Establishment for Improving Productivity of Cattle by Fecal Steroid and Milk Urea Nitrogen analysis - II. Application of Early Pregnancy Diagnosis and Monitoring Postpartum Ovarian Activity

  • Progesterone levels in the blood plasma or skim milk of cows are considered to be very useful indicator fur the detection of estrus cycle and early pregnancy diagnosis. During 13 to 14 days after estrus the level of progesterone in plasma or skim milk were not different between the inseminated arid non-inseminated cows. In the pregnant cows the peak level of progesterone reached on 14th day after artificial insemination (AI), but in the absence of conceptus the level declines after the 14th day slowly, and then very rapidly towards the basal level after the 17th day. This low level persists about 4 days, including those of estrus and ovulation a highly characteristic pattern which differs so markedly from that in the pregnant cows. Progesterone levels in blood plasma or skim milk can provided a reliable diagnosis of early pregnancy and monitoring ovarian activity in cows. The mean $\pm$ standard deviation of milk urea nitrogen(MUN) and protein concentration in the cows at 9 herds were 17.7$\pm$2.35 mg/dL and 3.2$\pm$0.17%, respectively. The days of nonpregnant after parturition was shorter in the cows in which the lower level of MUN than higher level of MUN concentration.

Effect of Estrus Synchronization Protocols and Gonadotropin Releasing Hormone Treatments on the Pregnancy and Fetal Loss Rate after Transfer of Korean Native Cattle Embryos to Holstein Recipients

  • This study examined pregnancy and fetal loss rates according to different estrus synchronization protocols and injection of gonadotropin releasing hormone (GnRH) after transfer of Korean Native Cattle embryos to Holstein recipients. In Experiment 1, recipients received no treatment (Control, n = 119); two injections of prostaglandin$F_{2{\alpha}}$ ($PGF_{2{\alpha}}$ ) 11 days apart (PGF group, n = 120); GnRH (day 0)-$PGF_{2{\alpha}}$ (day 7)-GnRH (day 9) (Ovsynch group, n = 120); and CIDR (day 0)-$PGF_{2{\alpha}}$ and CIDR removal (day 7)-GnRH (day 9) (CIDR group, n = 110). In Experiment 2, the control group was received no treatment of GnRH. The treatment groups were received GnRH at embryo transfer (ET) (day 0), 7 days later, 14 days later, ET and 7 days later, 7 and 14 days later, or ET, 7 and 14 days later. Recipients were assigned to treatment randomly and received two in vitro produced blastocysts. Pregnancy was diagnosed at day 60 by palpation per rectum. Fetal loss to term was determined by palpation every 90 days thereafter. In Experiment 1, the pregnancy rate in the CIDR group (59.1%) were higher than in the Control group (42.0%) (p<0.01); fetal loss rates were similar for all groups (12.0 to 18.5%). In Experiment 2, the pregnancy rate in Day 0+7+14 group was higher (60.2%) than the control (40.2%) (p<0.01) and resulted in a lower fetal loss (p<0.05) than the control (4.6 vs. 11.4%). There were no significant difference between other treatment and the control (p>0.05). These results show that pregnancy rates of bovine embryos can be enhanced by CIDR insertion or GnRH $3{\times}$ treatment. Additionally, fetal loss may be reduced with GnRH treatment after ET.

Studies on In Vivo Embryo Production by FSH-P and SUPER-OV in Korean Native Cattle III. The Factors Influencing In Vivo Embryo Production on Days and Seasons of Flushing (한우에서FSH-P와SUPER-OV에 의한 체내 수정란 생산에 관한 연구 II.계절 및 채란일에 따른 체내 수정란 생산에 영향을 미치는 요인)

