Common carp (Cyprinus carpio) and its aquaculture breed Israeli carp samples were obtained from two separate aquaculture facilities under the similar raising conditions during two years in the Kunsan National University, Korea. Genomic DNA was isolated from the common carp and Israeli carp for identification of genetic characteristics and genomic polymorphisms by polymerase chain reaction amplification of DNA using arbitrary primers. The arbitrary primer No.21 (ACTTCGCCAC) yielded the highest number of fragments with the average of 15.0 among the primers used in Israeli carp. A tota1 of 294 polymorphic products in common carp and 336 in Israeli carp were observed by random primers. The average number of polymorphic products generated by random RAPD primer No. 2 (GTAGAC-CCGT) showed 8.0 in Israeli carp. On average, each random RAPD primer produced 5.4 amplified polymorphic products in common carp and 6.2 in Israeli carp. An average genetic similarity (BS value) was 0.44$\pm$0.05 within the common carp and 0.32$\pm$0.04 within the Israeli carp. The degree of similarity frequency (BS) between two carps was 0.67 as generated by the primer No. 19 (GACGGATCAG). The average level of bandsharing was 0.57$\pm$0.03 between the two carps. Accordingly, the two carp populations were genetically a little distant. The electrophoretic analysis of PCR-RAPD products showed middle levels of variation between the two carp populations. This result implies that the genetic diversity among intra-population may be higher when compared with that between the two carps. The RAPD polymorphism generated by these random primers might be used as a genetic marker for populations or lines identification in important aquacultural carp.
Serum immunoglobulins (Igs) from Israeli carp were purified using affinity chromatography. Fish were immunized with purified mouse IgG, and the specific fish antibodies were purified from the immune serum on a mouse IgG-immobilized agarose gel. Rabbit anti-Israeli carp Igs (R $\alpha$ I. carp Igs) antibodies were produced following hyperimmunization with mouse IgG specific carp antibodies. SDS-PAGE analysis under reducing condition showed that Israeli carp Igs were composed of two $\mu$-like heavy chains with about 82 and 50 kd, respectively, and one light chain with about 25 kd. On immunoblotting analysis, however, R $\alpha$ I. carp Igs failed to react with the light chain. When both protein A and protein G-purified normal carp Ig were compared with mouse IgG-specific Israeli carp Ig, no significant structural differences among them were observed. To investigate if there is any homology between other fish Ig molecules, cross-reactivity of R $\alpha$ I. carp Igs against Ig molecules from 6 different fish sera and mouse control serum was checked on immunoblotting analysis. As a result, R $\alpha$ I. carp Igs responded to Israeli carp, common carp, and tilapia Ig molecules. In flow cytometry study, however, R $\alpha$ I. carp Igs appeared to recognize 42.0%, 35.8% and <5% of Israeli carp, common carp and tilapia $Ig^+$ head kidney cells, respectively. The result suggests the heterogeneity between receptor Igs on B-like lymphocytes and soluble Igs in serum. It is crucial to obtain pure fish Igs to produce reagent antibodies as tools for the study on their specific immune responses.
The cDNAs encoding glutathione peroxidase (GPx) were cloned and sequenced from the liver of three Chinese carps with different tolerance to hepatotoxic microcystins, phyto-planktivorous silver carp (Hypophthalmichthys molitrix) and bighead carp (Aristichthys nobilis), and herbivorous grass carp (Ctenopharyngodon idellus). Using genome walker method, a 750 bp 5'-flanking region of the silver carp GPx gene was obtained, and several potential regulatory elements were identified in the promoter region of the GPx gene. The silver carp GPx gene was widely expressed in all tissues examined. Despite phylogenetic analysis, assigning this newly described carp GPx to the group of mammalian GPx2, the carp GPx seems more similar to GPx1 from a physiological point of view. The constitutive expression pattern of the three carp liver GPx gene, shows a positive relationship with their tolerance to microcystins.

Parasitic infection is among the most common problems for carp cultivation. They are also important for the principal entrance of other hazardous infections as well. This study was carried out for determining of parasitic fauna of two major carp known as silver and grass carp with the comparison of prevalence value and intensity rate of parasites among them, alongside the relationship between the biometric characteristics and host sex with the infection level. For this purpose, a total of 94 fish samples were caught randomly using a fishing net, from Guilan ponds during spring and summer of the year 2018 and transported alive to the laboratory. Upon arriving, the biometric characteristics and genus of each carp were measured individually. Specimens were then acquired from the skin, gills, and eyes of the carp and examined according to standard parasitology methods. Recovered parasites were observed under a light microscope and then fixed for identification. As the result, the occurrence and intensity in the higher length group were comparatively greater than the lower one. Also, the prevalence and intensity of total parasites in male carp were higher than in females. In this research, Dactylogyrus hypophthalmichthys and Dactylogyrus aristhichtys were observed in silver carp and Dactylogyrus lamellatus was detected in grass carp. In the paper below, we found that the host specificity varies in different species of Dactylogyrus isolated from grass carp and silver carp.
In this study, carp edemavirus disease and herpesviral hematopoietic necrosis were examined in carp (Cyprinus carpio) and crucian carp (Carassius auratus) imported from China. From August 2016 to May 2017, 100 carp and 300 crucian carp were examined. There were no clinical symptoms in all the tested fish, and no internal or external parasitic infections were identified except for the detection of Argulus sp. in one leather carp. Carp edema virus disease was examined by PCR in C. carpio, but all were negative. In C. auratus, herpesviral hematopoietic necrosis was tested by PCR and 4 out of 300 were positive. The results of this study are expected to be useful as basic data for conducting risk analysis on the relevant organisms and counterpart countries or for designating new pathogens for quarantine.
