A STUDY ON THE MYCELIA CUL TURE AND GENETIC CHARACTERISTICS IN Lepista nuda (민자주방망이버섯(Lepista nuda)의 균사배양 및 유전적 특성에 관한 연구)

  • This study was carried out to investigate the cultural characteristics and the genetic characteristics in Lepists nuda. The mycelia cultural characteristics which include the specificity for pH, carbon source, nitrogen source and medium were studied using petridish culture and liquid media culture respectively. The genetic characteristics were also investigated using random amplified polymorphic DNA (RAPD) analysis. To find out the mycelial growth rate by various medium, mycelia was cultured for 25 days in PDA, YM, MEA, GPB and Yamanaka. The Yamanaka was superior to the other media in supporting the mycelial growth. pH6 produced the best result in the test of an optimal pH. In experiment for optimal carbon source, starch showa 46.8$\pm$1.7mm of diameter of mycelial colony as the best and for nitrogen source, yeast extract showed a good effect as well. In investigation for genetic characteristics, Lepists nuda was amplified by 7 primers among 10 primers and Pleurotus ostreatus was also amplified by 7 primers. From the RAPD analysis between species, the band patterns were different.

Study of Factors Influenced on denitrification in wastewater treatment (폐수처리 탈질 공정에 미치는 인자 연구)

  • In this study, the effects of several factors such as initial nitrate concentration, C/N ratio, biomass amount and external carbon source on denitrification process were investigated using synthetic wastewater and sludge obtained from wastewater treatment facility. As a result, the condition of lower initial nitrate concentration was increased to the removal rate of nitrate than that of high concentration. The increases of C/N ratio and initial biomass amount were linearly enhanced the removal rate. The use of ethanol as external carbon source was shown the highest removal yield than that of others.

A Study on the Removal of Nitrogen and Phosphorus by Operation Mode for Livestock Wastewater Treatment Post-process Using SBR (축산폐수의 후처리공정으로서 SBR 적용시 운전인자에 따른 질소와 인의 제거특성에 관한 연구)

  • This study examined the removal efficiency of the nitrogen and phosphorus in order to compensate for the combined process of ATAD(Autothermal Thermophilic Aaerobic Digestion) and EGSB(Expended Granular Sludge Bed), which are treatment methods for livestock wastewater, by introducing SBR(Sequencing Batch Reactor) as post-treatment process. The analysis on the treatment efficiency of each operation mode showed that, in the case of T-N, the treatment efficiency were 67.1% and 74.2% for Run-1 and Run-2, respectively, and in the case of T-P, the values were 71.2 and 74.1, respectively, which are indicating that the treatment efficacy is higher in the condition of Run-1, in which the time period of Anoxic and Aerobic segments were increased. In addition, the result of analyzing removal characteristics of nitrogen and phosphorus by Influx load showed that removal efficiency of nitrogen was better in the case of high influx load than in the case of low influx load. Regardless of Influx load, phosphorus showed constant influx concentration, so that removal efficiency of phosphorus was influenced littler by Influx load than that of nitrogen. This study also fed methanol as an external carbon source and performed experiment to induce denitrification under anoxic condition by using nitrate among nitrogen compounds of SBR reactor, and the results showed that intermittent feeding was more effective in Nitrogen Removal than composite feeding.

Discarded Egg Yolk as an Alternate Source of Poly(3-Hydroxybutyrate-co-3-Hydroxyhexanoate)

  • Many poultry eggs are discarded worldwide because of infection (i.e., avian flu) or presence of high levels of pesticides. The possibility of adopting egg yolk as a source material to produce polyhydroxyalkanoate (PHA) biopolymer was examined in this study. Cupriavidus necator Re2133/pCB81 was used for the production of poly(3-hydroxybutyrate-co-3-hydroxyhexanoate) or poly(3HHx), a polymer that would normally require long-chain fatty acids as carbon feedstocks for the incorporation of 3HHx monomers. The optimal medium contained 5% egg yolk oil and ammonium nitrate as a nitrogen source, with a carbon/nitrogen (C/N) ratio of 20. Time course monitoring using the optimized medium was conducted for 5 days. Biomass production was 13.1 g/l, with 43.7% co-polymer content. Comparison with other studies using plant oils and the current study using egg yolk oil revealed similar polymer yields. Thus, discarded egg yolks could be a potential source of PHA.

Effect of Medium Components on the Production of Lovastatin by Aspergillus terreus (Aspergillus terreus에 의한 Lovastatin 생산에 배지성분이 미치는 영향)

  • The biosynthesis of Lovastatin, a cholesterol lowering agent formed by the filamentous fungus Aspergillus terreus, was investigated in shaking flask. The effects of essential elements in the experimental medium such as carbon, nitrogen, phosphate sources, and amino acids were examined to increase Lovastatin productivity. Lovastatin production in shaking flasks was 68 mg/L in the used medium. Effect of carbon source on Lovastatin production was performed. As a carbon source in the medium, 45 mL/L of glycerol increased the Lovastatin production up to 256 mg/L, which was found to be improved almost 3.5 times in comparison with that in common medium. The optimum concventration of peptonized milk as nitrogen source was obtained 30g/L on Lovastatin production. The severe inhibition of the cell growth and the Lovastatin production were observed in shaking flasks conducted at the medium contained ammonium carbonate as a nitrogen source. Lovastatin production various concentrations of several phosphate compounds was also examined. The addition of either potassium phosphate diabsic or sodium phosphate dibasic increased the Lovastatin production and the optimal level of potassium phosphate dibasic was 6 g/L. Even though Lovastatin contain methionine-derived methyl group, L-methionine and DL-methionine tend to diminish the Lovastatin production. Among the amino acids, L-histidine and L-tryptophan had a remarkable enhancing effect on the Lovastatin production. The optimal concentration of L-histidine and L-tryptophan was 6g/L.

