Determination of Significance Threshold for Detecting QTL in Pigs (돼지의 QTL 검색을 위한 유의적 임계수준(Threshold) 결정)

  • Interval mapping using microsatellite markers was employed to detect quantitative trait loci (QTL) in the experimental cross between Berkshire and Yorkshire pigs. In order to derive critical values (CV) for test statistics for declaring significance of QTL, permutation test (PT) of Churchill and Doerge method(1994) and the analytical method(LK) of Lander and Kruglyak(1995) were used by each trait and chromosome. 525 $F_2$ progeny phenotypes of five traits(carcass weight, loin eye area, marbling score, cholesterol content, last back fat thickness) and genotypes of 125 markers covering the genome were used. Data were analyzed by line cross regression interval mapping with an F-test every by 1cM. PT CV were based on 10,000 permutations. CV at genome-wise test were 10.5 for LK and ranged from 8.1 to 8.3 for PT, depending on the trait. CV, differed substantially between methods, led to different numbers of quantitative trait loci (QTL) to be detected. PT results in the least stringent CV compared at the same % level.

Changes of Carcass Traits and Surface Meat Color of Korean Cattle (Hanwoo) Reared Different Altitudes or Transferred from Different Distance (고도 및 수송거리별 출하 한우의 도체특성 및 표면육색의 변화)

  • The purposes of the study were to examine the changes in carcass traits and surface meat color, and cortisol concentration by different altitudes and shipping distances for steer and cow. The experimental animals were shipped from Kangwondo, Kyunggido, Choongchungdo, and Kyungsangdo to Wonju LPC. The animals were examined for yield traits, quality traits, carcass grade, lightness, redness, yellowness, croma value, and cortisol concentration by different altitudes and shipping distances. The results showed that the carcass traits of steer like back fat thickness were not different by shipping distances of 100 km, 150 km, and 200 km. However, the fat thickness was higher in steers shipped from 250 km than 100 km, 150 km, and 200 km distance. Ribeye area was reduced significantly in 200 km and 250 km than 50 km. Yield index and yield grade were significantly low in 250 km than 50-200 km. However, meat color, fat color, texture, mature, and quality grade had no differences between shipping distances. Marbling score was not different in 50-200 km. However, the marbling score in 250 km was significantly lower than that of 50 km. In case of surface meat color by shipping distance, redness, yellowness, chroma value, and hue-angle were not different in shipping distance of 250 km. The lightness had similar result in 50-200 km. However, in case of 250 km the lightness was significantly low. The REA of cow carcass by shipping distance had no differences by shipping distance. The BET had similar results in 50-150 km. However, it had significantly thick in 200 km. The yield index and yield grade had no differences in 50-150 km. However, yield index and grade were significantly low in 200 km. The carcass trait of cow had no differences in all items by shipping distance. Although the carcass traits and the BET for steer by altitude had no differences between 100, 200, 300 and 500 m, but those were significantly thick in 400 m. And the yield index and the yield grade at altitude 400 m were lower than that of other altitudes. The quality traits and the quality grade had no differences between 100, 200, 300, 400, and 500 m altitudes. The yield traits, quality traits, yield grade, and quality grade had no significant differences by altitudes. In case of yield index of cow for 300 m was low than the cases of 100 m and 200 m. The surface meat color for steer and cow had no differences by altitudes. However, the lightness of cow had positive result in 100 m than 200 m and 300 m. In case of steer and cow the cortisol concentration by shipping distance was high as the shipping distances were longer. However, the cortisol concentrations of steer and cow by altitudes were decreased as the altitudes were increased. From the above results carcass traits and carcass grade were decreased and the cortisol concentration was increased as the altitudes were decreased for steer and cow.

  • The rapid development of the sheep genetic linkage map over the last five years has given us the ability to follow the inheritance of chromosomal regions. Initially this powerful resource was used to find markers linked to monogenic traits but there is now increasing interest in using the genetic linkage map to define the complex of genes that control multigenic production traits. Of particular interest are those production traits that are difficult to measure and select for using classical quantitative genetic approaches. These include resistance to disease where a disease challenge (necessary for selection) poses too much risk to valuable stud animals and meat and carcass qualities which can be measured only after the animal has been slaughtered. The goal for the new millennium will be to fully characterise the genetic basis of multigenic production traits. The genetic linkage map is a vital tool required to achieve this.

