Pinus rigida Allelochemicals가 차풀종자의 발아과정에서 동위효소의 활성에 미치는 영향 (Effects of Pinus rigida Allelochemicals on Isozyme Activities during Seed Germination of Cassia mimosoides var.nomame)

  • Eleven phenolic compounds including caffeic acid were identified through analyzing the aqueous extracts of Pinus rigida by HPLC. Among them, protocatechuic acid was the maximum amount of 6.84 ppm. Seed germination of Cassia mimosoides var. nomame was significantly stimulated by the extract of P. rigida leaves in the proportion ot concentration. However, root growth was elevalted at a threshold concentration below 25%, but it was inhibited at high concentrations. In 50% extract of P. rigida, upward root tip of C. mimosoides var. nomame showed negageotropism which the root end showed necrosis. New isozyme bands were induced indicating concentration activity of peroxidase from the extract of C. mimosoides var. nomame, especially in the cathodic region. Although it reduced the mumber of isozyme bands of esterase, esterase activities were stimulated in the anodic region of C. mimosoides var. nomame. The activity of amylase was not remarkably different between control and treatment.

  • The germination responses of Cassia mimosoides var. nomame seeds to temperature were examined under various conditions. The temperature range allowing germination was $20~40^{\circ}C$, and the linear relationship between the germination rate and temperature appeared to exist between $28~38^{\circ}C$ The total thermal time required for germination (10~80%) of C. mimosoides seeds ranged from 259 Kh (degree Kelvin$\times$hours) to 421 Kh and base temperature range was relatively constant, i.e., $23.39~26.68^{\circ}C$. In the increasing temperature (IT) regime, C. mimosoides seeds started to germinate at $16^{\circ}C$ and showed greater germination rate with increasing temperatures. The final germination rate was 64% at $36^{\circ}C$. On the other hand, in the decreasing temperature (DT) regime, the seeds began to germinate at $36^{\circ}C$, and the final germination rate was 52% at $20^{\circ}C$. An induced dormancy occurred at $16^{\circ}C$ in the DT regime.

Effect of Methanol Extract from Cassia mimosoides var. nomame on Ischemia/Reperfusion-induced Renal Injury in Rats

  • Objectives : The purpose of this study was to determine whether the methanol extract of Cassia mimosoides var. nomame Makino, a naturally growing plant in Korea, could prevent the renal-ischemia/reperfusion injury in a rat model or not. Methods : The radical scavenging activities of the extracts, and ascorbic acid as a positive control, were measured in vitro. At one hour after an intraperitoneal injection of the extract (400 mg/kg), renal ischemia/reperfusion injury was generated by 40 min clamping of the left renal artery in rats. After renal ischemia/reperfusion and 24 hr restoration of blood circulation, the serum creatinine concentration was measured. And the extent of epithelial cell injury and apoptosis was assessed by various staining technologies. The Bax/Bcl-2 ratio and activated caspase-3 were assessed by immunohistochemistry. Results : The extract showed a slightly lower level of radical scavenging activity than that of ascorbic acid. Compared to those of the vehicle-treated group, the extract-treated group displayed a significantly smaller tubular epithelial cell injury of 54% reduction in the outer medulla region and a lower serum creatinine concentration of 50% reduction. It seems that the reduction in cellular injury is due to the attenuation of the Bax/Bcl-2 ratio, and the inhibition of caspase-3 activation by the extract of Cassia mimosoides. Conclusions : Cassia mimosoides var. nomame Makino could be a good candidate for a prophylactic agent against the ischemia/reperfusion/induced kidney injury.

파종량 차이에 따른 차풀의 생육, 사초수량 및 사료가치 변화 (Effects of Seeding Rates on Growth, Forage Yield and Feed Value of Cassia mimosoides var. nomame)

