A Numerical Simulation of Hazardous Waste Destruction in a Dump Incinerator (덤프 소각기에서 유해폐기물 분해에 대한 수치해석)

  • A major source of the hazardous waste generated is from chemical industries producing plastics, herbicides, pesticides and chlorinated solvents. All of these processes produce a class if hazardous waste termed the chlorinated hydrocarbons(CHCs), either directly or from undesirable side reactions. In this study, we investigated the destruction characteristics of hazardous waste through incineration. A nonequilibrium combustion model was used to describe the effect of the chemical kinetics due to the flame inhibition characteristics of $CCl_4$ which was used as the surrogate of hazardous waste. A parametric screening studies was made in a dump incinerator proposed in this study. The dump incinerator showed high $CCl_4$ DRE(Destruction and Removal Efficiency) as 5 nines. $CCl_4$/CH$_4$ ratio appeared to be most important in the destruction of $CCl_4$ through incineration.

Protective Effect of Sachungwhan against CCl4-induced Hepatotoxicity

  • Sachungwhan reduced hepatotoxicity induced by carbon tetrachloride($CCl_4$). Improved liver function was observed by measuring the activities of aspartate aminotransferase(AST), alanine aminotransferase(ALT), alkaline phosphatase(ALP), blood urea nitrogen(BUN), creatinine(CRE), total cholesterol(TCHO), triglyceride(TG), low density lipoprotein cholesterol(LDL-CHO), high density lipoprotein cholesterol(HDL-CHO), total protein(TP), albumin(ALB) and total bilirubin(BIL) in serum. Hepatic parameters monitored were levels of cholesterol(CHO), triglyceride(TG), malondialdehyde(MDA), content of cytochrome P450(CYP), level of glutathione(GSH), and activities of NADPH-CYP reductase, superoxide dismutase(SOD), catalase(CAT), glutathione S-transferase(GST), glutathione reductase(GR), glutathione peroxidase(GPx). The histopathological examination showed that the treatment of Sachungwhan relieved the ballooning degeneration of hepatocytes which had been generated by $CCl_4$. The results suggested that hepatoprotective effects of Sachungwhan possibly are due to their promising antioxidative activity.

Mechanism and Effect of Corydalis ternata on the $CCl_4$-Induced$ Hepatotoxicity (사염화탄소에 의한 간손상에 미치는 현호색의 효과 및 그 기전)

  • Protective effect of Corydalis ternata against the carbon tetrachloride-induced toxicity was investigated. Carbon tetrachloride($CCl_4$) induces hepatotoxicity due to the reactive free radical(CCl$_3$ . ) generated by cytochrome P-450 enzyme. We examined effects of hexane, chloroform, butanol and water fractions prepared from the Corydalis ternata methanol extract. Rats were treated with those for 3 days, and liver microsomes and cytosols were prepared at 24 hour after last treatment. Hepatoprotective activity of the water fraction was higher than that of other fractions. To examine mechanism of the hepatoprotective effect of Corydalis ternuta, we measured contents of malondialdehyde(MDA), cytochrome P46O(CYP), glutathione, calcium as well as the activities of NADPH-CYP reductase, glutathione S-transferase(GST), superoxide dismutase(SOD), glutathione peroxidase(GPX) and catalase. The fraction inhibited production of MDA, content of CYP and calcium in liver of water fractions - treated rats as compared with those of CCl4-treated rats. The GST activity was increased. We speculate that the O2 radical scavenging activities of the water fraction might play a key role in the mechanism opposing the progression of $CCl_4$-induced hepatotoxicity, but the activities of SOD, GPX, CAT were decreased. These results suggest that the mechanism might be mainly due to the decrease of CYP contents, act as calcium channel blocker and increase of GST activity rather than $O_2$ radical scavenging activities.

