CS1-1의 각종 탄소원에서의 ${\beta}$-글루코시다아제 생성을 살피기 위하여 Quickfit FVIL 발효장치를 이용하고, 뉴트리엔트 이스트브로스, 카르복시멜틸셀룰로오스, 아비셀, 셀로비오스 등을 탄소원으로 한 배지에 뱃치로 배양하여 그 배양적 특성과 세포내 또는 세포외 ${\beta}$-글루코시다아제의 분포를 검토한 결과 : i)${\beta}$-글루코시다아제는 공시한 모든 배양 조건하에서 생성되었고 세포내 효소로서 배양여액에서는 검출되지 않았다. ii) 뉴트리엔트 이스트브로스와 카르복시메틸 셀룰로오스를 탄소원으로 할 때 보다 셀로비오스 및 아비셀을 탄소원으로 하는 경우 효소의 비활성도가 높았다. iii) 공시한 모든 기질에서 공히 균의 대수기(對數期)에 극대치의 비활성도를 나타내었다.
This study was done to investigate the protective effects of Iljungbogan-Tang on acute hepatotoxicity of rats induced by $CCL_4$ and acetaminophen. The subject animals were divided into 3 groups : control group(administrated 0.5% carboxymethyl cellulose), sample group(30, 100, 300, 600mg/kg administrated), positive control group (administrated silymarine), Acute hepatotoxicity of rats were induced by $CCL_4$ and acetaminophen, and the serum transaminase(AST, ALT) were measured for enzyme activities. The inhibitory effects on the serum AST activities were noted in sample group(100, 300, 600mg/kg administrated) on hepatotoxicity of rats induced by $CCL_4$. The inhibitory effects on the serum AST, ALT activities were noted in sample group(30mg/kg administrated) on hepatotoxicity of rats induced by acetaminophen. The inhibitory effects on the serum AST activities were noted in sample group(600mg/kg single dose administrated) on hepatotoxicity of rats induced by acetaminophen. It is considered that Iljungbogan-Tang has protective effects against hepatotoxicity in rats induced by $CCL_4$ and acetaminophen. So it is required to study about the actions of mutual relation of medicines and patho-mechanism through experiment.
The antioxidant activity of resveratrol in cholesterol-fed rats, along with its hypolipidemic effects was determined. Thirty two male Sprague-Dawley (SD) rats were randomly divided into three groups (Control, Res30 and Res70) and fed a hyperlipidemic diet for 4 weeks. Resveratrol was suspended in 0.3% carboxymethyl cellulose (CMC) solution and given to rats of the Res30 and Res70 groups once a day for 4 weeks by oral intubation at a dose of 30 and 70 mg/kg body weight, respectively. The control group received 0.3% CMC solution alone. Resveratrol significantly lowered serum lipid, hepatic cholesterol (TC) and triglyceride (TG) levels compared to the control. Excretion of bile acids was significantly enhanced by resveratrol. The overall potential of the antioxidant system was significantly enhanced by the resveratrol as plasma and hepatic thiobarbituric acid reactive substances (TBARS) levels were lowered while serum superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and catalase (CAT) activities were increased in the cholesterol-fed rats. These findings suggest that resveratrol maintains an antioxidant efficacy as well as its anti-hyperlipidemic effect.
Penicillium verruculosum 배양액으로부터 소단위체 분자량이 60,000(cellobiohydrolase I)과 66,000(cellobiohydrolase II) 및 76,000(cellobiohydrolase III)인 cellobiohydrolase를 분리 정제하여 그들의 일반적 특성을 검토하였다. 이들 세 효소들은 모두 당단백질로서 cellobiohydrolase I, II 그리고 III는 각각 $8.6\%$, $4.2\%$ 그리고 $8.5\%$의 당함량을 나타냈으며 세 효소 모두 pH 4.5 - 5.0, 온도 $50 - 60^{\circ}C$에서 최적조건을 나타냈다. Cellobiohydrolase I, II 및 III는 모두 Avicel, cotton, 여지 등의 결정성 섬유소 뿐만 아니라 carboxymethyl cellulose에도 활성도를 나타냈다. 정제된 cellobiohydrolase I, II 및 III의 Avicel에 대한 비 활성도는 각각 0.07, 0.10, 그리고 0.07 unit per mg. of protein 이었으며 Avicel 분해 생성물은 거의 cellobiose였다. 또한 p-Nitrophenyl-$\beta$-D-cellobioside를 기질로 하였을 때 이들 세 효소 모두 포도당에 의해 활성도가 저해되지 않은 반면 cellobiose에 의해서는 저해되었다. 아미노산조성, 트립신에 의한 펩타이드들의 용출양상 그리고 항체를 이용한 Immunoblotting 결과로부터 cellobiohydrolase II와 III는 동일 유전자산물이거나 1차 구조가 거의 유사한 단백질로 추정된다. Hydrocolloids have many applications in foods including their use in dysphagia diets. We aimed to evaluate whether hydrocolloids in foods affect the digestibility of starch and protein, and their effects on antioxidant capacity. The thickening hydrocolloids: locust bean gum and carboxymethyl cellulose, and the gel-forming agents: agar agar, konjacglucomannan, and Hot & Soft Plus were blended with corn starch and soy protein, skim milk, or grape juice and were examined for their in vitro-digestability by comparing the reducing sugar and trichloroacetic acid (TCA)-soluble peptide, for antioxidant capacity by total polyphenol contents and the 2,2-diphenyl-1-picrylhydrazyl radical scavenging activity. The hydrocolloids resulted in a decrease in starch digestibility with the gel-forming agents. Hydrocolloids diminished TCA-soluble peptides in skim milk compared to soy protein with the exception of locust bean gum and decreased free radical scavenging capacities and total phenolic contents in grape juice. Our findings may provide evidence for the use of hydrocolloids for people at risk of nutritional deficiencies such as dysphagia patients.
