This study was carried out to get the basic data for the mycelial growth of Lentinus lepideus in liquid culture. The optimal temperature and initial pH of mycelial growth of Lentinus lepideus were $25^{\circ}C$ and pH 5.5, respectively. The optimal medium was YMG medium. Among the carbon sources tested, glucose was effected to the mycelial growth and optimal glucose concentration was 4% (w/v). As nitrogen sources, malt extract and yeast extract appeared to be favorable and optimal malt extract and yeast extract [ratio (w/w) of 1:1] concentration was 1.5% (w/v).
This study was undertaken to examine the feasibility of black locust (Robinia pseudoacacia) as substrates for several edible mushrooms. For the cultivation of several edible and/or medicinal fungi on black locust, optimum bulk densities, synthetic or semisynthetic additives, natural additives and pretreatment methods were investigated. Fruit body yields of the fungi on various sawdust media composed of different wood species were also analyzed for testing the capability of black locust as a substrate for mushroom production. Mycelial growths decreased proportional when the bulk density increased. The most suitable carbon and nitrogen sources as additives to promote the mycelial growth were sucrose (2%, w/w) and ammonium phosphate (0.2%, w/w) respectively. When corn-powder and beer-waste as natural additives were added to sawdust of black locust showed the significant growth of mycelia. And the optimum mixing ratio was 10:2:1 (sawdust: corn-powder: beer-waste, w/w). Black locust after cold water treatment showed the outstanding mycelial growths. Any significant changes of pH, moisture content (%) and dry-weight losses (%) could not be found among culture substrates (sawdust of black locust, oak and poplar wood) examined before and after harvesting of fruit bodies. Yield of fruit bodies on black locust culture media were comparable with those culture media composed with oak and poplar wood. The present work indicated strongly the potentiality of black locust as raw materials for edible and medicinal mushrooms.
Hydrogenated amorphous carbon nitride[a-C:H(N)] films were deposited on p-type Si(100) at room temperature with bias voltage of 200 V by DC saddle-field plasma-enhanced chemical vapor deposition. Effects of the ratio of $N_2$ to $CH_4$($N_2/CH_4$), in the range of 0 and 4 on such properties as optical properties, microstucture, relative fraction of nitrogen and carbon, etc. of the films have been investigated. The thickness of the a-C:H(N) film was abruptly decreased with the addition of nitrogen, but at $N_2/CH_4$>0.5, the thickness of the film gradually decreased with the increase of the $N_2/CH_4$. The ratio of N to C(N/C) of the films was saturated at 0.25 with the increase of $N_2CH_4$. N-H, C≡N bonds of the films increased but C-H bond decreased with the increase of $N_2CH_4$.Optical band gap energy of the film decreased from 2.53 eV at the ratio of $N_2CH_4$=4.
BACKGROUND: Biochar-based fertilizers delay the nutrient release and feature a slow release effect for agricultural and environmental advantages. This experiment was conducted to evaluate agro-environmental effects of different application ratios of modified biochar pellets supplemented. METHODS AND RESULTS: The treatments consisted of the control, 40% N, 60% N and 60% N (0.07M MgO) of modified supplemented biochar pellets (MSBP), which were based on recommended ratio of nitrogen for rice cultivation. For the paddy water, the NH4-N and NO3-N concentrations in whole treatments rapidly increased at 84 days and 40 days after transplanting, respectively. The PO4-P concentrations in the MSBP were generally lower than those of the control. For the paddy soil, NH4-N concentrations in the MSBP were higher than those of the control at 5 days after transplanting, while NO3-N concentrations were not significantly different in the treatments through rice cultivation. P2O5 concentrations in the control were higher than those of the MSBP until 40 days after transplanting while K2O concentrations were not significantly different among the treatment. The highest carbon sequestration was 970 kg ha-1 in the 60% N (0.07M MgO), and the potential carbon storage in the 60% N (0.07M MgO) was higher at 222 kg ha-1 than the control during rice cultivation. It shown that the rice yield in the control was not significantly different from the 40% N and 60% N (0.07M MgO) application plots. CONCLUSION: Application of MSBP for rice cultivation was effective for carbon sequestration and agro-environmental effects even though nitrogen application ratio was reduced at 40% based on recommended application ratio of fertilizer.
Experiments were carried out to select an organic nitrogen source and optimize the culture conditions for the production of arachidonic acid by Mortierella alpina DSA-12. Corn steep powder(CSP) was selected as an organic nitrogen source based on arachidonic acid production and raw material price. The optimum C/N ratio was in the range of 15 to 17 with the medium containing glucose as carbon source and CSP as nitrogen source. The optimum culture conditions for arachidonic acid production showed 500 rpm agitation and 25$^{\circ}C$ culture temperature at 0.5 vvm aeration. Under the optimum conditions, the concentration of cell, total lipid and arachidonic acid were 21.8 g/L, 10.2 g/L and 3.70 g/L, respectively, from 50 g/L glucose and 18 g/L CSP. In the 500 L fermenter with 0.5 vvm aeration and 200 rpm agitation, the concentration of cell, total lipid and arachidonic acid were 19.8 g/L, 9.1 g/L and 3.67 g/L, respectively, from 50 g/L glucose and 18 g/L CSP. This result showed that an arachidonic acid production could be possible with a bench-scale fermenter using corn steep powder as a nitrogen source.

