Functional and Structural Characterization of Drosocin and its Derivatives Linked O-GalNAc at Thr11 Residue

  • Antimicrobial peptides have recently gained the much attention because of their ability to make defense system from attacking bacterial infections. Drosocin has been considered as very attractive antibiotic agents because of low toxicity against human erythrocytes and active at the low concentration. We have studied the structureactivity relationship of a glycopeptide drosocin focused on the N-acetyl-D-galactoside at $Thr^{11}$ residue. Based on the radial diffusion assay, we found that the acetylation of carbohydrate moiety increased the antimicrobial activity and the $Pro^{10}$, present in the middle of drosocin plays an important role in the antimicrobial activity. Our results provide a good lead compound for further studies on the design of drosocin-based analogues targeting glyco linked Thr site.

Photodegradation of Halogen Derivatives of Aliphatic Hydrocarbon in Aqueous Photocatalytic Suspensions (지방족 탄화수소의 할로겐 유도체 수용액의 광촉매-광분해)

  • The rates of photodegradation, reactivities, and mechanisms of photooxidation for the aqueous solution containing with halogen derivatives of aliphatic hydrocarbons have been discussed with respect to the kinds of photocatalysts, concentration of photocatalytlc suspensions, strength of radiant power, time of illumination, changes of pH of substrate solution, wavelength of radiation, and pressure of oxygen gas saturated In the solution. These aqueous solutions suspended with 0.5 $gL^{-1}$ $TiO_2$ powder have been photodecomposed in the range of 100 and 93.8% per 1 hour if it is illuminated with wavelength (λ $\geq$ 300nm) produced from Xe-lamp(450W). The photocatalytic abilities have been increased In the order of $Fe_2O_3$ < CdS < $CeO_2$ < Y_2O_3$ <$TiO_2$, and rates of photodegradation for the solution have maldmum values in the condition of pH 6 ~ 8 and 3 psi-$O_2$ gL^{-1}$. These rates for the Photoolddation Per 1 hour were dependent on the size of molecular weight and chemical bonding for organic halogen compounds and the rates of photodegadation were increased in the order of $C_2H_5Br$ < CH_2Br_2$ < C_5H_11Cl C_2H_4Cl_2$ < tracts-$C_2H_2Cl_2$ < cis-C_2H_2Cl_2$ The T_{1/2}$ and t99% for these solutions were 5~21 and 40~90 minutes. respectively, and these values were coincided with Initial reaction kinetics(ro). It was found that reaction of photodegradation has the pseudo first-order kinetics controlled by the amount of $h^+_{VB}$ diffused from a surface of photocatalysts.

Phytoremediation of Heavy-Metal-Contaminated Soil in a Reclaimed Dredging Area Using Alnus Species

  • To investigate the possible applications of plants to remediate heavy-metal-contaminated soil, a pilot experiment was performed for four years in a reclaimed dredging area using two Alnus species, i.e., Alnus firma and Alnus hirsuta. In a comparison of phytomass of the two species at two different planting densities, the phytomass of Alnus planted at low density was twice as high as that of Alnus planted at high density after four years. The Alnus species showed active acclimation to the heavy-metal-contaminated soil in a reclaimed dredging area. A. hirsuta showed greater accumulation of phytomass than A. firma, indicating that it is the better candidate for the phytoremediation of heavy-metal-contaminated soils. In the pilot system, Alnus plants took metals up from the soil in the following order; Pb > Zn > Cu > Cr > As > Cd. Uptake rates of heavy metals per individual phytomass was higher for Alnus spp. planted at low density than those planted at high density in the pilot system. Low plant density resulted in higher heavy metal uptake per plant, but the total heavy metal concentration was not different for plants planted at low and high density, suggesting that the plant density effect might not be important with regard to total uptake by plants. The quantity of leached heavy metals below ground was far in excess of that taken up by plants, indicating that an alternative measurement is required for the removal of heavy metals that have leached into ground water and deeper soil. We conclude that Alnus species are potential candidates for phytoremediation of heavy-metal- contaminated surface soil in a reclaimed dredging area.

