LNAPL에 의한 소수성 유기오염물질의 지하환경 내 이동성 변화가 위해성 증가에 미치는 영향: p-xylene과 benz[a]anthracene의 경우 (Enhanced Transport and Risk of a Highly Nonpolar Pollutant in the Presence of LNAPL in Soil-groundwater System: In Case of p-xylene and benz[a]anthracene)

  • 물질 간 상호작용이 위해도에 많은 영향을 미칠 수 있다는 것은 널리 알려져 있지만 이를 위해성평가에 적용할 수 있는 방안은 아직 미미한 실정이다. 본 연구에서는 p-xylene이 토양 내에 존재할 때, 대수층으로의 이동성이 극히 제한적인 benz[a]anthracene (BaA)의 지하수로의 이동성 증가를 확인하고 그 증가된 위해도를 산정하기 위하여, 지하 환경을 모사한 셀을 제작하여 실험을 수행하였으며, 이를 해석하는 간단한 물질이동 모델을 개발하였다. 셀 실험 결과 BaA의 이동성은, BaA와 p-xylene이 혼합물질로 동시에 존재하는 경우에, BaA만으로 오염되어 있는 경우보다 월등히 높았다. 후자의 경우는 강우에 의한 이동이 주요 이동기작이었으며 그 정도는 아주 미미하였다. Defined Time Steps (DTS)을 이용하여 개발된 오염물질 이동모델은 두 물질 모두 실험결과와 유사한 경향을 보이며 변하는 것을 확인하였으나, 더욱 정확한 예측을 위하여 모델의 수정, 보완작업이 계속되고 있으며 이는 다음 연구의 주제가 될 것이다. 실험실 규모의 셀 실험의 경우 모세관대가 물질이동에 중요한 역할을 하기 때문에 실험실 규모의 시뮬레이션에서는 모세관대 모듈을 적용하는 것이 보다 적합한 것으로 나타났다. 현장규모 오염에서의 모델링 결과를 토대로 위해도를 산정하고 비교한 결과, 지하수 섭취를 통한 BaA의 발암위해도는 NAPL이 존재할 때, 존재하지 않을 때보다, 약 15${\sim}$87배 크게 계산되었다. NAPL이 존재하지 않는 경우 BaA의 지중 이동속도는 매우 느리며, 실제 오염현장의 경우 이동 과정에서 물질분해가 일어나므로 NAPL의 존재 여부에 따른 실제 위해도 차이는 더 크게 발생할 것으로 예상된다.

Lack of Associations between Genetic Polymorphisms in GSTM1, GSTT1 and GSTP1 and Pancreatic Cancer Risk: A Multi-Institutional Case-Control Study in Japan

  • Background: We aimed to evaluate the role of genetic polymorphisms in tobacco carcinogen-metabolizing genes and their interactions with smoking in a hospital-based case-control study of Japanese subjects. Materials and Methods: We examine the associations of pancreatic cancer risk with genetic polymorphisms in GSTM1, GSTT1 and GSTP1, phase II enzymes that catalyze the conjugation of toxic and carcinogenic electrophilic molecules. The study population consisted of 360 patients and 400 control subjects, who were recruited from several medical facilities in Japan. Unconditional logistic regression methods were used to estimate odds ratios (ORs) and 95% confidence intervals (CIs) for the associations between genotypes and pancreatic cancer risk. Results: Among the control subjects, the prevalence of the GSTM1-null genotype and the GSTT1-null genotype was approximately 56% and 48%, respectively. Cases and controls were comparable in terms of GSTM1 and GSTT1 genotype distributions. Neither of the deleted polymorphisms in GSTM1 and GSTT1 was associated with the risk of pancreatic cancer, with an age- and sex-adjusted OR of 0.99 (95%CI: 0.74-1.32) for the GSTM1-null genotype, and 0.98 (95%CI: 0.73-1.31) for the GSTT1-null genotype. The OR was 0.97 (95%CI: 0.64-1.47) for individuals with the GSTM1 and GSTT1-null genotypes compared with those with the GSTM1 and GSTT1- present genotypes. No synergistic effects of smoking or GST genotypes were observed. Conclusions: Our results indicate no overall association between the GSTM1 and GSTT1 deletion polymorphisms and pancreatic cancer risk in the Japanese subjects in our study.

