The production of carotenoids, lipid content, and fatty acid composition were all studied in a strain of Sporobolomyces ruberrimus when using different concentrations of technical glycerol as the carbon source and ammonium sulfate as the nitrogen source. The total lipids represented an average of 13% of the dry weight, and the maximum lipids were obtained when using 65.5 g/l technical glycerol (133.63 mg/g). The optimal conditions for fatty acid production were at $27^{\circ}C$ using 20 g of ammonium sulfate and a pH range from 6 to 7, which produced a fatty acid yield of $32.5{\pm}1\;mg/g$, including $1.27{\pm}0.15\;mg$ of linolenic acid (LNA), $7.50{\pm}0.45\;mg$ of linoleic acid (LLA), $5.50{\pm}0.35\;mg$ of palmitic acid (PA), $0.60{\pm}0.03\;mg$ of palmitoleic acid (PAL), $1.28{\pm}0.11\;mg$ of stearic acid (SA), $9.09{\pm}0.22\;mg$ of oleic acid, $2.50{\pm}0.10\;mg$ of erucic acid (EA), and $4.25{\pm}0.20\;mg$ of lignoceric acid (LCA), where the palmitic, oleic, and linoleic acids combined formed about 37% of the total fatty acids. The concentration of total carotenoids was 2.80 mg/g when using 20 g of ammonium sulfate, and consisted of torularhodin (2.70 mg/g) and $\beta$-carotene (0.10 mg/g), at $23^{\circ}C$ and pH 6. However, the highest amount with the maximum specific growth rate was obtained (${\mu}_{max}=0.096\;h^{-1}$) with an ammonium sulfate concentration of 30 g/l.
A fungal strain, capable of solubilizing insoluble phosphate under diverse temperature, pH and salt conditions was isolated from Waste Mushroom bed of Agaricus bisporus in South Korea. Based on 18S rRNA analysis, the strain was identified as Aspergillus awamori bxq33110. The strain showed maximum phosphate solubilization in AYG medium (525 ${\mu}g\;mL^{-1}$) followed by NBRIP medium (515 ${\mu}g\;mL^{-1}$). The strain solubilized $Ca_3(PO_4)_2$ to a greater extent and rock phosphate and $FePO_4$ to a certain extent. However $AlPO_4$ solubilizing ability of the strain was found to be very low. Glucose at the rate of 2% ($561{\mu}g\;mL^{-1}$) was found be the best carbon source for Aspergillus awamori bxq33110 to solubilize maximum amount of phosphate. However, no significant difference ($P{\leq}0.05$) in phosphorus solubilization was found between 1% and 2% glucose concentrations. $(NH_4)_2SO_4$ was the best nitrogen source for Aspergillus awamori bxq33110 followed by $NH_4Cl$ and $NH_4NO_3$. At pH 7, temperature $30^{\circ}C$ and 5% salt concentration (674 ${\mu}g\;mL^{-1}$) were found to be the optimal conditions for insoluble phosphate solubilization. However, strain Aspergillus awamori bxq33110 was shown to have the ability to solublize phosphate under different stress conditions at $30-40^{\circ}C$ temperature, pH 7-10 and 0-10% salt concentrations indicating it's potential to be used as bio-inoculants in different environmental conditions.
A bacterium producing the extracellular xylanase and CMCase was isolated from soil and has been identified as Bacillus sp. The isolate, named Bacillus sp. A-7, was shown to be very similar to Bacillus licheniformis on the basis of its biochemical and physiological properties. The maximum xylanase and CMCase production were obtained when 2.0% (w/v) glucose and 0.3% (w/v) yeast extract were used as carbon source and nitrogen source, respectively. The best mineral conditions for xylanase and CMCase production were 0.1%(w/v) $CaC1_2$. Among the various feedstuffs, 1.0%(w/v) soybean meal was selected for the best xylanase and CMCase production.
Fungi which produce red pigment were isolated from tapioca chips, Korean Koji, air, and plant leaves. Among the Fungi, T-1 strain was selected as test strain. This strain was identified as a Monatcus sp. by the morphological propeties. Various culture conditions, and physical and physiological characteristics of red pigment were studied. According to the studies of culture conditions, optimum condition was found to be pH 6.5; 4 days of incubation; temperature, 32~33 c: 3.5% of Tapioca chips powder as carbon source, 0.2% of sodium nitrate as nitrogen source and 100 ml of medium in the 500 ml Erlenmyer flask at a rotary shaker (rpm180) as aeration condition. Also effective levels of vitamins, amino acids and inorganic compounds was found to be l$\mu\textrm{g}$/ml of folic acid and niacin; 0.3% of L-arginine, L-glutamic acid and L-proline; and 0.001% of manganese dioxide giving good results.
The bacterial strains, which utilizes 2,4,4'-trichloro-2'-hydroxydiphenyl ether(TCHDPE) as a sole carbon source, were isolated by selective enrichment culture from soil samples of industrial waste deposits. The bacterium that showed the highestt biodegradation activity was designated as EL-O47R The isolated strain EL-O47R was Identified as the genus Pseudomonas from the results of morphological, cultural, and biochemical tests. The optimum conditions of medium for the growth and the degradation of TCHDPE were TCHDPE 500 ppm, (NH4)2SO4 0.1% as the nitrogen source, initial pH 7.0±0.1, and 37℃, respectively. In this conditions, the regradation rate of TCHDPE was about 97%. Pseudomonas sp. EL-O47R was tested for resistance to several metal compounds and antibiotics. Pseudomonas sp. EL-O47R was moderately grown to Cd(NO3)2, ZnCl2, AgSO4, CuSO4 and HgCl2. This strain was sensitive to rifampicin and kanamycln but resistant to ampicillin, penicillin, tetracyclin and chloramphenlcol. Pseudomonas sp. EL-O47R was grown structurally related com- pounds and potential metabolites of TCHDPE, and has the stability on TCHDPE biodegradation.
