Serum Levels of Xanthine Oxidase Activities in Cyclohexanone-Treated Rats Pretreated with Carbon Tetrachloride

  • To investigate an effect of cyclohexanone (CHO) treatment on the serum levels of xanthine oxidase (XO) in liver damaged animals, the rats were intraperitoneally pretreated with 50% carbon tetrachloride ($CCl_4$) in olive oil (0.1 mL/ 100 g body weight) 14 times every other day. To the $CCl_4$-pretreated rats, CHO (1.56 g/kg body weight) was injected once and then the animals were sacrificed at 4 hours after CHO treatment. The increasing rate of serum and liver XO activities to the control was higher in CHO-treated animals pretreated with $CCl_4$ than the $CCl_4$-pretreated those. Concomitantly CHO injection to the $CCl_4$-pretreated animals showed somewhat higher Vmax and lower Km value in the kinetics of liver XO enzyme. Furthermore, increasing rate of hepatic malonedialdehyde content to the control was also higher in CHO-treated animals pretreated with $CCl_4$ than $CCl_4$-pretreated those. On the other hand, the injection of CHO to the $CCl_4$-pretreated animals showed the more enhanced liver damage on the basis of liver function finding; liver weight per body weight (%), serum levels of alanine aminotransferase activity and hepatic glucose-6-phosphatase activity. In conclusion, injection of CHO to the $CCl_4$-pretreated rats led to more increased activity of serum XO and it may be caused by acceleration of hepatocyte membrane permeability and induction of enzyme protein.

Curcuma Extract Enhances Hepatoprotective Action of Silymarin in Experimental Liver Damage Models (실리마린의 간보호 작용에 미치는 쿠쿠마 추출물의 영향)

  • Silymarin and curcumin have been used for supportive treatment of liver disease of difffrent etiology due to their hepatoprotective activities. The present study was carried out to investigate the hepatoprotective efffcts of silymarin and/or curcuma extract against hepatotoxins induced liver injury. To investigate hepatoprotective effects, the silymarin and/or curcuma extract were pre-treated orally to experimental animals. And thereafter a single dose of hepatotoxin, carbon tetrachloride ($CCl_4$) and acetaminophen were administered through oral or intraperitoneal route, respectively. Chronic liver damage was induced by subcutaneous injection of $CCl_4$ for 3 weeks (2 times/week). Hepatoprotective and therapeutic effects were monitored by estimating serurn ALT and AST levels and by measuring hepatic glutathione (GSH) and malondialdehyde (MDA)levels. Collagen type 1 was detected with irnrnunostaining to assess fibrosis. The results showed that the mix-ture of silymarin and curcuma extract significantly reduced serum biochemistry levels and MDA levels com-pared with those of control group in both acute and chronic animal models. In antifibrotic effect, the relative hepatic collagen content was significantly decreased by silymarin and/or curcuma extract treatment. It was concluded that the complex of silymarin and curcuma extract have a both hepatoprotective and therapeutic effect synergically in rat liver injury induced by heptotoxins.

Effect and Histopatbologic Examination of Calcium Channel Blocker on Carbon Tetrachloride-induced Hepatotoxicity in Rats (Calcium Channel Blocker가 사염화탄소에 의한 간손상에 미치는 영향 및 조직학적 검경)

  • Carbon tetrachloride (CCI$_4$) induces the hepatotoxicity due to the reactive free radicals generated by cytochrome P-450 (CYP-450) enzyme, which result in destabilization of cellular membrane. Diltiazem, a calcium channel blocking agent, has been known to suppress the CYP-450 enzyme activities. To study the effect of diltiazem in $CCl_4$-treated rats, we measured the activities of alanine aminotransferase (ALT), aspartate aminotransferase (AST) and glutathione S-transferase (GST), contents of bilirubin, albumin, total protein, cholesterol, triglyceride, blood urea nitrogen, creatinine malondialdehyde and calcium. Also we conducted liver histopathologic examinations. Diltiazem, when administered 1 hour prior to CCI$_4$ treaeent, significantly reduced the activities of ALT and AST, the contents of microsomal malondialdehyde and calcium in liver and microsome as compared with those of $CCl_4$-treated rats. In addition, histopathologic examination showed that diltiazem prevented the development of centrilobular necrosis induced by CCI$_4$ in liver tissue. Our results suggested that diltiazem could inhibit the formation of free radicals and the influx of calcium. Therefore diltiazem may be applied to suppress the liver damage caused by $CCl_4$.