Estimation of Gas-particle partitioning Coefficients (Kp) of Carcinogenic polycyclic Aromatic hydrocarbons in Carbonaceous Aerosols Collected at Chiang - Mai, Bangkok and hat-Yai, Thailand

  • To assess environmental contamination with carcinogens, carbonaceous compounds, water-soluble ionic species and trace gaseous species were identified and quantified every three hours for three days st three different atmospheric layer at the heart of chiang-Mai, bangkok and hat-Yai from December 2006 to February 2007. A DRI model 2001 Themal/Optical Carbon Analyzer with the IMPROVE thermal/optical reflectance (TOR) protocol was used to quantify the organic carbon(OC) and elemental carbon content in $PM_{10}$. Diurnal and vertical variability was also carefully investigated. In general, OC and EC contenttration shoeed the highest values at the monitoring period o 21.00-00.00 as consequences of human activities at night bazaar coupled with reduction of mixing layer, decreased wind speed and termination of photolysis nighttime. Morning peaks of carboaceous compounds were observed during the sampling period of 06:00 -09:00, emphasizing the main contribution of traffic emission in the three cities. The estimation of incremental lifetime partculate matter exposure (ILPE) raises concern of high risk of carbonaceous accumulation over workers and residents living close to the observatory sites. The average values of incremental lifrtime particulate matter exposure (ILPE) of total carbon at Baiyoke Suit Hotel and Baiyoke Sky Hotel are approsimately ten time shigher then those air sample collected at prince of songkla University Hat-Yai campus corpse incinerator and fish-can maufacturing factory but only slightly higher than those of rice straw burnig in Songkla province. This indicates a high risk of developing lung cancer and other respiratory diseases across workers and residents living in high buildings located in Pratunam area. Using knowledge of carbonaceous fractions in $PM_{10}$, one can estimate the gas-particle partitioning of polycyclic aromatic hydrocarbons (PAHs). Dachs-Eisenreich model highlights the crucial role of adsorption in gas-particle partitioning of low molecular weight PAHs, whereas both absorption and adsorption tend to account for gas-particle partitioning of high molecular weight PAHs in urban residential zones of Thailand. Interestingly, the absorption mode alone plays a minor role in gas-partcle partitiining of PAHs in Chiang-Mai, Bangkok and hat-Yai.

RTN4 3'-UTR Insertion/Deletion Polymorphism and Susceptibility to Non-Small Cell Lung Cancer in Chinese Han Population

  • Nogo protein, encoded by gene reticulon-4 (RTN4), includes three major isoforms by different splicing, named Nogo-A Nogo-B and Nogo-C. Nogo proteins play an important role in the apoptosis of cells, especially in tumor cells. RTN4 single nucleotide polymorphisms (SNPs) can influence the efficiency of transcription and translation thus being related with an individual's predisposition to cancer. The CAA insertion/deletion polymorphism (rs34917480) within RTN4 3'-UTR has been reported to be associated with many cancer types. In order to investigate the relationship between this polymorphism and susceptibility to non-small cell lung cancer (NSCLC) in the Chinese population, we conducted the present case-control study including 411 NSCLC patients and 471 unrelated healthy controls. The genotype distributions were significantly different between cases and controls (p=0.014). We found that the del allele could significantly increase NSCLC risk (ins/ins vs ins/del: p=0.007, OR 1.46, 95%CI=1.11-1.93; dominant model: p=0.004, OR 1.47, 95%CI=1.13-1.92 and allele model: p=0.008, OR 1.35, 95%CI=1.08-1.67). This association was stronger in participants over 60 years old, males and smokers. We therefore conclude that the CAA insertion/deletion polymorphism (rs34917480) contributes to non-small cell lung cancer risk in Chinese population. Age, sex and environmental exposure are also related to carcinogenic effects of rs34917480.