  • Cassia mimosoides var. nomame was grown at five seeding rates (10, 20, 30, 40 and 50 kg/ha) from March 31 to September 21, 1998 at Cheju to determine influence of seeding rate on agronomic characters, and forage yield and quality. Days to flowering increased from 126 to 129 days as seeding rate increased from 10 to 50 kg/ha. The plant height increased from 86 to 99 cm as seeding rate was increased from 10 to 30 kg/hac and then decreased to 93 cm at a rate of 50 kglha. With increasing seeding rate, stem diameter quadratically decreased and the number of branches and green leaves per plant linearly decreased but the number of withering leaves per plant linearly increased. As seeding rate increased &om 10 to 30 kg/ha, fresh forage, dry matter, crude protein, and total digestible nutrients (TDN) yield increased 'from 30.7 to 49.1, 7.5 to 12.2, 0.90 to 1.57 and 4.09 to 7.09 MTha, respectively, and then decreased with further increased seeding rates. Crude protein, ether extract, nitrogen free extract, and TDN wntent increased 12.1 to 14.2% 2.2 to 2.9%, 35.1 to 39.2%, 54.8 to 60.3% respectively, but crude fiber wntent decreased 39.8 .to 33.3% and crude ash declined 4.9 to 3.8% as seeding rate increased from 10 to 50 kgiha. The optimum seeding rate to obtain the highest TDN yield was estimated to be 36kglha. (Key words : Cassia mimosoides var. nomame, Seeding rate, Forage yield, Forage quality)

Methanol Extract of Cassia mimosoides var. nomame Attenuates Myocardial Injury by Inhibition of Apoptosis in a Rat Model of Ischemia-Reperfusion

  • Interruption of blood flow through coronary arteries and its subsequent restoration triggers the generation of a burst of reactive oxygen species (ROS), leading to myocardial cell death. In this study, we determined whether a methanol extract of Cassia mimosoides var. nomame Makino could prevent myocardial ischemia-reperfusion injury. When radical scavenging activity of the extract was measured in vitro using its ${\alpha}$,${\alpha}$-diphenyl-${\beta}$-picrylhydrazyl (DPPH) radical quenching ability, the extract showed an activity slightly lower than that of ascorbic acid. Three days after oral administration of the extract (400 mg/kg/day) to rats, myocardial ischemia/reperfusion injury was generated by 30 min of ligation of the left anterior descending coronary artery (LAD), followed by 3 hr reperfusion. Compared with the vehicle-treated group, administration of the extract significantly reduced infarct size (IS) (ratio of infarct area to area at risk) in the extract-treated group by 28.3%. Reduction in the cellular injury was mediated by attenuation of Bax/Bcl-2 ratio by 33.3%, inhibition of caspase-3 activation from procaspase-3 by 40%, and subsequent reduction in the number of apoptotic cells by 66.3%. These results suggest that the extract attenuates myocardial injury in a rat model of ischemia-reperfusion by scavenging ROS, including free radicals, and consequently blocking apoptotic cascades. Therefore, intake of Cassia mimosoides var. nomame Makino might be beneficial for preventing ischemic myocardial injury.

차풀 종자로부터 Daucosterol과 Naphthalene glucoside의 분리 (Isolation of Daucosterol and Naphthalene glucoside from Seeds of Cassia mimosoides var. nomame Makino)

  • 본 연구는 속리산국립공원 내 쌍곡계곡에 분포하는 식물상을 조사하기 위해 수행되었고 조사기간은 2006년 3월부터 2007년 6월까지 조사하였다. 식물상을 조사한 결과, 88과 242속 342종 52변종 7품종 등 총 401분류군이 확인되었다. 한국특산식물은 개비자나무, 키버들, 개족도리, 할미밀망, 민산초나무, 청괴불나무, 병꽃나무, 분취 등 총 8분류군이 조사되었다. 환경부 지정 멸종위기식물은 망개나무 등 1분류군이며, 산림청 지정 희귀식물은 개족도리, 고란초, 망개나무, 백작약, 태백제비꽃, 꼬리진달래, 말나리 등 7분류군이 조사되었다. 이들의 보전을 위해서는 향후 희귀식물에 대한 구체적인 보전방안과 번식방법 등이 논의되어야 할 것으로 판단된다. 자원식물은 관상용 식물 220분류군, 식용 식물 239분류군, 약용 식물 281분류군, 기타용 식물 206분류군으로 나타났다. 귀화식물은 오리새, 소리쟁이, 취명아주, 아까시나무, 붉은토끼풀, 토끼풀, 큰달맞이꽃, 비짜루국화, 붉은서나물, 개망초, 망초, 뚱딴지, 서양민들레 등 13분류군이 관찰되었으며, 귀화율은 약 3.24%, 자연파괴도는 약 4.53%로 나타났다.