  • This study was devised to investigate the effects of flavonoids on carbon tetrachloride toxicity of rats. Through the experiments, the contents of total cholesterol, transaminase(GOT, GPT) and alkaline phosphatase(Alk. P) activities in the liver and serum were determined. The results obtained are summarized as follows; (1) The liver cholesterol concentration of flavonoid injected groups were remarkably increased up to 45% at 2 days after $CCl_4$ injection and then the values were almost recovered to the level of control group at 4 days after $CCl_4$ injection. The increase of serum cholesterol content was inhibited by the flavonoids after $CCl_4$ injection, but the result was not significant. (2) Flavonoids injection was found to have the ability to decrease the elevated serum GOT, Alk. P activities resulting from injection of $CCl_4$ and induce rapid recovery from such an elevated level and the extent of such a decreasing action was greatest in hesperidin injection group. The liver microsomal Alk. P and GOT activities were not affected by the treatment of flavonoids. (3) The elevated serum and liver microsomal GPT activities induced by $CCl_4$ injection were inhibited by the injection of flavonoids.

  • Carbon tetrachloride (CCl4) is known to cause liver injury characterized by centrilobular necrosis. The toxicity is thought to be exerted via cytochrome P-450-mediated metabolism of $CCl_4$ to trichloromethyl and trichloro-methylperoxy radicals, which initiates lipid peroxidation, leading to hepatocellular membrane damage [l, 2]. Our study demonstrates a therapeutic efficacy of YCY, extract of a cricket, Gryllus bimaculatus, on liver injuries induced by $CCl_4$. (omitted)

  • The effect of hepatic injury produced by CCL, was studied on rats receiving a low protein-high carbohydrate (7% casein), standard protein (20% casein) and a high protein diet (30% casein). The rats fed low protein diet are resistant to CCl$_4$ in its effects on the liver as judged by histology, serum enzymes(guanase, ALT) and the content of hepatic protein. On the other hand, the pretreatment of hydrocortisone before injection of CCl$_4$ to the rats fed a standard diet, slightly decreased both serum ALT and guanase activities. In the pretreatment of actinomycin D, the liver and serum guanase activities were significantly decreased. It indicates that the cause of increasing serum guanase is based on the alteration of membrane permeability and the result of accelerated enzyme synthesis in liver cells of CCl$_4$ intoxicated rats.

Isorhamnetin-3-O-galactoside Protects against CCl4-Induced Hepatic Injury in Mice

  • This study was performed to examine the hepatoprotective effect of isorhamnetin-3-O-galactoside, a flavonoid glycoside isolated from Artemisia capillaris Thunberg (Compositae), against carbon tetrachloride ($CCl_4$)-induced hepatic injury. Mice were treated intraperitoneally with vehicle or isorhamnetin-3-O-galactoside (50, 100, and 200 mg/kg) 30 min before and 2 h after $CCl_4$ (20 ${\mu}l/kg$) injection. Serum aminotransferase activities and hepatic level of malondialdehyde were significantly higher after $CCl_4$ treatment, and these increases were attenuated by isorhamnetin-3-O-galactoside. $CCl_4$ markedly increased serum tumor necrosis factor-${\alpha}$ level, which was reduced by isorhamnetin-3-O-galactoside. The levels of inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2), and heme oxygenase-1 (HO-1) protein and their mRNA expression levels were significantly increased after $CCl_4$ injection. The levels of HO-1 protein and mRNA expression levels were augmented by isorhamnetin-3-O-galactoside, while isorhamnetin-3-O-galactoside attenuated the increases in iNOS and COX-2 protein and mRNA expression levels. $CCl_4$ increased the level of phosphorylated c-Jun N-terminal kinase, extracellular signal-regulated kinase and p38, and isorhamnetin-3-O-galactoside reduced these increases. The nuclear translocation of nuclear factor kappa B (NF-${\kappa}B$), activating protein-1, and nuclear factor erythroid 2-related factor 2 (Nrf2) were significantly increased after $CCl_4$ administration. Isorhamnetin-3-O-galactoside attenuated the increases of NF-${\kappa}B$ and c-Jun nuclear translocation, while it augmented the nuclear level of Nrf2. These results suggest that isorhamnetin-3-O-galactoside ameliorates $CCl_4$-induced hepatic damage by enhancing the anti-oxidative defense system and reducing the inflammatory signaling pathways.