대기중 다환 방향족 탄화수소류의 분석에 관한 연구 (A Study on the Analysis of Polycyclic Aromatic Hydrocarbons in Air)

  • 환경중 다환 방향족 탄화수소류(PAHs)는 유기물의 연소중의 열합성에 의해서 주로 발생하며 강한 발암성물질로 알려져 있다. 또한 도시 대기중의 PAHs는 주로 자동차 배기가스 및 주택연료의 불완전연소에 의해서 발생하는 것으로 알려져 있으며, 국내에서도 자동차의 수요 급중과 도시난방의 서구화에 따라 도시 대기중 PAHs의 증가가 우려되며 이에따라 대기중 PAHs의 인체 위해성 평가가 시급한 실정이다. 본 연구에서는 위해성 평가를 위한 기초 조사로서 1996년 4월부터 1997년 1월가지 1년에 4차례에 걸쳐 서울을 비롯한 6대 도시 14개 지역에서의 대기중의 23종의 PAHs농도를 측정하였다. 분석 결과 총 263종의 거의 모든 시료에서 14종의 PAHs가 검출되었으며 1년간 총 PAHs의 평균 농도는 $28.72ng/m^3$이었고, 겨울철(평균 $47.76ng/m^3$)에 가장 높은 농도로 검출되었다. 각 시료에서 추출된 총 유기물질(EOM ; Exoacted Organic Material)과 총 PAHs 농도는 대체로 비례하였으며 EOM에 함유된 총 PAHs는 평균 0.28%로 조사되었다. 본 실험 결과 PAH의 위해성 평가가 수행되어야 할 것으로 기대되며 계절별 시료 분석을 지속적으로 수행하여 보다 신뢰성 있는 데이타 구축이 필요하다.

국내 유통 김치 중 N-Nitrosodimethylamine과 그 전구물질의 함량 분석 및 안전성 평가 (Analysis and Risk Assessment of N-Nitrosodimethylamine and Its Precursor Concentrations in Korean Commercial Kimchi)

  • Dimethylamine(DMA), 질산염, 아질산염, biogenic amines은 발암성 니트로사민의 전구물질이다. 본 연구는 국내 상품 김치로 유통되고 있는 국내산 배추김치, 깍두기, 총각김치, 맛김치, 묵은김치, 백김치의 김치 내 DMA, 질산염, 아질산염, biogenic amines의 오염도를 조사하였다. 배추김치 내 DMA, 질산염, 아질산염 함량은 각각 29.7, 2,178.8, 3.0 mg/kg이었다. 김치 종류 중에서 깍두기의 DMA와 질산염 함량이 가장 낮았다. Tryptamine, putrescine, cadaverine, tyramine, spermidine이 백김치와 총각김치를 제외한 분석된 김치 내 검출되었다. 백김치 내 tryptamine이 단지흔적량 존재하고 spermidine은 총각김치에서는 검출되지 않았다. 배추김치 내 tryptamine, putrescine, cadaverine, tyramine, spermidine의 함량은 각각 15.0, 64.6, 18.0, 44.0, 7.8 mg/kg이었다. 깍두기 내 tyramine의 함량이 낮았다. 깍두기 내 NDMA의 오염도를 분석하였고, 그 평균 함량은 $1.38{\mu}g/kg$이었다. 깍두기 1일 평균 섭취량과 체중을 근거로 깍두기 섭취군의 NDMA 인체노출량을 산출한 결과 깍두기 섭취로 인한 NDMA 노출량은 $2.31{\times}10^{-7}mg/kg\;b.w./d$로 나타났으며, 노출안전역(MOE)은 259,924로 위해 정도가 매우 낮은 안전한 수준으로 나타났다.

