Aquatic animal raised in hatcheries play an important role in supplying seedling to stock enhancement and seed quality, especially, seed health is the key factor for survival in the field after release and for stocking effectiveness. We have inspected the hatchery-reared seeds of 33 marine species and 12 freshwater species for legally designated diseases in stock enhancement program in 2010. Results showed that abalone was the most abundant as 20.0% in the marine species group and then sea cucumber (15.6%), olive flounder (8.4%), rockfish (6.7%), black sea bream (6.3%) and swimming crab (6.1%) were followed. Crucian carp was the most abundant as 19.4%, and then eel (11.8%), Korean bullhead (10.9%), mandarin fish (10.8%), melanian snail (8.4%), catfish (7.7%) were followed in the freshwater species group. The total number of inspection cases for eight pathogens in this study were 2,105 and disqualification cases were 30 by detection of aquatic animals pathogens such as koi herpesvirus (KHV), red sea bream iridovirus (RSIV), white spot syndrome virus (WSSV) or viral haemorrhagic septicemia virus (VHSV).
Cell size, DNA content, chromosome and PCR-based mitochondrial 12S rRNA gene analyses were conducted to obtain basic informations for genetic stock identification of spotted flounder (Verasper variegatus) from Yeocheun, Korea. The mean erythrocytic and nuclear volumes of spotted flounder were $211.10{\mu}m^3$ and $23.03{\mu}m^3$, respectively. The haploid DNA content of this species was 0.79 pg/cell which correspond to $46.5\%$ of carp and to $22.6\%$ of mammals. Spotted flounder had the 2n = 46 acrocentric chromosomes but no heteromorphic sex chromosomes was found. Mitochondrial DNA gene for 12S ribosomal RNA was amplified by polymerase chain reaction (PCR) and the PCR products were subjected to digestion with 15 restriction endonucleases. Restriction enzyme analyses revealed that Ava I, Mae II, Sma I and Xba I had one restriction site in the mitochondrial 12S rRNA gene segment of spotted flounder, while Mae I had two. Segments of 12S rRNA gene from mitochondria in spotted flounder were sequenced and compared with channel catfish and human as controls. The 12S rRNA gene of this species was more similar to that of channel catfish than to human's.

Endogeneous activities of ${\beta}-galactosidase$-like enzyme in various tissues from several finfishes and shellfishes were examined by histochemical analysis based on X-gal staining and by fluorimetric measurement using 4-methylumbelliferyl-${\beta}$-D-galactoside (4-MUG). Species used in this study were 3 freshwater fishes, mud loach (Misgurnus mizolepis), common carp (Cyprinus carpio) and tilapia (Oreochromis niloticus) ; 3 marine fishes, olive flounder (Paralichthys olivaceus), stone flounder (Kareius bicoloratus) and marbled sole (Limanda yokohamae) ; and 4 shellfishes, abalone (Haliotis discus hannai), Pacific oyster (Crassoskra gigas), pearl oyster (Pinctada fucata martensii) and ark shell (Anadara broughtonii). The activities of ${\beta}-galactosidase$-like enzyme in all finfishes examined were significantly different among species, with the wide variations between tissues in a species. In general, the tissues such as kidney, intestine and liver were ones which showed the significantly higher values in 4-MUG fluorimetry and deeper staining patterns in X-gal analysis compared to other tissues. On the other hand, serum and muscle revealed the significantly lower activities than others did, regardless of species. Shellfishes were also found to have endogenous activities of ${\beta}-galactosidase$-like enzyme which were significantly varied depending on both species and organs in a species. Hepatopancreas from all shellfishes examined showed the deepest pattern in X-gal staining and also the highest value in 4-MUG analysis, while activities of ${\beta}-galactosidase$-like enzyme in adductor muscles and mantle muscles from all shellfish species in this study except Pacific oyster were negligible : Pacific oyster had the significant activity of this enzyme in muscle tissues. Putative endogenous lacZ fragment was amplified from both finfishes and shellfishes by polymerase chain reaction (PCR). The molecular size of PCR products was about 510 bp, and there was no difference in size among species examined.