The Effects of Puerariae Radix Catechins Administration on Liver Function in Carbon Tetrachloride-Treated Rats (사염화탄소를 투여한 흰쥐에 있어서의 간 기능에 미치는 칡 카테킨의 효과)

  • These studies were carried out to investigate the effects of Puerariae radix catechins(PRC) administration on the biochemical parameters of liver function in liver of carbon tetrachloride(CCl4)-treated rats. Thirty six healthy Sprague-Dawley rats weighing about 120g were used for this experiment and divided intot he following 3 groups : normal control group(NCON), $CCl_4$ control group(CCON), PRC treated group(PRC). Fifty percent $CCl_4$ in oil was administered(I.P.) by 2ml per kg body weight two times a week for 3 weeks. PRC treated groups were administered orally at the leaves of 1% per day in distilled water for 8 weeks. Lipid hydroperoxides were analyzed by using chemiluminescence-high performance liquid chromatography(CL-HPLC) method as a phosphatidylcholine hydroperoxide value(PCOOH) in liver tissues. $CCl_4$ treatment significantly(p<0.05) resulted in an increase in GPT & GOT activities and liver hydroperoxide values comparing with those of the untreated control, while administration of PRC to the $CCl_4-treated$ rats significantly(p<0.001) decreased GPT & GOT activities and liver hydroperoxide value. Their ultrastructual changes of hepatocellular organelles were shown to clarify the morphologic nature of protective effects of PRC on hepatocytic injuries. $CCl_4$ treatment observed to change the ultrastructual nature of outer membrane of hepatocytes. However, the hepatic changes on PRC treatment to $CCl_4$ group was not found. PRC administration may inhibit the formatiion of liver lipid hydroperoxides in vivo and were very effective in recovering the liver function in $CCl_4-treated$ rats.

  • In order to test relationships between hepatotoxicity and transmethylation, activities of protein methylases and SAM (S-adenosyl-L-methionine)-synthetase were examined in liver tissues of rats treated with $CCl_4$. Also the concentrations of SAM and SAH were measured by HPLC in rat liver. The results are as follows. (1). Activities of protein methylases were not significantly changed in 24 hours after $CCl_4$ treatment. However, in 48 hours, activities of protein methylases were significantly increased in comparison with that of control. (2). Activity of SAM-synthetase was increased steadily in the time course after $CCl_4$ treatment. (3). S-adenosyl-L-methionine concentration of liver tissues in $CCl_4$-treated group was elevated in 24 hours, and then declined thereafter. But the SAH concentration was slightly decreased in the time course after $CCl_4$ treatment. These results indicate that SAM was very actively used in transmethylation reactions of $CCl_4$ damaged rat liver, suggesting the strong relationships between hepatotoxicity and transmethylation phenomena.

Protective Effects of Monosodium-L-Glutamate on the Fatty Liver induced by Carbon Tetrachloride in Rat (사염화탄소-유발지방간에 대한 L-글루탐산 일나트륨의 보호작용)

  • To achieve better understanding of the effects of monosodium-L-glutamate(MSG) against $CCl_4$ fatty liver in Wister male rats, 5% MSG solution was given as drinking water and $CCl_4$ 0.1 ml/kg was injected subcutaneously twice a week for four weeks. It was showed that increased hepatic phospholipid and hepatic triacylglycerol levels by $CCl_4$ challenge were significantly decreased by additionnal MSG, respectively. However, MSG had no apparent effect on the elevated hepatic cholesterol level in the presence of $CCl_4$. Histologically, additional MSG markedly inhibited fatty degeneration, spotty necrosis, inflammation and periportal vascular proliferation manifested by $CCl_4$. respectively. These results indicated that effects of MSG against $CCl_4$ induced-fatty liver appeared to be involved with partial restoration of altered hepatic lipid composition.