Effectiveness of edible coatings containing soy protein isolate (SPI), in reducing oxidative browning and moisture loss during storage ($4^{\circ}C$) of cut apples, potatoes, carrots, and onions was investigated. The SPI coatings were shown to have antioxidative activity. Furthermore, addition of carboxymethyl cellulose (CMC) to the formulations significantly improved its antioxidative activity. Oxidative discoloration, as determined by Commission Internationale De I'Eclairage (CIE) lightness ($L^*$), redness ($a^*$), and yellowness ($b^*$) color scale, was significantly reduced (p <0.05) by SPI coating treatments over a storage time of 120 min. Loss of lightness was reduced by SPI coatings with and without CMC. These respectively showed 4.03 and 3.71% change of $L^*$ value compared to 8.56% for control. Browning of the control in cut potatoes was significantly increased by 106.6% in contrast to 34.3 and 35.2% for SPI coatings with and without CMC, respectively. The $b^*$ values also reflected effectiveness of SPI. Moisture barrier effect was significantly better for the treatments, compared to the control. SPI coatings reduced moisture loss in apples and potatoes, respectively, by 21.3 and 29.6% over the control. Cut onions did not show any treatment effect both in terms of browning and moisture loss. SPI coatings prove to be good moisture barrier and antioxidative property.
The recent outbreak of renal failure in infants in China and in animals in USA and Europe has been determined to be caused by melamine adulterated in the food. In the course of the investigation, cyanuric acid was identified in addition to melamine in the offending food. Fish feeds were also recently found to be contaminated with melamine. The purpose of this study was to characterize the histopathological effect and toxicity potential of different concentrations of melamine and cyanuric acid in the kidney of Japanese catfish (Silurus asotus). The fish were administered melamine and cyanuric acid in combination at the concentrations of 12.5, 25, 50, 100 and 200 mg/kg/day for 3 days by single oral administration dissolved in carboxymethyl cellulose. The results showed that renal crystals were observed in renal tubules and collecting ducts at the concentration over 25 mg/kg dose group and the number of crystals in kidney were in proportion to the concentrations of melamine and cyanuric acid.
Soy protein isolate (SPI)-based edible coating, with and without carboxymethyl cellulose (CMC), were used to reduce oxidative degradation of cut pork stored at $4^{\circ}C$ for 5 days. The SPI coating reduced (p<0.05) thiobarbituric acid-reactive substances (TBARS) and peroxide value (PV), compared with controls. The inhibition of TBARS and PV for SPI-coated porks with and without CMC, compared with the control was 19.1 and 23.9, and 25.7 and 37.7%, respectively. The SPI coating prevented loss of $L^*$ and $a^*$ values of porks compared to the control. The ability of the SPI coating to provide a moisture barrier for the porks was reduced (p<0.05). The SPI-coated porks with and without CMC reduced moisture loss by 37.3 and 44.6%, respectively, over the control. However, SPI coating of porks did not inhibit the growth of either total plate counts or L. monocytogenes. The result revealed that SPI can effectively be used as a natural antioxidative coating to extend quality and shelf life of pork.
The effect of Kundur (oleo-gum resin of Boswellia serrata) and its fractions viz: methanol soluble (MS) and methanol insoluble (MI) were investigated on mercuric chloride induced nephrotoxicity in rats. The animals of group I and II were administered with 1% carboxymethyl cellulose (CMC) (1,000 mg/kg, p.o.) and the animals of groups III, IV and V were administered with Kundur (1,000 mg/kg, p.o.), MS (650 mg/kg, p.o.) and MI (350 mg/kg, p.o.) respectively for ten days. On 10th day a single dose of the mercuric chloride (3 mg/kg, 5.c.) was also administered to all groups except the group I which received only 1% CMC (10 ml/kg, p.o.). After two days of mercuric chloride administration the blood samples of each animal were collected and analyzed for blood urea nitrogen and creatinine concentration. Rats fed with Kundur and MI fraction showed a significant prevention in the rise of serum markers while MS failed to prevent the rise of these serum makers. These results suggest that Kundur and MI fraction may have potential to reduce the nephrotoxicity in rats.
This study was conducted to examine the development of a new anti-inflammatory substance with potent anti-inflammatory activities that was derived from the Platycodi Radix butanol fraction. To accomplish this, the chemical structures and anti-inflammatory activities of the components were elucidated. Upon column chromatography of the tertiary subfraction, fractions 8-4-1 and 8-4-2 were identified as platycodin D and D3, respectively, following recrystallization, based on melting point (MP), infrared (IR), and positive fast atom bombardment (FAB)-mass and nuclear magnetic resonance (NMR) spectral data. Platycodin D and D3 exhibited strong anti-inflammatory activities in rats when administerd at oral doses of 12 mg/kg and 36 mg/kg, p.o., respectively. Platycodin D and D3 induced inhibitory effects on capillary vascular permeability in rats at oral doses of 16 mg/kg and 24 mg/kg, p.o., respectively, and potent inhibition of leukocyte emigration in a carboxymethyl cellulose (CMC)-pouch when administered at doses of 3 mg/rat and 7 mg/rat, s.c., respectively. These results verified the high antiinflammatory potency of the platycodin D and D3 components in Platycodi Radix.