  • This study was carried out to establish an effective and practical system for commercialization of embryo production techniques by analyzing several factors influencing in vivo embryo production on days and seasons of flushing in Korean native cattle. In vivo embryos were flushed 226 times from 128 donors. The results obtained for the factors influencing in vivo embryo production on days and seasons of flushing were as follows :1.The percentages of fertilized, transferable and freezable embryos by seasons were significantly different in both FSR-P and SUPER-OV(P<0.01). The percentages of them were highest in sunrrner with FS H-P and highest in autumn with SUPER -OV.2. The production of transferable and freezable embryos by flushing days was highest in 8 days with FSH-P, and there was no difference between 7 and 8 days for SUP ER-OV. 3. The failure rates of recovery were 17.0% in SUPER-OV and 21.2% in FSH-P, respectively. The donors superovulated but failed recovery were 8.5% in SUPER-OV and 12.9% in FSH-P, respectively. Nonsuperovulated donors was 8.4% and donors giving less than 2 eggs at recovery was 8.4% in both FSH -P and SUPER-OV 4. The donors returned to normal estrus after superovulation were 34.1% after 1 cycle,39.4% after 2 cycles, and 16.7% after 3 cycles by FSH-P, respectively. For SUPER-OV, they were 55.3, 33.0 and 9.6%, respectively. Generally, normal estrus after the treatment of superovulation was earlier and the occurrence of ovarian cyst was also lower in SUP ER-OV than in FSH-P.5.The percentages of blastocyst in embryos flushed at 7~8 days after estrus were 21. 9% and 54.3% in FSH -P and SUPER-OV, respectively. Programmed Reproductive Management Including Presynch-Ovsynch-Resynch Protocol in Dairy Cows (젖소에서 일련의 Presynch-Ovsynch-Resynch Protocol을 이용한 번식관리 예)

  • This study was to evaluate the usefulness of a programmed reproductive management including Presynch-Ovsynch-Resynch protocol in lactating dairy cows. Nine hundred and thirty four cows calved during February to May 2008 were used for this study. Cows received im injections of 25 mg $PGF_{2{\alpha}}$ at $45{\pm}3$ and $59{\pm}3$ days in milk (DIM). Fourteen days later, Ovsynch was initiated with an im injection of $100\;{\mu}g$ GnRH ($73{\pm}3$ DIM), 25 mg $PGF_{2{\alpha}}$ 7 days later ($80{\pm}3$ DIM), $100\;{\mu}g$ GnRH 56 h later and timed artificial insemination (TAI) 16 h after the GnRH injection. Cows showed estrus during the Presynch-Ovsynch, were inseminated artificially according to am-pm rule. Pregnancy was determined on 32 days after AI using ultrasonography. Cows diagnosed pregnant were re-examined for pregnancy by rectal palpation at 60 days after AI. Non-pregnant cows were resynchronized by receiving $100\;{\mu}g$ GnRH, 25 mg $PGF_{2{\alpha}}$ 7 days later, $100\;{\mu}g$ GnRH 56 h later and TAI 16 h after. Cows with estrus since the first AI before the completion of Resynch protocol were also inseminated artificially according to am-pm rule. Pregnancy was determined by the same manner as following the first AI. Fifty five percents of the cows treated showed estrus and received AI before completion of the Presynch-Ovsynch protocol, while 45% received TAI, regardless of estrus exhibition following the completion of the protocol. The pregnancy rate following the first AI was higher in cows that showed estrus and received AI (43.3 and 38.5%) during the Presynch-Ovsynch than cows with TAI (34.6 and 29.6%) on 32 and 60 days after AI (P < 0.01). Sixty six percents of cows diagnosed non-pregnant following the first AI showed estrus before the completion of Resynch protocol and received AI, while 34% received TAI, regardless of estrus exhibition following the completion of the protocol. The pregnancy rate following the second AI was higher in cows that showed estrus before the completion of Resynch protocol and received AI (40.2 and 36.8%) than cows with TAI (21.2 and 18.2%) on 32 and 60 days after AI (P < 0.01). The cumulative pregnancy rates following the first and second AIs were 60.8 and 53.9% on 32 and 60 days after AI, respectively. These data suggest that inclusion of Presynch-Ovsynch-Resynch protocol and estrus detection into the programmed reproductive management might be an alternative option to control dairy cattle breeding.