According to the 5th Climate Change Report, global average temperature in 2081~2100 will increase 1.8℃ based on RCP 4.5 and 3.7℃ based on RCP 8.5 from the current climate value (IPCC Working Group I AR5). As temperature is expected to increase due to global warming and the intensity and frequency of rainfall are expected to increase, damage to crops is expected, and countermeasures must be taken. This study intends to evaluate rice growth in terms of nitrogen utilization efficiency according to future climate change conditions. In this experiment, Oryza sativa cv. Shindongjin were planted at the SPAR facility of the NICS in Wanju-gun, Jeollabuk-do on June 10, and were planted and grown according to the standard cultivation method. Cultivation conditions are high temperature, high CO2 (current temperature+4.7℃·CO2 800ppm), high temperature (current temperature+4.7℃·CO2 400ppm), current climate (current tempreture·CO2 400 ppm). Nitrogen was varied as 0, 9, 18 kg/10a. The N content and C/N ratio of all rice leaves, stems, and seeds increased at high temperature, and the N content and C/N ratio decreased under high temperature and high CO2 conditions com pared to high temperature. Compared to the current climate, NUE increases by about 8% under high temperature and high CO2 conditions and by about 2% under high temperature conditions. This seems to be because the increase in temperature and CO2 induced the increase in biomass. ANUE related to yield decreased by about 70% compared to the current climate under high temperature conditions, and decreased by about 45% at high temperature and high CO2, showing a tendency to decrease compared to high temperature. This appears to be due to reduced fertility and poor ripening due to high temperature stress. However, as the nitrogen increased, the number of ears and the number of grains increased, slightly offsetting the production reduction factor.
Basic informations for direct somatic embryo formation in Rehmannja glutinosa Lib. were obtained in 500ml erlenmyer flask. The ratio of ammonium to nitrate nitrogen of 825(mg/l) : 1900(mg/l) was proper condition for somatic embryo formation from stem and petiole explants and 3% sucrose was the most effective carbon source. Full strength MS medium with 2mg/l BA was better than LS medium for somatic embryogenesis. The initial pH 5.7 of medium(full strength MS with 2.0mg/l BA and 0.1mg/l NAA) was good for embryo production. Potassium ion was taken up rapidly within 2 weeks. while $Ca^{++}$ and $Mg^{++}$ ion contents were almost constant during culture period. Sucrose hydrolysis occurred throughout the culture, while glucose and fructose were absorbed simultaneously from the third week of culture.
Mycelial expansion and fruit body development of caterpillar fungi, Cordyceps sobolifera(Hill.) Berk. et Br. was examined on various carbon and nitrogen sources, some substrates with various supplements. Best temperature for the mycelial expansion was $25^{\circ}C$, while optimal pH was 7. Glucose and polypeptone were the best sources of carbon and nitrogen, respectively, as well as 1:1 was the best ratio of carbon and nitrogen. The unpolish rice and bean curd lees were used as core substrates combined individually with five various supplements. In substrates based, all the supplements with all combinations were found to exhibit the mycelial growth and fruit body formation. Likewise, yolk was found as a superior supplement and also clarified quite high polysaccharide content in the fruit body.
A new precess which consists of pre-dewatering device, post composting for solid phase and post sequencing batch reactor(SBR) for liquid phase was designed. Nitrogen in supernatants of dewatering device was removed by sequencing batch reactor. Experiments were carried out to investigate the SBR operation modes such as fill ratio, SRT, and operation cycle. The optimum fill ratio, SRT and aeration/non-aeration time were 1/12, 15days, and 2hr aeration / 1hr non aeraion, respectively. Methanol as an external carbon source increased denitrification when step feeding method was applied, not single feeding method.
This study was conducted to establish a database of high solid manure(HSM) in domestic. Theoretical methane potential and HSM characteristics was evaluated using breef and dairy manure (n=156). Total solids and Volatile solids of HSM increased depending on time flow, the results showed $20.4{\pm}3.2$ and $17.4{\pm}2.8%$. respectively. C/N ratio of breef HSM was higher than dairy HSM C/N ratio. In theoretical methane potential, the result of breef and dairy HSM was showed $505.2{\pm}25.3$ and $493.5{\pm}20.2$, respectively. Nitrogen content of total HSM increased depending on time flow, the result of breef and dairy nitrogen content was showed $1.9{\pm}0.3$ and $2.8{\pm}0.2$, respectively. Carbon content of total HSM showed approximately 10% reduction. The optimal time of bed replacement was indicated between 29 amd 31 days based on the optimal C/N ratio. Therefore, this study was considered that it has high utilization for livestock manure recycling and basis of relevant research.