Methanol Extract of Cassia mimosoides var. nomame and Its Ethyl Acetate Fraction Attenuate Brain Damage by Inhibition of Apoptosis in a Rat Model of Ischemia-Reperfusion

  • Ischemic stroke, a major cause of death and disability worldwide, is caused by occlusion of cerebral arteries that, coupled with or without reperfusion, results in prolonged ischemia (hypoxia and hypoglycemia) and, ultimately, brain damage. In this study, we examined whether methanol extract of the whole plant of Cassia mimosoides var. nomame Makino that grows naturally in Korea, as well as Japan and China, and some of its fractions obtained by partitioning with organic solvents could protect human hepatocellular carcinoma cells (HepG2) under hypoxic condition by inhibiting apoptosis. We also investigated if these extracts could attenuate brain damage in a rat model of 2 hr of ischemia, generated by middle cerebral artery occlusion, and 22 hr of reperfusion. The whole extract ($100{\mu}g$/mL) maintained the cell number at more than half of that initially plated, even after 24 hr of cell culture under hypoxic condition (3% $O_2$). In the absence of the whole extract, almost all of the cells were dead by this time point. This improvement of cell viability came from a delay of apoptosis, which was confirmed by observing the timing of the formation of a DNA ladder when assessed by gel electrophoresis. Of fractions soluble in hexane, ethyl acetate (EA), butanol and water, EA extracts were selected for the animal experiments, as they improved cell viability at the lowest concentration ($10{\mu}g$/mL). The whole extract (200 mg/kg) and EA extract (10 and 20 mg/kg) significantly reduced infarct size, a measure of brain damage, by 34.7, 33.8 and 45.2.0%, respectively, when assessed by 2,3,5-triphenyl tetrazolium chloride staining. The results suggest that intake of Cassia mimosoides var. nomame Makino might be beneficial for preventing ischemic stroke through inhibition of brain cell apoptosis.

Effect of Phytolacca americana Extracts on the Activities of AsPOX and GuPOX during Germination Process of Cassia mimosoides var. nomame

  • After Cassia mimosoides var. nomame was treated with Phytolacca americana extracts during the germination process, its effect on the activities of AsPOX and GuPOX were examined. As the concentration of P. americana extract increased. the activity of AsPOX decreased while that of GuPOX increased. During the germination process, the activity of AsPOX was lower than the control, while the activity of GuPOX was over 2 times higher than the control. Activity of total peroxidase by IEF was extremely high at pI 6.4 and pI 6.6 when treatment was made with a 30% concentration of P. americana extract. The treatment with phenolic compounds, caffeic acid and benzoic acid did not show much difference from the control although a slight increase was observed at pI 6.6. Activity of GuPOX in C. mimosoides was over 4 times higher in roots than in shoots. Namely, because GuPOX activity of C. mimosoides was increased by extracts of P. americana, defense enzyme, GuPOX. was generated against external stress, and we could certified the activity increase at pI 6.4, especially in root.

자생식물과 귀화식물의 수용 추출액이 Zoysiagrass의 발아와 유근생장에 미치는 영향 (Effects of Aqueous Extracts from Naturalized and Korean Wild Plants on Seed Germination and Seedling Growth of Zoysiagrass)

  • 자생종과 귀화종의 토양 pH를 측정한 결과 자생종의 토양 pH범위는 5.33~7.20인 반면 귀화 종의 토양 pH는 3.95~6.48로 자생종에 비하여 귀화종의 pH가 낮았다. 귀화식물 추출액 50% 농도 이상부터는 잔디의 발아율이 60% 미만으로 급격하게 억제되었으며 망초추출액이 가장 현저한 발아억제를 보인 반면, 자생식물인 차풀의 30%와 50% 추출액농도에서 발아율이 증가하였다. 자생식물과 귀화식물 추출액에 의한 잔디의 유근생장 억제는 각 농도에 따라 큰 차이를 보이지 않았지만 차풀의 경우는 15% 이상의 추출액 농도에서 유근생장이 60% 이상 현저하게 억제되었다. HPLC에 의한 차풀추출액의 성분 분석 결과는 254nm에서 protocatechuic acid, $ ho$-hydroquinone, $\rho$-coumaric acid, ferulic acid가 분석되었고 284nm에서는 vanillic acid, hydroquinone, benzoic acid, cinnamic acid가 분석되었다. Phenolic compound 각 농도로 잔디의 발아와 유근생장을 조사한 결과 ferulic acid와 vanillic acid는 잔디의 발아율을 증가시켰고 $\rho$-coumaric acid는 유근생장을 억제시켰다.