Estimation of Genetic Parameters for Ultrasound and Carcass Traits in Hanwoo (한우의 초음파 측정 형질과 도체 형질의 유전모수 추정)

  • This study was conducted to estimate genetic parameters for ultrasound and carcass traits in Hanwoo. Heritabilities and genetic and phenotypic correlations were estimated for carcass and ultrasound measurements collected from Hanwoo cows (n=312) born at Hanwoo experiment station. Traits evaluated were eye muscle area (EMA), backfat thickness (BF), marbling score (MS) from carcass, and ultrasound eye muscle area (UEMA), ultrasound backfat (UBF), and ultrasound marbling score (UMS). Parameters were estimated using multi-trait animal models byderivative-free restricted maximum likelihood procedures. Estimated heritabilities for UBF, UEMA and UMS were 0.43, 0.23 and 0.32, while heritabilities for BF, EMA and MS were 0.33, 0.13 and 0.33 in fattened cows, respectively. Genetic correlations between ultrasound and carcass measurements were estimated to -0.19, -0.61, and -0.36 for UBF: UEMA, UBF: UMS, and UEMA: UMS in fattened cows, respectively. Phenotypic correlations between ultrasound and carcass measurements were 0.03, 0.13 and 0.26 for UBF: UEMA, UBF: UMS, and UEMA: UMS in fattened cows, respectively. As for the steer, genetic correlations between ultrasound and carcass measurements were 0.36, -0.80 and 0.27 for UBF: UEMA, UBF: UMS, and UEMA: UMS in steers, respectively. Phenotypic correlations between ultrasound and carcass measurements were 0.13, 0.07 and 0.41 for UBF: UEMA, UBF: UMS, and UEMA: UMS in steers, respectively. In conclusion, this finding would be very useful to implement into Hanwoo breeding program.

Effects of Geographic Locations and Year-Seasons of Birth on Ultrasound Scanned Measures and Carcass Traits of Hanwoo Steers (한우 거세우의 초음파 생체진단형질과 도체형질에 대한 지역과 출생년도 및 계절 효과 분석)

  • Ultrasound measures of back fat thickness (UBF), eye muscle area (UEMA) and marbling score (UMS) and carcass measures of carcass weight (CW), backfat thickness (BF), eye muscle area (EMA) and marbling score (MS) were available on 26,129 Hanwoo steers. Statistically significant differences by regions of the farms location and birth years-seasons for the steers taken ultrasound measures and their carcass measures (p<0.01) were found. Steers in Gyeonggi province showed the highest values in ultrasound measures and carcass traits except in BF compared to steers in the other provinces. Comparing between ultrasound and carcass measures, UBF was thicker in general than BF in all regions except in Daejeon city. UEMA was higher than EMA in all regions except in Gyeonggior in Jeju provinces. Especially, the difference in Jeonnam province between UEMA and EMA was $7cm^2$ while the differences between UMS and MS ranged from 0.9 to 2.26 depending on the regions of steers located. Steers born in spring showed greater ultrasound or carcass values than those born in autumn. However, carcass measures of steers born in autumn were greater than those born in spring, 2009 except MS. The pearson and residual correlations were 0.63 and 0.65 between UBF and BF, 0.31 and 0.32 between UEMA and EMA and 0.56 and 0.56 between UMS and MS, respectively.

Repeated Records Animal Model to Estimate Genetic Parameters of Ultrasound Measurement Traits in Hanwoo Cows (반복모형을 이용한 한우 초음파 측정형질의 유전모수추정)

  • The present study data were obtained from 36,894 cows in Korea Animal Improvement Association from 2001 to 2009 which was subjected for ultrasound measurements (eye muscle area, back-fat thickness, marbling score) and descent. Repeated record models were carried out using 7,913 of 36,894 of total animal traits. The ultrasound measured traits and performance test data were used to study the chest girth, body condition score, eye muscle area, back-fat thickness and marbling score with genetic correlation and parameters for the ultrasound measured traits using REMLF90 program. Genetic correlation of eye muscle area with back-fat thickness, marbling score and back-fat thickness with marbling score were noticed in repeated records animal model as 0.69, 0.54, and 0.59, whereas in multiple trait animal model method were 0.07, 0.66, and 0.39, respectively. Repeated records of animal models were used as positive correlation of traits. Multiple trait animal models were used as negative correlation of eye muscle area with marbling score. The analysis on repeat records of animal models using ultrasound measurements about Korean cattle showed positive effects for each traits. In comparison differences between the repeat records of animal models and multiple trait animal models was found with higher traits of her, the heritability and repeatability was found higher in repeat records animal models. In light of these assessments, carcass traits by ultrasound measurements are expected to help and improve an accurate analysis of each trait and if the research analysis using repeat records of animal models continue when we estimate genetic ability of these traits.