Human Papillomavirus Genotype Distribution and E6/E7 Oncogene Expression in Turkish Women with Cervical Cytological Findings

  • Background: Infection with certain human papillomavirus (HPV) genotypes is the most important risk factor related with cervical cancer. The objective of the present study was to investigate the prevalence of HPV infection, the distribution of HPV genotypes and HPV E6/E7 oncogene mRNA expression in Turkish women with different cervical cytological findings in Mersin province, Southern Turkey. Materials and Methods: A total of 476 cytological samples belonging to women with normal and abnormal cervical Pap smears were enrolled in the study. For the detection and genotyping assay, a PCR/direct cycle sequencing approach was used. E6/E7 mRNA expression of HPV-16, 18, 31, 33, and 45 was determined by type-specific real-time NASBA assay (NucliSENS EasyQ$^{(R)}$HPV v1.1). Results: Of the 476 samples, 106 (22.3%) were found to be positive for HPV DNA by PCR. The presence of HPV was significantly more common (p<0.001) in HSIL (6/8, 75%) when compared with LSIL (6/14, 42.9%), ASC-US (22/74, 29.7%) and normal cytology (72/380, 18.9%). The most prevalent genotypes were, in descending order of frequency, HPV genotype 66 (22.6%), 16 (20.8%), 6 (14.2%), 31 (11.3%), 53 (5.7%), and 83 (4.7%). HPV E6/E7 oncogene mRNA positivity (12/476, 2.5%) was lower than DNA positivity (38/476, 7.9%). Conclusions: Our data present a wide distribution of HPV genotypes in the analyzed population. HPV genotypes 66, 16, 6, 31, 53 and 83 were the predominant types and most of them were potential carcinogenic types. Because of the differences between HPV E6/E7 mRNA and DNA positivity, further studies are required to test the role of mRNA testing in the triage of women with abnormal cervical cytology or follow up of HPV DNA positive and cytology negative. These epidemiological data will be important to determine the future impact of vaccination on HPV infected women in our region.

  • Food safety is a term broadly applied to food quality that may adversely affect human health. These include zoonotic diseases and acute and chronic effects of ingesting natural and human-made xenobiotics. There are two major areas of concern over the presence of residues of antibiotics in animal-derived foodstuffs with regard to human health. The first is allergic reactions. Some antibiotics, such as penicillins can evoke allergic reactions even though small amounts of them are ingested or exposed by parenteral routes. The second is development of antibiotic resistance in gut bacteria of human. Recently multi-resistant pneumococcal, glycopeptide-resistant enterococci and gram negative bacteria with extended-spectrum $\beta$-lactamases have spread all over the world, and are now a serious therapeutic problem in human. Although it is evident that drugs are required in the efficient production of meat, milk and eggs, their indiscriminate use should never be substituted for hygienic management of farm. Drug should be used only when they are required. In addition to veterinary drugs, environmental contaminants that were contaminated in feed, water and air can make residues in animal products. Mycotoxins, heavy metals, pesticides, herbicides and other chemicals derived from industries can be harmful both to animal and human health. Most of organic contaminants, such as dioxin, PCBs and DDT, and metals are persistent in environment and biological organisms and can be accumulated in fat and hard tissues. Some of them are suspected to have endocrine disrupting, carcinogenic, teratogenic, immunodepressive and nervous effects. The governmental agencies concerned make efforts to prevent residue problems; approval of drugs including withdrawal times of each preparation of drugs, establishment of tolerances, guidelines regarding drug use and sanitation enforcement of livestock products. National residue program is conducted to audit the status of the chemical residues in foods. Recently HACCP has been introduced to promote food safety from farm to table by reducing hazardous biological, chemical and physical factors. Animal Production Food Safety Program, Quality Assurance Programs, Food Animal Residue Avoidance Databank are para- or non-governmental activities ensuring food safety. This topic will cover classification and usage or sources of chemical residues, their adverse effects, and chemical residue status of some countries. Issues are expanded to residue detection methodologies, toxicological and pharmacokinetic backgrounds of MRL and withdrawal time establishments, and the importance of non-governmental activities with regard to reducing chemical residues in food.