Pregnancy of Monozygotic Twins by Bisection of Korean Native Cattle Embryos (수정란 분할에 의한 한우 쌍태의 임신)

  • This study was carried out to produce monozygotic twin calves by transfer of bisected embryos. Four Korean native cattle donors were superovulated with FSH and flushed to collect embryos on day 6 or 7 of the estrus cycle. Morula and early blastocyst embryos showed 1 or 2 grade were bisected with microblade and each set of demi-embryos without zona pellucida were transferred nonsurgically to 10 recipients respectively. The results obtained were as follows; 1. Twenty four demi-embryos (92.3%) were separated from 13 original embryos and among them 20 demi-embryos (83.3%) had normal appearance without severe damage. 2. Four sets of fresh demi-embryos were transferred to 4 recipients and one recipient was twin pregnant 3. Six sets of frozen-thawed demi-embryos were transferred to 6 recipients. Two recipients were pregnant, one of them twin.

Fertility of Holstein Cows in Chengdu, China

  • Data on the use of breeding records of dairy cattle farm of Fenghuang-shan in Chengdu areas during a period of six years is systematically analyzed. The results show that Holstein heifers have their 1st estrus at an average age of $513.6{\pm}46.7$ d which is not related to the calving season. Estrus mostly occurs in the months with mild ambient temperature (March, April, May, November and December). There is a very poor rate of estrus detection; only 30.0% successive estrus is observed within 24 d, 29.3% within 25-48 d, 40.6% over 48 d. The average number of insemination per conception is 1.50 for heifers and 1.74 for cows, but conception rate (CR) is lower in the relatively warm months (July, August and September) ranged from 48.1% to 51.9% compared with 58.1% to 68.5% in other months. High temperature is the most important factor affecting fertilization in warm months, but neither did CR decline immediately with the increased air temperature in June, nor did it increase immediately with the declined air temperature in September. Post partum anestrous period is $119.5{\pm}60.5$ d. The average interval from calving to conception is $159.4{\pm}85.6$ d with only 19.8% of the cows conceived within 85 d of calving. Cows with high milk yield have longer acyclic periods and lower conception rates. Improvement of efficient managements must be a precedent condition in Chengdu areas.

Effect of Dietary Supplementation with Vitamin and Mineral Complexes on Estrus Rate, Pregnancy Rate, Gestation Length, and Birth Weight of Calves in Multiparous or Primiparous Korean Native Cows (한우 경산우 및 미경산우에 비타민과 미네랄 복합제의 급여가 발정발현율, 임신율, 임신 기간 및 송아지 체중에 미치는 효과)

  • The increase in the total number of cows on farms, which breed Korean Native Cattle (KNC), is associated with many problems. In particular, the services per conception and calving interval have increased. In this study, we examined the effect of dietary supplementation with vitamin and mineral complex on the estrus rate, pregnancy rate, gestation length, and birth weight of KNC calves. Multiparous or primiparous KNC were divided into 3 groups with 40 heads per group. Experimental group 1 was administered a consisting of 35,000 IU vitamin A, 100 IU vitamin E, 200 mg $\beta$-carotene, 200 mg Zn methionine, and 1.5 mg Se. Experimental group 2 was administered a vitamin complex (100 g/day) consisting of 100 IU vitamin E and 200 mg $\beta$-carotene. In the case of multiparous KNC, the estrus detection rate in the control group was 90.0% and those in the experimental groups were 75.0% to 95.0%. However, the first-service pregnancy rate after parturition in the control group was 41.2%, which was significantly lower than that in the experimental groups (71.0% to 76.7%; p<0.05). The average duration of pregnancy in the group supplemented with the vitamin complex was days, which was similar to that in the case of the control group. The birth weight of calves from cows fed with vitamin complex was 25.3 to 27.0 kg, which was similar to that in the case of the control group (25.2 to 26.0 kg). In the case of primiparous KNC, no differences in the estrus rate, pregnancy rate, gestation length, or the birth weight of calves were noted between the groups. Thus, dietary supplementation vitamin and mineral complex have no effect on the reproductive efficiency in primiparous cows, but the pregnancy rate was observed to have increased in multiparous KNC with these supplements.

