Bioactive Functions of Detoxified Puffer Liver Oil (제독처리한 복어 간유의 생리활성)

  • In this study, we investigated the effects of detoxified puffer liver (PL) oil on fatigue, hepatotoxicity and hyperlipidemia. There are no toxicities in both raw and purified PL oil. The test of swimming time was extended in detoxified PL oil pretreated group compared to the non-treated group. When rats treated with PL oil, the hepatic injuries induced by carbon tetrachloride or DL-galactosamine were reduced. The increased serum triglyceride and total cholesterol by poloxamer-407 were lowered by treating with PL oil remarkably. Also the bleeding time of hyperlipidemic animals was extended and plasma clotting time was delayed by PL oil.

Pharmaceutical Study on Pranoprofen Argininate and Pranoprofen Lysinate (Pranoprofen Argininate 및 Pranoprofen Lysinate의 약제학적 연구)

  • Novel pranoprofen algininate and lysinate salts were manufactured and their salt formation was confirmed by melting point, infrared spectroscopy, nuclear magnetic resornance spectroscopy, differential scanning calorimetry and powder X-ray diffractometry. The physical properties of pranoprofen lysinate and argininate salts were compared with those of pranoprofen through in vitro and in vivo tests. Solubility, $pK_a$ and lipid-water partition coefficient were measured through in vitro experiments, while antiinflammatory efficacy, analgesic effect, acute toxicity and in situ absorption were tested through in vivo experiments. The results obtained were as follows: 1) The solubilities of pranoprofen argininate and lysinate salts were increased markedly in pH 6.8 and pH 7.5 phosphate buffer solutions, comparing with that of pranoprofen itself. 2) $pK_a$ values of pranoprofen, pranoprofen argininate and lysinate salts were 6.34, 7.99 and 7.56 in carbon tetrachloride, and 5.86, 6.69 and 7.92 in chloroform, respectively by liquid-liquid partition method. 3) The lipid-water partition coefficients of pranoprofen argininate and lysinate salts were increased more than that of pranoprofen in carbon tetrachloride, chloroform, or benzene-pH 6.8 buffer system, but were nearly identical using pH 1.2 buffer as water phase. 4) Antiinflammatory effects of pranoprofen argininate and lysinate salts were remarkably increased and analgesic effects of the salts were as same as that of pranoprofen. 5) Pranoprofen argininate and lysinate salts were safer than pranoprofen itself in acute toxicity, and the in situ absorption rates of pranoprofen, pranoprofen argininate and lysinate salts were 0.392, 0.960 and $0.762\;hr^{-1}$, respectively according to the rat intestine recirculation experiment.

Changes of Liver Function In Korean Black Goats Dosed wit Carbon Tetrachloride and 1-naphthylisothiocyanate (한국흑염소에 있어서 사염화탄소와 1-naphthylisothiocyanate 투여시의 간기능 변화)

  • In order to study the effects of administration of carbon tetrachloride(CCI$_4$) and 1-naphthylisothiocyanate(ANIT) on the liver of Korean black goats, some liver function tests and liver biopsy were done on 4 Korean black goats dosed with CCI$_4$(0.4m1/kg of body weight) in-traruminally and 4 Korean black goats dosed with ANIT(400mg/kg of body weight) by stomach tube. BSP Tl/2 and serum total bilirubin concentration in goats dosed with CCI$_4$ were increased gradually, reached to maximum value on 2nd and 1st day, respectively, and then began to decrease in normal range, gradually. In goats dosed with ANIT, BSP Tl/2 and serum total bilirubin concentration were increased rapidly, reached to maximum value on 0.5 and 1st day, respectively, and then returned to normal ragne, rapidly. Serum SDH, AST and GGT activities in goats dosed with CCI$_4$ were increased rapidly and reached to maximum value on 3rd, 1st and 2nd day, respectively. Thereafter, the serum enzyme activities began to decrease in normal range gradually. In goats dosed with ANIT, however, serum SDH, AST and GGT activities were not changed. The histopathologic changes in goats dosed with CCI$_4$ were lipidosis and centrilobular nee-rosis of the hepatic parenchyma. In goats dosed with ANIT, hyperplasia of bile duct epithelium was noticeable, but pathologic changes in liver parenchyma were not noticed. Conclusively, in Korean black goats dosed with CCI$_4$, main finding was necrosis of hepatic parenchyma. In Korean black goats dosed with ANIT, main finding was cholestasis.

