{-1}$ for Pb and $3.9{\pm}0.37{\mu}gL^{-1}$ Cd in NaCl solution. When the pH increased from 5.5 to 8.5, the peak currents of Pb and Cd decreased. At a pH of 8.3, the ratio of the current drop compared with that at a pH of 5.5 was 0.6 for Pb and 0.73 for Cd. The concentrations corrected by the current drop are in agreement with the concentrations obtained with ICP (inductively coupled plasma). This system demonstrates the reliable detection of heavy metals in aqueous media and, at a high $Na^+$ concentration, the successful application for the determination of Pb and Cd in sea salts.
