Ethanol Effect of Nelumbo nucifera Root on Carbohydrate Methabolism Related Enzyme Activities and Antioxidative Effect in Streptozotocin-Induced Diabetic Rats (Streptozotocin으로 유발된 당뇨 흰쥐에 대한 연근 에탄올 추출물의 당대사 효소활성과 항산화 작용에 미치는 영향)

  • This study was done to investigate the carbohydrate metabolism related enzyme activities and antioxidative effects of Nelumbo nucifera(N.N) Root in streptozotocin (STZ)-induced diabetic rats. The contents of serum glucose, triglyceride (TG) and Total cholesterol were significantly decreaed in N.n treated group compared to the those of STZ-control group. The activity of glucose-6-pase(G-6-Pase) was significantly decreased in N.N treated group. Also the activity of glucokinase(Gk) was significantly increaed in N.N treated group. The content of hepatic glycogen was significantly increaed in N.N treated group, in addition, content of malondialdehyde(MDA) was significanly decreased in N.N treated group. Also, content of glutathione(GSH) was significanly increased in N.N treated froup. whereas, activity of catalase(CAT) was significantly decreaed in N.N treated group compared to the those of STZ-control group. activity of glutathione peroxidase(GSH-Px) was inecreaed. In conclusion, these results indicated that ethanol extract of N.N would have carbohydrate metabolism antioxidative effects in STZ-induced diabetic rats.

Phosphorus Significance in Alleviating Oxidative Stress Induced by Drought in Kentucky Bluegrass (Poa pratensis L.)

  • The objective of this study was to determine phosphorus effects on drought stress-induced oxidative stress in Kentucky bluegrass. Drought stress was induced by reducing of water to plants in pots. Two types of phosphorus were applied as potassium phosphate (P) or potassium phosphonate (PA). Application of phosphorus was efficient to ameliorate the adverse effects of drought. Osmotic potential, total chlorophyll and carotenoid content were significantly decreased by drought stress, but was relieved by P or PA application. Superoxide (O2•-) concentration was significantly increased more than 14-fold under drought-stressed plants, was accompanied with increase of hydrogen peroxide (H2O2) and lipid peroxidation (MDA). However, malondialdehyde (MDA) was much less in P or PA applied plants under drought stress condition. Activities of catalase (CAT), ascorbate peroxidase (APX) and guaiacol-peroxidase (GPX) were largely increased by drought stress and its increase rate was much higher in P or PA applied plants except APX. These results indicate that drought stress-induced oxidative stress is alleviated by P or PA application due to the increase of activities of antioxidant enzymes.

Alleviating Effects of Nitric Oxide on Cadmium Toxicity in White Poplar (Populus alba)

  • Cadmium (Cd) is non-essential heavy metal that negatively affects plant metabolism. Nitric oxide (NO) is an increasingly important molecule for plant metabolism that makes signaling. In this study, it was aimed to investigate the alleviating effect of sodium nitroprusside (SNP) application as NO donor in white poplar (Populus alba) under Cd stress conditions. SNP and without SNP treatments increased the Cd accumulation in root tissue. While photosynthetic pigments (Chl a, Chl b, Chl a+b, and carotenoid) content decreased by only Cd application, SNP+Cd application decreased the rate of photosynthetic pigments reduction. When the results of Cd and Cd+SNP applications were evaluated for mineral (Fe, Zn, Mn and Cu) uptake, it was found that the positive effect of SNP was heterogeneously affected. Depending on SNP application, it was found that malondialdehyde (MDA) amount decreased in leaf in 100 µM Cd applications while hydrogen peroxide (H2O2) amount decreased in 100 and 500 µM Cd applications. When antioxidant enzyme activities were examined, it was found that catalase (CAT) and ascorbate peroxidase (APX) enzyme activities increased with 100 µM SNP applications under all Cd applications. As a result, it was found that SNP application under Cd stress generally supports physiological processes positively in white poplar, suggesting that NO molecule plays important alleviating roles in plant metabolism.