Many specialty seafood restaurants have several tanks from which you may choose a fish. Popular fish for this dish may include Korean rockfish, flounder, yellow corvina, codfish, croakers, pollacks, and even fresh water fish like carp and trout. In addition, other shell fish such as crabs, clams and oysters can be also added to this soup to compliment and enhance its spicy but refreshing flavor. This soup is one of Korean's most popular dishes while drinking soju. If you order sushi or sashimi, the soup is made from the left-over parts of the fish. The objective of this study is to determine a standardized recipe of Maeun-Tang and to investigate consumption so as to increase its intake opportunities for consumers through its development. Fish species used in Maeuen-Tang's recipes were flounder, Korean rockfish and other fish. Onion, radish, chilies, crown daisy, garlic and some times, pumpkin and bean curds were added to the mixture to absorb the kochujang which is the base flavor of this dish. Most of the stock used in Maeuen-Tang's recipes is pure water, fish stock and vegetable stock, 1.0-1.5 Korean rock fishes and 0.5-1.0 flounders, with its head and bone, which were used in 3-4 portions of Maeun-Tang. Most of the seafood restaurants used seasoning which didn't age and was made by the restaurant. The cooking equipment used in the Maeuen-Tang's recipes were a cook-port and earthen bowl.
Environmental Specimen Banks (ESBs) are playing pivotal role in monitoring the effect of environmental pollution on the ecosystem based on the retrospective analysis of the representative samples collected regularly and stored in cryogenic condition. In Korea, National Environmental Specimen Bank (NESB) was established in 2009 and the standard operating procedures (SOPs) for sampling, and cryogenic milling and storage had been prepared during 2007-2010. Since then, the tentative SOPs for the seven kinds of specimens (shoots of Red Pine (Pinus densiflora) and Korean Pine (Pinus koraiensis), leaves of Mongolian Oak (Quercus mongolica) and Zelkova Tree (Zelkova serrata), eggs of Feral Pigeon (Columba livia var. domestica), muscles and organs of Common Carp (Cyprinus carpio), and Freshwater Bivalve (Unio (Nodularia) douglasiae)) have been put to test in the field and laboratory as well against the practicality and feasibility. The SOPs were improved by reflecting the findings from the research and the following discussion regarding the selection of specimen (Feral Pigeon suffering from a control management), sample size (a problem of decreasing number of sampling trees related to increasing sampling time) and period (a problem related to a bud growth), and sampling methods etc.. In addition, barcoding system for the management of the specimen information, and monitoring system of the cryogenic storage to regulate the optimum temperature and the liquid nitrogen level were also developed for the efficient and effective control of the samples. Lastly, the safety guide and emergency protocol were augmented to guarantee a safe work environment with the cryogenic facility. These improvements of the SOPs are expected to contribute to more stable operation of the NESB.

Environmental Specimen Banks (ESBs) have been established widely in the world since 1979 for monitoring long-term bio-accumulation of environmental pollutants. The ESBs perform the retrospective analysis of representative biological samples collected regularly and store them in cryogenic condition. In Korea, National Environmental Specimen Bank (NESB) was established in 2009. Since then, NESB had prepared the standard operating procedures (SOPs) for the seven kinds of specimens (Red Pine (Pinus densiflora), Common Carp (Cyprinus carpio), and etc.) for monitoring the effect of environmental pollution on the terrestrial ecosystem and river ecosystem. In 2012, NESB added Black-tailed Gull (Larus crassirostris)'s eggs to the list of the environmental specimen for monitoring marine environmental pollution. In this study, we sampled the eggs of the Black-tailed Gull on two islands (Baekryeongdo of the West Sea and Hongdo of the South Sea). Especially, we selected eggs which remained in the early stages of embryo development for certifying the consistent and stable monitoring for environmental pollutants in egg contents. However, it was not considered to classify an order of eggs per clutch in this study. It is known that the concentration variations of pollutants exist among eggs in one clutch. Therefore, it is needed to consider the positions of eggs in the laying sequences to meet the objective of bioaccumulation monitoring. We collected 30 eggs in each site but the amount may be insufficient to achieve storage target (over 2,000 g). Therefore, we need to consider an optimal sampling size.