Effect of Solanum lyratum Extract on the Hepatotoxicity of Carbon Tetrachloride in Rats (백모등 엑스의 간기능개선 효과)

  • The studies were attempted to investigate the therapeutic effect of Solanum lyratum extract(SLE) on the liver damage induced by carbon tetrachloride in rats. Male rats (Sprangue-Dawley) were treated with subchronic concentration of $CCl_4$, which was 1 mg/kg of $CCl_4$ via per oral administration daily for three days. The level of serum cholesterol, transaminase, bilirubin and BSP were measured. Treatment of SLE significantly decreased the increment in serum transaminase activities, total cholesterol and enlargement of liver but serum bilirubin was not reduced. The protective effect was observed when SLE was given daily for 4 weeks after $CCl_4$ treated for 3 days. SLE administration to rats prevented the development of fatty change, necrosis, and fibrosis induced by $CCl_4$. The results suggest that SLE has appreciable therapeutic effect on $CCl_4$ induced hepatotoxicity.

Studies on Protective Effect of Da-9601, Artemisia asiatica Extract, on Acetaminophen- and $CCI_{4}$induced Liver Damage in Rats

  • The hepatoprotective effect of DA-9601, a quality-controlled extract of artemisisa asiatica, on liver damage induced by acetaminophen (APAP) and carbon tetrachloride ($CCI_{4}$) was investigated by means of serum-biochemical, hepatic-biochemical, and histopathological examinations. Doses of Da-9601 (10, 30, or 100 mg/kg) were administered intragastrically to each rat on three consecutive days i.e. 48 h, 24 h and 2 h before a single administration of APAP (640 mg/kg, i.p.) or $CCI_{4}$ (2 ml/kg, p.o.). Four h and 24 h after hepatotoxin treatment, the animals were sacrificed for evaluation of liver damage. Pretreatment of Da-9601 reduced the elevation of serum ALT, AST. LDH and histopathological changes such as centrilobular necrosis, vacuolar degeneration and inflammatory cell infiltration dose-dependently. Da-9601 also prevented APAP- and $CCI_{4}$-induced hepatic glutathione (GSH) depletion and $CCI_{4}$-induced increase of hepatic malondialdehyde (MDA), a parameter of lipid peroxidation, in a chemically induced liver injury by complex mechanisms which involve prevention of lipid peroxidation and preservation of hepatic GSH.

Antimicrobial activity and toxicity of Quisqualis indica

  • The plant Quisqualis indica (Compositae) has ehnopharmcological reputation of being used as a healing agent in Bangladesh. In this study, preliminary screenings were conducted to look at the antimicrobial susceptibility and cytotoxicity of the plant extract. The extractives of the plant were subjected to screening for inhibition of microbial growth by the disc diffusion method. The zones of inhibition demonstrated by the n-hexane, carbon tetrachloride, chloroform and aqueous soluble partitionates of the methanolic extract ranged from 8 - 15 mm, 8 - 18 mm, 12 - 20 mm and 10 - 16 mm, respectively at a concentration of 400 ${\mu}g$/disc. All the extractives were also subjected to brine shrimp lethality bioassay for primary cytotoxicity evaluation. Here, the carbon tetrachloride soluble materials demonstrated the highest cytotoxicity with $LC_{50}$ of 0.826 ${\mu}g$/ml, while n-hexane, chloroform and aqueous soluble partitionates of the methanolic extract revealed the $LC_{50}$ of 1.254, 3.866 and 5.366 ${\mu}g$/ml, respectively. This is the first report of the antimicrobial activity and cytotoxicity Q. indica.

