발달 단계에 따른 한국산 메기(Silurus asotus) 난모세포의 미세구조적 변화 (Ultrastructural Changes of Oocyte in Korean Catfish, Silurus asotus)

  • This study was carried out to investigate the histomorphological changes and the electrophoretic patterns of egg components, obtained from 100 of 1-year-old female catfish(Silurus asotus). Especially, the light microscopic and ultrastructural changes of ooplasm and follicular membranes of oocytes, were observed by light and transmission electron microscope. All data were collected from October in 1992 to May in 1993. The size of the nucleoli and number of the yolk granules increased as the oocyte grew. Yolk granules were deposited in the oocyte as fluid. Due to the presence of large early and late maturing oocytes, their ovaries were large, transparent, granular, and greenish in color. As the percentages of fish in LMO and RO stage increased from March to April, mean of GSI values(19.95%) increased. Follicle cells such as granulosa cell and thecal cell change a squamous into cuboid shape in LPO and EMO stage. Processes, microvilli, from the granulosa cells and from the oocyte grow and make contact with each other in the pore canals of the zona radiata during vitellogenesis, but are withdrawn as the zona radiate becomes more compact and devoid of pore canals during oocyte maturation. The electrophoretic pattern of major band in mature stage was much thicker(21k, 24k, 32k, 45k, 67∼110k, 170k dalton) than that in previtellogenic phase.

Effect of Dietary Lipid Sources on Body Fatty Acid Composition of Chinese Longsnout Catfish Leiocassis longirostris

  • We investigated the effects of dietary lipid sources on growth and fatty acid composition of juvenile Chinese longsnout catfish. Triplicate groups of fish (initial average weight, 3.8 g) were fed four diets containing either fish oil (FO), soybean oil (SO), linseed oil (LO) and lauric acid (LA) for 10 weeks. There were no differences among the groups in body weight, feed intake, feed efficiency, protein efficiency ratio, and body proximate composition of fish fed the diets containing different lipid sources (P > 0.05) during the study. However, fatty acids compositions of the whole body were influenced by dietary lipid source. Fish fed the SO diet had high concentration of linoleic acid, whereas those of fish fed the LO diet were rich in linolenic acid and arachidonic acid. Fish fed the FO diet had significantly (P < 0.05) higher levels of monounsaturated fatty acids such as 18:1n-9 and 20:1n-9 than those of fish fed the SO and LO diets. Eicosapentaenoic acid (EPA; 20:5n-3) and docosahexaenoic acid (DHA; 22:6n-3) composition of body were not influenced by dietary lipid source. The results suggest that each of FO, SO, LO or LA can be used as a lipid source in the diets of Chinese longsnout catfish without any negative effects on growth and feed utilization and these data demonstrate the potential impact which dietary fat composition can change the body fatty acid profile.

  • 메기의 산란행동을 관찰하기 위하여 1994년 5월부터 1995년 7월까지 산란시기에 HCG, 초어의 뇌하수체 등의 호르몬을 주사하여 이들의 행동을 관찰하였다. 이들의 산란행동은 기본적으로 chasing-clinging-embracing (enfolding) 순으로 이루어 졌는데, 주사후 15-16시간이 경과하면 산란행동을 시작하였고, 2-3시간 동안 산란행동을 하다가 19시간이 지나면 모든 행동이 끝났다. 그러나 지금까지 알려진 일본에서 메기의 자연산란 행동과는 수컷이 접근하는 방법과 수컷이 암컷을 감싸 조일 때 수컷의 뒷지느러미의 방향이 달랐다. 그리고 한국산 메기는 수컷이 암컷에 접근할 때 등쪽에서 접근하여, 배쪽에서 접근하는 S. biwaensis 와도 차이를 보였다.