  • This study was to investigate the preventive effects in anti-oxidation of Ulva lactuca extract fractions (ULEF) against $CCl_4$ toxification in liver total homogenate and mitochondrial fraction of ULEF-pretreated and carbon tetrachloride $(CCl_4)-posttreated$ rats. ULEF was intraperitoneally administered into rats at dose of 1 ml/kg for14 days. On the day 15, 3.3 ml/kg of $CCl_4$ dissolved in olive oil (1:1) was injected 12 hr before anesthetization. Activities of superoxide dismutase(SOD) in mitochondrial fraction were measured and catalase (CAT), glutathione peroxidase (GPx), malondialdehyde (MDA) in liver total homogenate. SOD, CAI and GPx were higher in the ULEF-pretreated and $CCl_4-posttreated$ group than those in the $CCl_4-posttreated$ group, and the pretreatment of ULEF decreased MDA. Hepatoprotective Effects of Aqueous Extract from Aerial Part of Agrimmony (선학초 추출물의 간보호 효과)

  • Hepatoprotective effects of an aqueous extract prepared from the aerial part of Agrimonia eupatoria L., a species of agrimmony, were investigated in experimental liver-damaged models. To investigate hepatoprotective effects, the agrimmony extract were fed orally to experimental animals. Thereafter a single dose of hepatotoxin, carbon tetrachloride $(CCl_4)$ or D-galac-tosamine was orally administrated. Chronic liver damage was induced by oral administration of $CCl_4$ for 2 weeks (1 time/day). Hepatoprotective effects were monitored by estimating serum AST and ALT levels. The results showed that the agrimmony extract significantly reduced AST and ALT levels compared with those of control group in both acute and chronic animal models. It was concluded that the agrimmony extract have hepatoprotective effects against rat liver injury induced by $CCl_4$ or D-galactosamine.

Effects of Larval Extracts from P. brevitarsis seulensison Cytokine and Diagnostic Marker in Carbon Tetrachloride-Administered SD Rats

  • This study was designed to determine the effects of larval extracts from Protaetia brevitarsis seulensis shade dried larva from Korea and China on liver tissues of hepatic injury groups. The experimental groups are divided into five groups; Normal, carbon tetrachloride single injection ($CCl_4$), Pb-CHI ($CCl_4$ + P. brevitarsis shade dried larva from China), Pb-KOR ($CCl_4$ + P. brevitarsis shade dried larva from Korea) and SIL ($CCl_4$ + 0.35% silymarin) groups. Sprague Dawley rats were oral injected with $CCl_4$ at a dose l mg/kg (20% in corn oil) for induction of liver damage for 4 weeks. Each experimental group was fed with a dose 50 mg/kg of larval extracts based on medicinal preparations from 3 weeks to 4 weeks after $CCl_4$ treatment. At the end of 4 weeks, we evaluated the serum levels of glutamate-oxaloacetate transaminase (GOT), glutamate-pyruvate transferase (GPT), alkaline phosphatase (ALP) and blood urea nitrogen (BUN) in serum and the cytokine levels of tumor necrosis factor-${\alpha}$) (TNF-${\alpha}$) and transforming growth factor-${\beta}$ (TGF-${\beta}$) in the cells isolated from spleen and liver. The histological analysis was also conducted. The $CCl_4$ injection reduced body weight, induced congestion of middle lobe and hepatocytic degeneration, resulting in disintegration of hepatic cords, and increased biochemical markers of blood related to hepatic injury. On the other hand, the Pb-CHI and Pb-KOR group decreased the levels of biochemical markers in blood and cytokine levels in spleen and liver. Especially, the Pb-KOR group facilitated the recovery of biochemical values of blood related to hepatic injury, hepatic lesions and fibrosis. Taken together, larval extract from P. brevitarsis might prevent acute hepatotoxicity and enhance the recovery of liver fibrosis and cirrhosis induced by $CCl_4$, and the ingredients could be a promising candidate for the prevention and treatment of hepatic disorders.

Protective Effect of Galgeun-Tang Against $CCl_4$ Induced Hepatotoxicity (갈근탕의 사염화탄소에 의한 간세포 독성 억제효과)

  • Galgeun-tang (GGT) has been a great source for treating cold diseases in the folk medicine recipe. Carbon tetrachloride ($CCl_4$) is one type of hepatotoxin that can eventually cause liver injury. During the experiment, we first studied the protective effects of GGT against $CC_4$-induced hepatotoxicity. GGT was pretreated for 3 h, and 1% $CCl_4$ was added to mouse primary liver cells. After 4 h, ROS generation and expression of antioxidant enzymes (catalase (CAT), superoxide dismutase (SOD) and glutathione peroxidase (GPx)) were analyzed by FACS and real time PCR. Also, the activities of ALT and LDH were measured using cultured medium. The hepatic levels of TNF-alpha and iNOS, which are related to inflammation and stress response gene, HSP72 and HO-1 were analyzed by PCR or real time PCR. Liver tissues were analyzed by HE stain. From the observation, we discovered that GGT treatment protects $CCl_4$-induced hepatotoxicity, and that GGT pretreatment decreases ROS generation, TNF-alpha and iNOS expression. However, gene expression of CAT, SOD, GPx, HSP72 and HO-1 were increased by GGT. These results lead to the conclusion that GGT has protective effects against $CCl_4$-induced hepatotoxicity.