Effect of Whaganjeon on the hepatoxicity of carbon tetrachloride in rats (화간전(化肝煎)이 carbon tetrachloride로 유도(誘導)한 간중독(肝中毒) 흰쥐에 미치는 영향(影響))

  • The purpose of this study is to observe the effect of Whaganjeon on serum reaction in $CCl_4$ treated rats. In this study, the experimental rats divided four group(Control group, $CCl_4$ group, Whaganjeon group, and Sample group) : Under the same condition, control group were administered water, sample group were administered Whaganjeon for 7days. And then, both $CCl_4$ group and sample group were injected to abdomen with $CCl_4$ for 1days. The change of GOT, GPT, ALP, LDH, and bile acid activity in blood serum. The obtained results are summarised as follows : 1. In the change of SGOT, SGPT contents, as compared with control group, sample group was significantly decreased. 2. In the change of serum ALP contents, as compared with control group, sample group was significantly decreased. 3. In the change of serum bile acid contents, as compared with control group, sample group was significantly decreased. 4. In the change of serum LDH contents, as compared with control group, sample group was significantly decreased.

  • The Transient State Theory of Significant Liquid Structure is applied to a binary liquid solution of benzene and carbon tetrachloride, which gives slightly positive deviation from Raoults law. The partition function for the solution is derived according to the proposed theory. The various thermodynamic properties such as total and partial vapor pressures, molar volumes, entropies of mixing, and compressibilities are calculated at three different temperatures, 298.15^{\circ}K$, 313.15^{\circ}K$ and 343.1515^{\circ}K$. The calculated values agree satisfactorily with experimental data.

  • Degradations of phenol and chlorinated compounds mixtures were studied with ultrasound of 20 kHz and 0.57, 1.14 W/mL. In presence of carbon tetrachloride (CT), degradation rate of phenol is faster than chloroform (CF), dichloromethane (DCM) and phenol solution. It is due to that CT generates of free chlorine (HOCl and $OCl^-$) from the sonochemical degradation and plays a role of hydrogen atom scavenger. CF and DCM are react with free chlorine, so amount of free chlorine is smaller than CT solution. The degradation rates of chlorinated compounds decreased with co-presence of phenol in the solution due to the distribution ultrasonic energy to both compounds. The measured chloride ion was lower than the theoretical concentration assuming complete degradation. This means not all the contaminants destructed went through complete degradation.

The Effect of Electro-acupuncture at ST36 on Carbon Tetrachloride-induced Chronic Hepatic Damage in Rats (족삼리 전침자극이 Carbon Tetrachloride 장기투여로 유발된 흰쥐의 만성 간손상에 미치는 영향)

  • Objectives : The purpose of this study is to investigate the effect of electro-acupuncture(EA) at ST36 on $CCl_4{^-}$ induced chronic hepatic damage in rats. Methods : The authors performed several experimental items, including measurements of liver index, hematological analysis for WBC, lymphocytes, neutrophils, monocytes, hemoglobin, and biochemical assays for ALT, AST, ALP in serum, and histological analysis of liver tissue. Results : Liver index(liver weight/body weight), the levels of WBC, Neutrophils and Monocytes in blood, the levels of ALT, AST and ALP in serum which were increased by $CCl_4{^-}$ intoxication were reduced significantly by EA at ST36. The levels of lymphocyte and hemoglobin in blood which were decreased by $CCl_4{^-}$ intoxication were significantly increased by EA at ST36. Conclusions : EA at ST36 has a hepatoprotective effect on chronic hepatic damage in $CCl_4{^-}$ intoxicated rats.