Genetic Parameter Estimation on the Growth and Carcass Traits in Hanwoo(Korean Cattle) (한우의 성장 및 도체형질에 대한 유전모수 추정)

  • This study was conducted to investigate the genetic correlations among the traits used to select young bulls and proven bulls in Hanwoo Performance and Progeny Test Program in Korea. For the estimation of heritabilities and correlations among the growth traits of bulls and carcass traits of progeny steers, 2,532 records of performance tested bull calves and 1,819 records of progeny tested steers were collected from Livestock Improvement Main Center (LIMC), National Agricultural Cooperative Federation (NACF). Fixed effects of mixed model for each traits were selected by using stepwise regression analysis and prior values of variance components were estimated by MTDFREML. The prior values of variance components were estimated with pairwise 2 traits model followed by single trait analysis. The estimated heritability of backfat thickness(BF), dressing percentage(DP), loin-eye muscle area(LMA), marbling score(MS) and weight at 12 months(WT12) was 0.51, 0.32, 0.27, 0.33, 0.50 and 0.26, respectively. Genetic correlation of WT12 of bull calves with backfat thickness, carcass weight and loin-eye muscle area of steers was positive correlation as 0.05, 0.35 and 0.21, respectively. However genetic correlation of WT12 with DP and MS showed negative correlation as 󰠏0.09 and 󰠏0.27, respectively and these negative genetic correlations implies that bulls that may be superior in carcass traits can be lost at the first step of selection and current selection method should be modified to solve this problem.

  • Leptin, the hormone product of the obese gene, is secreted predominately from white adipose tissue and regulates feed intake, energy metabolism and body composition. It has been considered a candidate gene for performance, carcass and meat quality traits in beef cattle. The objective of this study was to identify SNPs in the promoter region of the leptin gene and to evaluate the possible association of the SNP genotypes with carcass and meat quality traits in Korean cattle. We identified a total of 25 SNPs in the promoter region (1,208-3,049 bp upstream from the transcription start site) of the leptin gene, eleven (g.1508C>G, g.1540G>A, g.1545G>A, g.1551C>T, g.1746T>G, g.1798ins(G), g.1932del(T), g.1933del(T), g.1934del(T), g.1993C>T and g.2033C>T) of which have not been reported previously. Their sequences were deposited in GenBank database with accession number DQ202319. Genotyping of the SNPs located at positions g.2418C>G and g.2423G>A within the promoter region was performed by direct sequencing and PCR-SSCP method to investigate the effects of SNP genotypes on carcass and meat quality traits in Korean cattle. The SNP and SSCP genotypes from the two mutations of the leptin promoter were shown to be associated with the BF trait. The average BF value of animals with heterozygous SNP genotype was significantly greater than that of animals with the homozygous SNP genotypes for the g.2418C>G and g.2423G>A SNPs (p<0.05). Analysis of the combined genotype effect in both SNPs showed that animals with the AC SSCP genotype had higher BF value than animals with BB or AA SSCP genotypes (p<0.05). These results suggest that SNP of the leptin promoter region may be useful markers for selection of economic traits in Korean cattle.

Association Study Between Polymorphisms of Inositol 1,4,5-triphosphate Receptor Type 1 (IP3R1) Gene and Carcass Traits in Korean Cattle (Hanwoo) (한우 Inositol 1,4,5-triphosphate Receptor Type 1 (IP3R1) 유전자의 다형성 및 형질 관련성 분석)

  • Inositol 1,4,5-triphosphate receptor type 1 (IP3R1) is a $Ca^{2+}$ release channel that responds to the second messenger IP3 and that modulates diverse cellular functions such as contraction/excitation, secretion, gene expression and cellular growth. We discovered single nucleotide polymorphisms (SNPs) within IP3R1 gene and analyzed associations between gene polymorphisms and carcass traits in Korean cattle (Hanwoo) in order to develop novel DNA markers at genomic level. Three SNPs were detected at the position of g.1428617A>G, g.1418843C>T and g.1414377C>T with 24 unrelated Hanwoo samples by direct sequencing of the PCR products. We found that genotype of g.1414377C>T SNP was associated with live weight (P<0.05) and carcass weight (P<0.01) using the general linear model of SAS package. These results suggest that polymorphism of IP3R1 gene was associated with weight-related traits in Hanwoo.