정상세포와 암세포의 증식 억제에 대한 에탄올의 영향 (Anti-Proliferative Effect of Ethanol on Normal and Cancer Cells)

  • 에탄올은 사람에 대한 발암물질로 잘 알려져 있다. 또한 여러 조직이나 세포에서의 에탄올에 의한 세포증식억제효과도 잘 알려져 있다. 본 연구에서는 여러 암세포에서 에탄올에 의한 세포증식억제 효과를 조사하였는데 특히 발암원성 $ras$로 형질전환되거나 미세주입된 세포에서의 영향을 조사하였다. 에탄올은 여러 정상세포들의 증식을 억제하였다. 반면에 여러 암세포나 발암원성 Ras에 의한 세포증식은 억제하지 못 하였다. 또한 발암원성 단백질의 세포내 미세주사에 의한 DNA합성 유도도 에탄올에 의해 억제 되지 않았다. 이러한 에탄올의 세포증식억제 효과는 $N$-acetylcysteine이나 4-methylpyrazole과 같은 항산화제에 의해 제거되었다. 이러한 실험 결과는 에탄올에 의한 세포증식억제 효과는 Ras단백질의 upstream에 있거나 또는 Ras와 독립적으로 작용하며, 활성산소 형성과 밀접한 관계가 있다는 것을 알려준다.

EFFECTS OF 50Hz CIRCULARLY POLARIZED MAGNETIC FIELDS ON SPONTANEOUS MAMMARY TUMORS IN RATS

  • Several epidemiological studies have suggested that residential or occupational exposure to power frequency magnetic fields (MF) might increase the risk of cancer. The objective of this study is to elucidate the possible carcinogenic effects of MF exposure using female Sprague-Dawley (Crj:CD)rats. A total of 360rats was randomly divided into 6 groups of 60 rats each. Two groups were served as a negative control (vehicle: sesame oil only) or a positive control (single oral administration of 7, 12dimethylbenz(a)anthracene; DMBA, 90mg/kg body weight at 50-52days of age). Other four groups were simultaneously exposed to 0 (sham-exposed) 7, 70 or 350 $\mu$T(rms), continuous circularly polarized 50HzMF, 22 hrs/day, 7 days/weeks for 30weeks from 8weeks of age. Experiment was conducted under SPF condition and in a blinded manner, Ten animals in each grout were served as satellite animals and their several hormonal concentrations in sera, such as melatonin and prolactin, collected at the midnight were measured. In addition, complete histopathological examination were performed in other 50 animals per each group. In the positive control group, the first mammary nodule was palpated at the 7th weeks of experiment in 5 out of 59 animals. Afterward, the incidence of palpable mammary nodules increased steadily and reached at 76% and 98% of live animals at 14weeks and the end of experiment in sham and 350$\mu$ T-exposed groups were not significantly different from those in the sham-exposed and negative control groups. Histopathologocally, most of palpable nodules were mammary tumors. The incidences of animals with mammary tumors per animals survived ant the end of experiment were 4.1 and 100% in the negative and positive control groups, and 0.0, 6.0, 8.0 and 6.0% in the sham-, 7, 70and 350 $\mu$T-exposed groups, respectively. These incidences in three MF-exposed groups were not significantly different from those in both the sham- exposed and negative control groups. Based on these results, it was not supported that continuous circularly polarized 50 Hz magnetic fields at up to 350$\mu$T affect the incidence of spontaneous mammary tumors in female SD rats under the present experimental conditions.