Study on the Hepatoprotective Effect and Cytochrome P450 Regulation of Scutellaria Radix (황금의 간세포 보호활성 및 cytochrome P450 발현 조절에 관한 연구)

  • Carbon tetrachloride $(CCl_4)-induced$ liver injury depends on a toxic agent that has to be metabolized by the liver NAPDH-cytochrome P450 enzyme system to a highly reactive intermediate. Although several isoforms of cytochrome P450 may metabolize $CC1_4$, attention has been focused largely on the cytochrome P450 2E1 (CYP2E1), which is ethanol-inducible. Alternations in the activity of CYP2E1 affect the susceptibility to hepatic injury from $CC1_4$. In this study, the liver protective effect of the hot water extracts of Scutellaria radix (SR) was investigated. The SR exhibited a hepatoprotective activity against $CCl_4-induced$ liver damage in Chang liver cells. The expression of CYP2E1, measured by RT-PCR and Western blot analysis, was significantly decreased by SR treatment in Chang cells. Based on these findings, it is suggested that hepatoprotective effect of SR possibly related to downregulation of CYP2E1 expression.

Hepaprotective Effect of Standardized Ecklonia stolonifera Formulation on CCl4-Induced Liver Injury in Sprague-Dawley Rats

  • The liver is an essential organ for the detoxification of exogenous xenobiotics, drugs and toxic substances. The incidence rate of non-alcoholic liver injury increases due to dietary habit change and drug use increase. Our previous study demonstrated that Ecklonia stolonifera (ES) formulation has hepatoprotective effect against alcohol-induced liver injury in rat and tacrine-induced hepatotoxicity in HepG2 cells. This present study was designated to elucidate hepatoprotective effects of ES formulation against carbon tetrachloride ($CCl_4$)-induced liver injury in Sprague Dawley rat. Sixty rats were randomly divided into six groups. The rats were treated orally with ES formulation and silymarin (served as positive control, only 100 mg/kg/day) at a dose of 50, 100, or 200 mg/kg/day for 21 days. Seven days after treatment, liver injury was induced by intraperitoneal injection of $CCl_4$ (1.5 ml/kg, twice a week for 14 days). The administration of $CCl_4$ exhibited significant elevation of hepatic enzymes (like AST and ALT), and decrease of antioxidant related enzymes (superoxide dismutase, glutathione peroxidase and catalase) and glutathione. Then, it leaded to DNA damages (8-oxo-2'-deoxyguanosine) and lipid peroxidation (malondialdehyde). Administration of ES formulation inhibited imbalance of above factors compared to $CCl_4$ induced rat in a dose dependent manner. Real time PCR analysis indicates that CYP2E1 was upregulated in $CCl_4$ induced rat. However, increased gene expression was compromised by ES formulation treatment. These findings suggests that ES formulation could protect hepatotoxicity caused by $CCl_4$ via two pathways: elevation of antioxidant enzymes and normalization of CYP2E1 enzyme.

Antioxidative and Hepatoprotective Effects of Lythrum salicaria (털부처꽃의 항산화 및 간장보호활성)

  • Several parts of Lythrum salicaria were used for this study. Scavenging activities on radicals, inhibitory activity on linoleic acid peroxidation and total phenol contents of extracts from root, flower, and aerial part were evaluated. Flower and root selected from in vitro assay were subjected to in vivo assay on $CCL_4$-induced liver injury rat model for two weeks. Carbon tetrachloride intoxication on rats produced large amounts of hepatic lipid peroxidation product, thiobarbituric acid reactive substance (TBARS) compared with normal rats. Treatment with root extract of L. salicaria (LSR) showed effective inhibitory activity on lipid peroxidation product. Administration with LSR extract significantly alleviated $CCL_4$-induced increase in GPT activity which were more effective than silymarin. The results of this study suggest that root and flower of L. salicaria have antioxidant and liver protecting activities, and root part is the most effective candidate to develop a new functional material.