Protective Effects of the Postbiotic Levilactobacillus brevis BK3 against H2O2-Induced Oxidative Damage in Skin Cells

  • Postbiotics have various functional effects, such as antioxidant, anti-inflammatory, and anti-obesity. Levilactobacillus brevis BK3, the subject of this study, was derived from lactic acid bacteria isolated from Kimchi, a traditional Korean fermented food. The antioxidant activity of BK3 was confirmed through the measurements of 2,2-diphenyl-1-picryl-hydrazyl (DPPH), 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), and total antioxidant capacity (TAC). The wrinkle improvement effect was validated by assessing elastase inhibitory activity and collagenase inhibitory activity. The intracellular activity was confirmed using human keratinocytes (HaCaT) and human fibroblasts (HFF-1). BK3 protects skin cells from oxidative stress induced by H2O2 and reduces intracellular reactive oxygen species (ROS) production. In addition, the expressions of the antioxidant genes superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GPx) were upregulated. Meanwhile, matrix metalloproteinase-1 (MMP-1) and collagen type I alpha 1 (COL1A1), involved in collagen degradation and synthesis, were significantly regulated. These results suggest the possibility of utilizing BK3 as a functional ingredient with antioxidant and wrinkle-improving effects.

Effect of Pyroligneous Liquor on Oxygen Radicals and Their Scavenger Enzymes in Liver of CD Rats (흰쥐 간조직의 활성산소 및 제거효소에 미치는 목초액의 영향)

  • This study was designed to investigate the effects of pyroligneous liquor on oxygen radicals and their scavenger enzymes in the liver of Cri/Bgi CD rats (7 rats per group). Male rats were fed a basic diet prepared in our Lab., PL-0 (Control), PL-1, PL-25, PL-50 and PL-75 groups were Prepared to be 0%, 1%, 25%, 50% and 75%with distilled water using pyroligneous liquor (35% of Choa Co. Ltd.), and were administrated orally for 8 weeks. Superoxide radical contents in liver mitochondria and microsomes were significantly decreased to 12-14%, 11-15%, respectively, in these PL-25 and PL-50groups compared with the control group. Hydroxyl radical content in mitochondria and microsomes were markedly decreased to 12-20% and 17%, respectively, in these PL-25 and PL-50% groups compared with the control group. Hydrogen peroxide content in mitochondria and microsomes were significantly decreased about 15-12% and 22-20% in liver of PL-25 and PL-50 groups compared with the control group. Mn-SOD and Cu/Zn-SOD activities in liver of PL-25 and PL-50 groups were remarkably increased to 15-25%, 11-16%, respectively, compared with the control group. GPx activities in mitochondria and microsomes were significantly increased in the liver of PL-25 and PL-50 groups compared with the control group. CAT activities in mitochondria and cytosol were significantly increased to 12-14%, 15-27%, respectively, in the liver of PL-25 and PL-50 groups compared with the control group. These results suggest that long term administration orally of 25 and 50% pyroligneous liquor may effectively inhibit the formation of oxygen free radicals, and also scavenger enzyme activities significantly increase through the administration orally.

Individual Differences of Ozone Resistance for Seed Germination and Seedling Development of Pinus thunbergii (해송의 종자 발아 및 유묘 생장에 대한 오존저항성의 개체간 차이)