Mineralogical Studies for the Standadization of the Kaolinitum (백석지(白石脂)의 품질표준(品質標準) 설정(設定)을 위한 약용광물학적(藥用鑛物學的) 연구(硏究))

  • Objectives : This study was carried out to characterize the 'Kaolinitum' used as mineral medicine before and after physical and chemical manipulation. Methods : The mineralogy of the original raw kaolinitumes with different localities was identified by X-ray diffraction analysis. The concentrations of major, minor and trace elements of samples, furthermore, measured by the X-ray fluorescence spectrometer (XRF) and inductively coupled plasma-mass spectrometer (ICP-MS). In order to understand the variety of kaolinitum after thermal treatment, the differential thermal analysis (DTA) and the thermo-gravimetric analysis (TGA) were carried out. Results : According to the result of X-ray diffraction analysis, the kaolinitumes are composed ofcalcite, alunite and quartz. Si and Al are the major ions and heavy metals such as Cd, Cr, Pb, Zn, As, Ni and Cu are included in the kaolinitumes as trace ions. The concentration of these metals in the samples decreased after physical and chemical manipulation. The weight of kaolinitum was also decreased at $500^{\circ}C$ and about $700^{\circ}C$during experiment of TGA. Conclusion : The results of this study can be used to provide the scientific basis of the medical usage of kaolinitum.

  • This study was aimed ultimately to develop an adsorption process treating heavy metal industrial wastewater by utilizing domestically abundant natural zeolite and the study was conducted in a series of investigations. Presented if1 this paper are the results of the preliminary batch mode test. Factors affecting an adsorption process of heavy metals of aqueous waste stream by zeolite are numerous. Factors such as hydrogen ion concentration and temperature are taken into consideration in the investigation to evaluate adsorptive capacity. The mechanisms of adsorption may better be described by an evaluation of adsorption isotherm andi of adsorption kinetics. It is observed from the preliminary investigation that an optimum adsorption occurs at higher pH's than 4. It is further demonstrated that $Cd^{+2}$ adsorption by zeolite follows the BET model better than the Freundlich and the Langmuir model and that the reaction time of at least 10 minutes is required. It is interesting to note that higher adsorptive capacity was found at higher temperature, suggesting that the adsorption is not only due to simple physisorption but also due to chemisorption.

Ginsenoside Rd alleviates mouse acute renal ischemia/reperfusion injury by modulating macrophage phenotype

  • Background: Ginsenoside Rd (GSRd), a main component of the root of Panax ginseng, exhibits anti-inflammation functions and decreases infarct size in many injuries and ischemia diseases such as focal cerebral ischemia. M1 Macrophages are regarded as one of the key inflammatory cells having functions for disease progression. Methods: To investigate the effect of GSRd on renal ischemia/reperfusion injury (IRI) and macrophage functional status, and their regulatory role on mouse polarized macrophages in vitro, GSRd (10-100 mg/kg) and vehicle were applied to mice 30 min before renal IRI modeling. Renal functions were reflected by blood serum creatinine and blood urea nitrogen level and histopathological examination. M1 polarized macrophages infiltration was identified by flow cytometry analysis and immunofluorescence staining with $CD11b^+$, $iNOS^+$/interleukin-12/tumor necrosis factor-${\alpha}$ labeling. For the in vitro study, GSRd ($10-100{\mu}g/mL$) and vehicle were added in the culture medium of M1 macrophages to assess their regulatory function on polarization phenotype. Results: In vivo data showed a protective role of GSRd at 50 mg/kg on Day 3. Serum level of serum creatinine and blood urea nitrogen significantly dropped compared with other groups. Reduced renal tissue damage and M1 macrophage infiltration showed on hematoxylin-eosin staining and flow cytometry and immunofluorescence staining confirmed this improvement. With GSRd administration, in vitro cultured M1 macrophages secreted less inflammatory cytokines such as interleukin-12 and tumor necrosis factor-${\alpha}$. Furthermore, macrophage polarization-related pancake-like morphology gradually changed along with increasing concentration of GSRd in the medium. Conclusion: These findings demonstrate that GSRd possess a protective function against renal ischemia/reperfusion injury via downregulating M1 macrophage polarization.

Estimation of polycyclic aromatic hydrocarbons emission from sewage sludge of sewerage treatment plants in Korea

  • Polycyclic aromatic hydrocarbons (PARs) are an important group of organic contaminants present in sewage sludge, due to their persistence, toxic, bioaccumulative and long range transfer. These characters make themselves as Persistent Organic Pollutants(POPs) in Long Range Transboundary Air Pollutions convention(LRT AP) of Europe. A method of the gas chromatographic-mass spectrometric (GC-MS) determination of PARs present in sewage sludge was developed and applied to analyzed samples from five sewerage treatment plants (SWTPs), having different treatment types. PARs were extracted from freeze-dried samples by toluene 16 hours in a soxhlet extraction system. The sludge extracts were cleaned-up by an activated silica gel column chromatography. The sum of the 16 US Environmental Protection Agency PARs sewage sludge samples varied from 2.44 to 4.82 ${\mu}g$/g. Concentration of emission carcinogen PARs(PARcarc), such as Benzo(a)anthracene, Benzo(b)fluoranthene, Benzo(k)fluoranthene, Benzo(a)pyrene, Dibenzo(a, h)anthracene and Indeno(1, 2, 3-cd)pyrene ranged from 0.62 to 1.03 ${\mu}g$/g. The total amount of PAHs emission from sewage sludge in Korea was calculated as a top-down approach. PARs and $\sum$PAHcarc from sewage treatment plants had several pathway each by-products. In the ocean dumping, PAHs and $\sum$PAHcarc emissions were 1155.95 kg/year and 5040.32 kg/year. In recycle, PAHs and $\sum$PAHcarc emissions were 98.36 kg/year and 428.87 kg/year. In the landfill, PAHs and $\sum$PAHcarc emissions were 190.40 kg/year and 830.21 kg/year. In the incineration, PAHs and $\sum$PAHcarc emission were 33.10 kg/year and 830.21 kg/year. (In case of incineration, the whole provisions of PARs and $\sum$PAHcarc contained to flowed in sludge was supposed to be exhausted to environment through exhaust after incineration.)