  • As a basic study to elucidate nutritional physiology and composition of synthetic medium of red rotting bacteria, Erwinia carotovora, of ginseng, the effects of hydrogen ion concentration, various kinds of carbon sources, nitrogen source, micrometallic salts and it's concentration on the gorwth of the bacteria were investigated and the results were as follows. Optimal pH in the basal medium for the growth of the bacteria was 6.5. After incubation the pH in culture media was neutralized. Among the various kinds of carbon sources, sucrose, glucose mannitol, but organic acids were not utilized effectively as nutrients. After incubation the pH turned acidic. Alanine as organic nitrogen sources nad ammonium sulfate as inorganic nitrogen promoted the growth, but L-valine and sodium nitrite were the least effective. Ferric chloride 1.0mg/dl and ferrous sulfate 100mg/dl were the most effective as micrometallic sources. Control and boric acid were the least effective. New synthetic medium based on the above results was follows ; Alanine 1.0g, $KH_2PO_4\;1.0g, \;sucrose\;30.0g, \;MgSo_4{\cdot}7H_2O\;0.5g, \;FeCl_3{\cdot}6H_2O\;1.0mg\;thiamine\;200{\gamma}g$, and distilled water 1000ml, pH6.5.

Influence of Physiological and Environmental Factors on Growth and Sporulation of an Antagonistic Strain of Trichoderma viride RSR 7

  • Influence of physiological and environmental factors on an antagonistic strain of Trichoderma viride RSR7 were studied optimize its biocontrol potential. The growth and sporulation of T. viride was greatly influenced by various carbon and nitrogen sources, and the environmental factors such as pH and temperature. The best growth and sporulation of T. viride was observed when sucrose, peptone and trehalose were supplemented in the medium as sole carbon sources. Rhamnose, pyruvic acid and sorbitol also supported a good growth. However, with these carbon sources the sporulation was poor. Growth and sporulation was also affected by various nitrogen sources. Growth and sporulation both were favoured by ammonium forms of nitrogen compared to nitrite or nitrate forms. Urea did not support either growth or sporulation. Among amino acids, glutamic acid, asparagine, leucine, aspartic acid, glutamic acid and alanine supported good growth as well as sporulation. T. viride was able to utilize large number of amino acids as sole nitrogen source. Proline was good for growth, but not for sporulation. Maximum growth and sporulation of T. viride was between pH 4.5 to 5.5. Temperatures between $20^{\circ}C\;and\;37^{\circ}C$ were good for both growth and sporulation of T. viride. At lower temperatures(i.e. below $20^{\circ}C$) growth and sporulation were inhibited. Based on the present study it may be concluded that T. viride RSR7 is capable of growing and sporulating with varied nutritional and environmental conditions and, therefore, this strain of T. viride may be useful as a biocontrol agent under diverse physiological and environmental conditions.

  • This study was conducted to analyze the operating conditions of predenitrification process to improve the treatment efficiency in low organic loading sewage plant in use today, and to investigate the treatment efficiency of pilot plant added night soil as well as the nitrogen removal characteristics of pilot plant added carbon sources. In the operation under the condition of $BOD_{5}$ sludge load 0.03-0.28kg $BOD_{5}$/kg VSS/d and oxic ammoniac nitrogen sludge load 0.02-0.24 $kgNH_{4}^{+}$-N/kg MLVSS/d, nitrification efficiency is higher than 95%. In order to achieve 70% nitrogen removal at the T-N sludge loading 0.06kg T-N/kg VSSㆍd and the SRT 6~11 days, optimum operating factors were revealed to $CODc_{r}$/T-N ratio 9, recycle ratio 2.6, and denitrification volume ratio 0.33. At this time, denitrification capacity was approximately 0.09 kg $NO_{3}^{-}$-N/kg $CODc_{r}$; specific nitrification rate was 3.4mg $NH_{4}^{+}$-N/g MLVSS/hr; and specific denitrification rate was 4.8mg $NO_{3}^{-}$-N/g MLVSS/hr.

  • Production and properties of invertase from Aspergillus niger were investigated. Inulin and sucrose were best carbon source and yeast extract was most suitable for the production of the enzyme among tested carbon and nitrogen sources. The enzyme among tested carbon and nitrogen sources. The enzyme was maximally produced by cultivating the organism at medium of pH 4.5 and temperature of 3$0^{\circ}C$ The optimum pH and temperature for the enzyme activity were pH 5.0 and temperature of 5$0^{\circ}C$, respectively. Among tested metal ions. Hg++, Cu++ and Ag+ ions Inhibited the enzyme activity drastically.