In this study, the cultural medium used for the efficient production of $\gamma$-PGA with a newly isolated Bacillus sp. RKY3 was optimized. It was necessary to supplement the culture medium with L-glutamic acid and an additional carbon source in order to induce the effective production of $\gamma$-PGA. The amount of $\gamma$-PGA increased with the addition of L-glutamic acid to the medium. The addition of 90 g/L L-glutamic acid to the medium resulted in the maximal yield of $\gamma$-PGA (83.2 g/L). The optimum nitrogen source was determined to be peptone, but corn steep liquor, a cheap nutrient, was also found to be effective for $\gamma$-PGA production. Both the $\gamma$-PGA production and cell growth increased rapidly with the addition of small amounts of $K_2HPO_4$ and $MgSO_4\cdot7H_{2}O$. Bacillus sp. RKY3 appears to require $Mg^{2+}$, rather than $Mn^{2+}$, for $\gamma$-PGA production, which is distinct from the production protocols associated with other, previously reported bacteria. Bacillus sp. RKY3 may also have contributed some minor $\gamma$-PGA depolymerase activity, resulting in the reduction of the molecular weight of the produced $\gamma$-PGA at the end of fermentation.
To produce polyhydroxyalkanoate (PHA) from inexpensive substrates by bacteria, vegetable-oil-degrading bacteria were isolated from a rice field using enrichment cultivation. The isolated Pseudomonas sp. strain DR2 showed clear orange or red spots of accumulated PHA granules when grown on phosphate and nitrogen limited medium containing vegetable oil as the sole carbon source and stained with Nile blue A. Up to 37.34% (w/w) of intracellular PHA was produced from corn oil, which consisted of three major 3-hydroxyalkanoates; octanoic (C8:0, 37.75% of the total 3-hydroxyalkanoate content of PHA), decanoic (C10:0, 36.74%), and dodecanoic (C12:0, 11.36%). Pseudomonas sp. strain DR2 accumulated up to 23.52% (w/w) of $PHA_{MCL}$ from waste vegetable oil. The proportion of 3-hydroxyalkanoate of the waste vegetable-oil-derived PHA [hexanoic (5.86%), octanoic (45.67%), decanoic (34.88%), tetradecanoic (8.35%), and hexadecanoic (5.24%)] showed a composition ratio different from that of the corn-oil-derived PHA. Strain DR2 used three major fatty acids in the same ratio, and linoleic acid was the major source of PHA production. Interestingly, the production of PHA in Pseudomonas sp. strain DR2 could not occur in either acetate- or butyrate-amended media. Pseudomonas sp. strain DR2 accumulated a greater amount of PHA than other well-studied strains (Chromobacterium violaceum and Ralstonia eutropha H16) when grown on vegetable oil. The data showed that Pseudomonas sp. strain DR2 was capable of producing PHA from waste vegetable oil.
About 100 bacterial strains producing proteolytic enzymes were isolated from Korean traditional soy paste Doenjang. Among them, strain SYG3 producing the highest level of angiotensin converting enzyme (ACE) inhibitor into the culture medium was selected and identified as Bacillus pumilus according to the Bergey's mannual of systematic bacteriology. Soybean powder as a nitrogen source and glucose as a carbon source supported high level of ACE inhibitor production. The presence of 3% NaCl also enhanced the production of ACE inhibitor in the medium. The optimum initial pH of the medium and culture temperature for the production of ACE inhibitor were 7.0 and $32^{\circ}C$, respectively. The maximal level of ACE inhibitory effect was obtained after 36 hours of cultivation under the optimized conditions, which was about 98% of inhibition ratio.
A yeast strain producing fruity-floral aroma was isolated from soil and identified as Hansenula saturnus var. saturnus. Glucose was found to be the best carbon source and sodium nitrate or phenylalanine as nitrogen source in terms of the nature and the intensity of the aroma produced by the isolated yeast. Seventeen compounds, mainly esters and alcohols, were identified in the ether-pentane extract of the culture broth by gas chromatography and/or coupled gas chromatography-mass spectrometry. Ethyl alcohol, isobutyl alcohol, isoamyl alcohol, phenethyl alcohol and their acetate esters together with ethyl caprylate were the major compounds in the aroma concentrate. Three unusual compounds, dibutyl disulfide, 3-methyl pentanoic acid and methyl pentanoate were also tentatively identified in the culture broth of the isolated yeast.
Three marine bacteria producing pigments were isolated from seawater of Jeju-Do and local solar saltern in Korea. Based on phenotypic characteristics and 16S rRNA sequence analysis, the strains were identified as Pseudoalteromonas spp., which produced red (Ju11-1), yellow (Ju14), and orange (TA20) pigments. The pigments showed UV absorption maxima at 537, 378 and 387 nm, respectively. The strains were growing well on Marine broth 2216 culture medium. The productivity of pigments reached the maximum value after 28 hours (Ju11-1, Ju14) and 24 hours (TA20) at $30^{\circ}C$, 2% NaCl and pH 6-7. The best pigment production of strains were supported by 1% of lactose (Ju11-1) and maltose (Ju14, TA20) as a carbon source and 1% of beef extract as a nitrogen source.