  • In construction of a large cross section NATM tunnel, to keep the tunnel face stability by the ground itself bench cut method is commonly used. In order to necessity of partial face excavation method, we have to look for more enhanced method that can maintain better stress intensity. This paper presents a stress distribution of the Center Diaphragm Method from the partial face excavation methods, with the numerical analysis, and induced the optimal face distance, which is minimizing stress concentration and the optimal excavation step. Commerical 3 dimensional continuum analyzing FLAC-3D Ver. 2.1 program is used for the analysis. Analyses were performed to investigate ground behavior for tunnels with variable bench-length varying from 2m to 40m.

EFFECTS OF bFGF AND PDGF-BB ON OSTEOBLAST DIFFERENTIATION OF BONE MARROW-DERIVED MESENCHYMAL STEM CELL IN RAT (bFGF, PDGF-BB가 백서 골수기원 간엽 줄기세포의 조직골세포 분화에 미치는 영향에 관한 연구)

  • In this study we evaluate the effects of bFGF-BB and PDGF on in vitro proliferation, differentiation and mineralization of mesenchymal stem cells (MSCs) from rat. MSCs were prepared from the bone marrow of 6 or 7-week-old male rats with a technique previously described by Maniatopoulos et al. in 1988. Lineage differentiation to osteogenesis, chondrogenesis and adipogenesis were performed. At first, we characterized the cultured cell on passage 1, 3, 5, 7 with immunocytochemical staining using CD29, 44, 34, 45, ${\alpha}$-SMA and type I collagen. And to study the effects of bFGF and PDGF-BB on proliferation, differentiation and mineralization, we seeded the expanded cell at a density of 6 $6{\times}10^3\;cells/cm^2$ to 100-mm dish for evaluation of cell proliferation and MTT assay was carried out on day 2, 4, 7, 9. We also resuspended the cells with same density $(6{\times}10^3\;cells/cm^2)$ to 24 well plates for subculture. On the following day, the attached cells were exposed to 2.5ng/ml bFGF and/or 25ng/ml PDGF-BB daily during 5 days. The osteocalcin (OC) level was assessed and mineral contents were evaluated with alizarin red S staining on subculture day 2, 7, 14, 21. We identified the mesenchymal stem cell from the bone marrow derived cells of rat through their successful multi-differentiation and stable display of its phenotype. And bFGF and PDGF-BB showed the effect that inhibited osteoblastic differentiation and mineralization mildly in above concentration at in vitro culture. Characterization of β-Glucosidase Produced by the White Rot Fungus Flammulina velutipes

  • β-Glucosidase production by the white rot fungus Flammulina velutipes CFK 3111 was evaluated using different carbon and nitrogen sources under submerged fermentation. Maximal extracellular enzyme production was 1.6 U/ml, corresponding to a culture grown in sucrose 40 g/land asparagine 10 g/l. High production yield was also obtained with glucose 10 g/land asparagine 4 g/l medium (0.5 U/ml). Parameters affecting the enzyme activity were studied using p-nitrophenyl-β-D-glucopyranoside as the substrate. Optimal activity was found at 50℃ and pHs 5.0 to 6.0. Under these conditions, β-glucosidase retained 25% of its initial activity after 12 h of incubation and exhibited a half-life of 5 h. The addition of MgCl2, urea, and ethanol enhanced the β-glucosidase activity up to 47%, whereas FeCl2, CuSO4, Cd(NO3)2, and cetyltrimethylammonium bromide inflicted a strong inhibitory effect. Glucose and cellobiose also showed an inhibitory effect on the β-glucosidase activity in a concentration-dependent manner. The enzyme had an estimated molecular mass of 75 kDa. To the best of our knowledge, F. velutipes CFK 3111 β-glucosidase production is amongst the highest reported to date, in a basidiomycetous fungus.