  • Individual differences of ozone ($O_3$) resistance for seed production, seed germination and seedling development were examined in this study. Five in each healthy and damaged trees of Pinus thunbergii growing in air polluted area for 12 years were chosen based on visible foliar injury and growth. The cones of P. thunbergii, which were collected from healthy and damaged trees, were analyzed for physical characteristics and seeds from the cones were used to test germination percentage under $O_3$ treatment. The germinated seeds were continuously exposed to $O_3$ treatment and the lipid peroxidation and activities of antioxidative enzymes were determined for both seeds and seedlings. The $O_3$ treatment for seed germination and seedling development were conducted at three conditions: control, 150 ppb and 300 ppb of $O_3$. The non-treated seeds from the damaged trees showed 21.6% lower germination than those from the healthy ones. On the $O_3$ treatment of 300 ppb, seed germination decreased approximately 10% for the healthy trees and 19% for the damaged trees compared to that on the control. The seeds from the healthy trees showed significantly higher activities of superoxide dismutase (SOD), glutathione reductase (GR), and catalase (CAT) than those from the damaged trees. The activities of GR, ascorbate peroxidase (APX), and CAT decreased along with the increasing $O_3$ concentration in two tree grades. Malondialdehyde (MDA) content of seeds was not influenced by $O_3$ treatment for two tree grades. In seedling development, there were no significant differences for length and biomass of needle and root of two tree grades at both the control and 150 ppb of $O_3$. At 300 ppb of $O_3$ treatment, however, the length and biomass of needle and stem decreased for two tree grades but no significant differences was detected in root. The seedlings from the damaged trees were more sensitive to the $O_3$ treatment, showing higher activities of SOD, APX, and CAT and content of MDA compared to those from the healthy tree seedlings. Our results indicate that seed germination and seedling development are vulnerable to increasing $O_3$ concentrations and that attention must be paid to the individual selection of tree species for reforestation.

The Correlativity of Ulva lactuca Fractions, LPS, Enzymatic Activity and the Evaluation of Water Fraction (갈파래 분획, LPS, 효소활성의 상관성과 수층분획의 가치평가)

  • Lipopolysaccharide(LPS) was posttreated after the 14 day-pretreatment of Ulva lactuca fractions(ULF), and their correlativity to enzymatic activity alteration was investigated in the liver of rats. ULF was intraperitoneally administered into rats at dose of $1m{\ell}/kg$ of 100 mg/kg concentration for 14 days. On the day 15, $1m{\ell}/kg$ of LPS was injected. The corelativity was examined by measuring the changed values of superoxide dismutase(SOD) in mitochondrial fraction and catalase(CAT), glutathione peroxidase(GPx) in liver homogenate. The results showed that LPS treatment decreased the high values of SOD, CAT, GPx to the low values, but ULF pretreatment increased the low values of SOD, CAT, GPx to the high values. It was suggested that ULF, LPS and antioxidative enzymes like SOD, CAT, GPx had the corelativity of the high-low-high pattern and that the ULF pretreatment played the proper preventive role in the protection against the LPS treatment-induced enzymatic inactivity in the water fraction.

In vivo Physiological Activity of Mentha viridis L. and Mentha piperita L. (박하의 in vivo 생리활성)

  • Alcohol metabolizing and antioxidant activity of Mentha species were investigated in rat liver. Fifty six Sprague Dawley rats were randomly divided into seven groups such as normal (ethanol excluded), negative control (40% ethanol (10 g/kg of body weight/day) fed), positive control (1 g Silymarin/kg of body weight/day with ethanol fed), two Mentha viridis extracts (0.2 g & 1 g M. viridis methanol ext./kg of body weight/day with ethanol fed) and two M piperita extracts (0.2 g & 1 g M. piperita methanol ext./kg of body weight/day with ethanol fed) groups. After 2 weeks, rats were sacrificed under ether. The activities of alcohol dehydrogenase (ADH), aldehyde dehydrogenase (ALDH), catalase (CAT), manganese superoxide dismutase (Mn-SOD), glutathione peroxidase (GAH-px) and the content ofthiobarbituric acid reactive substance (TBARS) in the rat livers and the activity of glutamate pyruvate transferase (GPT) in serum were evaluated. From the analyses, 1 g M. viridis and 0.2 g M. piperita administrated groups showed higher ADH and ALDH activity than the other groups. Groups fed with 0.2 g and 1 g M. viridis ext. and 0.2 g M. piperita ext. showed higher CAT activity than the other groups. All the Mentha extract fed groups exhibited more effective in recovering Mn-SOD, GSH-px and GPT acitivities to a similar degree of normal group. TBARS contents of two M. viridis ext. fed group and 0.2 g M. piperita ext. fed group were higher than those of the other groups. M. viridis extract fed groups showed more effective in CAT and Mn-SOD activities than M. piperita extract groups at p < 0.05. Finally, it is concluded that both Mentha species have alcohol metabolizing and antioxidant activity and M viridis is more effective than M. piperita.