The purpose of this study is to understand the living culture of transnational married women and to analyze the out door play of children in their hometown. The data was collected through observation from 27th June to 7th July 2008 in Jian, Jilin Province China. The children's play and lifestyles were observed, and data pertaining to the culture of the people were collected by a teacher and staff. We also visited the residents for housing information. The results are given below. 1. They dressed in Korean clothes on festive days and the boys put on a hood. They had eating habits which included cooking for themselves or buying semi-manufactured goods but did not use, instant food. The housing habits involved a combination of cooking and heating by Korean floor heating system(Ondol). They utilized outdoor space to grow vegetables. Those with a fulltime job(teacher) preferred to live in an apartment but an apartment was too expensive. Public utility charges and traffic expenses were cheap. 2. The main festive days are the lunar New Year's Day and Chuseok. The children returned home and enjoyed the festive day with their parents. The language used are Korean language and Chinese. Some Korean words and phrases in Jian Joseonjok have different meanings as compared to how they are used in Korea. A capping ceremony did not to celebrate becoming an adult from an adolescent. Couples performed a wedding ceremony at a wedding hall attended by their parents and invited relatives from both families. The relatives gave the couple a wedding gift. They did not go on a wedding trip as it was not affordable but instead spent their wedding night at a hotel in this culture. When someone dies, they bury the body after cremation. They perform a memorial service for three years on the birthday of the departed. They have a banquet on the 60th birthdays with their relatives and neighbours and are typically presented with a carp for longevity. 3. They understand capitalism and therefore send their children to school to improve their social position. The Korean and Chinese languages are required subjects in school. The students choose a second language(English or Russian). They prefer English class but at the time of this study an English class was not offered at the school in Jian Joseonjok. Therefore the children entered a Chinese school. 4. The children play outdoor games such as Y$\acute{a}$o J$\grave{i}\bar{a}$(要家), X$\grave{i}$ang g$\grave{i}$(象棋), T$\grave{i}\grave{a}$o p$\acute{i}$ j$\grave{i}$n(r)(跳皮節), D$\grave{o}$uch ing g$\grave{u}$n 凍冷根, B$\bar{e}$i B$\bar{e}$i 背背, and soccer. In this study, WUA (Weighted Usable Area) was calculated to determine ecological flow at the confluence of Nakdong River and Gumho River by using River2D. To calibrate River2D, simulation results of River2D were compared with calibrated HEC-RAS simulation results and the optimum parameters were determined. After parameter calibration, WUA of Zacco platypus and Zacco temmincki which are dominant species in Nakdong River was calculated with changing upstream flowrate. From the result, WUA is changed according to flowrate and growth stage. In the flowrate-WUA/A graph, ecological flow can be determined as $33.3m^3/s{\sim}39.96m^3/s$ in Nakdong River and $3.6m^3/s{\sim}4.32m^3/s$ in Gumho River. After dredging for Four major rivers restoration project, WUA of Zacco platypus and Zacco temmincki were calculated by using the ecological flow. The results show that WUA after dredging are decreased when compared with undredged condition. WUA of Common carp is 2~3 times bigger than WUA of Zacco platypus and Zacco temmincki at the dredged condition in Nakdong River.

A search for contraction and relaxation responses on the smooth muscles, antimicrobial and antioxidant activity in different body parts of 4 freshwater fish and 4 marine fish were conducted. The four freshwater fish studied were Sarotherodon niloticus (tilapia), Channa arus (snake head), Cyprinus carpio (Israel carp) and Siluru osotus (catfish), and the four marine fish were Scyliorhinus torazame (tiger shark), Ateiepus japonicus (tadpole fish), Mugil cephalus (gray mullet) and Thamnaconus modestus (file fish). Frozen samples were extracted with distilled water containing $1{\%}$ acetic acid. Antimicrobial activity against Bacillus subtilis and Eschrrichia coli was detected in extracts from several tissues in all species tested. Relatively high antimicrobial activity could also be detected in the bile extracts from C. carpio, M. cephalus, and T. modestus. Contraction and relaxation responses on smooth muscles could be detected in all species tested, especially in the intestine extracts. Antioxidant activity was also detected in extracts from several tissues in all species tested, while in the extracts from S. osotus, excluding livers and spleens, no antioxidant activity was detected. Results from this study suggest that fish are a potential source for the discovery of novel bioactive materials.