Behavioral Characteristics of Bos indicus Cattle after a Superovulatory Treatment Compared to Cows Synchronized for Estrus

  • The intensity and duration of sexual behavior in Bos indicus was assessed through the continuous observation of sexual receptivity. Two groups of cows were formed: only synchronized (n = 50) and other group further superovulated (n = 20). An intravaginal implant that released progesterone over 9 d was used. After removing the implant, 25 mg of $PGF_{2{\alpha}}$ was administered. In the superovulated group, the administration of 280 mg (Follicle stimulant hormone) FSH-P1 per cow with a decreasing dosage over 4 d was utilized. In both groups, behavioral observations began at the moment of implant removal. Sexual behavior was analyzed using a Kruskal-Wallis test to compare the mean of hours in estrus, effective mountings and number of mounts/hour during estrus. A nonparametric survival analysis was performed using the time in two ways: i) when an event happened it was placed in a 24 h timeframe and, ii) the time of observation in continuous form (96 h) assessing the difference between curves by the log rank test Chi-square. The only significant difference was the number of mounts/h during receptivity (p<0.05). In the superovulated group three periods of sexual activity during the day were identified, with these events being of greater frequency and duration than the synchronized group (p = 0.02); besides, the superovulated group began estrus before the synchronized group (p = 0.0035) when using the total period. In a simulation study, when the number of observations went from two (06:00-18:00) to three periods (06:00, 12:00 and 18:00) cows detected accurately (<6 h after the onset) increased more than 20%. The results show that superovulated cows presented greater intensity and duration of sexual activity in contrast with only synchronized animals.

  • The objective of this work was to determine the effect of corpus luteum (CL) grade on pregnancy rate after embryo transfer in Korean cattle and we found that CL development was linked to pregnancy rate. The in vivo derived blastocyst-stage embryos were transferred to 15 recipients synchronized in the estrus cycles. Based on size and palpable characteristics, CLs were categorized into three grade. The grade three CL is not to be identified by rectal palpation. The pregnancy rates tended to increase with the increase in CL size of recipients. In grade one, two, and three, the pregnancy rates were 62.5%, 50.0%, and 0%, respectively. This result suggests that pregnancy rates after embryo transfer might be affected by the CL status of recipients.

Studies on Embryo Transfer in Cattle (소의 수정란이식에 관한 연구)

  • This study was carried out to produce superior dairy cattle by embryo transfer. Seven dairy cows were superovulated with divided injection of FSH 4Omg for 5 days started on day 9 to 14 of the estrus cycle and injection of PGF$_2$$\alpha$ 45mg on day 4 of FSH injection. Donor cows were flushed to collect embryos on day 7 or 8 of the estrus cycle. Fresh embryos collected were transferred to synchronized dairy recipients or frozen using glycerol 3 step method to he equilibrated. And 35 embryos which were frozen using glycerol 6 step method were imported from U.S.A. After glycerol dilution of frozen embryos was done by reverse density during freezing. frozen-thawed embryos were transferred to synchronized dairy or beef recipients. The results obtained were as follows; 1. Total of 24 embryos were collected from 7 donor cows flushed and transferable embryos were 18 (75.0%). 2. Among 24 embryos. morula, early blastocyst, blastocyst, expanded blastocyst and unfertilized ova were 3 (12.5%), 1 (4.2%), 10 (41.6%), 4 (16.7%) and 6 (25.0%), respectively. 3. Heat inducing rate after 1st and 2nd injections of PGF$_2$$\alpha$ in Holstein and beef cattle was 83.3% and 71.4% and 62.5% and 69.2%, respectively. 4. Among 56 recipients, 23 head were pregnant (41.1%). The pregnancy rate of fresh embryos was 50.0% (1/2 heads) and the pregnancy rate of frozen embryos which were frozen using glycerol 3 step and using glycerol 6 step imported from U.S.A. was 52.6%(l0/19 heads) and 34.3%(12/35 heads), respectively. 5. The pregnancy rate of blastocyst (60.0%) was higher than that of morula (39.0%), early blastocyst (25.0%) and expanded blastocyst (0%). 6. The pregnancy rate of grade I embryos (52.2%) was higher than that of grade 2 (34.6%) and grade 3 (28.6%). 7. The pregnancy rate according to synchrony of recipient with donor was higher in simultaneous recipient (55.0%) and +l2hrs' (53.8%) than -24hrs' (23.5%), -l2hrs' (20.0%) and +24hrs' (0%).