Comparison of Antioxidant Activities of Water Extract from Dandelion (Taraxacum officinale) Aerial Parts, Roots, and Their Mixtures (서양민들레(Taraxacum officinale) 지상부, 지하부 및 혼합 추출물의 항산화 활성 비교)

  • The present investigation evaluated the antioxidant activities of water extracts from dandelion (Taraxacum officinale) aerial parts, roots, and mixed extracts. Mixed extract of T. officinale was a mixture of aerial parts and roots at 9:1 and 8:2 weight ratios. Extracts from aerial parts (DAE), roots (DRE), and mixture of aerial parts and roots (DME) were measured for cell viability and catalase activity in HepG2 cells, 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, and lipid peroxidation inhibitory activity. Cell viabilities of HepG2 cells treated with DAE, DRE, DME 8:2, and DME 9:1 against $H_2O_2$-induced oxidative damage were 63.4%, 54.6%, 76.7% and 83.4% at a concentration of $400{\mu}g/mL$, respectively. Catalase activity was highest in DME 9:1 (12.2 mU/min/mg protein) compared with DAE (9.0 mU/min/mg protein) and DRE (9.7 mU/min/mg protein). DPPH radical scavenging activity of DME showed a significantly lower $EC_{50}$ value than DAE ($EC_{50}$ value of DME $9:1=163.3{\mu}g/mL$, DME $8:2=172.4{\mu}g/mL$, and $DAE=173.7{\mu}g/mL$). Lipid peroxidation inhibitory activity of DME showed a significantly lower $EC_{50}$ value than DAE [$EC_{50}$ values of DME $(9:1)=454.4{\mu}g/mL$, DME $(8:2)=426.6{\mu}g/mL$, and $DAE=654.7{\mu}g/mL$]. The results indicate that a small amount of T. officinale roots increased antioxidant activity of aerial parts. Especially, a 9:1 mixture was more valuable than 8:2 mixture for industry.

Effect of Pine (Pinus densiflora) Needle Hot Water Extract on Antioxidant Activity in Rats Treated with Carbon Tetrachloride (적송잎 열수 추출물이 사염화탄소를 투여한 흰쥐의 항산화 활성에 미치는 영향)

  • Pine (pinus densiflora) needles have long been used as a traditional health-promoting medicinal food in Korea. This study was conducted to investigate the effects of pine needle extracts on the hepatic antioxidant system in the damaged liver of carbon tetrachloride ($CCl_4$)-treated rats. Nine-week-old Sprague Dawley rats were divided into four groups: normal group (NOR), $CCl_4$-treated group (CCL), pine needle hot water extract and $CCl_4$-treated group (CCL-P), and Vitamin C and $CCl_4$-treated group (CCL-V). The enzyme activities and antioxidant effects of the pine needle hot water extracts were investigated at the levels of liver homogenates and serum of rats intoxicated with $CCl_4$. Serum GOT and GPT activities by $CCl_4$ treatment increased compared to those of the NOR group. However, they tended to decrease in the hot water extract-administered group. Liver SOD activity in the CCL group was significantly lower than the NOR group (p<0.05). However, they increased in the CCL-P group compared to the CCL group. Further, the CAT and GPx activities of serum treated with $CCl_4$ were higher compared to those of the NOR group but lower in the CCL-P group compared to CCL group. These results suggest that pine needle hot water extract increases antioxidant activities.