The Annual Reproductive Cycle of Silurus microdorsalis, a Korean Endemic Species

  • The annual reproductive cycle of the Korean endemic slender catfish, Silurus microdorsalis, was examined histologically regarding water temperature and day length of habitat, gonadosomatic index (GSI), and development characteristics of female and male gonads. The maximum GSI value was found in May, $1.23{\pm}0.33$ and $11.77{\pm}3.23$ for male and female respectively (habitat water temperature $21.5^{\circ}C/13.59hr$ day length). On the other hand, the minimal level was $0.63{\pm}0.10$ in July ($26.5^{\circ}C/14.17$) for male and $1.36{\pm}0.08$ in October ($20^{\circ}C/11.2hr$) for female. We compared and calculated the stages of testis and ovary development process in order to determine the germ cell development characteristics and the reproductive cycle. According to results, we classified the annual reproductive cycle of the slender catfish into five stages: Growing phase (December-February), Mature phase (March-April), Ripe and spawning phase / Releasing phase in male (May-June), Degenerative phase (July-August), and Resting phase (September-November).

내분비 장애물질 검출을 위한 In Vitro Bioassay 개발 : 어류 혈청을 이용한 간세포 단층배양 (Development of In Vitro Bioassay for Detection of Estrogenic Activity of Xenobiotics : Monolayer Culture of Hepatocytes using Fish Serum)

  • 본 연구는 내분비 장애물질의 검출을 위하여 간세포의 단층 형성, 생존 및 기능에 미치는 어류 혈청의 영향에 대해 검토하였다. 한국산 메기의 간세포는 자신의 혈청 및 뱀장어, 틸라피아 등 타어종의 혈청에 의해 부착 및 단층이 형성되었으나, FBS는 메기 간세포의 단층을 형성시키지 못했다. 0.5에서 3%의 어류 혈청으로 메기 간세포의 단층을 형성 시킬 수 있는데, 이것은 FBS(5~20%) 사용의 1/10 이하로 적은 양이며, 어류 혈청이 FBS를 대체할 수 있고, FBS보다 간세포의 형태 및 기능 유지에 효과적인 것으로 나타났다. 어류 혈청이 첨가된 배양액에서 메기 간세포는 적어도 10일 이상 단층 형성을 유지할 수 있어, 내분비 장애물질 연구에 이용될 수 있을 것이다. 결론적으로 본 연구에서 개발된 어류 혈청을 사용한 메기 간세포 배양시스템과 효소면역측정법(ELISA)은 bisphenol A 등의 내분비 장애물질의 검출 및 연구를 위한 유용한 도구로서 이용될 수 있다고 생각된다.

메기(Silurus asotus) 기아 시 외부형태 및 체 절단면 변화 (The effect of starvation on the characteristics of morphometric measurement and body-sectioned shape in Far Eastern catfish (Silurus asotus))

  • 메기(Silurus asotus)를 대상으로 210일간 기아 시 계측형질 및 체 절단면 형질을 측정하여 그 기아 효과를 조사하였다. 실험 종료시 섭이군의 생존율은 92.2±0.47%였으며, 기아군에서는 74.4±2.59%의 생존율을 보였다. Ls, DALPO/Ls, DADOP/Ls, ED/Ls, CH/Ls, BDAA/Ls, BDMA/Ls 및 BWAA/Ls의 8개 항목에서 유의한 차이를 보였으며(p<0.05), 다른 16개 항목 DALAD/Ls, DPDPL/Ls, HPLAA/Ls, HALAV/Ls, HALOP/Ls, DAUPO/Ls, DADAV/Ls, DADAA/Ls, Max BL/Ls, Man BL/Ls, LD/Ls, HWOP/Ls, DAUF/HL, DAUS/HL, IW/HL 및 DAUE/HL에서는 초기군, 섭이군 및 기아군의 세 그룹 간 유의한 차이가 없었다(p>0.05). 섭이군은 각 체 절단면에서 기아군보다 더 높은 전체 높이(THX), 더 넓은 너비(WX) 및 더 넓은 면적(AX)을 보였다. 본 연구는 메기의 기아 상태에 대한 외형 및 체 절단면 형질에 대한 정보를 제공한다. 이는 메기에 대한 먹이 조절에 대한 방법을 보조하고 영양상태를 나타내는 기본 기초자료로 활용될 수 있을 것이다. 또한, 본 연구에서 사용한 조사 분석 방법을 타 어종에서 적용 시 그 영양 상태에 대한 정보를 파악하는 데 유용하게 활용될 수 있을 것으로 사료된다.