The full-length cDNA of grass carp (Ctenopharyngodon idellus) and silver carp (Hypophthalmichthys molitrix) uncoupling protein 2 (UCP2) was obtained from liver. The grass carp UCP2 cDNA was determined to be 1152 bp in length with an open reading frame that encodes 310 amino acids. Five introns (Intron 3, 4, 5, 6 and 7) in the translated region, and partial sequence of Intron 2 in the untranslated region of grass carp UCP2 gene were also obtained. Gene structure comparison between grass carp and mammalian (human and mouse) UCP2 gene shows that, the UCP2 gene structure of grass carp is much similar to that of human and mouse. Partial UCP2 cDNA sequences of bighead carp (Aristichthys nobilis) and mud carp (Cirrhinus molitorella), were further determined. Together with the common carp (Cyprinus carpio) UCP2 sequence from GenBank (AJ243486), multiple alignment result shows that the nucleotide and amino acid sequences of the UCP2 gene, were highly conserved among the five major Chinese carps that belong to four subfamilies. Using beta-actin as control, the ratio UCP2/beta-actin mRNA (%) was determined to be $149.4{\pm}15.6$ (common carp), $127.4{\pm}22.1$ (mud carp), $96.7{\pm}12.7$ (silver carp), $94.1{\pm}26.8$ (bighead carp) and $63.7{\pm}16.2$ (grass carp). The relative liver UCP2 expression of the five major Chinese carps, shows a close relationship with their food habit: benthos and detrituseating fish (common carp and mud carp) > planktivorious fish (silver carp and bighead carp) > herbivorious fish (grass carp). We suggest that liver UCP2 might be important for Chinese carps to detoxify cyanotoxins and bacteria in debris and plankton food.
잉어(Cyprinus carpio) 암컷과 붕어(Carassius auratus) 수컷간 유도된 잡종에 대한 세포 유전학적 연구를 수행하였다. 유도된 잡종의 세포 및 핵의 크기는 양친의 중간값을 나타내었고, 염색체의 경우 잉어, 붕어 및 유도된 잡종 모두 2n=100 으로 나타났다. Flow cytometry를 이용, 잡종의 genome size를 분석한 결과, 유도된 잡종은 3.7pg/cell를 갖음으로서 잉어(3.6pg/cell)와 붕어(3.8pg/cell)의 중간치를 나타내었다.
Metallothioneins (MTs) are low molecular weight, cysteine-rich, and heavy metal binding proteins, which could be induced with heavy metals such as Cd, Hg, Zn and Cu in liver, kidney, and in cultured cells. By using ion exchange chromatography on DE-52, MT was purified from the serum of carp induced with cadmium in order to produce antibody against MT. Polyclonal antibody produced against purified carp MT reacted well with MT in the serum of carp induced with cadmium, whereas control serum did not. This may indicate that the polyclonal antibody against the carp MT could be used for the preparation of biosensors to detect MT in fishes like carp.
방글라데시 미멘싱에 소재하는 양식어장에서 pangasiid catfish (Pangasius hypophthalmus), silver carp (Hypophthalmichthys molitrix)와 catla (Catla catla)의 조합과 혼합비를 달리하여 16주간 복합양식하였을 때, 이들 어종의 성장률을 조사하였다. 헥타르당 30,000개체의 밀도로 복합양식하여 성장률을 측정하였으며, 혼합조성비는 다음과 같다: 실험구(treatment) 1은 pangasiid catfish와 silver carp을 1:1비율로, 실험구 2는 pangasiid catfish와 cata를 1:1비율로, 실험구 3과 4는 pangasiid catfish, silver carp과 catla를 각각 2:1:1과 3:2:1비율로 복합양식하였다. 양식환경에 영향을 주는 수온, 투명도, 용존산소량, pH, $NO_{3}-N$, $PO_{4}-P$, 엽록소 a와 같은 몇몇 물리화학적, 생물학적 요인과 플랑크톤의 군집을 정기적으로 조사하였다. 어류의 성장속도는 비 성장율(specific growth rate), 일간 체중 증가(daily weight gain)와 체중증가 비율(percent weight gain)을 측정하였다. Pangasiid catfish의 성장률과 사료이용률은 모든 실험구에서 유사하게 나타났다. Silver carp의 성장률은 모든 실험구에서 유의한 차이를 나타내었는데(p<0.05), 양식밀도가 증가할수록 성장률이 현저하게 감소하였으며, Silver carp의 밀도가 가장 낮은 복합양식 조성인 pangasiid catfish, silver carp과 catla의 비율이 각각 2:1:1일 때, silver carp의 성장률이 가장 높게 나타났다. Silver carp의 밀도가 증가할수록 catla의 성장률이 현저하게 감소하는 것으로 나타나, catla의 성장률은 silver carp의 유무에 영향을 받는 것으로 판단되었다. Catla의 성장률은 pangasiid catfish와 calta의 비율이 1:1일 때, 가장 높은 것으로 나타났다. 양식수율과 경제적 이익은 실험구에 따라 현저하게 다른 양상을 보였다(p<0.05). 가장 높은 양식수율과 경제적 이익은 pangasiid catfish, silver carp과 catla를 각각 2:1:1 비율로 양식한 결과에서 얻어졌는데, 이는 silver carp의 높은 성장률과 catla의 시장 단가가 높은 것에 기인하는 것으로 나타났다. 모든 실험구의 수질 상태는 양식기간 동안 적절한 수준을 유지하였으며, 각각의 값은 모든 실험구에서 뚜렷한 차이를 보이지 않았다(p<0.05).