Genetic Divergence between Two Marine Catfish of Family Ariidae - Arius maculatus and Osteogeneiosus militaris

  • Two species of marine catfish, Arius maculatus and Osteogeneiosus militaris, belonging to family Ariidae were analysed electrophoretically for genetic variation in 6 enzymes, alcohol dehydrogenase (ADH), malate dehydrogenase (MDH), lactate dehydrogenase (LDH), glucose dehydrogenase (GDH), malic enzyme (ME) and superoxide dismutase (SOD). Eighteen individuals of each species were studied. Two loci MDH and ADH were polymorphic in both. Average heterozygosity in A. maculatus was 1.46, while it was 2.5 in O. militaris. The allele frequencies were used to estimate Nei's genetic distance (D). The D value was calculated to be 0.6879. Two isozyme loci, ME and SOD, were found to be the most reliable species specific markers. No tissue specific loci were observed for the enzymes studied, the bands being identical in each case. The genetic distance observed between O. militaris and A. maculatus in this study suggests that they would be more appropriately classified as species of the same genus rather than being assigned separate genera.

Introduction of bacterial and viral pathogens from imported ornamental finfish in South Korea

  • Background: Live fish import may lead to the unintended introduction of pathogens. We examined the monthly distribution of microbial pathogens in ornamental finfish imported into South Korea over a 6-month period. Results: Vibrio alginolyticus was detected in one lemon damsel in June and July; V. vulnificus was detected in one lemon damsel, one caerulean damsel, and one pearl-spot chromis and one ocellaris clownfish in July, April, and May, respectively; Photobacterium damselae was detected in one ocellaris clownfish and one caerulean damsel in June and July, respectively; V. anguillarum was detected in one pearl-spot chromis in February; V. harveyi was detected in one ocellaris clownfish and two mandarin fish in February and April, respectively; Yersinia ruckeri was detected in a pearlscale goldfish group in June and July and in two colored carp groups in July; and Lactococcus garvieae was detected in a lemon damsel group and a sutchi catfish group in July and May, respectively. European catfish virus, the only viral pathogen detected, was found in two sutchi catfish groups in May. Conclusion: This study is the first to identify pathogenic species and the presence or absence of pathogens (non-quarantine diseases) in imported ornamental finfish. These results demonstrate that various pathogens with the potential to harm indigenous fish populations can accompany ornamental finfish imported into South Korea.

메기의 껍질로부터 항균성 펩타이드의 정제 (Purification of Antibacterial Peptide from the Skin of the Catfish Silurus asotus)

  • 메기(Silurus asotus)의 껍질로부터 항균성 펩타이드를 정제하였다. 메기 껍질의 산 추출물은 단계적 농도구배조건으로 Sep-Pak C18에 의해 부분적으로 정제되었으며, 그 중에서 특히 60% 메탄올 분획(RM60)이 Escherichia coli D31에 대해 가장 좋은 항균활성을 나타내었다. 이 RM60을 사용하여 이온교환 및 5단계의 연속적인 역상 HPLC로 정제하였다. 정제된 펩타이드의 분자량과 아미노산 서열분석은 MALDI-TOF MS와 에드만 분해법으로 분석하였다. 이 펩타이드의 분자량은 약 4182.1 [M+H]+이었으며, 분석된 이 물질의 부분적인 일차구조서열은 다음과 같다; PALXXKARREAKVKF. 이러한 결과는 메기의 껍질에서 존재하는 이 펩타이드가 메기의 껍질에서 반응하는 선천성 방어 시스템에서 중요한 역할을 하고 있다고 여겨진다.
